Neoplastic cells from 9 patients affected with a "histiocytic" lymphoma were studied with 5 membrane markers of B or T lymphocytes. In 2 patients a monoclonal B-cell proliferation was found; they had been affected previously with well documented B-cell proliferations: chronic lymphocytic leukemia or Waldenström's macroglobulinemia. The blast cells of 2 other patients had T-cell features; in a fifth case, the abnormal cells carried only a strong receptor for the Fc fragment of IgG, which suggested their truly monocytic origin. In 4 patients, the cells had no detectable surface markers. These fi

A two-year-old female leopard gecko presented with a mass in the oral cavity with apparent exophthalmos. Despite medical management, the animal died. Grossly, the firm and tan to white mass extended from the oropharyngeal region to the external surface overlying the cornea of the left eye. Histologically, the mass was a poorly-demarcated, densely cellular, invasive neoplasm. The neoplasm was composed of solid sheets and interlacing bundles of round, polygonal, and short spindle-shaped cells. Immunohistochemically, the neoplastic cells were positive for Iba-1 and CD204. These findings were cons

Histiocytic sarcoma (HS) is a rare and aggressive hematolymphoid neoplasm that frequently presents as extranodal disease. Intra-abdominal involvement is exceedingly rare and may clinically resemble abdominal tuberculosis. We present the case of a 29-year-old woman with abdominal pain, unintentional weight loss of 15 kg (25% of her baseline body weight) over four months, and altered bowel habits. CT demonstrated a large intra-abdominal mass measuring 14 × 11 × 7 cm with internal calcifications and mass effect on adjacent structures. Initial diagnostic evaluation included image-guided fine-needl

A 7-month-old intact female bearded collie dog was admitted after a 2-week history of progressive cough, inappetence, and lethargy, with no response to previous treatment with doxycycline and steroids. Mild attenuation of lung sounds in the right middle hemithorax was the only abnormality detected on physical examination. Abdominal ultrasound and thoracic radiographs were performed and revealed multifocally distributed nodules and masses, well-circumscribed and of variable size in the kidneys and pulmonary parenchyma. Ultrasound-guided fine needle aspirates of the renal and pulmonary masses we

Here we characterize 6 cases (4 autopsies and 2 biopsies) of histiocytic sarcoma in the CNS of cats. All affected cats had chronic, progressive clinical signs. Three autopsied cats were euthanized because of a poor prognosis, and one died. The clinical outcome for the biopsy cases remains unknown. Tumors occurred in the brain (4 cases), spinal cord (1 case), and brain and spinal cord (1 case). Neoplasms were restricted to the CNS in 3 cases. Reported gross changes in the 4 autopsy cases consisted of neuroparenchymal swelling with or without tissue pallor or gray discoloration (2 cases) and a y

Histiocytic sarcoma (HS) is a rare tumor that may result from the transdifferentiation of preexisting hematolymphoid neoplasms in a subset of patients. There are instances of correlation or concurrence between HS and a number of cancers, particularly B-cell-associated hematopoietic tumors. Only three cases of HS occurring subsequent to or concurrently with gastrointestinal stromal tumors (GIST) have been recorded. Our main objective was to give an overview of demographics, clinical signs and symptoms, histopathological findings, and immunohistochemical and molecular analysis when HS develops s

BACKGROUND: Haematological malignancies, such as lymphoma and leukaemia, can have a variety of clinical manifestations. The most frequent cause of death from haematological malignancies is multiple organ failure due to neoplastic organ infiltration and/or septic shock. Histiocytic sarcoma (HS) is a rare malignant nodal or extranodal tumour with histiocytic immunophenotype that originates from a lymphohematopoietic precursor. The patients with HS usually have a poor prognosis due to its aggressive clinical behaviour. Rare cases of undiagnosed sudden HS death have been described in the literatur

Histiocytic sarcoma (HS) is a rare malignant lymphohematopoietic neoplasm; it has been cited in the recent World Health Organization (WHO) classification as a malignant proliferation of cells exhibiting morphological and immunophenotypic features of mature histiocytes. To our knowledge, the present case is the first to be described in the bladder of a patient without a history of lymphoma. Only one case has been reported so far regarding a secondary bladder presentation in the setting of a previous diffuse large B-cell lymphoma. We discuss the case of a 68-year-old male who presented with hema

