Ginsenoside Rg3 Promotes Healthy Aging Through Phosphoinositide-Associated Lysosomal Regulation in Caenorhabditis elegans.

Lysosomal decline and proteostasis collapse are defining features of aging and major contributors to neurodegenerative vulnerability, yet interventions that preserve neuronal lysosomal function across aging remain poorly defined. Here, we identify 20(S)-ginsenoside Rg3, a ginseng-derived natural product, as a geroprotective small molecule in Caenorhabditis elegans. Rg3 extended lifespan and preserved key physiological outputs, including locomotion and pharyngeal pumping, indicating that Rg3 delays functional aging. Rg3 also maintained mechanosensory neuron integrity and touch sensitivity, supp

Chrononutrition in the aging era: Circadian regulation of metabolism, resilience, and healthy longevity.

The increasing prevalence of obesity, type 2 diabetes, cardiovascular disease, and age-related disorders has been linked to a growing mismatch between modern lifestyles and the biological rhythms that regulate human physiology. Chrononutrition, which explores the interaction between meal timing, circadian rhythms, and metabolism, has emerged as a promising strategy to improve metabolic health and support healthy aging. This review examines the molecular and physiological foundations of chrononutrition, focusing on the interplay between central and peripheral circadian clocks, metabolic regulat

Low genetic risk for coronary artery disease underlies multigenerational longevity and healthy aging.

Aging is a major risk factor for chronic diseases. Unlike the general population, members of long-lived families maintain exceptional health as they age, with over 10 years delayed onset of their first chronic disease. We therefore hypothesize that one of the key features explaining healthy survival up to high ages (longevity) is the absence of chronic disease risk alleles. We investigated this hypothesis in the Leiden Longevity Study, a cohort with data from more than 420 long-lived families in three generations and the Leiden 85-plus study. To analyze our data, we constructed a set of polyge

Alzheimer's Disease Accelerates Cerebral Atrophy by Over a Decade Compared to Healthy Aging.

Brain aging is accompanied by progressive morphological and neurobiological changes, which are significantly accelerated in neurodegenerative diseases, such as Alzheimer's disease. Detecting and differentiating these changes early is crucial for diagnosis, treatment planning, and therapeutic development. In this work, we present a computational multiphysics framework that couples protein biomarker propagation with tissue-level atrophy to distinguish between cognitively normal aging, mild cognitive impairment, and Alzheimer's disease. Our model integrates a network-based simulation of amyloid b

Early Life Nutrition: The First 1000 Days and Healthy Aging in Cystic Fibrosis.

Achievement of optimal nutritional status within the first 1000 days of life for a child with cystic fibrosis (CF) is of paramount importance, with an emphasis on favorable early life growth trajectories that best optimize pulmonary and extrapulmonary health. The 'first 1000 days' framework emphasizes that environmental, sociocultural and nutritional exposures during this period can have life-long consequences for physical, cognitive, social and emotional health and development. Optimal nutrition encompasses not just physical growth, but the provision of nutrients and optimal feeding throughou

Inflammaging and the role of micronutrients as immunomodulators: a pathway to healthy aging.

BACKGROUND: Healthy aging is increasingly challenged by inflammaging, a chronic, low‑grade inflammatory state primarily exacerbated by gut microbiota dysbiosis and declining immune function. Persistent digestive and systemic inflammation, along with immunosenescence, contributes to multiple age‑related diseases. Micronutrients regulate key components of immune system and support the composition and function of the gut microbiota, underscoring their emerging role as modulators of inflammaging. MAIN BODY: This narrative review synthesizes current evidence insights on how micronutrients regulate

Anti-inflammatory phytochemicals from medicinal herbs based on senoinflammation schema for healthy aging.

Aging is characterized by chronic inflammation involving a continuous, multifarious, low-grade inflammatory response that contributes to age-related chronic diseases. Recently, researchers have focused on age-related chronic inflammation and anti-inflammatory phytochemicals to ameliorate age-related degenerative conditions. As proposed earlier, senoinflammation is a broader view of age-related inflammation with an insightful perspective, which differs from the traditional phenomenal view. Recent advances have identified the senescence-associated secretory phenotype (SASP) as a central mediator

Plasma Metabolomic Signature Predicting Deviation from Healthy Vascular Aging in Two European Cohorts.

INTRODUCTION: Biological rather than chronological age (C-age) drives deviation from healthy aging. This study aimed to identify a plasma metabolomic signature indicative of age-related cardiovascular risk (pMTB-age) predicting vascular morbidity and mortality. METHODS: Nuclear magnetic resonance identified 38 plasma metabolites in two population cohorts: the Flemish cohort examined as discovery (N = 719 [2005-2010]) and internal replication cohort (N = 580/719 [2009-2013]) and the external replication Spanish Hortega cohort (N = 811 [2001]). RESULTS: The trained model (pMTB-age), relating C

Biases in an artificial intelligence image-generator's depictions of healthy aging and Alzheimer's.

OBJECTIVE: This content analysis study investigates potential biases in image generation by 2 artificial intelligence (AI) tools, DALL-E 3 and Midjourney, in portraying older adults and individuals living with dementia. Despite widespread use of generative AI in various sectors, there is limited research on how these models might perpetuate stereotypes and stigmatization through their images. MATERIALS AND METHODS: 1056 images were generated using specified prompts categorized into 3 groups: general older adults, dementia-related, and control. Each prompt began with "photorealistic portrait"

Pursuing healthy aging and longevity through natural product and stem cell-driven rejuvenation.

