Conditions for success of engineered underdominance gene drive systems.

Engineered underdominance is one of a number of different gene drive strategies that have been proposed for the genetic control of insect vectors of disease. Here we model a two-locus engineered underdominance based gene drive system that is based on the concept of mutually suppressing lethals. In such a system two genetic constructs are introduced, each possessing a lethal element and a suppressor of the lethal at the other locus. Specifically, we formulate and analyse a population genetics model of this system to assess when different combinations of release strategies (i.e. single or multip

The molecular basis of CO2 reception in Drosophila.

CO(2) elicits a response from many insects, including mosquito vectors of diseases such as malaria and yellow fever, but the molecular basis of CO(2) detection is unknown in insects or other higher eukaryotes. Here we show that Gr21a and Gr63a, members of a large family of Drosophila seven-transmembrane-domain chemoreceptor genes, are coexpressed in chemosensory neurons of both the larva and the adult. The two genes confer CO(2) response when coexpressed in an in vivo expression system, the "empty neuron system." The response is highly specific for CO(2) and dependent on CO(2) concentration. T

Effectiveness of orally-delivered double-stranded RNA on gene silencing in the stinkbug Plautia stali.

Development of a reliable method for RNA interference (RNAi) by orally-delivered double-stranded RNA (dsRNA) is potentially promising for crop protection. Considering that RNAi efficiency considerably varies among different insect species, it is important to seek for the practical conditions under which dsRNA-mediated RNAi effectively works against each pest insect. Here we investigated RNAi efficiency in the brown-winged green stinkbug Plautia stali, which is notorious for infesting various fruits and crop plants. Microinjection of dsRNA into P. stali revealed high RNAi efficiency-injection o

Mode of action of nanoparticles against insects.

The employment of nanoparticles obtained through various synthesis routes as novel pesticides recently attracted high research attention. An impressive number of studies have been conducted to test their toxic potential against a wide number of arthropod pests and vectors, with major emphasis on mosquitoes and ticks. However, precise information on the mechanisms of action of nanoparticles against insects and mites are limited, with the noteworthy exception of silica, alumina, silver, and graphene oxide nanoparticles on insects, while no information is available for mites. Here, I summarize cu

The Adaptive Roles of Active Transposable Elements in Insect Hosts.

Active transposable elements (TEs) are capable of generating new insertions in genomes and have historically been viewed as genomic parasites due to their largely detrimental or neutral effects. However, emerging evidence suggests that these elements also play a crucial role in driving adaptive evolution in insects. This mini-review synthesizes recent findings on how active TEs contribute to insect adaptation through various mechanisms, including regulation of gene expression, structural variation, and epigenetic effects. Notable examples of adaptation driven by active TEs include their roles

"Plus-C" odorant-binding protein genes in two Drosophila species and the malaria mosquito Anopheles gambiae.

Olfaction plays a crucial role in many aspects of insect behaviour, including host selection by agricultural pests and vectors of human disease. Insect odorant-binding proteins (OBPs) are thought to function as the first step in molecular recognition and the transport of semiochemicals. The whole genome sequence of the fruit fly Drosophila melanogaster has been completed and a large number of genes have been annotated as OBPs, based on the presence of six conserved cysteine residues and a conserved spacing between the cysteines. These proteins can be divided into three distinct subgroups; thos

A drug-inducible sex-separation technique for insects.

Here, we describe a drug-inducible genetic system for insect sex-separation that demonstrates proof-of-principle for positive sex selection in D. melanogaster. The system exploits the toxicity of commonly used broad-spectrum antibiotics geneticin and puromycin to kill the non-rescued sex. Sex-specific rescue is achieved by inserting sex-specific introns into the coding sequences of antibiotic-resistance genes. When raised on geneticin-supplemented food, the sex-sorter line establishes 100% positive selection for female progeny, while the food supplemented with puromycin positively selects 100%

Transposition of the piggyBac element in embryos of Drosophila melanogaster, Aedes aegypti and Trichoplusia ni.

The Lepidopteran transposable element piggyBac is being recognized as a useful vector for genetic engineering in a variety of insect species. This transposon can mediate transformation in the Dipteran species Ceratitis capitata, and can potentially serve as a versatile vector for transformation of a wide variety of insect species. Using a plasmid-based interplasmid transposition assay, we have demonstrated that this transposon, of the short inverted terminal repeat type, is capable of transposition in embryos of three different insect species, Drosophila melanogaster, the yellow fever mosquito

A repertoire of the dominant transcripts from the salivary glands of the blood-sucking bug, Triatoma dimidiata, a vector of Chagas disease.

Triatoma (T.) dimidiata is a hematophagous Hemiptera and a main vector of Chagas disease. The saliva of this and other blood-sucking insects contains potent pharmacologically active components that assist them in counteracting the host hemostatic and inflammatory systems during blood feeding. To describe the repertoire of potential bioactive salivary molecules from this insect, a number of randomly selected transcripts from the salivary gland cDNA library of T. dimidiata were sequenced and analyzed. This analysis showed that 77.5% of the isolated transcripts coded for putative secreted protein

A ventral nerve cord enriched major allergen family gene maintains muscle function under heat stress in Bactrocera dorsalis.

