GABA transporters mediate glycine release from cerebellum nerve endings: roles of Ca(2+)channels, mitochondrial Na(+)/Ca(2+) exchangers, vesicular GABA/glycine transporters and anion channels.

GABA transporters accumulate GABA to inactivate or reutilize it. Transporter-mediated GABA release can also occur. Recent findings indicate that GABA transporters can perform additional functions. We investigated how activation of GABA transporters can mediate release of glycine. Nerve endings purified from mouse cerebellum were prelabeled with [(3)H]glycine in presence of the glycine GlyT1 transporter inhibitor NFPS to label selectively GlyT2-bearing terminals. GABA was added under superfusion conditions and the mechanisms of the GABA-evoked [(3)H]glycine release were characterized. GABA stim

Glycine attenuates sepsis-induced white matter injury by modulating gut microbiota.

Sepsis poses a significant threat to preterm infants and is a leading cause of white matter injury (WMI); however, effective therapeutic strategies remain limited. Recent studies suggest that gut microbiota dysbiosis contributes to sepsis-induced systemic inflammation and neurological damage. After treating mice with LPS-induced sepsis with glycine, we evaluated pathological changes in the brain and ileum by HE staining and analyzed gut microbiota composition by 16S rRNA gene sequencing. Inflammatory cytokine levels in brain and ileal tissues were quantified by ELISA. Transcriptomic profiling

Glycine-Functionalized Polycaprolactone Electrospun Nanofibers as Bioactive Scaffolds for Skin Repair.

Chronic cutaneous ulcers pose a major clinical challenge due to persistent inflammation, impaired angiogenesis, and limited regenerative capacity, underscoring the need to develop bioactive scaffolds that mimic the extracellular matrix (ECM) and promote skin repair. In this study, electrospun Poly(ε-caprolactone) (PCL) scaffolds functionalized with 10% (PCLGLI10) and 20% (PCLGLI20) glycine were fabricated, physicochemically and mechanically characterized, and evaluated in combination with human Wharton's jelly mesenchymal stromal cells (hWJ-MSCs). Glycine incorporation reduced fiber diameter t

Highly Efficient Electrosynthesis of Glycine over an Atomically Dispersed Iron Catalyst.

Glycine is a nonessential amino acid that plays a vital role in various biological activities. However, the conventional synthesis of glycine requires sophisticated procedures or toxic feedstocks. Herein, we report an electrochemical pathway for glycine synthesis via the reductive coupling of oxalic acid and nitrate or nitrogen oxides over atomically dispersed Fe-N-C catalysts. A glycine selectivity of 70.7% is achieved over Fe-N-C-700 at -1.0 V versus RHE. Synergy between the FeN3C structure and pyrrolic nitrogen in Fe-N-C-700 facilitates the reduction of oxalic acid to glyoxylic acid, which

Biological Investigations and Spectroscopic Studies of New Moxifloxacin/Glycine-Metal Complexes.

Two novel ligand-metal complexes were prepared through the reaction of Zn(II) and Sn(II) with moxifloxacin (MOX) in the presence of glycine (Gly) to investigate their biological activities. IR, UV/VIS and 1 H-NMR analysis have been carried out for insuring the chelation process. Results suggested that MOX and Gly react with the metal ions through the carbonyl oxygen atom and the oxygen atom of the carboxylic group of MOX and Gly. The antimicrobial activity was carried out against some common bacterial and fungal pathogens and the radical scavenging activity (RSA%) was evaluated using DPPH and

Glycine receptor antibody-associated epilepsy in a boy aged 4 years.

Our patient was a previously normal boy who presented to his local hospital with an explosive onset of prolonged seizures and encephalopathy. He was treated for a presumed central nervous system infection and initial neuroimaging was normal. Despite treatment with antibiotics and antiepileptic drugs (AEDs), he remained encephalopathic and became ataxic over the next 48 hours, not related to medication. The seizures also proved resistant to treatment despite polytherapy with AEDs, and he required immune-modulatory treatment, intravenous methylprednisolone and intravenous immunoglobulin, in addi

Hypoventilation in glycine-receptor antibody related progressive encephalomyelitis, rigidity and myoclonus.

