P2X7 receptor-mediated depression-like reactions arising in the mouse medial prefrontal cortex.

Major depressive disorder is a frequent and debilitating psychiatric disease. We have shown in some of the acute animal models of major depressive disorder (tail suspension test and forced swim test) that depression-like behavior can be aggravated in mice by the microinjection into the medial prefrontal cortex of the P2X7R agonistic adenosine 5'-triphosphate or its structural analog dibenzoyl-ATP, and these effects can be reversed by the P2X7R antagonistic JNJ-47965567. When measuring tail suspension test, the prolongation of immobility time by the P2YR agonist adenosine 5'-[β-thio]diphosphate

Salmon Calcitonin Exerts an Antidepressant Effect by Activating Amylin Receptors.

Depressive disorder is defined as a psychiatric disease characterized by the core symptoms of anhedonia and learned helplessness. Currently, the treatment of depression still calls for medications with high effectiveness, rapid action, and few side effects, although many drugs, including fluoxetine and ketamine, have been approved for clinical usage by the Food and Drug Administration (FDA). In this study, we focused on calcitonin as an amylin receptor polypeptide, of which the antidepressant effect has not been reported, even if calcitonin gene-related peptides have been previously demonstrat

Hippocampal Neurogenesis Is Enhanced in Adult Tau Deficient Mice.

Tau dysfunction is common in several neurodegenerative diseases including Alzheimer's disease (AD) and frontotemporal dementia (FTD). Affective symptoms have often been associated with aberrant tau pathology and are commonly comorbid in patients with tauopathies, indicating a connection between tau functioning and mechanisms of depression. The current study investigated depression-like behavior in Mapt-/- mice, which contain a targeted deletion of the gene coding for tau. We show that 6-month Mapt-/- mice are resistant to depressive behaviors, as evidenced by decreased immobility time in the f

Apelin-13 exerts antidepressant-like and recognition memory improving activities in stressed rats.

Apelin is the endogenous ligand for the G-protein-coupled receptor (APJ). The localization of APJ in limbic structures suggests a potential role for apelin in emotional processes. However, the role of apelin in the regulation of stress-induced responses such as depression and memory impairment is largely unknown. In the present study, we evaluated the role of apelin-13 in the regulation of stress-induced depression and memory impairment in rats. We report that repeated intracerebroventricular injections of apelin-13 reversed behavioral despair (immobility) in the forced swim (FS) test, a model

Early life adversity and serotonin transporter gene variation interact to affect DNA methylation of the corticotropin-releasing factor gene promoter region in the adult rat brain.

The interaction between childhood maltreatment and the serotonin transporter (5-HTT) gene linked polymorphic region has been associated with increased risk to develop major depression. This Gene × Environment interaction has furthermore been linked with increased levels of anxiety and glucocorticoid release upon exposure to stress. Both endophenotypes are regulated by the neuropeptide corticotropin-releasing factor (CRF) or hormone, which is expressed by the paraventricular nucleus of the hypothalamus, the bed nucleus of the stria terminalis, and the central amygdala (CeA). Therefore, we hypot

A study on evalution of antidepressant effect of imipramine adjunct with Aswagandha and Bramhi.

Depressive disorders increase the risks of self-harm or even suicide in patients. Indigenous drugs are being tried to treat such patient along with conventional antidepressant drugs. This study was planned to investigate the antidepressant action of Ashwagandha and Bramhi and also to confirm its efficacy in the behavioural despair animal model of depression. Normal saline as control (5 ml/kg), Imipramine as standard (16, 32, 64 mg/ kg) and Ashwagandha (50, 100, 150 mg/kg), Bramhi (20, 40, 80 mg/kg) as test drugs were introduced to the albino rats weighing between 200-250 gm for 2 weeks, 1 hr b

Pharmacological activity of hyperforin acetate in rats.

Hyperforin, the main antidepressant component of Hypericum extract, is not stable with regard to heat and light. Therefore, we investigated a newly synthetized derivative, hyperforin acetate. Herein we demonstrate its efficacy in animal models sensitive to antidepressant and anxiolytic drugs. In the forced swimming test, triple administration of hyperforin (5-20 mg/kg) significantly reduced the immobility time of rats, while in the learned helplessness test a daily treatment of 10 mg/kg for seven consecutive days was necessary to elicit an antidepressant effect. In the elevated plus-maze and i

The potential use of GABA agonists in psychiatric disorders: evidence from studies with progabide in animal models and clinical trials.

Progabide, a new antiepileptic GABA agonist of moderate affinity for GABA receptors, has been studied in a number of psychiatric disorders and the results compared with the action of this drug in animal models. In an animal model for anxiety (the aversive response to periaqueductal grey stimulation in the rat) progabide had a similar action to that of diazepam. However in clinical trials to date the effect of the GABA agonist was inferior to that of benzodiazepines. As progabide diminishes both the nigrostriatal dopamine neuron activity and the effects of striatal dopamine receptor activation,

Antidepressant effect and mechanism of TMP269 on stress-induced depressive-like behavior in mice.

TMP269, a class IIA histone deacetylase inhibitor with selectivity, that has a protective effect on the central nervous system, yet its specific mechanism of action remains ambiguous. Although major depressive disorder (MDD) is highly prevalent, its pathophysiology is poorly understood. Recent evidence suggests that histone deacetylase 5 plays a key role in the pathological process of depression and the fact that preclinical studies have shown HDAC5 to be a potential antidepressant target, the search for natural drugs or small molecule compounds that can target HDAC5 may be a potential therape

Stereotypic behaviour in standard non-enriched cages is an alternative to depression-like responses in C57BL/6 mice.

