Modulation of ghrelin O-acyltransferase expression in pancreatic islets.

BACKGROUND: Ghrelin, the only identified circulating orexigenic signal, is unique in structure in which a specific acyl-modification of its third serine occurs. This acylation is necessary for ghrelin to bind to its receptor and to exert its biologic activity, which is catalyzed by ghrelin O-acyltransferase (GOAT). Although ghrelin is mainly secreted from gastric X/A like endocrine cells, it is also expressed in pancreatic islet cells and regulates insulin secretion. In this study, we examined the expression and regulation of GOAT in pancreas. METHODS: GOAT mRNA and immunoreactivity were exam

Ghrelin expression in human testis and serum testosterone level.

Ghrelin, which is the endogenous ligand for the growth hormone (GH) secretagogue receptor (GHS-R), has been primarily linked to the central neuroendocrine regulation of GH secretion and food intake, although additional peripheral actions of ghrelin have also been reported. Recent research has suggested that ghrelin also affects testosterone (T) secretion in vitro. To investigate the role of ghrelin in human testicular function, we examined the expression of ghrelin in testicular tissues by immunohistochemistry. Testicular tissue samples were collected from the testes of 5 fertile volunteers, 8

Energy homeostasis and gastric emptying in ghrelin knockout mice.

To elucidate the role of endogenous ghrelin in the regulation of energy homeostasis and gastric emptying, ghrelin knockout mice (ghrelin(-/-)) were generated. Body weight, food intake, respiratory quotient, and heat production (indirect calorimetry), and gastric emptying ((14)C breath test) were compared between ghrelin(+/+) and ghrelin(-/-) mice. In both strains, the effect of exogenous ghrelin on gastric emptying and food intake was determined. Ghrelin(-/-) mice showed some subtle phenotypic changes. Body weight gain and 24-h food intake were not affected, but interruption of the normal ligh

Ghrelin/Leptin-imbalance in patients with primary biliary cirrhosis.

BACKGROUND: The recently discovered peptide hormone ghrelin mainly produced in gastric oxyntic cells may act as a counterpart to leptin in the regulation of food intake and fat utilization. Leptin, involved in the stimulation of proinflammatory cytokines and catabolic energy balance, is elevated in patients with liver cirrhosis. In the present study, we evaluated serum ghrelin and bound leptin levels in patients with primary biliary cirrhosis (PBC) in relation to C-peptide and glucose concentration. METHODS: In 22 female patients with PBC (Child-Pugh stage A) and in 36 female controls we meas

Ghrelin, hypothalamus-pituitary-adrenal (HPA) axis and Cushing's syndrome.

Ghrelin, a peptide predominantly produced by the stomach, has been discovered as a natural ligand of the GH Secretagogue receptor type 1a (GHS-R1a), known as specific for synthetic GHS. Ghrelin has recently attracted considerable interest as a new orexigenic factor. However, ghrelin exerts pleiotropic actions that are explained by the widespread distribution of ghrelin and GHS-R expression. Besides strong stimulation of GH secretion, the neuroendocrine ghrelin actions also include significant stimulation of both lactotroph and corticotroph secretion; all these actions depend on acylation of gh

Helicobacter pylori has no effect on plasma ghrelin levels.

OBJECTIVE: Helicobacter pylori is the major etiologic agent for chronic active gastritis, and it also plays a crucial role in gastric and duodenal ulcer disease, as well as in gastric carcinoma. H. pylori infection has been shown to decrease plasma somatostatin (SST) and increase plasma gastrin concentrations. Ghrelin is a recently discovered peptide produced mostly in the stomach of rodents and humans and is secreted into the bloodstream. There is no data in the literature about the relationship between H. pylori and ghrelin. DESIGN: Thirty-nine age- and BMI-matched H. pylori infection posit

Nicotinic acetylcholine receptors and their role in ghrelin-producing cells.

Gastric orexigenic peptide ghrelin has been implicated in tobacco addiction, but there are conflicting results regarding the effects of acute smoke on the plasma ghrelin levels. In this study, we screened the effects of tobacco-related chemicals, especially focusing on the effects of nicotine on ghrelin regulation at cellular level. Nicotine alone modestly stimulated ghrelin secretion, and enhanced the stimulatory effects of isoproterenol and oxytocin on ghrelin secretion from ghrelin-producing MGN3-1 cells evaluated by a newly developed ghrelin-HiBiT assay system. MGN3-1 cells expressed high

Ghrelin's role in sleep and sleep deprivation: a narrative review.

Deprivation of sleep (DS) is widespread in modern societies and is associated with cardiometabolic, cognitive, and psychiatric disturbances. Acute DS has also been reported to produce short-lived improvements in mood in some individuals with depression, suggesting the involvement of specific biological mediators. Ghrelin, a stomach-derived peptide with central actions in hypothalamic and limbic circuits, has emerged as a candidate linking DS with alterations in sleep, circadian regulation, mood, and cognition. Both acylated and unacylated isoforms exhibit distinct biological activities, and ac

The structure and function of the ghrelin receptor coding for drug actions.

