[Color perception disorders in therapeutic glycoside serum concentrations].

Vision deficiencies are dominant symptoms from digitalis intoxication. Twenty-nine and twenty-eight patients have been examined by the Farnsworth-Munsell-100-Hue-test under common maintenance therapy with digitoxin respectively digoxin or beta-acetyldigoxin. This was done to prove how glycosides may create colour vision disturbances even in "therapeutic" glycoside serum concentrations, and in which degree the lipophile digitoxin differs in its effect from the less lipophile digoxin. To exclude any congenital colour vision deficiencies the study was restricted to females. No definite difference

Combinatorial Biosynthesis of Antithrombotic Flavonoid 6-O-Glycosides From Carthamus tinctorius.

Flavonoid 6-O-glycosides from Carthamus tinctorius (safflower) represent a unique class of potential antithrombotic agents. However, their pharmaceutical development has been hindered by limited natural resources. In this study, we characterized four key biosynthetic enzymes from C. tinctorius: Ct6OGT (6-O-glycosyltransferase), Ct7OGT (7-O-glycosyltransferase), Ct3OGT (3-O-glycosyltransferase), and CtFLS (flavonol synthase). Among them, Ct6OGT is the first identified high-regioselectivity flavonoid 6-O-glycosyltransferase. By combining these enzymes with upstream genes identified from safflowe

Rare quinochalcone C-glycosides with vasoprotective activities from Carthamus tinctorius L.

Myocardial ischemia/reperfusion injury (MI/RI) is an inevitable consequence of reperfusion therapy for ischemic myocardium. Endothelial protection and therapeutic angiogenesis are pivotal strategies for attenuating MI/RI and restoring myocardial perfusion. To investigate the chemical basis underlying the vasoprotective potential of Carthamus tinctorius, we utilized both in vivo (PTK787-induced intersegmental vessel injury in zebrafish) and in vitro (OGD/R-injured HUVECs) assays. The 30% EtOH eluate obtained from D101 macroporous resin chromatography of the Carthamus tinctorius extract exhibite

Anticancer activity of triterpene glycosides from the sea star Solaster pacificus.

Marine triterpene glycosides are known to exhibit significant anticancer activity. We investigated pacificusoside C and cucumariosides C1 and C2 isolated from a sea star Solaster pacificus in prostate cancer models with varying drug resistance and in non-cancerous cells in vitro. Cucumarioside C1 showed selectivity comparable to cisplatin, whereas the other compounds were less selective. Cucumarioside C1 induced apoptosis and enhanced cytotoxic effects of cisplatin, carboplatin, docetaxel, and cabazitaxel, making it a potential candidate for combination therapy. All three glycosides were activ

Structurally diverse sesquiterpenoid glycosides with anti-inflammatory activity from Cissampelopsis spelaeicola.

Sixteen previously undescribed sesquiterpenoid glycosides (1-16), bearing eudesmane-type (1-4) and cadinane-type skeletons (5-16), were obtained from the stems of Cissampelopsis spelaeicola. The structural elucidation and absolute configuration assignment of compounds 1-16 were accomplished through multiple spectroscopic analyses, computed ECD/NMR, and X-ray crystallography. Furthermore, anti-neuroinflammatory effects of all isolates were assessed by NO inhibition in LPS-stimulated BV-2 microglia. Compounds 12 and 14-16 exhibited potent NO inhibitory effects (IC50 = 5.62-19.29 μM), outperformi

The mechanisms of tripterygium glycosides-induced reproductive toxicity and detoxification strategies.

Tripterygium glycosides (TG) is a widely used preparation in the treatment of rheumatoid arthritis (RA), nephrotic syndrome and diabetic nephropathy. Although the clinical efficacy is definite, the side-effects on reproductive system limit its wide application. It is of great significance to take measures to alleviate its reproductive toxicity and expand its clinical use. The mechanism of TG-induced reproductive toxicity involves oxidative stress, inflammation, apoptosis, and metabolism imbalance, which lead to adverse effects on male and female reproductive organs. To mitigate these effects,

Ten new resin glycosides, ipoalbins I-X, from Ipomoea alba seeds.

Ipomoea alba L. (Convolvulaceae) is an annual vine native to tropical America that is cultivated primarily for ornamental purposes. Its seeds are used in traditional medicine as a laxative, and young shoots are consumed as food. In this study, ten new resin glycosides, ipoalbins I (1)-X (10), were isolated from I. alba seeds. The structures of 1-10 were determined based on spectroscopic data. All compounds had intramolecular cyclic ester structures (jalapins). The sugar moieties in 1-10 were partially acylated by organic acids, including isobutyric, (E)-2-methylbut-2-enoic, and 2S-methyl-3S-hy

Stilbene glycosides alleviate atherosclerosis partly by promoting lipophagy of dendritic cells.

Atherosclerosis (AS) is a chronic inflammatory disease resulting from lipid metabolism disorders and immune imbalances. Dendritic cells (DCs) are key cells that regulate adaptive and adaptive immunity. When DCs engulf excessive amounts lipids, their function is altered, thereby, accelerating the inflammatory process of AS. Cellular lipophagy serves to reduce lipid accumulation and maintain cellular lipid metabolism balance. In this study, we investigated the effectiveness of 2,3,5,4'-tetrahydroxystilbene 2-O-β-D-glucoside (TSG) in intervening in the promotion of DCs lipid accumulation by ox-LD

Enzymatic Biosynthesis of Simple Phenolic Glycosides as Potential Anti-Melanogenic Antioxidants.

Simple phenolics (SPs) and their glycosides have recently gained much attention as functional skin-care resources for their anti-melanogenic and antioxidant activities. Enzymatic glycosylation of SP aglycone make it feasible to create SP glycosides with updated bioactive potentials. Herein, a glycosyltransferase (GT)-encoding gene was cloned from the fosmid libraries of Streptomyces tenjimariensis ATCC 31603 using GT-specific degenerate PCR followed by in silico analyses. The recombinant StSPGT was able to flexibly catalyze the transfer of two glycosyl moieties towards two SP acceptors, (hydro

Factors influencing the variability of antioxidative phenolic glycosides in Salix species.

Phenolic glycosides, especially the salicylates, are important secondary metabolites in the bark of willows (Salix spp.). Because of their anti-inflammatory, analgesic, and fever-reducing properties, they are of particular interest to society. Compared to the fabrication of synthetic salicylacetylic acid, the commercial production of willow bark extracts with adequate amounts of salicylate is very difficult due to several biological and technical reasons. Therefore, one of the objectives was to identify salicylate-rich clones from three species, Salix daphnoides , Salix purpurea , and Salix pe

Hypoglycemic effects of sesquiterpene glycosides and polyhydroxylated triterpenoids of Eriobotrya japonica.

The effects of the constituent sesquiterpene glycosides 1-3 and polyhydroxylated triterpenoids 5-6 isolated by MeOH extraction of Eriobotrya japonica were studied in genetically diabetic mice (C57BL/KS-db/db/Ola) and normoglycemic rats. The sesquiterpene glycoside 3 and the polyhydroxylated triterpenoids 5 and 6 produced a marked inhibition of glycosuria. Furthermore, 5 and 6 were able to reduce blood glucose levels in normoglycemic rats. While there are already some data reported on hypoglycemic activity of polyhydroxylated triterpenoids, there are no previous data showing hypoglycemic activi
