Influence of various feeding conditions, the migrating myoelectric complex and cholinergic drugs on antral slow waves in sheep.

The presented study was designed to elucidate whether the cholinergic mechanisms control ovine antral slow waves in various physiological conditions, including feeding and various phases of migrating myoelectric complex (MMC). The investigations were carried out on six adult sheep of Polish Merino breed with seven bipolar electrodes surgically implanted onto the antral and small intestinal wall. In the course of chronic experiments, the myoelectric activity was recorded from these regions using the multichannel electroencephalograph. Experiments were performed on 48 h fasted and non-fasted ani

Chlorisondamine, a non-competitive ganglionic blocker, antagonizes the cardiovascular effects of cocaine in conscious squirrel monkeys.

Cocaine (0.3 mg/kg i.v.) produced prolonged pressor and tachycardiac responses in conscious squirrel monkeys. Peak pressor and tachycardiac responses following cocaine were 32.7 +/- 3.3 mm Hg and 78.8 +/- 7.4 beats/min, respectively. Pretreatment with 1 mg/kg chlorisondamine, a noncompetitive ganglionic blocker, attenuated the pressor (18.3 +/- 3.8 mm Hg) and tachycardiac (63.7 +/- 10 beats/min) effects of cocaine, while 5 mg/kg chlorisondamine completely prevented these effects of cocaine. This finding supports the conclusion that the cardiovascular effects of cocaine are centrally mediated.

Capsaicin-sensitive nerves modulate reactive hyperemia in rat gut.

Reactive hyperemia (RH) is a local, vascular response that occurs following release from mechanical occlusion of an artery, with restoration of intra-arterial pressure. The mechanism of this postocclusion hyperemia in the gut has not been identified, although metabolic, myogenic, and neurogenic mediators of this response have been proposed. The present study was conducted to evaluate a possible modulatory role for sensory innervation of the intestinal vasculature in RH, using acute and chronic treatment with capsaicin applied in different ways. In anesthetized rats, the velocity of flowing blo

Accelerated norepinephrine turnover in peripheral tissues after ventromedial hypothalamic stimulation in rats.

To obtain evidence for a functional connection between the ventromedial hypothalamic nucleus (VMH) and the sympathetic nervous system, effects of electrical stimulation of the VMH, the lateral hypothalamic area (LH) and the paraventricular hypothalamic nucleus (PVN) on norepinephrine (NE) turnover in the heart, liver, pancreas, spleen, submandibular gland and the interscapular brown adipose tissue were examined in anesthetized rats. Stimulation of the VMH elicited a 3-8-fold increase in the rate of NE turnover in all organs examined, whereas stimulation of the LH or the PVN had no appreciable

The mode of action of 4-aminopyridine on the chronotropic and inotropic responses in the isolated, blood-perfused dog heart.

The effects of 4-aminopyridine (4-AP, 0.3 to 300 micrograms) on atrial rate and atrial muscle contractile force were investigated in the isolated, blood-perfused dog atrial preparation. Low doses (less than 30 micrograms) of 4-AP injected into the sinus node artery caused a dose-dependent positive inotropic response and a negligible positive chronotropic response. A higher dose (300 micrograms) of 4-AP induced a transient positive chronotropic response followed by a negative response and positive and negative inotropic responses. The negative cardiac responses to 4-AP were completely inhibited

Nicotine-induced salivation in cats: effects of various drugs.

The effect of an intracerebroventricular injection of an antimuscarinic drug, ganglionic blocking agent, alpha and beta adrenergic blocking drug, antiserotonin agent or antihistamine upon salivation produced by nicotine similarly injected in the cat was investigated. Atropine and hexamethonium abolished the salivation evoked by nicotine. On the other hand, salivation induced by nicotine was not significantly altered by yohimbine, practolol, methysergide and antazoline. It is concluded that the salivation produced by nicotine is mediated via central receptors which have mixed nicotinic and musc

Cisapride induces clustered spikes and irregular spiking of the small intestine of the rat.

