Extended recombinant human growth hormone treatment after renal transplantation in children.

Recombinant human growth hormone (rhGH) was administered to 13 pediatric renal allograft recipients, ages 7.6 to 17.7 yr, who were 14 to 92 months posttransplant and growth retarded as manifested by either a standard deviation (SD) more negative than -2.00 or a height velocity index of less than 25%. The rhGH was given either daily or thrice weekly (0.375 mg/kg/wk) for a period of 12 to 36 months. Growth velocity increased from 2.7 +/- 2.1 SD for the 12-month period before the initiation of treatment of 6.3 +/- 2.9 SD (P less than 0.00005) and 5.2 +/- 2.9 SD (P less than 0.02) after 12 and 24

Human growth hormone-variant is a biologically active somatogen and lactogen.

The human GH-variant (hGH-V) gene, a member of the GH-PRL gene family, is expressed by the placenta during the second and third trimesters of gestation. The secreted hGH-V protein differs from pituitary GH (hGH-N) by only 13 amino acids. We have previously demonstrated that hGH-V can bind to both somatogen and lactogen cell surface receptors in vitro, but that the ratio of its somatogen to lactogen receptor-binding affinities is substantially higher than that of hGH-N. We now characterize the somatogen and lactogen bioactivities of hGH-V and contrast them to the bioactivity of hGH-N. Somatogen

Biosynthetic human growth hormone: subchronic toxicity studies in rats and monkeys.

Biosynthetic human growth hormone was injected subcutaneously in rats for 90 days and in cynomolgus monkeys for 30 days. The daily doses were 0.5, 3.3 and 25 IU kg-1 (rats) and 0.3 and 15 IU kg-1 (monkeys). The growth hormone was tolerated well in both rats and monkeys. No drug related deaths occurred and all animals appeared to be normal and also behaved normally throughout the dosing period. Increased body weight gain, increased food utilisation and increased organ weights were seen in the rats in the high and intermediate dose groups. The higher doses of human growth hormone (3.3 and 25 IU

Pituitary growth hormone from human cadavers: neurologic disease in ten recipients.

Creutzfeldt-Jakob disease (CJD) has been reported in three US patients previously treated with human growth hormone derived from large pools of human cadaver pituitary glands (pit-hGH). Neurologic disorders other than CJD occurred in 10 growth hormone-deficient patients treated with pit-hGH. These 10 cases could have been chance events or true syndromes; some cases may have been caused by transmissible agents contained in the pooled growth hormone product.

Effect of human growth hormone treatment on adipose tissue in children.

Body fat and the size and number of adipose cells were determined in 22 children with growth hormone deficiency before and after 6 months of replacement therapy with human growth hormone (HGH). A marked reduction in body fat and adipose cell size was found, and the total number of adipose cells increased at the normal rate during treatment. HGH does not appear to induce catch-up growth in the number of adipose cells.

Stimulatory effect of thyroid hormone on growth hormone gene expression in a human pituitary cell line.

A cell line (GX) derived from a pituitary tumor of a boy with gigantism, which exhibits morphological characteristics of epithelial cells, has been established and maintained in culture for over 2 yr. Initially, GX cells secreted GH which was stimulated by T3. However, after 2 yr in culture, GH production in GX cells is no longer detectable, though GH messenger RNA (mRNA) synthesis has been maintained. The GX cell line is a somatomammotroph line as indicated by the presence of both GH and PRL mRNAs. GX cells were used to study the effect of T3 on GH gene expression. A nearly 3-fold increase in

Potential effects of recombinant human growth hormone (r-hGH) on somatic growth in uremic rats.

In this study, we investigated both the efficacy of growth hormone (GH) in the improvement of growth disturbances in uremia and the effects on the remaining kidney, using uremic rats. Twenty 5/6 nephrectomized uremic rats were divided into two groups. One half of them were injected intraperitoneally with 2IU of human recombinant growth hormone (r-hGH) twice daily. The others were injected only with the vehicle. Sham operated control rats were also injected with the vehicle. These rats were pairfed and observed for as long as 4 weeks, during which, the growth in stature in uremic rats without r

[Safety considerations for the clinical application of recombinant human growth hormone].

