Modulation of tryptase and histamine release from human lung mast cells by protease inhibitors.

Inhibition of IgE dependent histamine release from human mast cells by protease inhibitors has been observed in skin, tonsil and synovial tissues. However, little is known about the actions of protease inhibitors on tryptase release from human lung mast cells. We therefore examined the ability of protease inhibitors to modulate tryptase and histamine release from human lung mast cells. IgE dependent tryptase release from dispersed lung mast cells was inhibited to a maximum of approximately 53.8% and 44.5% by N-a-tosyl-L-lysine chloromethyl ketone (TLCK) and N-p-Tosyl-L-phenylalanine chlorometh

Early and late histamine release induced by albumin, hetastarch and polygeline: some unexpected findings.

OBJECTIVE: The perioperative use of colloidal plasma substitutes is still under discussion. We therefore conducted a prospective randomised study with three commonly used plasma substitutes to examine their histamine releasing effects in 21 volunteers. MATERIAL OR SUBJETS: 21 male volunteers were enrolled in this prospective, randomised, controlled clinical study. Endpoints were the incidence of early and late histamine release and the time course of the release kinetics. Normovolemic hemodilution technique was used with hydroxyethyl starch (n = 6), human albumin (n = 6) and polygeline (n = 9)

Basophil histamine release in atopic dermatitis and its relationship to disordered cyclic nucleotide metabolism.

Maximal histamine release (HR) from leucocytes, in response to Concanavalin A (Con A) was significantly higher in a group of 16 adults with moderate to severe atopic dermatitis (AD) when compared to 13 non-atopic adults. In a further 4 adults with AD, HR was similar to that in the normals, suggesting the existence of 'high releaser' and 'low releaser' subsets within the AD group. Leucocyte cyclic AMP phosphodiesterase (PDE) activity was significantly higher in the 'high releaser' group compared to the 'low releaser' and normal groups. High and low HR responses showed strong correlations with h

[Histamine release due to ganglion blocking agent and muscle relaxants from rat mast cells].

The mechanisms of histamine release due to d-tubocurarine from rat mast cells have long been known to anesthesiologists, such as degranulatory process, Ca++ dependent process and energy coupled process. However, not much is known about histamine release due to some drugs such as trimethaphan and muscle relaxants other than d-tubocurarine, like alcuronium, pancuronium or succinylcholine, although many clinical reports related to hypersensitivity reactions due to muscle relaxants can be found. In this experiment, the histamine releasing activities of trimethaphan and muscle relaxants were examin

Relationship between serum IgE levels and anaphylactic histamine release from isolated rat mast cells.

Inbred Hooded Lister rats were immunized with egg albumin with B. pertussis vaccine used as an adjuvant. The serum levels of total IgE and IgE antibody (egg albumin specific) were determined by radioimmunoassay techniques before and after immunization. The basic level of total IgE in serum was 560 +/- 110 ng/ml. After immunization a maximal peak at day 11 of 1940 +/- 160 ng/ml was registered. Anti-egg albumin IgE antibody showed a maximum around day 13 of 75 +/- 11 units/ml. Pleural mast cells were isolated on Ficoll between day 14 and 20 after immunization. A significant negative correlation

Potentiation of IgE-mediated cutaneous reactivity and blood leucocyte histamine release by deuterium oxide.

A previous study (Gillespie & Lichtenstein, 1972) demonstrated that there was potentiation of histamine release from human peripheral blood leucocytes following exposure to antigin or anti-IgE in deuterium oxide (D2O). The current study confirms the results with human leucocytes and indicates that the degree of histamine release due to anti-IgE or its potentiation by D2O appeared independent of the serum IgE concentration of the cell donor. Further studies demonstrated that the peripheral blood leucocytes from monkeys with a sufficient degree of IgE-mediated reactivity to Ascaris antigen-relea

Antigen-indued histamine release from peritoneal mast cells of mice producing reagin-like antibody.

The time course of production in the mouse circulation, after different schemes of immunization, of reagin-like and γ1 antibodies, as measured by the ability of the antisera to induce thermolabile long-latence and thermostable short-latence PCA reactions, has been studied. γ1 antibody was obtained in all schemes of immunization and its amount in antiserum increased with time. Reagin-like antibody appeared on the 7th day after immunization with sufficient dose of ovalbumin and adjuvant, and on later days with less efficient antigens such as DNP—BSA and DNP—BGG. Maximal amounts of the reagin-lik

Massive release of the histamine-degrading enzyme diamine oxidase during severe anaphylaxis in mastocytosis patients.

BACKGROUND: Histaminolytic activity mediated by diamine oxidase (DAO) is present in plasma after induction of severe anaphylaxis in rats, guinea pigs, and rabbits. Heparin released during mast cell degranulation in the gastrointestinal tract might liberate DAO from heparin-sensitive storage sites. DAO release during anaphylaxis has not been demonstrated in humans. METHODS: Plasma DAO, tryptase, and histamine concentrations of four severe anaphylaxis events were determined at multiple serial time points in two patients with systemic mastocytosis. The histamine degradation rates were measured i

Anaesthetic agents inhibit gastrin-stimulated but not basal histamine release from rat stomach ECL cells.

By mobilizing histamine in response to gastrin, the ECL cells in the oxyntic mucosa play a key role in the control of the parietal cells and hence of gastric acid secretion. General anaesthesia suppresses basal and gastrin- and histamine-stimulated acid secretion. The present study examines if the effect of anaesthesia on basal and gastrin-stimulated acid secretion is associated with suppressed ECL-cell histamine secretion. A microdialysis probe was implanted in the submucosa of the ventral aspect of the acid-producing part of the stomach (32 rats). Three days later, ECL-cell histamine mobiliz

Induction of histamine release in parasitized individuals by somatic and cuticular antigens from Onchocerca volvulus.