The aim of the present study was to investigate the effect of cisapride on the myoelectric activity of the small intestine in the rat. Intestinal myoelectric activity of fasted conscious rats was monitored by three bipolar electrodes chronically implanted at 5, 15 and 25 cm distal to the pylorus and connected to an EEG amplifier. In the basal state, regularly occurring migrating myoelectric complexes (MMCs) and clustered spikes were registered. Cisapride at doses of 0.5-4.0 mg-1 i.v. did not affect the MMCs, but at 8 mg kg-1 i.v. the regular MMCs were replaced by irregular spiking activity (p

Effects of stimulating the sympathetic innervation in bursts on submandibular vascular and secretory function in cats.

1. The effects of continuous stimulation of the peripheral end of the ascending cervical sympathetic nerve were compared with those of intermittent stimulation, so arranged as to deliver the same total number of impulses, in cats under chloralose anaesthesia. 2. Continuous stimulation caused a flow of saliva at 5-10 Hz, but not at 2 Hz. In contrast, the same total number of impulses delivered intermittently in bursts elicited a prompt secretion at a frequency as low as 20 Hz for 1 s at 10 s intervals (corresponding to 2 Hz continuously) and a significantly higher rate of secretion at 50 Hz in

Possible sensory receptor of nonadrenergic inhibitory nervous system.

To determine the sensory receptor of the nonadrenergic inhibitory nervous system (NAIS), 22 cats were anesthetized and serotonin was continuously administered (50-250 micrograms.kg-1.min-1 iv) to increase pulmonary resistance (RL) to 377 +/- 57% (SE) of the control value. We then 1) mechanically irritated the trachea, 2) intravenously administered capsaicin (5 micrograms/kg), or 3) induced hypoxia (arterial PO2 30-40 Torr) to stimulate irritant and bronchial C-fiber receptors, pulmonary C-fiber receptors, or the carotid body (chemoreceptors), respectively. Effect and mechanism of vagal nerve stimulation on somatostatin secretion in dogs.

To investigate the effect of vagal nerve stimulation on the release of pancreatic somatostatin, we electrically stimulated (10 Hz, 5 ms, 13.5 mA, and 10 min) the thoracic vagi just below the heart in halothane anesthetized dogs (n = 15). The stimulation increased the pancreatic output of somatostatinlike immunoreactivity (SLI) (delta = +248 +/- 81 fmol/min, P less than 0.005; base-line levels = 455 +/- 150 fmol/min). min). Fast and slow synaptic potentials produced in a mammalian sympathetic ganglion by colon distension.

Radial distension of the large intestine produced a slow depolarization in a population of neurons in the inferior mesenteric ganglion of the guinea pig. The slow potentials often occurred simultaneously with cholinergic fast potentials [( excitatory postsynaptic potentials (EPSPs]) yet persisted in the presence of nicotinic and muscarinic cholinergic antagonists when all fast EPSPs were absent. The amplitude of the distension-induced noncholinergic slow depolarization increased with increasing distension pressure. For distensions of 1-min duration at pressures of 10-20 cm of water, the mean d

[Receptive relaxation of the canine stomach].

Receptive relaxation of the stomach by feeding was studied in 5 conscious dogs by means of chronically implanted force transducers on the gastric body and antrum. It was found that only the gastric body but not antrum relaxed by feeding, and magnitude of the relaxation of the gastric body was linearly related to the volume (5, 10 and 20 g/kg) of test meals, given once or in multiple times. Secondly, in the study of pharmacological blockers, it was found that atropine, hexamethonium and phentolamine significantly inhibited relaxation, while naloxone and proglumide significantly suppressed relax

Effect of ganglionic blockade on catecholamine secretion in exercised dogfish.

A brief bout of vigorous exercise results in significant increases in plasma epinephrine (E) and norepinephrine (NE) in the dogfish, Squalus acanthias. Since the presence of a functioning sympathetic nervous system in dogfish is in doubt, experiments were undertaken to show whether or not exercise-induced catecholamine (CA) secretion is under autonomic neurogenic control. Changes in plasma E and NE in a control group of exercised fish were compared with changes in fish exercised while under the influence of ganglionic blockade. Ganglionic blockade was induced in dogfish by hexamethonium infusi

Drug administration into the third ventricle of the brain of anesthetized and conscious cats.