Recombinant human growth hormone (rhGH) is an effective therapeutic drug for improving short stature. Currently, rhGH can be used for various causes of short stature, including growth hormone deficiency, and the expansion of its clinical application has raised concerns about its safety. Based on existing evidence, when rhGH is used in a standardized manner for physiological replacement therapy, its safety profile is favorable. In clinical practice, attention should be focused on short-term safety during rhGH treatment, with the combination of literature evidence and clinical experience. There

Hypercorticosteronemia induces hyperphagia and obesity in human growth hormone transgenic rats.

Hyperphagia and subsequent obesity are important public health issues due to the associated risks of developing serious diseases. Certain stressors play a major role in the development of hyperphagia. In previous studies, we established a line of human growth hormone transgenic (TG) rats that exhibit hyperphagia and obesity from a young age. We recently demonstrated that voluntary running on a running wheel alleviates hyperphagia in TG rats. Wheel running provides environmental enrichment for rodents and plays a role in relieving stress. These results suggested that stress is the major factor

Human growth hormone and glutamine for patients with short bowel syndrome.

BACKGROUND: There has been clinical enthusiasm for treating short bowel patients with human recombinant growth hormone and/or glutamine in hopes of reducing parenteral nutrition dependency. It has been more than a decade since Byrne and colleagues reported enhanced absorption of nutrients, improved weight gain, and reduction in parenteral nutrition requirements with the administration of a combination of human growth hormone (HGH) and glutamine in patients with short bowel syndrome. Other studies have reported inconsistent results. OBJECTIVES: The purpose of this systematic review was to eval

Long-term safety of recombinant human growth hormone in turner syndrome.

CONTEXT: Turner syndrome (TS) affects more than 50,000 girls and women in the United States. The National Cooperative Growth Study (NCGS) has collected efficacy and safety data for 5220 TS children treated with recombinant human GH (rhGH) during the last 20 yr. OBJECTIVES: Our objective was to determine frequencies of specific targeted adverse events (AEs) and additional AEs of interest in TS patients. Corresponding safety data in non-TS patients or normal populations were compared for selected AEs. METHODS: Patients may be enrolled at rhGH initiation and followed until discontinuation. Inve

Recombinant human growth hormone therapy in autosomal recessive polycystic kidney disease.

Patients with autosomal recessive polycystic kidney disease (ARPKD) may have growth retardation that is disproportionate to the degree of renal dysfunction. We treated growth-retarded ARPKD patients with recombinant growth hormone (rhGH) and document the response to therapy and effect of rhGH on the rate of progression of renal failure. The diagnosis of ARPKD and congenital hepatic fibrosis was made on the basis of clinical findings and by abdominal ultrasound examinations. Seventeen patients (6 girls/11 boys) aged 0.3-18.3 years were studied. Diagnosis was made prenatally in 6, after birth in

Iatrogenic Creutzfeldt-Jakob disease following human growth hormone therapy: case report.

We report the case of a 41-year-old man with iatrogenic Creutzfeldt-Jakob disease (CJD) acquired after the use of growth hormone (GH) obtained from a number of pituitary glands sourced from autopsy material. The incubation period of the disease (from the midpoint of treatment to the onset of clinical symptoms) was rather long (28 years). Besides the remarkable cerebellar and mental signs, the patient exhibited sleep disturbance (excessive somnolence) from the onset of the symptoms, with striking alteration of the sleep architecture documented by polysomnography. 14-3-3 protein was detected in

Human growth hormone induces system B transport in short bowel syndrome.

BACKGROUND: After massive enterectomy, remnant intestine undergoes compensatory adaptation. A combination of human growth hormone (hGH) and a glutamine-enriched modified diet induces further adaptation in patients with short bowel syndrome (SBS) on long-term total parenteral nutrition. The specific actions of each component, however, are not well-defined. METHODS: New Zealand White rabbits were randomized to control, sham operation, or SBS (70% midjejunoileal resection) groups and treated with either hGH or saline. Sodium-dependent uptake of glucose, glutamine, alanine, leucine, and arginine

Effects of recombinant human growth hormone in adults receiving maintenance hemodialysis.

Administration of recombinant human growth hormone stimulates protein synthesis, decreases urea generation, and improves nitrogen balance in individuals with normal renal function. However, little information is available concerning the effects of growth hormone in patients with renal disease. This pilot study evaluated urea kinetics and clinical/metabolic responses to short-term growth hormone administration in five clinically stable adult patients requiring maintenance hemodialysis for end-stage renal failure. The dialysis prescription, medications, and oral calorie and protein intake of eac
