Clinical, histopathological, and immunohistochemical studies of histiocytic sarcoma in four-toed hedgehogs (Atelerix albiventris): A retrospective study.

Histiocytic sarcoma was investigated histopathologically and immunohistochemically in 17 four-toed hedgehogs (Atelerix albiventris), along with a review of their clinical data. Cases were histopathologically classified into two types: round-polygonal cell type (6 cases) and spindle cell type (11 cases). Round-polygonal cell type was found in visceral organs such as the spleen, lymph nodes, and more, and most cases of this type were consistent with disseminated histiocytic sarcoma. On the other hand, spindle cell type occurred mainly in skin, and almost all cases were consistent with localized

Paraechinus aethiopicus (Ehrenberg 1832) and Atelerix algirus (Lereboullet 1842) hedgehogs: Possible reservoirs of endemic leishmaniases in Tunisia.

Rodents and dogs are the confirmed leishmaniases reservoir hosts in Tunisia. Recently, we described hedgehog Leishmania (L.) major and L. infantum infection in an L. infantum endemic area in the North-West. In order to assess if the observation could extend to other endemic areas and to highlight the potential role of hedgehogs as reservoir host, we aimed here at investigating their Leishmania infection in different foci in Tunisia located along a North-South transect, during and outside different transmission seasons. Based on morphological criteria, 2 hedgehogs' species, Atelerix algirus and

Comparative population dynamics of a generalist (Ixodes ricinus) and specialist tick (I. hexagonus) species from European hedgehogs.

Although the population dynamics of the tick Ixodes ricinus are relatively well studied, those of other Western European tick species are largely unknown. Moreover, there is very little information related to the interactions between I. ricinus and other ticks. Such knowledge, however, is of special interest in respect to the epidemiology of tick-borne pathogens such as Borrelia spp. We compared the dynamics of the generalist I. ricinus with the nest-dwelling hedgehog specialist, I. hexagonus. Both species were collected from hedgehogs from a naturally infested experimental population between

Sequencing of Betacoronavirus erinacei from faeces of pet hedgehogs demonstrates a continuity of MERS-CoV like viruses in European and Eurasian hedgehog species.

Hedgehogs have been recently identified as carriers of Betacoronavirus erinacei (also known as Erinaceus coronavirus, EriCoV) a virus closely related to B. cameli responsible for human Middle East Respiratory Syndrome (MERS), raising questions about the risk of hedgehog-to-human transmission and suggesting the need for coronavirus (CoV) surveillance in hedgehogs. This study investigated the presence of CoVs in fecal samples of hedgehogs kept as pets in Italy in 2021-2022. A pan-CoV nested RT-PCR targeting the RdRp gene was used for screening and positive samples were sequenced and phylogenetic

Hedgehog Inhibitor and Paclitaxel: A Phase I Trial of the Oral Hedgehog inhibitor Taladegib in Combination with Weekly Paclitaxel in Patients with Advanced Solid Tumours.

The hedgehog (Hh) signalling pathway is aberrantly activated in solid tumours. Inhibition of Hh signalling by SMO inhibitors is effective in the treatment of basal cell carcinomas and medulloblastomas whereas combination treatments may be required in other solid tumours. When given with an SMO inhibitor in preclinical models, the efficacy of paclitaxel is enhanced and there is potential reversal of mechanisms of resistance to paclitaxel supporting clinical investigation of the combination. In this phase I trial, the safety and feasibility of the oral SMO inhibitor, taladegib, was evaluated in

The Role of the Hedgehog Pathway in Alcohol-Induced Birth Defects.

In this chapter, we review the current literature that focuses on the role of Sonic hedgehog (Shh) signaling, a major morphogenic pathway regulating embryonic development, in the pathology of fetal alcohol spectrum disorder (FASD), caused by prenatal alcohol exposure (PAE). Ethanol exposure disrupts Shh signaling during development across a range of species including in mice, chick, and zebrafish. Ethanol appears to interfere with Shh signaling through multiple mechanisms, both direct and indirect, including reduced Shh ligand production, impaired post-translational processing, and inducing ap

Hearing and anatomy of the ear of the European hedgehog Erinaceus europaeus.

A major threat to the declining European hedgehog (Erinaceus europaeus) is road traffic. Devising methods to reduce the number of collisions would increase hedgehog welfare in an urbanized world and serve to protect this flagship species, and this goal might be advanced by an understanding of their hearing. This study investigates the auditory capabilities and anatomy of the ear of the European hedgehog. Using auditory brainstem response testing on 20 live hedgehogs from Danish wildlife rescue centres, we measured hearing thresholds across 4-85 kHz and found a peak sensitivity around 40 kHz, r

Numb positively regulates Hedgehog signaling at the ciliary pocket.