Depressive-like forms of waking inactivity have been recently observed in laboratory primates and horses. We tested the hypotheses that being awake but motionless within the home-cage is a depression-like symptom in mice, and that in impoverished housing, it represents an alternative response to stereotypic behaviour. We raised C57BL/6 ('C57') and DBA/2 ('DBA') females to adulthood in non-enriched (n=62 mice) or enriched (n=60 mice) cages, observing home-cage behaviour during the active (dark) phases. We predicted that being still but awake would be reduced by environmental enrichment; more pr

Brain injury caused by HIV protease inhibitors: role of lipodystrophy and insulin resistance.

HIV-associated neurocognitive disorders (HAND) remain prevalent even with widespread use of combination antiretroviral therapy (ART), suggesting a potential role for co-morbidities in neurologic decline. Indeed, it is well established that ART drugs, particularly HIV protease inhibitors, can induce hyperlipidemia, lipodystrophy, and insulin resistance; all of which are associated with neurologic impairment. This study was designed to determine how metabolic dysfunction might contribute to cognitive impairment and to reveal specific metabolic co-morbidities that could be targeted to preserve br

How valuable are animal models in defining antidepressant activity?

Animal models of depression have been utilised to screen novel compounds with antidepressant potential although uncertainty lingers concerning their clinical relevance. In order for a model to be considered of any value, it must possess predictive validity (does drug action in the model correspond to that in the clinic?), face validity (are there phenomenological similarities between the model and the clinic?) and construct validity (does the model possess a strong theoretical rationale?). On the one hand, there are models based on stress such as the learned helplessness model, the forced swim

Patient perspective and self-help.

Effective adaptation to PD demands that patients and their caregivers elaborate coping strategies that empower them and promote a more salutary orientation to formidable psychosocial difficulties and thus, perhaps, blunt or prevent depressive responses. Patients and their loved ones should be encouraged to seize the initiative in terms of promoting an acceptable life-style: in short, "breaking through" rather than "breaking down." The notion of a "Работает" PD patient is not necessarily a contradiction in terms if the patient and family can be encouraged to view organic disease as a learning op

Learned illness behavior and adjustment to arthritis.

OBJECTIVE: To evaluate the effects of parents' avoidant illness behaviors on their adult children's adjustment to arthritis. METHODS: Men and women attending a rheumatology clinic completed arthritis health status questionnaires and described how their parents generally responded to their own minor illnesses. Some participants rated their parents as never avoiding routine activities when confronting minor illness and were assigned to the non-avoidant group. Participants in the avoidant group reported that their parents avoided routine activities in response to minor illness. Those in the mixe

Evaluation of depression in rats exposed to chronic (unpredictable) electric shock.

The present study was undertaken to evaluate the applicability of a proposed behavioral stress paradigm as an animal model for depression. Rats were trained to press a lever under a fixed ratio (FR) 5 schedule in a Skinner box for 10 days and were subsequently exposed to a daily regimen of 20 cycles of FR 5 and 10 cycles of variable ratio (VR) 10 for about a week. This exposure resulted in a reduction of the number of lever presses and successful escapes compared to the level achieved after training. In addition, weight gain was significantly suppressed compared with other treatments. Acute an

Paradoxical punishment as it relates to the battered woman syndrome.

The main goal of this article is to suggest some processes by which the behavioral interactions of an abusive couple can be developed and maintained. A summary of these processes as they have been adapted from the animal analog and human research follows. First, the pairing of abuse (punishment) with love and affection (reinforcement) could indicate that the battered woman responds to the reinforcement that follows an abusive incident. Second, the battered woman may seek to escape the fear of the tension-building phase but during the escape behavior she is punished (physical abuse), which fort

Attributional change during psychotherapy.

Previous research suggests that causal attributions for life-events are involved in the process of recovery from depression. The study assessed depressive symptoms and attributions before and after treatment in 40 clients. Each client received two forms of therapy in a cross-over design: Exploratory (relationship-oriented, interpersonal) therapy, and Prescriptive (cognitive-behavioural) therapy. During the course of treatment, attributions became significantly more unstable, specific and controllable. There was a positive relation between attributional change and change in depression symptoms,

Depression as a consequence of inadequate neurochemical adaptation in response to stressors.

Stressors induce behavioural disturbances and neurochemical changes in animals, some of which are reminiscent of the symptoms and presumed neurochemical concomitants of depression in humans. Just as in humans, where considerable inter-individual variability is evident in the symptom profile of depression, there is marked inter-individual and inter-strain variability in the behavioural effects of stressors in animals. It is proposed that stressors induce adaptive neurochemical changes, failure of which may engender behavioural disturbances. Variability in the symptoms of depression and in the e

Lived Experiences of Primary Caregivers of Children with Dyslexia: A Phenomenological Study.

BACKGROUND: Caregivers play an important role in children's development. Therefore, they need to recognize the learning difficulties that their children face and support them to ensure learning gains and prevent secondary complications. Nevertheless, supporting a child with a learning disability can be challenging for caregivers in many ways.1 This study investigated caregivers' knowledge of dyslexia, their challenges when raising children with dyslexia, and the support they seek to raise such children. METHODS: Ten individual in-depth interviews (IDIs) and four focus group discussions (FGDs)

Motivational withdrawal in college English learning: a mixed-methods investigation of diminished motivational support and perceived effort ineffectiveness.

INTRODUCTION: This study investigates the latent motivational structure and qualitative manifestations of motivational withdrawal among some Chinese higher vocational college students learning College English within exam-oriented instructional contexts. Drawing on Self-Determination Theory (SDT) and Learned Helplessness Theory (LHT), the study investigates how diminished self-determined motivational support and perceived effort ineffectiveness are reflected in students' reported experiences and observable disengagement. METHODS: It employed a multiple-phase mixed-methods design involving an e