Glycine receptor (GlyR) antibodies have been identified in patients with rigidity and hyperekplexia, but the clinical phenotype associated with these antibodies has not been fully elucidated. The clinical features in two additional patients with GlyR antibodies are described. A 55-year-old man presented with stimulus-induced hyperekplexia and rigidity in the lower limbs and trunk. He initially responded to benzodiazepines, but presented after 18 months with severe, painful, prolonged spasms associated with supraventricular and ventricular arrhythmias, hypoventilation and oxygen desaturation re

Spontaneous glycine-induced calcium transients in spinal cord progenitors promote neurogenesis.

Glycine and GABA are depolarizing during early development, but the purpose of this paradoxical chloride-mediated depolarization remains unclear, especially at early stages. It was previously reported that suppressing glycine signaling from the beginning of development in zebrafish embryos caused an abnormal maintenance of the progenitor population and a specific reduction of spinal interneurons but not of other cell populations. Here, we show that cells including progenitors in the embryonic spinal cord had occasional spontaneous, glycine-mediated calcium transients that were blocked by the g

GABA/glycine signaling during degeneration and regeneration of mouse hypoglossal nerves.

In the adult central nervous system (CNS), GABA and glycine (Gly) are predominant inhibitory neurotransmitters, negatively regulating glutamatergic transmission. In the immature CNS, on the other hand, they act as trophic factors, mediating morphogenesis. In the present study, to investigate their involvement in axonal regeneration, we morphologically examined changes in their signaling in mouse hypoglossal nuclei during degeneration and regeneration of hypoglossal nerves. We found that (1) expression and localization of presynaptic elements were not changed, (2) localization of gephyrin, whic

Repetitive light stimulation inducing glycine receptor plasticity in the retinal neurons.

Neurotransmitter receptor plasticity is a mechanism that can regulate the temporal and intensity encoding of a synapse. While this has been extensively studied as a mechanism of learning, less is known about such processes in sensory systems. This study examines modulation of glycine receptor function at the first synapse in the retina. It was found that horizontal cells, which are postsynaptic to photoreceptors, have glycine receptor currents that are enhanced when internal calcium is elevated. This can be achieved by glutamatergic synaptic input or by activation of voltage-gated calcium chan

Glycine induced calcium concentration changes in vestibular type I sensory cells.

Glutamate is the neurotransmitter of the synapse between vestibular type I hair cells and the afferent nerve calyx. This calyx may also be involved in local feedback, which may modify sensory cell activity via N-methyl-D-aspartate (NMDA) receptors. Glycine is the co-agonist of glutamate in NMDA receptor activation. Both agents have been detected by immunocytochemistry in the nerve calyx. Glutamate and NMDA stimulations cause changes in the intracellular calcium concentration ([Ca(2+)](i)) of isolated type I sensory cells. We investigated the effect of glycine stimulation on [Ca(2+)](i) in guin

Protein kinases modulate two glycine currents in salamander retinal ganglion cells.

1. Protein kinase modulation of glycine-activated currents was examined in acutely dissociated ganglion cells from tiger salamander retina using whole-cell voltage-clamp techniques. 2. Glycine (100 microM) induced an outward chloride current in cells clamped at 0 mV. Co-application of 50 microM forskolin made the glycine-induced current more transient. The combination of forskolin and glycine reduced the later portion of current response without changing the initial peak amplitude. 3. 3-Isobutyl-1-methylxanthine (IBMX) or 8-bromoadenosine 3',5'-cyclic monophosphate (8-Br-cAMP) produced effects

ECG and cardiac enzymes after glycine absorption in transurethral prostatic resection.

The electrocardiogram (ECG) was recorded and the serum levels of creatine kinase (CK), aspartate transaminase (ASAT) and prostatic acid phosphatase (PAP) was measured in 22 patients undergoing transurethral resection of the prostate (TURP) under spinal or epidural anaesthesia. The irrigating fluid consisted of 1.5% glycine and 1% ethanol, and absorption of the fluid was monitored by detection of alcohol in the expired breath. The results show that nine of the 11 patients (82%) who absorbed more than 1,000 ml of irrigating fluid had developed ECG changes 24 h after the operations. The most comm

Glycine and betaine improve the quality of cryopreserved sika deer sperm.

Sperm cryopreservation imposes osmotic stress due to the addition and removal of high-concentration cryoprotectants. Whether organic osmolytes can alleviate this stress to improve cryopreservation efficiency remains unclear. Here, we evaluated the effects of glycine and betaine, supplemented as organic osmolytes throughout the cryopreservation process, on the quality and developmental potential of cryopreserved sika deer sperm. Pooled semen samples from five healthy adult stags were randomly allocated to four groups: a control (base extender) and three treatment groups supplemented with glycin
