The significance of Hippo pathway protein expression in oral squamous cell carcinoma.

INTRODUCTION: The Hippo pathway consists of mammalian sterile 20-like kinase 1/2 (MST1/2), large tumor suppressor 1/2 (LATS1/2), and yes-associated protein (YAP)1. Herein, we present the first report on the significance of major Hippo pathway protein expression in oral squamous cell carcinoma (OSCC). METHODS: The analyses included oral epithelial dysplasia (OED, n = 7), carcinoma in situ (CIS, n = 14), and oral squamous cell carcinoma (OSCC, n = 109). RESULTS: Cytoplasmic expression of MST1, LATS1, and LATS2 was low in OED, CIS, and OSCC. The cytoplasmic expression of MST2 was high in OED (5

Signaling pathway networks in frozen shoulder fibrosis: from inflammatory initiation to fibrotic progression.

Frozen shoulder, also known as adhesive capsulitis, is characterized by fibrosis, thickening, and contracture of the shoulder joint capsule. Through a systematic review of existing research, we have analyzed the complex molecular network of shoulder fibrosis. Previous studies have found that the disease begins with inflammation of the nuclear factor kappa B (NF-κB) and leptin/Janus kinase/signal transducer and activator of transcription (JAK-STAT) pathways, and the transforming growth factor-beta (TGF-β) pathway is the key cause of fibrosis. Wingless/Int (WNT) and potentially Hippo pathways ma

Metabolic stress-induced dysregulation of the Hippo signaling pathway inhibits the occurrence of hepatocarcinoma.

Liver fibrosis is the natural stress response of the liver to injury and a critical intermediate stage in the progression of most liver diseases. Here, we first demonstrated via a retrospective clinical study that the incidence of advanced hepatocellular carcinoma (HCC) was significantly higher in patients infected with hepatitis B virus (HBV) than in those with hepatolenticular degeneration (HLD). Further analyses involving micro RNA (miRNA) and proteomics were conducted to investigate the distinct mechanisms underlying liver fibrosis induced by HBV and HLD. Results showed significant changes

AMPK stimulation inhibits YAP/TAZ signaling to ameliorate hepatic fibrosis.

Hepatic fibrosis is driven by the activation of hepatic stellate cells (HSCs). The Hippo pathway and its effectors, YAP and TAZ, are key regulators of HSC activation and fibrosis. However, there is a lack of mechanistic understanding of YAP/TAZ regulation in HSCs. Here we show that AMPK activation leads to YAP/TAZ inhibition and HSC inactivation in vitro, while the expression of a kinase-inactive mutant reversed these effects compared to wild type AMPKɑ1. Notably, the depletion of LATS1/2, an upstream kinase of YAP/TAZ signaling, rescues YAP/TAZ activation, suggesting that AMPK may be mediatin

MicroRNA-132-3p, Downregulated in Myeloid Angiogenic Cells from Hereditary Hemorrhagic Telangiectasia Patients, Is Enriched in the TGFβ and PI3K/AKT Signalling Pathways.

BACKGROUND: Hereditary hemorrhagic telangiectasia (HHT) is a rare, autosomal dominant genetic disorder characterized by life-threatening vascular dysplasia. Myeloid angiogenic cells (MACs), alternatively called early endothelial progenitor cells or circulating angiogenic cells, do not directly incorporate into developing blood vessels, but augment angiogenesis in a paracrine manner. MAC dysfunction has been reported in HHT. MicroRNAs (miRNAs) regulate cellular function by modulating gene expression post-transcriptionally. To date, the role of miRNAs in HHT MAC dysfunction has not been document

Decoding cadmium toxicity in spiders: F-actin and Hippo pathway insights.

Heavy metals have notable effects on arthropods, but their regulatory mechanisms remain unclear. This study reveals the potential regulatory roles and mechanisms of F-actin and Hippo signaling pathway in response to cadmium (Cd) toxicity of spider. The actin gene encoding G-actin (F-actin subunit) in Pardosa pseudoannulata (P. pseudoannulata) was cloned and found to be up-regulated in Cd-exposed spiders, particularly in the 3rd and 5th instars. RNA interference (RNAi) of this gene led to increased Cd levels and suppressed physiological traits, affecting antioxidant, detoxification, immune enzy

Targetable pathways for drug repurposing in gastric cancer.

Gastric cancer (GC) is both the fifth most common cancer worldwide and the fifth in mortality. Owing to a lack of symptoms in the early stages and unspecific clinical presentation in the later stages, GC is usually diagnosed at advanced stages. This means that only approximately 60% of patients are eligible for curative treatment, and overall, GC patients have a 5-year survival rate of only 28.3%, underscoring the importance of developing new treatment strategies. Drug repurposing involves identifying new therapeutic uses for approved drugs and is a promising strategy for cancer treatment beca

Gypensapogenin I ameliorates liver fibrosis by targeting MST1, restoring Hippo pathway homeostasis, and inhibiting HSCs activation.

Gypensapogenin I (GI) is a new dammarane type triterpenoid saponin extracted from Gynostemma pentaphyllum. This study explored the therapeutic potential and mechanism of GI on liver fibrosis (LF). LF is a key pathological stage of chronic liver disease, and the current treatment is limited. In the mouse LF model induced by carbon tetrachloride (CCl4) and the LX-2 hepatic stellate cell model activated by transforming growth factor-β1 (TGF-β1), GI showed significant antifibrotic effects in vitro and in vivo, and could reduce liver injury, inflammation, collagen deposition and cell activation. An updated patent review of TEAD modulators (2022-present).