Drugs targeting the ghrelin receptor hold therapeutic potential in anorexia, obesity and diabetes. However, developing effective drugs is challenging. To tackle this common issue across a broad drug target, this study aims to understand how anamorelin, the only approved drug targeting the ghrelin receptor, operates compared to other synthetic drugs. Our research elucidated the receptor's structure with anamorelin and miniGq, unveiling anamorelin's superagonistic activity. We demonstrated that ligands with distinct chemical structures uniquely bind to the receptor, resulting in diverse conforma

To eat or not to eat: A role for ghrelin and LEAP2 in eating disorders?

Ghrelin is an orexigenic hormone that orchestrates many diverse behaviours of relevance for feeding control. It appears to operate as a hunger hormone, organizing food intake into meals and ensuring that we seek out and consume a variety of foods. With this physiological biography, the ghrelin system, including the pathways through which it operates, has been interrogated for its role in the aetiology, pathology and treatment of eating disorders. While common obesity does not appear to be a hyperghrelinemic state, it would be difficult to completely rule out enhanced ghrelin signalling. At the

Molecular Mechanisms and Health Benefits of Ghrelin: A Narrative Review.

Ghrelin, an endogenous brain-gut peptide, is secreted in large quantities, mainly from the stomach, in humans and rodents. It can perform the biological function of activating the growth hormone secretagogue receptor (GHSR). Since its discovery in 1999, ample research has focused on promoting its effects on the human appetite and pleasure-reward eating. Extensive, in-depth studies have shown that ghrelin is widely secreted and distributed in tissues. Its role in neurohumoral regulation, such as metabolic homeostasis, inflammation, cardiovascular regulation, anxiety and depression, and advanced

Insights Into the Regulation of Offspring Growth by Maternally Derived Ghrelin.

The regulation of fetal development by bioactive substances such as hormones and neuropeptides derived from the gestational mother is considered to be essential for the development of the fetus. On the other hand, it has been suggested that changes in the physiological state of the pregnant mother due to various factors may alter the secretion of these bioactive substances and induce metabolic changes in the offspring, such as obesity, overeating, and inflammation, thereby affecting postnatal growth and health. However, our knowledge of how gestational maternal bioactive substances modulate of

The Role of the Gastric Hormones Ghrelin and Nesfatin-1 in Reproduction.

Ghrelin and nesfatin-1 are enteroendocrine peptide hormones expressed in rat X/A-like and human P/D1cells of the gastric mucosa. Besides their effect on food intake, both peptides are also implicated in various other physiological systems. One of these is the reproductive system. This present review illustrates the distribution of ghrelin and nesfatin-1 along the hypothalamus-pituitary-gonadal (HPG) axis, their modulation by reproductive hormones, and effects on reproductive functions as well as highlighting gaps in current knowledge to foster further research.

Cachexia: Pathophysiology and Ghrelin Liposomes for Nose-to-Brain Delivery.

Cachexia, a severe multifactorial condition that is underestimated and unrecognized in patients, is characterized by continuous muscle mass loss that leads to progressive functional impairment, while nutritional support cannot completely reverse this clinical condition. There is a strong need for more effective and targeted therapies for cachexia patients. There is a need for drugs that act on cachexia as a distinct and treatable condition to prevent or reverse excess catabolism and inflammation. Due to ghrelin properties, it has been studied in the cachexia and other treatments in a growing n

Prader-Willi syndrome: A model for understanding the ghrelin system.

Subsequent to the discovery of ghrelin as the endogenous ligand of growth hormone secretagogue receptor 1a, this unique gut peptide has been found to exert numerous physiological effects, such as appetite stimulation and lipid accumulation via the central regulating mechanisms in the hypothalamus, stimulation of gastric motility, regulation of glucose metabolism and brown fat thermogenesis, and modulation of stress, anxiety, taste sensation, reward-seeking behaviour and the sleep/wake cycle. Prader-Willi syndrome (PWS) has been described as a unique pathological state characterised by severe o

Meta-analysis of ghrelin alterations in schizophrenia: Effects of olanzapine.

OBJECTIVE: Schizophrenia is associated with an increased prevalence of the metabolic syndrome. Patients receiving antipsychotic medications, including olanzapine, are at further risk. Ghrelin is an appetite-stimulating peptide hormone, although whether blood levels are altered by antipsychotic treatment, remains unclear. We performed a systematic review and meta-analysis comparing blood ghrelin levels in patients with schizophrenia before and after treatment with olanzapine. METHOD: Two authors independently searched major electronic databases from inception until February 2018 for studies me
