Antipathogenic properties of camel milk have been investigated to substitute for drugs hence overcome drug resistance. The main objective of this present study was to investigate the anthelmintic activity of camel milk in relationship to its chemical composition. In vitro anthelmintic effects of camel milk against Haemonchus contortus from sheep were ascertained by egg hatching and worm motility inhibitions in comparison to milks from cow, ewe and goat as well as a reference drug albendazole. Chemical composition revealed that camel milk has higher contents of protective protein (lactoferrin)

Anthelmintic resistance is an increasing problem that threatens livestock production worldwide. Understanding of the genetic basis of benzimidazole resistance recently allowed the development of promising molecular diagnostic tools. In this study, isolates of Haemonchus contortus obtained from goats, sheep and buffaloes raised in Brazil were screened for presence of the polymorphism Phe200Tyr in the β-tubulin 1 gene, which confers resistance to benzimidazole. The allelic frequency of the mutation conferring resistance ranged from 7% to 43%, and indicated that resistance to benzimidazole could

Infections with Haemonchus contortus are a major constraint on ruminant health world-wide. Young lambs are very sensitive to Haemonchus infection. Older lambs and sheep acquire immunity after a continuous or seasonal exposure to the parasite. The mechanisms underlying immunity are still not completely understood. Antibodies, in particular local IgA and IgE, certainly play a role. The role of IgG is less clear. Lymphocyte proliferation responses seem to correlate to immunity. Sheep that have high antigen-induced lymphocyte responses have a low susceptibility to infection. Furthermore, several s

The distribution of granule-specific sheep mast cell proteinase (SMCP), was assayed by immunocytochemistry and quantified by immunoassay in sheep immune to Haemonchus contortus. Repeated infection with Haemonchus larvae over 10-12 weeks induced a pronounced mucosal mastocytosis, including intraepithelial globule leukocytes (GL), which, 7 days after ceasing this dosing regime, was associated with the inability of incoming larvae to establish within the abomasal mucosa. Loss of this resistance, due to the cessation of stimulation with Haemonchus larvae 84 days previously or to treatment of sheep

Gastrointestinal nematodes (GINs), particularly Haemonchus contortus and Trichostrongylus colubriformis, threaten sheep health and welfare globally, while anthelmintic resistance drives the need for sustainable alternatives. Essential oils (EOs) from Citrus species, rich in bioactive terpenes, represent a promising option, especially as locally sourced by-products. Five furocoumarin-free (FCF) Citrus EOs from Calabria (bergamot, sweet orange, lemon, mandarin, grapefruit) were chemically characterised by GC-MS. Anthelmintic activity against H. contortus and T. colubriformis was evaluated using

BACKGROUND: Antiparasitic resistance in sheep necessitates non-chemical strategies for gastrointestinal strongylosis control. We have evaluated the potential of three predatory mite species-Macrocheles robustulus, Macrocheles muscaedomesticae and Rhabdocarpais consanguineus-to reduce transmission of the nematode Haemonchus contortus by preying on infective larvae.
METHODS: Under laboratory conditions, mites were exposed to mixed prey environments containing housefly Musca domestica eggs and H. contortus third-stage (L3) larvae. Predation was first monitored over 8 h. This was followed by a mo

The macrocyclic lactone eprinomectin has a null milk withdrawal period, and plays a key role in limiting infection by gastro-intestinal nematodes (GIN) in dairy sheep. Starting in 2019, a cluster of suspicions of lack of effectiveness of eprinomectin (EPN) was observed in the western area of the French Pyrenees. First, we conducted fecal egg count reduction tests in 47 dairy sheep farms of this area. They revealed that 29/47 (62 %) farms were facing resistance to eprinomectin. Post-treatment GIN species identification conducted in 21 of the 29 resistance cases showed Haemonchus contortus was t

Due to differences in their feeding behavior, sheep and goats are often assumed to respond differently to helminth infections. The present study compared Haemonchus contortus infection profile between sheep and goats under the same experimental setting. Experimental infection was conducted using a randomized block design in four groups of intact sheep (InfSH and ConSH) and goats (InfG, and ConG). Groups InfSH and InfG (N = 7 each) received 10,000L3 of H. contortus whereas the control groups ConSH and ConG (N = 7 each) remained uninfected. Faecal egg counts and PCV were measured from Day 0 to d

