Isolation of Peptide Inhibiting SGC-7901 Cell Proliferation from Aspongopus chinensis Dallas.

Aspongopus chinensis Dallas is used as a traditional Chinese medicine as well as an edible insect. Although its anti-tumor effects have been observed, the anti-tumor active component(s) in the hemolymph of A. chinensis remains unknown. In this study, a combination usage of ultrafiltration, gel filtration chromatography, FPLC and RP-HPLC to separate and purify active peptides was performed based on the proliferation of the human gastric cancer SGC-7901 cell line treated with candidates. One peptide (MW = 2853.3 Da) was isolated from the hemolymph of A. chinensis. A total of 24 amino acid residu

Effectiveness of orally-delivered double-stranded RNA on gene silencing in the stinkbug Plautia stali.

Development of a reliable method for RNA interference (RNAi) by orally-delivered double-stranded RNA (dsRNA) is potentially promising for crop protection. Considering that RNAi efficiency considerably varies among different insect species, it is important to seek for the practical conditions under which dsRNA-mediated RNAi effectively works against each pest insect. Here we investigated RNAi efficiency in the brown-winged green stinkbug Plautia stali, which is notorious for infesting various fruits and crop plants. Microinjection of dsRNA into P. stali revealed high RNAi efficiency-injection o

The genome of the water strider Gerris buenoi reveals expansions of gene repertoires associated with adaptations to life on the water.

BACKGROUND: Having conquered water surfaces worldwide, the semi-aquatic bugs occupy ponds, streams, lakes, mangroves, and even open oceans. The diversity of this group has inspired a range of scientific studies from ecology and evolution to developmental genetics and hydrodynamics of fluid locomotion. However, the lack of a representative water strider genome hinders our ability to more thoroughly investigate the molecular mechanisms underlying the processes of adaptation and diversification within this group. RESULTS: Here we report the sequencing and manual annotation of the Gerris buenoi (

Plant growth stage-specific injury and economic injury level for verde plant bug, Creontiades signatus (Hemiptera: Miridae), on cotton: effect of bloom period of infestation.

Verde plant bugs, Creontiades signatus Distant (Hemiptera: Miridae), were released onto caged cotton, Cossypium hirsutum L., for a 1-wk period to characterize the effects of insect density and bloom period of infestation on cotton injury and yield in 2011 and 2012, Corpus Christi, TX. When plants were infested during early bloom (10-11 nodes above first white flower), a linear decline in fruit retention and boll load and a linear increase in boll injury were detected as verde plant bug infestation levels increased from an average of 0.5 to 4 bugs per plant. Lint and seed yield per plant showed

Entomo-venomics: The evolution, biology and biochemistry of insect venoms.

The insects are a hyperdiverse class containing more species than all other animal groups combined-and many employ venom to capture prey, deter predators and micro-organisms, or facilitate parasitism or extra-oral digestion. However, with the exception of those made by Hymenoptera (wasps, ants and bees), little is known about insect venoms. Here, we review the current literature on insects that use venom for prey capture and predator deterrence, finding evidence for fourteen independent origins of venom usage among insects, mostly among the hyperdiverse holometabolan orders. Many lineages, inc

RNAi-Mediated Knockdown of Tektins Does Not Affect Male Fertility in Lygus hesperus.

Mirid plant bugs (Hemiptera: Miridae), including Lygus hesperus (western tarnished plant bug), are key pests of numerous agricultural crops. While management of this pest relies heavily on chemical insecticides, the evolution of resistance and environmental concerns underscore the need for new and more effective approaches. Genetic-based strategies that target male fertiliy are currently being evaluated for population suppression. However, a lack of candidate gene targets with appropriate function, specifically in non-model species like L. hesperus, has hindered progress in the development and

Evaluation of soybean germplasms for resistance to stay-green syndrome.

Stay-green syndrome (SGS) resistant germplasms serve as the cornerstone for soybean improvement. A comprehensive assessment was conducted on a panel of 1553 germplasms to evaluate their resistance to SGS through natural inoculation. Over a three-year period, one landrace, ZaoShuHeiDou, emerged as resilient to SGS, displaying a significantly reduced risk of SGS (p < 0.05) compared to its counterparts. The broad-sense heritability of SGS resistance was 0.86. Notably, landraces demonstrated a substantially lower incidence of SGS compared to improved cultivars. Furthermore, the severity of SGS cor

A new species of Neoadoxoplatys Kormilev, 1956 (Hemiptera: Pentatomidae: Discocephalinae) from Brazil.