A method is described for the administration of drugs into the third ventricle of the brain of conscious and anesthetized cats. Noradrenaline administered by this route produced cardiovascular stimulant effects as against depressor effects when administered into a lateral cerebral ventricle. The pressor responses evoked by third ventricular administration of noradrenaline were antagonized by spinal section in anesthetized cats and by hexamethonium in conscious animals. Conscious cats were more sensitive to the pressor effects of third ventricular noradrenaline than chloralose-anesthetized anim

[Pontine control of the internal urethral sphincter activity on decerebrate dogs].

The present study was performed to investigate the pontine control of the internal urethral sphincter (IUS) activity using 13 supracolicular decerebrate female dogs. The lower urinary tract function was evaluated by measuring the bladder and the IUS pressure and the external urethral sphincter (EUS) EMG. Bilateral ureters were cannulated and urine was drained extracorporeally. The bladder neck was ligated to induce a isovolumetric bladder contraction, and the urethra was separated from the bladder to evaluate the function of the bladder and the urethra separately. Electrical stimulation (ES) (

Vasoconstricting effect of vasopressin on regional vascular beds in conscious rats after ganglionic blockade.

We analyzed the vasoconstrictor effects of arginine vasopressin (AVP) on regional arteries before and after ganglionic blockade with hexamethonium. Simultaneous measurements of mean arterial pressure and regional flows were obtained in conscious rats, using chronically implanted electromagnetic flow probes. Regional vascular resistance was calculated as mean arterial pressure divided by regional flow. AVP was applied intravenously as a bolus, at doses ranging from 5 x 10(-11) to 5 x 10(-8) g/kg. AVP increased mean arterial pressure, decreased superior mesenteric, renal and terminal aortic flow

The role of central and autonomic neural mechanisms in the cardiovascular effects of cocaine in conscious squirrel monkeys.

The effects of cocaine on cardiovascular function were studied in six conscious squirrel monkeys. Cocaine (0.01-3 mg/kg i.v.) increased mean arterial blood pressure and heart rate (HR) in a dose-dependent manner. The effect of cocaine on HR reached a maximum at 0.3 mg/kg. Doses of cocaine up to 3 mg/kg did not evoke cardiac rhythm disturbances. Pentobarbital or halothane anesthesia attenuated the pressor and tachycardiac effects of 3 mg/kg of cocaine. Antagonism of the pressor response to cocaine by halothane was significantly greater than that by pentobarbital. Halothane, but not pentobarbita

Evidence for a respiration-modulated cholinergic action on the activity of medullary respiration-related neurons in the rabbit. An iontophoretic study.

Effects of the iontophoretically administered cholinergic agonists acetylcholine, bethanechol and DMPP on the activity of medullary respiration-related neurons were examined in urethane-anaesthetized rabbits. Inhibitory effects prevailed over excitatory effects. Analysis of cholinergic effects by cycle-triggered averaging revealed three major types of neuronal responses: (i) constant alterations of spike-density throughout the whole period of activity ("constant effects"), (ii) effects increasing during the progression of the burst of discharge or effects restricted to a particular fraction of

Stimulation of lacrimal secretion by sympathetic nerve impulses in the rabbit.

Direct electrical stimulation of the preganglionic nerve trunk of the ipsilateral superior cervical sympathetic ganglion induced fluid secretion from the cannulated main excretory duct of the non-stimulated rabbit lacrimal gland. The optimum strength and frequency of stimulation were 4 volts and 25 Hz. At this stimulus parameter, the rate of secretion was 2.1 +/- 0.2 microliter/10 min. The sympathetic-induced lacrimal secretion was markedly depressed after intravenous administration of hexamethonium (1 mg/kg) and by post-ganglionic neurectomy. The results suggest that, in addition to parasympa