Histiocytic sarcoma is a rare malignant neoplasm that may occur de novo or in the context of a previous hematologic malignancy or mediastinal germ cell tumor. Here, we performed whole exome sequencing and RNA-sequencing (RNA-Seq) on 21 archival cases of primary histiocytic sarcoma. We identified a high number of genetic alterations within the RAS/RAF/MAPK pathway in 21 of 21 cases, with alterations in NF1 (6 of 21), MAP2K1 (5 of 21), PTPN11 (4 of 21), BRAF (4 of 21), KRAS (4 of 21), NRAS (1 of 21), and LZTR1 (1 of 21), including single cases with homozygous deletion of NF1, high-level amplific

OBJECTIVES: To describe a series of miniature schnauzers diagnosed with histiocytic sarcoma and assess for possible breed predisposition.
MATERIALS AND METHODS: Medical records of miniature schnauzers with a diagnosis of histiocytic sarcoma between January 2008 and April 2015 were reviewed. Data collected included signalment, body weight, presenting complaint, date of diagnosis, clinicopathologic and diagnostic imaging findings, treatment, therapeutic response, date of death or last follow-up and necropsy findings. Breed predisposition was assessed with odds ratios, using breed-matched dogs w

A 67-year-old woman presented with post-menopausal bleeding. Physical examination and imaging revealed a cervical mass with extensive pelvic involvement and supraclavicular lymphadenopathy. Clinical diagnosis was stage IVB cervical carcinoma. Cervical biopsy revealed high-grade squamous intraepithelial lesion and epithelioid tumor cells in the stroma. Lymph node biopsy revealed metastasis by similar epithelioid tumor cells. The epithelioid tumor cells were positive for p16, CD68, and CD163 on immunohistochemical study. The diagnosis of histiocytic sarcoma (HS) was rendered. This case demonstra

BACKGROUND: Histiocytic sarcoma (HS) is a rare malignant neoplasm showing morphologic and immunophenotypic features of histiocytes. Molecular characteristics of HS and fine-needle aspiration (FNA) criteria for its diagnosis have not been established.
METHODS: A case series of HS in 8 FNA samples from 6 patients was reviewed along with histopathologic and clinical data. Immunohistochemistry was performed on cell blocks (3 cases), core biopsies (5 cases), and surgical specimens (4 cases). Targeted-exome next-generation sequencing (NGS) was performed on surgical resection specimens in 4 cases.

We report an unusual case of histiocytic sarcoma with bladder involvement. An 80 year-old man with a previous history of diffuse large B-cell malignant lymphoma presented with hematuria and back pain. Serial urine cytologies revealed no urothelial malignant cells, but cystoscopy showed a large intravesical mass. The patient underwent transurethral resection (TUR) of the tumor. The bladder TUR specimen showed a widely infiltrating epithelioid neoplasm, with intense immunohistochemical positivity for CD45 and histiocytic markers (CD68, lysozime and fascin). Histopathological diagnosis was histio

Histiocytic sarcoma (HS) is an extremely rare but aggressive malignancy of hematopoietic origin. The diagnosis of HS mainly relies on pathological morphology and immunohistochemical staining combined with clinical symptoms. However, whether histiocytic sarcoma can be detected using flow cytometry (FCM) is not clear. We report the case of a 61-year-old female patient with a group of abnormal cells identified primarily by FCM. Largeabnormal cells -CD68+ CD11c+ CD14+ HLA-DR+ CD123+ CD45+ presented in upper forward scatter and side scatter, indicating that a malignancy originally derived from mono

Histiocytic sarcoma is a rare hematologic malignant neoplasia originating from histiocytic or dendritic cell clones. The lesions may be in nodal or extranodal sites, most commonly in the gastrointestinal tract. A small number of cases presents as unique cutaneous lesions. The definitive diagnosis is made by positivity for the immunohistochemical markers CD163, CD68, CD4 and lysozyme. The treatment is controversial, often with combined systemic chemotherapy. This is a case of cutaneous histiocytic sarcoma in an 82-year-old patient presenting two nodular lesions in the breast and right arm which

Histiocytic sarcoma (HS) is an extremely rare true histiocytic malignancy. We report a case of HS arising from thyroid gland in a 69 year-old man. Following subtotal thyroidectomy, a histopathologic, immunohistologic, and genotypic examination revealed HS. This tumor was composed of large spindle or round epithelioid cells with abundant eosinophilic cytoplasm. The neoplastic cells were positive for macrophage-associated antigen CD68, CD163, and lysozymes, as well as CD45, HLA- DR, DP, DQ, and S100, most consistent with a diagnosis of HS. The BIOMED-2 multiplex PCR analysis showed polyclonal B-
