Tumour cells commonly exhibit aerobic glycolysis and produce lactate despite oxygen availability. Lactate dehydrogenase (LDH) catalyses pyruvate-lactate interconversion and regulates intracellular lactate levels. Endothelial cells also depend on glycolysis for ATP production, which prompted us to investigate LDH in canine hemangiosarcoma (HSA), a malignant endothelial tumour. Medical records for 79 dogs with confirmed splenic hemangiosarcoma (HSA) following splenectomy were reviewed for information regarding either the presence or absence of macroscopic liver lesions and the histopathological characteristics of the liver. Only 29 of 58 dogs (50%) with grossly abnormal livers had HSA metastasis. No dogs with grossly normal livers had metastasis detected on liver pathology. OBJECTIVE: To identify microRNAs (miRNAs) differentially expressed in the serum of dogs with splenic hemangiosarcoma between preoperative and postoperative samples.
METHODS: RNA was extracted from serum collected from dogs with hemangiosarcoma preoperatively, 24 to 48 hours postoperatively, and 10 to 30 days postoperatively. Quantitative reverse transcription PCR was performed with differential expression analysis between sample time points for miRNA (miR)-126-5p, miR-150-5p, miR-203a-3p, miR-214-3p, miR-452-5p, miR-494-3p, miR-497-5p, and miR-543 using miR-16-5p and miR-93-5p as controls.

The hypothesis that neutered dogs in the Veterinary Medical Database (VMDB) are at increased risk for developing hemangiosarcoma (HSA) was tested. Dogs (n = 5736) were diagnosed with HSA from a population of 2 106 324 dogs in the VMDB from 1964 to 2003. A case-control design matched on age and time period was created for general, cardiac, and splenic HSAs. A logistic regression analysis was performed including breed.