The gastrointestinal nematode infection poses a covert threat to both humans and domestic animals worldwide, eliciting a type 2 immune response within the small intestine. Intestinal tuft cells detect the nematode and activated group 2 innate lymphoid cells. Tuft cell-derived leukotrienes (one of the metabolites of arachidonic acid) were found to drive rapid anti-helminth immunity, but it is still poorly understood whether the tuft cell-mediated type 2 immune circuit and arachidonic acid metabolism modulate anti-parasitic immunity in the gastric epithelium. This study was designed to evaluate

Haemonchus contortus is the most pathogenic nematode in small ruminants and anthelmintic resistance (AR) hampers its efficient control. Early detection of AR status is required to reduce selection for AR and cannot be achieved using phenotypic tests. For benzimidazoles (BZs), the detection of AR-associated alleles characterised by single nucleotide polymorphisms (SNPs) in the isotype 1 β-tubulin gene allows early AR detection in strongyles. The F200Y, F167Y, E198A and E198L polymorphisms have been described in BZ-resistant populations with a clear variation in frequencies between regions. A no

A population of Haemonchus contortus that was highly resistant to benzimidazoles and avermectin/milbemycins with a subpopulation that was resistant to levamisole, was replaced with a susceptible laboratory isolate of H. contortus in a flock of sheep. The anthelmintic susceptibility and population genetics of the newly established population were evaluated for 3.5 years using in vivo, in vitro, and molecular methods. Successful replacement of the resistant population with a susceptible population was confirmed using phenotypic and genotypic measurements; larval development assay indicated full

It is increasingly difficult to manage and control gastrointestinal nematode parasites in pasture-based ruminant livestock operations because of the high prevalence of anthelmintic resistance. Anthelmintics should be combined with alternative forms of control. Sustainable tools include copper oxide wire particles and condensed tannin-rich plants, which target primarily Haemonchus contortus in small ruminants. Nematophagous fungi reduce larvae on pasture and target nematode larvae in feces of most livestock species. In addition, and perhaps most importantly, genetic selection focuses on parasit

Managing infections of sheep with anthelmintic resistant gastrointestinal nematodes (GIN) is a major challenge for sheep producers in Western Europe. New methods of grazing management have been poorly explored as a component of an integrated and sustainable control of these parasites. Therefore, the purpose of this study was to evaluate the effect of two different types of grazing systems of sheep (intensive cell grazing versus conventional rotational grazing) on GIN infections over two years in a farm located in a temperate environment of Western France. When considering the whole study, the

In a context of nematodicidal resistance, anthelmintic combinations have emerged as a reliable pharmacological strategy to control gastrointestinal nematodes in grazing systems of livestock production. The current work evaluated the potential drug-drug interactions following the coadministration of two macrocyclic lactones (ML) ivermectin (IVM) and abamectin (ABM) to parasitized cattle using a pharmacokinetic/pharmacodynamic (PK/PD) approach. The kinetic behavior of both compounds administered either separately or coadministered was assessed and the therapeutic response of the combination was

There is no effective natural alternative control for gastrointestinal nematodes (GIN) of small ruminants, with Haemonchus contortus being the most economically important GIN. Despite frequent reports of multidrug-resistant GIN, there is no new commercial anthelmintic to substitute failing ones. Although trematocidal activity of artemisinin analogs has been reported in sheep, neither artemisinin nor its plant source (Artemisia annua) has been evaluated for anthelmintic activity in ruminants. This study evaluated the anthelmintic activity of A. annua crude extracts in vitro and compared the mos

Haemonchosis is a very common disease in small ruminants caused by H. contortus, a blood sucking parasite causing anaemia that may be fatal particularly to young animals. Therefore, detection of the infection during prepatent period is important for early treatment. Excretory-Secretory (ES) protein of H. contortus was purified through immunoaffinity chromatography. Dot -ELISA was performed with crude ES antigen as well as immunoaffinity purified fraction (F-1) with experimental and natural sera of sheep infected with H. contortus. Solid dot formation took place with 4 day, 1, 2 and 3 weeks pos

In all, 2 groups of lambs were infected either with Oestrus ovis first-instar larvae or with 10,000 third-stage larvae (L3) of Haemonchus contortus. Another group of lambs was infected with both parasites. Fecal nematode egg counts, plasma pepsinogen concentrations, specific O. ovis enzyme-linked immunosorbent assay (ELISA) antibodies, and blood eosinophil counts were monitored and compared to with the values recorded for a control group of uninfected lambs. There was no significant difference between the burden of H. contortus found in mixed and single infections. However, the nematode egg pr