Hedgehog (Hh) signaling relies on the primary cilium, a cell surface organelle that serves as a signaling hub for the cell. Using proximity labeling and quantitative proteomics, we identify Numb as a ciliary protein that positively regulates Hh signaling. Numb localizes to the ciliary pocket and acts as an endocytic adaptor to incorporate Ptch1 into clathrin-coated vesicles, thereby promoting Ptch1 exit from the cilium, a key step in Hh signaling activation. Numb loss impedes Sonic hedgehog (Shh)-induced Ptch1 exit from the cilium, resulting in reduced Hh signaling. Numb loss in spinal neural

A simple and scalable zebrafish model of Sonic hedgehog medulloblastoma.

Medulloblastoma (MB) is the most common malignant brain tumor in children and is stratified into three major subgroups. The Sonic hedgehog (SHH) subgroup represents ∼30% of all MB cases and has significant survival disparity depending upon TP53 status. Here, we describe a zebrafish model of SHH MB using CRISPR to create mutant ptch1, the primary genetic driver of human SHH MB. In these animals, tumors rapidly arise in the cerebellum and resemble human SHH MB by histology and comparative onco-genomics. Similar to human patients, MB tumors with loss of both ptch1 and tp53 have aggressive tumor h

The Hedgehog Pathway as a Therapeutic Target in Chronic Myeloid Leukemia.

Despite the development of therapeutic agents that selectively target cancer cells, relapse driven by acquired drug resistance and resulting treatment failure remains a significant issue. The highly conserved Hedgehog (HH) signaling pathway performs multiple roles in both development and tissue homeostasis, and its aberrant regulation is known to drive the pathogenesis of numerous human malignancies. However, the role of HH signaling in mediating disease progression and drug resistance remains unclear. This is especially true for myeloid malignancies. The HH pathway, and in particular the prot

Hedgehog-GLI mediated control of renal formation and malformation.

CAKUT is the leading cause of end-stage kidney disease in children and comprises a broad spectrum of phenotypic abnormalities in kidney and ureter development. Molecular mechanisms underlying the pathogenesis of CAKUT have been elucidated in genetic models, predominantly in the mouse, a paradigm for human renal development. Hedgehog (Hh) signaling is critical to normal embryogenesis, including kidney development. Hh signaling mediates the physiological development of the ureter and stroma and has adverse pathophysiological effects on the metanephric mesenchyme, ureteric, and nephrogenic lineag

Hedgehog Autoprocessing: From Structural Mechanisms to Drug Discovery.

Hedgehog (Hh) signaling plays pivotal roles in embryonic development. In adults, Hh signaling is mostly turned off but its abnormal activation is involved in many types of cancer. Hh signaling is initiated by the Hh ligand, generated from the Hh precursor by a specialized autocatalytic process called Hh autoprocessing. The Hh precursor consists of an N-terminal signaling domain (HhN) and a C-terminal autoprocessing domain (HhC). During Hh autoprocessing, the precursor is cleaved between N- and C-terminal domain followed by the covalent ligation of cholesterol to the last residue of HhN, which

Hedgehog signalling in bone and osteoarthritis: the role of Smoothened and cholesterol.

Hedgehog signalling is essential for development, crucial for normal anatomical arrangement and activated during tissue damage repair. Dysregulation of hedgehog signalling is associated with cancer, developmental disorders and other diseases including osteoarthritis (OA). The hedgehog gene was first discovered in Drosophila melanogaster, and the pathway is evolutionarily conserved in most animals. Although there are several hedgehog ligands with different protein expression patterns, they share a common plasma membrane receptor, Patched1 and hedgehog signalling pathway activation is transduced

Role of the hedgehog signaling pathway in rheumatic diseases: An overview.

Hedgehog (Hh) signaling pathway is an evolutionarily conserved signal transduction pathway that plays an important regulatory role during embryonic development, cell proliferation, and differentiation of vertebrates, and it is often inhibited in adult tissues. Recent evidence has shown that Hh signaling also plays a key role in rheumatic diseases, as alterations in their number or function have been identified in rheumatoid arthritis, osteoarthritis, ankylosing spondylitis, systemic sclerosis, and Sjogren's Syndrome. As a result, emerging studies have focused on the blockade of this pathogenic

The development of zebrafish pancreas affected by deficiency of Hedgehog signaling.

The pancreas development depends on complex regulation of several signaling pathways, including the Hedgehog (Hh) signaling via a receptor complex component, Smoothened, which deficiency blocks the Hh signaling. Such a defect in birds and mammals results in an annular pancreas. We showed that in developing zebrafish, the mutation of Smoothened or inhibition of Hh signaling by its antagonist cyclopamine caused developmental defects of internal organs, liver, pancreas, and gut. In particular, the pancreatic primordium was duplicated. The two exocrine pancreatic primordia surround the gut. This p

Hedgehog signaling underlying tendon and enthesis development and pathology.

Hedgehog (Hh) signaling has been widely acknowledged to play essential roles in many developmental processes, including endochondral ossification and growth plate maintenance. Furthermore, a rising number of studies have shown that Hh signaling is necessary for tendon enthesis development. Specifically, the well-tuned regulation of Hh signaling during development drives the formation of a mineral gradient across the tendon enthesis fibrocartilage. However, aberrant Hh signaling can also lead to pathologic heterotopic ossification in tendon or osteophyte formation at the enthesis. Therefore, th