Neoadoxoplatys Kormilev, 1956 (Pentatomidae: Discocephalinae) is a Neotropical genus with four known species poorly represented in entomological collections. The genus is placed in the Janeirona Distant, 1911 clade of Ochlerini, and is easily distinguished by the femora armed distally with two rows of spines, and the labium with an intercalary segment between labiomeres I-II. In this study, we describe a new species of Neoadoxoplatys from Brazil, similar in size to Neoadoxoplatys longirostra Ruckes, 1958, and provide a differential diagnosis to distinguish the new species from the remaining fo

Polycomb group proteins confer robustness to aposematic coloration in the milkweed bug, Oncopeltus fasciatus.

Aposematic coloration offers an opportunity to explore the molecular mechanisms underlying canalization. In this study, the role of epigenetic regulation underlying robustness was explored in the aposematic coloration of the milkweed bug, Oncopeltus fasciatus. Polycomb (Pc) and Enhancer of zeste (E(z)), which encode components of the Polycomb repressive complex 1 (PRC1) and PRC2, respectively, and jing, which encodes a component of the PRC2.2 subcomplex, were knocked down in the fourth instar of O. fasciatus. Knockdown of these genes led to alterations in scutellar morphology and melanization.

Seasonal Reproductive Performance and Pre-diapausing Mating Status of Female Riptortus pedestris (Hemiptera: Alydidae) Collected in Fields.

Two unexplained phenomena are found in Riptortus pedestris (Fabricius) (Hemiptera: Alydidae): the first is that it tends to enter reproductive diapause much earlier in the season and to occur on host plants until late fall before finding hibernation site. The second is that they emerge in early spring when primary food sources such as leguminous plants are unavailable. To understand these phenological trends, the reproductive seasonality of both field-collected and laboratory-reared R. pedestris were compared under conditions of with/without food or access to mates. Females collected in spring

The Apple Snail Pomacea maculata (Caenogastropoda: Ampullariidae) as the intermediate host of Stomylotrema gratiosus (Trematoda: Stomylotrematidae) in Brazil: the first report of a mollusc host of a Stomylotrematid Trematode.

Trematodes belonging to the family Stomylotrematidae are intestinal parasites of birds. Despite the worldwide distribution and diversity of host species, the first intermediate host remains unknown. For a survey of parasites of Pomacea maculata , snails were collected from the municipality of São Vicente Férrer, state of Maranhão, northeastern Brazil. In the present study, the xiphidiocercariae shed from these snails were used in the experimental infection of the water bug Belostoma plebejum. The insect mortality was observed 30 days post-infection, and the metacercariae recovered in the body

Monogamy and polygamy in two species of mirid bugs: a functional-based approach.

Multiple mating in females is widespread among insects in spite of the risk of predation, disease acquisition and/or physical injury that may occur. One common consequence of female polyandry is competition among sperm from two or more males within the female to fertilize the ova. This competition is an evolutionary driving force that determines a series of adaptations in both males and females. In this work, we examine some behavioral, morphological and physiological characteristics of males and females of two Heteropteran species that are related to their monoandrous/polyandrous mating behav

Aquatic assassins: Proteo-transcriptomic and functional profiling of giant water bug and water scorpion venoms.

Aquatic heteroptera possess highly specialized salivary venoms that they use to immobilize their prey; however, their evolutionary origins, molecular composition, and functional diversity remain poorly understood. Here, we integrate proteo-transcriptomics with biochemical analysis to compare venoms from three different ecologically important taxa: giant water bug (Lethocerus indicus), electric light bug (Diplonychus rusticus), and water scorpion (Laccotrephes maculatus). The proteo-transcriptome profiling revealed that peptidases are the dominant toxin family across all three species, establis

Harnessing Leguminosae Typical Scents for a More Effective and Eco-Friendly Pheromone Trapping of Pest.

The utilization of pheromone-based products for insect pest management has been a longstanding practice. However, mounting evidence suggests that pheromones of insect pests could also inadvertently attract natural enemies, leading to their unintentional entrapment by pheromone baits. In East Asia, pheromone traps of the bean bug Riptortus pedestris capture several species of natural enemies, which reduces top-down control of the pest and could lead to failures of pest management programs. Here, we identified novel fragrances that could be incorporated into standard pheromone traps to enhance p

Unveiling detoxifying symbiosis and dietary influence on the Southern green shield bug microbiota.

The Southern green shield bug, Nezara viridula, is an invasive piercing and sucking pest insect that feeds on crops and poses a threat to global food production. Insects live in close relationships with microorganisms providing their host with unique capabilities, such as resistance to toxic plant metabolites. In this study, we investigated the resistance to and detoxification of the plant metabolite 3-nitropropionic acid (NPA) by core and transient members of the N. viridula microbial community. Microbial community members showed a different tolerance to the toxin and we determined that six o
