The lignan, (-)-sesamin reveals cytotoxicity toward cancer cells: pharmacogenomic determination of genes associated with sensitivity or resistance.

(-)-Sesamin is a lignan present in sesam oil and a number of medicinal plants. It exerts various pharmacological effects, such as prevention of hyperlipidemia, hypertension, and carcinogenesis. Moreover, (-)-sesamin has chemopreventive and anticancer activity in vitro and in vivo. Multidrug resistance (MDR) of tumors leads to fatal treatment outcome in many patients and novel drugs able to kill multidrug-resistant cells are urgently needed. P-glycoprotein (MDR1/ABCB1) is the best known ATP-binding cassette (ABC) drug transporter mediating MDR. ABCB5 is a close relative to ABCB1, which also med

Multidrug-resistant Enterobacteriaceae colonising the gut of adult rural population in South India.

BACKGROUND: Multidrug-resistant (MDR) colonisers act as a reservoir for transmission of antibiotic resistance and are a source of infection. Exposure to antibiotics by the commensal flora renders them resistant. Antibiotic consumption and hospitalisation are two major factors influencing this. We studied, antibiotic-resistant bacteria colonising rural adult population who had restricted access to health care and presumably had low consumption of antibiotics. AIM: Detection of multidrug resistance genes of extended spectrum β-lactamase (ESBL-CTX-M), AmpC β-Lactamase (CIT), Klebsiella pneumonia

Involvement of the efflux pumps in chloramphenicol selected strains of Burkholderia thailandensis: proteomic and mechanistic evidence.

Burkholderia is a bacterial genus comprising several pathogenic species, including two species highly pathogenic for humans, B. pseudomallei and B. mallei. B. thailandensis is a weakly pathogenic species closely related to both B. pseudomallei and B. mallei. It is used as a study model. These bacteria are able to exhibit multiple resistance mechanisms towards various families of antibiotics. By sequentially plating B. thailandensis wild type strains on chloramphenicol we obtained several resistant variants. This chloramphenicol-induced resistance was associated with resistance against structur

Zinc finger proteins and ATP-binding cassette transporter-dependent multidrug resistance.

BACKGROUND: Multidrug resistance (MDR) remains a significant challenge in cancer treatment, leading to poor clinical outcomes. Dysregulation of ATP-binding cassette (ABC) transporters has been identified as a key contributor to MDR. Zinc finger proteins (ZNPs) are key regulators of transcription and have emerged as potential contributors to cancer drug resistance. Bridging the knowledge gap between ZNPs and MDR is essential to understand a source of heterogeneity in cancer treatment. This review sought to elucidate how different ZNPs modulate the transcriptional regulation of ABC genes, contri

Comparative genomics reveals the evolution of antimicrobial resistance in Bacteroides nordii.

Bacteroides nordii, is an understudied member of the pathogenic B. fragilis group which comprises several multidrug-resistant (MDR) strains. Thus, it is of great interest to study the genome biology of Bacteroides nordii. However, no detailed study is available that characterized B. nordii at the genetic level and explored its role as a potential pathogen. We isolated an MDR strain viz., B. nordii PGMM4098 from the pus sample and subjected it to whole genome sequencing using Illumina technology. The draft genome was de-novo assembled and annotated, followed by comprehensive comparative genomic

Equisetin targets membrane integrity and transcriptional networks to combat multidrug-resistant Enterococcus faecalis.

This study evaluates the natural product Equisetin as a potent agent against multidrug-resistant Enterococcus faecalis (MDR E. faecalis). Equisetin exhibited strong antibacterial activity (MIC = 5.84 μg/mL) by disrupting membrane integrity, inducing protein/DNA leakage, depolarizing membrane potential, elevating ROS, and depleting ATP. It also inhibited biofilm formation and showed minimal hemolytic activity without inducing resistance. Transcriptomics revealed upregulation of DNA repair/stress genes and downregulation of metabolic/protein synthesis genes, indicating a multi-target mechanism.

Relationships between multidrug resistance (MDR) and stem cell markers in human chronic myeloid leukemia cell lines.

The K562 cell line (chronic myeloid leukemia), sensitive to chemotherapy (non-MDR), and the Lucena cell line, resistant to chemotherapy (MDR) were investigated. The results suggest that both cell lines possess CD34+CD38- profiles of hematopoietic stem cell markers. The promoter regions of ABCB1, ABCG2 and ABCC1 genes contain binding sites for the Oct-4 transcripton factor, which is also considered a marker of tumor stem cells. Lucena cells showed an over-expression of the ABCB1 gene and a high expression of the Oct-4, ABCG2 and ABCC1 genes as compared to K562 cells.
