Targeting PSAT1 in diabetic kidney disease: a ferroptosis-driven strategy for precision therapy.

Diabetic kidney disease (DKD) involves mitochondrial dysfunction and ferroptosis, yet the convergence of these processes at the gene level remains unclear. We first analyzed human renal transcriptomic data (GSE30528) to identify differentially expressed genes (DEGs). DEGs associated with both mitophagy and ferroptosis (DEMFRGs) were extracted. Protein-protein interaction (PPI) networks, the MCC algorithm, and LASSO regression were used to screen hub genes. Immune infiltration was assessed via CIBERSORT. Diagnostic performance of hub genes was evaluated using ROC curves in GSE30528 and validate

Enhanced allergic Eustachian tube response in surfactant protein D knockout mice induced by ovalbumin.

BACKGROUND: Allergic reactions have been widely recognized as closely associated with the development of Eustachian tube dysfunction (ETD). However, the precise pathophysiological mechanisms remain unclear, and effective models to elucidate the role of allergic responses in ETD are lacking. Surfactant protein D (SPD), an important immune defense factor in the Eustachian tube (ET), not only reduces surface tension but also exhibits anti-adhesion and inflammation-regulating properties. Nevertheless, the role of SPD in allergic inflammation of the ET has not been fully explored. OBJECTIVE: This

Context-Aware Multilevel Classification of Semantic Relations in Drug-Adverse Drug Reaction (ADR) Networks-Predicting Drug-Induced Liver Injury (DILI) as a Case Study.

Adverse drug reactions (ADRs) are a major concern for public health and patient safety and a burden in drug discovery, with drug-induced liver injury (DILI) as a prototypical case. Next to computational models such as deep learning, knowledge graph models can help capture biological systems and pathways underlying toxic effects. We present a hierarchical, context-aware knowledge graph model to detect ADRs, with DILI as a case study. The model builds on a commercial knowledge graph integrating over 200 life-science databases and millions of publications and patents. It combines translation- and

Machine learning-assisted prediction of PANoptosis-related molecular targets and precise screening of neuroprotective drugs for spinal cord injury.

Spinal cord injury (SCI) is a highly disabling central nervous system disease with complex pathology, and targeted neuroprotective drugs remain clinically lacking. However, traditional molecular target screening and drug prediction methods are inefficient, costly, and poorly targeted, failing to meet clinical precision treatment needs. To address this, we introduced machine learning to construct a multi-dimensional data integration framework. First, we established normal, acute- and subacute-phase SCI mouse complete transection models, and RNA-seq combined with single-cell sequencing revealed

Regulation of Polyomavirus Transcription by Viral and Cellular Factors.

Polyomavirus infection is widespread in the human population. This family of viruses normally maintains latent infection within the host cell but can cause a range of human pathologies, especially in immunocompromised individuals. Among several known pathogenic human polyomaviruses, JC polyomavirus (JCPyV) has the potential to cause the demyelinating disease progressive multifocal leukoencephalopathy (PML); BK polyomavirus (BKPyV) can cause nephropathy in kidney transplant recipients, and Merkel cell polyomavirus (MCPyV) is associated with a highly aggressive form of skin cancer, Merkel cell c

An intrachromosomal insertion causing 5q22 deletion and familial adenomatous polyposis coli in two generations.

We report familial adenomatous polyposis coli (FAPC) with epidermoid cysts, osteomata, and areas of congenital hypertrophy of the retinal pigment epithelium (CHRPEs) in a male patient and his maternal aunt, both of whom suffered a mild to moderate degree of mental handicap. Both had an interstitial deletion of the long arm of chromosome 5 (del(5)(q22q23.2)). Two other normal family members had the underlying direct insertion of chromosome 5(dir ins(5)(q31.3q22q23.2)). Molecular genetic and fluorescent hybridisation studies have shown that loci D5S37 and D5S98 are outside the deletion whereas l

QTL Mapping Using a High-Density Genetic Map to Identify Candidate Genes Associated With Metribuzin Tolerance in Hexaploid Wheat (Triticum aestivum L.).

Tolerance to metribuzin, a broad-spectrum herbicide, is an important trait for weed control in wheat breeding. However, the genetics of metribuzin tolerance in relation to the underlying quantitative trait loci (QTL) and genes is limited. This study developed F8 recombinant inbred lines (RILs) from a cross between a highly resistant genotype (Chuan Mai 25) and highly susceptible genotype (Ritchie), which were used for QTL mapping of metribuzin tolerance. Genotyping was done using a diversity arrays technology sequencing (DArTseq) platform, and phenotyping was done in controlled environments. H