Aging is an inevitable biological process associated with progressive physiological decline and increased disease susceptibility. Cellular senescence stands as a key mechanism among the hallmarks of aging, which is characterized by irreversible cell-cycle arrest, chromatin remodeling, and a pro-inflammatory senescence-associated secretory phenotype (SASP). Importantly, SASP drives inflammaging and propagates senescence via a bystander effect, exacerbating tissue dysfunction. Recent advances in senolytic therapies and senostatics offer promising strategies to eliminate or rejuvenate senescent c

Astrocytes phenomics as new druggable targets in healthy aging and Alzheimer's disease progression.

For over a century after their discovery astrocytes were regarded merely as cells located among other brain cells to hold and give support to neurons. Astrocytes activation, "astrocytosis" or A1 functional state, was considered a detrimental mechanism against neuronal survival. Recently, the scientific view on astrocytes has changed. Accumulating evidence indicate that astrocytes are not homogeneous, but rather encompass heterogeneous subpopulations of cells that differ from each other in terms of transcriptomics, molecular signature, function and response in physiological and pathological con

Deciphering the influence of gut and oral microbiomes on menopause for healthy aging.

Menopause is characterized by the cessation of menstruation and a decline in reproductive function, which is an intrinsic component of the aging process. However, it has been a frequently overlooked field of women's health. The oral and gut microbiota, constituting the largest ecosystem within the human body, are important for maintaining human health and notably contribute to the healthy aging of menopausal women. Therefore, a comprehensive review elucidating the impact of the gut and oral microbiota on menopause for healthy aging is of paramount importance. This paper presents the current un

Plasma markers of neurodegeneration, latent cognitive abilities and physical activity in healthy aging.

Blood-based biomarkers of neurodegeneration demonstrate great promise for the diagnosis and prognosis of Alzheimer's disease. Ultra-sensitive plasma assays now allow for quantification of the lower concentrations in cognitively unimpaired older adults, making it possible to investigate whether these markers can provide insight also into the early neurodegenerative processes that affect cognitive function and whether the markers are influenced by modifiable risk factors. Adopting an exploratory approach in 93 healthy older adults (65-75 years), we used structural equation modelling to investiga

The bottlenose dolphin (Tursiops truncatus): a novel model for studying healthy arterial aging.

Endothelial function declines with aging and independently predicts future cardiovascular disease (CVD) events. Diving also impairs endothelial function in humans. Yet, dolphins, being long-lived mammals adapted to diving, undergo repetitive cycles of tissue hypoxia-reoxygenation and disturbed shear stress without manifesting any apparent detrimental effects, as CVD is essentially nonexistent in these animals. Thus, dolphins may be a unique model of healthy arterial aging and may provide insights into strategies for clinical medicine. Emerging evidence shows that the circulating milieu (bioact

Are adverse childhood experiences associated with trajectories of healthy aging? Evidence from China.

Adverse childhood experiences (ACEs) have life-course impact and may be associated with healthy aging. This study aimed to explore the association between ACEs and healthy aging trajectories among middle- and older-aged adults in China. The data were obtained from the China Health and Retirement Longitudinal Study from 2011 to 2018. A total of 8906 respondents were enrolled in the analysis. Through latent growth mixture model, five categories of healthy aging trajectories which defined as 'stable type with high starting point', 'stable type at the middle level', 'stable type with low starting

A Cross-sectional Study of Perceptual and Acoustic Voice Characteristics in Healthy Aging.

PURPOSE: The human voice qualitatively changes across the lifespan. Although some of these vocal changes may be pathologic, other changes likely reflect natural physiological aging. Normative data for voice characteristics in healthy aging is limited and disparate studies have used a range of different acoustic features, some of which are implicated in pathologic voice changes. We examined the perceptual and acoustic features that predict healthy aging. METHOD: Participants (N = 150) aged between 50 and 92 years performed a sustained vowel task. Acoustic features were measured using the Multi

Alpha desynchronization during simple working memory unmasks pathological aging in cognitively healthy individuals.

Our aim is to explore if cognitive challenge combined with objective physiology can reveal abnormal frontal alpha event-related desynchronization (ERD), in early Alzheimer's disease (AD). We used quantitative electroencephalography (qEEG) to investigate brain activities during N-back working memory (WM) processing at two different load conditions (N = 0 or 2) in an aging cohort. We studied 60-100 year old participants, with normal cognition, and who fits one of two subgroups from cerebrospinal fluid (CSF) proteins: cognitively healthy (CH) with normal amyloid/tau ratio (CH-NAT, n = 10) or path

Voxel-wise deviations from healthy aging for the detection of region-specific atrophy.

The identification of pathological atrophy in MRI scans requires specialized training, which is scarce outside dedicated centers. We sought to investigate the clinical usefulness of computer-generated representations of local grey matter (GM) loss or increased volume of cerebral fluids (CSF) as normalized deviations (z-scores) from healthy aging to either aid human visual readings or directly detect pathological atrophy. Two experienced neuroradiologists rated atrophy in 30 patients with Alzheimer's disease (AD), 30 patients with frontotemporal dementia (FTD), 30 with dementia due to Lewy-body