INTRODUCTION: The Transcriptional Enhanced Associated Domain (TEAD) family has attracted increasing attention due to its crucial role in the Hippo signaling pathway, which regulates cell growth, division, survival, differentiation, and tissue homeostasis in multicellular organisms. Inhibition of TEADs with small molecules has been shown to be an effective strategy for the treatment of tumors, and several compounds have entered clinical trials as monotherapy and/or combination therapy. AREAS COVERED: Due to the explosion of patent disclosures on TEAD modulators (inhibitors or degraders) in the

Multi-omics characterization of the monkeypox virus infection.

Multiple omics analyzes of Vaccinia virus (VACV) infection have defined molecular characteristics of poxvirus biology. However, little is known about the monkeypox (mpox) virus (MPXV) in humans, which has a different disease manifestation despite its high sequence similarity to VACV. Here, we perform an in-depth multi-omics analysis of the transcriptome, proteome, and phosphoproteome signatures of MPXV-infected primary human fibroblasts to gain insights into the virus-host interplay. In addition to expected perturbations of immune-related pathways, we uncover regulation of the HIPPO and TGF-β

Genomic abnormalities and signal transduction dysregulation in malignant mesothelioma cells.

Malignant mesothelioma (MM) is a tumor with poor prognosis associated with asbestos exposure. While it remains to be clarified how asbestos fibers confer genetic/epigenetic alterations and induce cellular transformation in normal mesothelial cells, the understanding of key molecular mechanisms of MM cell development, proliferation, and invasion has progressed. MM shows frequent genetic inactivation of tumor suppressor genes of p16(INK4a)/p14(ARF) and neurofibromatosis type 2 (NF2) which encodes Merlin, and epigenetic inactivation of RASSF1A. However, no frequent mutations of well-known oncogen

PMEPA1 modulates YAP1 nuclear translocation to disrupt EMT subtypes and promote metastasis in Biliary tract cancer.

Biliary tract cancer (BTC) is an aggressive tumor with poor prognosis and limited treatment options. Epithelial-mesenchymal transition (EMT) significantly contributes to BTC metastasis, yet the critical regulators and underlying molecular mechanisms of EMT remain poorly unclear. In our study, we conducted an integrative analysis of single-cell RNA sequencing (scRNA-seq) data from 47 BTC samples, identifying two EMT-enriched epithelial subpopulations and a BTC specific EMT gene set. Using the EMT gene set, we classified BTC patients and noticed that those with elevated EMT scores had worse prog

An unrecognized mechanism of self-protection in degenerating neurons mediated by astrocytic YAP through Wnts/β-catenin/EAAT2 signaling in C9orf72-poly-GA mice.

Rationale: Amyotrophic lateral sclerosis (ALS) is a fatal neurodegenerative disorder characterized by the progressive loss of motor neurons in the central nervous system (CNS). Non-neuronal cells, particularly astrocytes, have been recognized as pivotal contributors to ALS onset and progression. However, the underlying mechanisms of interactions between astrocytes and motor neurons during ALS remain unclear. Recent studies have identified the neuronal Hippo kinase mammalian sterile 20-like kinase 1 (MST1) as a key regulator of neurodegeneration in ALS. Yes-associated protein (YAP), a major dow

Lead exposure promotes NF2-wildtype meningioma cell proliferation through the Merlin-Hippo signaling pathway.

BACKGROUND: Lead is a persistent inorganic environmental pollutant with global implication for human health. Among the diseases associated with lead exposure, the damage to the central nervous system has received considerable attention. It has been reported that long-term lead exposure increases the risk of meningioma; however, the underlying mechanism remains poorly understood. Clinical studies have indicated that loss-of-function and mutations in the neurofibromin-2 (NF2) gene play a crucial role in promoting meningioma formation. METHODS: The effect of Pb on meningioma were tested in-vitro

The interplay of signaling pathway in endothelial cells-matrix stiffness dependency with targeted-therapeutic drugs.

Cardiovascular diseases (CVDs) have been one of the major causes of human deaths in the world. The study of CVDs has focused on cell chemotaxis for decades. With the advances in mechanobiology, accumulating evidence has demonstrated the influence of mechanical stimuli on arterial pathophysiology and endothelial dysfunction that is a hallmark of atherosclerosis development. An increasing number of drugs have been exploited to decrease the stiffness of vascular tissue for CVDs therapy. However, the underlying mechanisms have yet to be explored. This review aims to summarize how matrix stiffness

A novel form of necrosis, TRIAD, occurs in human Huntington's disease.

We previously reported transcriptional repression-induced atypical cell death of neuron (TRIAD), a new type of necrosis that is mainly regulated by Hippo pathway signaling and distinct from necroptosis regulated by RIP1/3 pathway. Here, we examined the ultrastructural and biochemical features of neuronal cell death in the brains of human HD patients in parallel with the similar analyses using mutant Htt-knock-in (Htt-KI) mice. LATS1 kinase, the critical regulator and marker of TRIAD, is actually activated in cortical neurons of postmortem human HD and of Htt-KI mouse brains, while apoptosis pr

USP25 aggravates liver cancer development and impairs chemosensitivity by limiting LATS1 activation.

Hepatocellular carcinoma (HCC) is a primary liver malignancy with high rates of morbidity and mortality, yet effective treatment options remain limited. Accumulating evidence indicates that large tumour suppressor kinase 1 (LATS1), a core kinase in the Hippo pathway, mediates the phosphorylation of downstream YAP/TAZ, thereby suppressing HCC progression. Although LATS1 protein stability has been reported to be modulated by ubiquitination, the specific mechanism controlling LATS1 kinase activity remains unknown. Here, we identify USP25 as a deubiquitinase that regulates LATS1 activity, independ