The host immune response in onchocerciasis is believed to contribute to the clinical manifestations of infection. Mazzotti and chronic inflammatory reactions might be mediated by mechanisms involving specific IgE and reactivity of mast cells and basophils to the parasite antigens. In this report, we show that Onchocerca volvulus antigens are capable of inducing histamine release. Three types of extracts were prepared from the parasite: soluble total, surface, and cuticular collagen. Soluble extracts released histamine in all individuals with onchocerciasis at significantly higher levels (P < 0

Histamine release from the hypothalamus induced by gravity change in rats and space motion sickness.

Freely moving rats were exposed to 2 g hypergravity in an animal centrifuge device to produce motion sickness. Histamine release from the anterior hypothalamus of the rats was measured in vivo with a microdialysis technique. After a 2-h load of 2 g hypergravity, rats ate kaolin. Because pica, eating a nonnutritive substance such as kaolin, is a behavioral index of motion sickness in rats, this finding indicates that the rats suffered from motion sickness. During 2 g hypergravity for 2-h, histamine release from the hypothalamus was transiently increased. In contrast, neither the transient incre

Spontaneous histamine release from washed leukocytes and whole blood in atopic and non-atopic individuals.

The ability of basophils from patients with allergic rhinitis, extrinsic asthma, chronic idiopathic urticaria and atopic dermatitis to release histamine spontaneously in vitro was studied. Spontaneous release in the presence and absence of 30% D2O was investigated from both washed leukocytes and whole blood. Compared with controls histamine release (HR) from washed leukocytes was significantly enhanced in allergic rhinitis patients only, whereas in the other groups only a certain percentage of patients was found to have high spontaneous HR. In whole blood experiments HR in all groups was withi

[Changes in degranulation and histamine release from mast cells in rats sensitized by penicilloyl-protein].

In this paper, the mechanism of the penicillin G anaphylactic reaction was studied. Rats were sensitized to penicilloyl (BPO)-protein. The incidences of anaphylactic reaction and death were 100 and 40%, respectively. After being challenged, the degranulation rates of rat cranial periosteous and peritoneal mast cells were 91 +/- 5 and 44 +/- 10%, respectively. In the control group, they were 11 +/- 5 and 7 +/- 2%, respectively. The histamine release rates from sensitized and control rat peritoneal mast cells were 13 +/- 2 and 3.0 +/- 0.4%, respectively. The passive cutaneous anaphylaxis (PCA) t

Allergy to suxamethonium: persisting abnormalities in skin tests, specific IgE antibodies and leucocyte histamine release.

Twenty-one patients, who had previously experienced an anaphylactic reaction to suxamethonium during general anaesthesia, were selected for this study. Initially, skin tests with muscle relaxants were carried out in the twenty-one patients, detection of specific anti-choline IgE in nineteen, and leucocyte histamine release in seventeen. These three tests were then repeated between 1 year and 4 years after the initial evaluation. In the majority of patients, sensitization to the muscle relaxants persisted for more than 1 year after the anaphylactic reaction. Only three patients out of twenty-on

Characterization of primate bronchoalveolar mast cells. I. IgE-dependent release of histamine, leukotrienes, and prostaglandins.

Large numbers of functional mast cells were obtained by bronchoalveolar lavage (BAL) of Macaca arctoides monkeys that had been infected with the nematode Ascaris suum. These lavage cells, of which 21% were mast cells, released histamine, LTC4, and PGD2 in a concentration-dependent fashion when challenged with ascaris antigen or antibody to human IgE. However, there was no release of histamine when these cells were challenged with compound 48/80. The amount of mediator released was highly dependent on the sensitivity of the cells to immunologic challenge, but was generally in the range of 2 to

A mathematical model to quantitate histamine release from basophils challenged in vitro with insect venom.

Using a two parameter mathematical model, it was possible to describe clinically obtained data from histamine release (HR) in vitro as a response to basophils challenged by insect venom. The Gompertz function used in this model not only reflects the observed HR data but can be used to predict maximal HR values from initial lower antigen concentrations HR determinations. The model was used to calculate the concentration of venom antigens releasing 25% of total intracellular histamine (HR25). This is clinically significant in that while knowledge of the HR25, or any other reference fraction, fac

Drug inhibition of anaphylactic histamine release from peritoneal cells of rats infected with Toxocara canis.

An experimental model for studying anaphylactic histamine release from rat peritoneal cells in vitro is described. Rats were sensitized by feeding the infective eggs of ttoxocara canis, the antigen being prepared from the infective eggs. The characteristics of histamine release are comparable with those found for other anaphylactic systems. Addition of 0.2% inactivated normal rat serum to the incubation media reduced the spontaneous histamine release from 11.5 +/- 1.9% to 3.1 +/- 1.6% (mean +/- SD) of total histamine, whereas the corresponding figure for anaphylactic histamine release was 41.3

Porcine mast cells infected with H1N1 influenza virus release histamine and inflammatory cytokines and chemokines.

Mast cells reside in many tissues, including the lungs, and might play a role in enhancing influenza virus infections in animals. In this study, we cultured porcine mast cells from porcine bone marrow cells with IL-3 and stem cell factor to study the infectivity and activation of the 2009 pandemic H1N1 influenza virus of swine origin. Porcine mast cells were infected with H1N1 influenza virus, without the subsequent production of infectious viruses but were activated, as indicated by the release of histamines. Inflammatory cytokine- and chemokine-encoding genes, including IL-1α, IL-6, CXCL9, C