Heat stress threatens insect survival by disrupting tissue homeostasis and physiological performance, yet how insects preserve tissue function under high temperature remains poorly understood. Members of the major allergen gene family are widely distributed in insects, but whether and how they contribute to heat stress adaptation remains unclear. Here, using the tropical and subtropical horticultural pest Bactrocera dorsalis, a species capable of surviving temperatures above 40 °C, as a model, we identified a previously uncharacterized heat-responsive gene, matl2, which encodes a secreted lipi

Partial Substitution of Meat with Insect (Alphitobius diaperinus) in a Carnivore Diet Changes the Gut Microbiome and Metabolome of Healthy Rats.

Insects are suggested as a sustainable protein source of high nutritional quality, but the effects of insect ingestion on processes in the gastrointestinal tract and gut microbiota (GM) remain to be established. We examined the effects of partial substitution of meat with insect protein (Alphitobius diaperinus) in a four-week dietary intervention in a healthy rat model (n = 30). GM composition was characterized using' 16S rRNA gene amplicon profiling while the metabolomes of stomach, small intestine, and colon content, feces and blood were investigated by 1H-NMR spectroscopy. RNAi-Mediated Knockdown of Halloween Genes in Lygus hesperus Has Varied Effects on Oogenesis and Fertility.

For many insects, the ecdysteroid biosynthetic pathway that converts cholesterol to ecdysone includes multiple cytochrome P450 monooxygenases, a Rieske oxygenase, and a short-chain dehydrogenase. Collectively, the genes encoding these enzymes are referred to as Halloween genes. While the importance of these enzymes in oogenesis is well-documented in holometabolous insects, less is known about their oogenic role in hemipterans. To address this, we used RNA interference-mediated knockdown to determine the role that the Halloween transcripts neverland, spookiest, shroud, phantom, disembodied, sha

Fruitless alternative splicing and sex behaviour in insects: an ancient and unforgettable love story?

Courtship behaviours are common features of animal species that reproduce sexually. Typically, males are involved in courting females. Insects display an astonishing variety of courtship strategies primarily based on innate stereotyped responses to various external stimuli. In Drosophila melanogaster, male courtship requires proteins encoded by the fruitless (fru) gene that are produced in different sex-specific isoforms via alternative splicing. Drosophila mutant flies with loss-of-function alleles of the fru gene exhibit blocked male courtship behaviour. However, various individual steps in

Beyond molting disruption: Tebufenozide modifies gut immunity and antiviral responses in Helicoverpa armigera (Noctuidae).

The insect growth regulator tebufenozide, an ecdysone agonist, is widely used to control Helicoverpa armigera through disrupting molting and development. However, its other effects on the insect physiology remain poorly understood. This study investigates how tebufenozide modulates gut immunity, microbial homeostasis, and antiviral defense mechanisms in H. armigera larvae. Using RT-qPCR, we analyzed the expression of key immune genes following exposure to lethal (LC₅₀) and sublethal (LC₁₀, LC₂₅) concentrations of tebufenozide. Our results demonstrate that tebufenozide induced the IMD pathway t

The down-regulation of salivary protein gene expression by etofenprox partially contributed to reducing the risk of increased fecundity in the brown planthopper.

Etofenprox is a pyrethroid insecticide that acts on the nervous system of insects. Due to its low toxicity to aquatic animals, it is permitted for use in controlling insect pests in rice fields. The brown planthopper (BPH), Nilaparvata lugens, a significant piercing-sucking pest feeding on rice exclusively, secretes various salivary components when feeding. Salivary proteins are essential for BPH feeding, but their response to etofenprox is not well understood. The application of etofenprox down-regulated the expression of 21 salivary protein genes, among which 9 genes (NlShpa, Salivap 3, CA,

A novel needleless delivery of Cas9 ribonucleoprotein complex in Bemisia tabaci embryos for gene editing.

The CRISPR/Cas9 system, particularly the Cas9-sgRNA ribonucleoprotein (RNP) complex, offers a highly efficient platform for gene editing. However, traditional microinjection methods for RNP delivery in insect embryos are labor-intensive, technically demanding, and often reduce embryo viability, especially in species with fragile, microscopic embryos. In this study, a novel, non-invasive delivery method for the RNP complex in tiny insect embryos has been optimized. Whitefly, Bemisia tabaci, an invasive insect pest of agricultural importance and vector of plant diseases, was considered as a mode

Knockout of a testis-specific gene cluster impairs male fertility in the fall armyworm, Spodoptera frugiperda.

BACKGROUND: The function of some testis-specific genes (TSGs) in model insects have been studied, but their function in non-model insects remains largely unexplored. In the present study, we identified several TSGs in the fall armyworm (FAW), a significant agricultural pest, through comparative transcriptomic analysis. A testis-specific gene cluster (TSGC) comprising multiple functional genes and long non-coding RNAs was found. RESULTS: Quantitative PCR analysis validated the testis-specific expression of genes within this cluster across different developmental stages. Interspecies collineari

Biotin synthesis and assimilation are vital for pathobiology of the insect pathogenic fungus Beauveria bassiana.

Biotin is essential for metabolic processes. Bioinformatics analysis revealed that there was a gene cluster consisting of three biotin synthesis genes (BbBIOB, D, and F) in the insect pathogenic fungus Beauveria bassiana. When the cluster was ablated, the protein biotinylation was significantly decreased, and the disruptant displayed phenotypic defects in radial growth, conidial germination, development, and virulence. However, exogenous biotin could restore the protein biotinylation and phenotypic defects. Notably, B. bassiana growth on asparagine (as nitrogen source) was independent of the b
