There is an urgent need for new, potent antituberculosis (anti-TB) drugs with novel mechanisms of action that can be included in new regimens to shorten the treatment period for TB. After screening a library of carbostyrils, we optimized 3,4-dihydrocarbostyril derivatives and identified OPC-167832 as having potent antituberculosis activity. The MICs of the compound for Mycobacterium tuberculosis ranged from 0.00024 to 0.002 μg/ml. It had bactericidal activity against both growing and intracellular bacilli, and the frequency of spontaneous resistance for M. tuberculosis H37Rv was less than 1.91

A study has been made of a number of interferences observed in the trace determination of beryllium by atomic-absorption in the nitrous oxide-acetylene flame. The major negative interference caused by the presence of excess of aluminium salts may be overcome by the use of 8-hydroxyquinoline. Magnesium and silicon also depress the Be signal, but most other metals cause enhancement. In most instances the enhancements may be made uniform by the addition of potassium ions to the sample solution.

A series of 1-(quinoliloxypropyl)-4-aryl-piperazines has been synthesized and the target compounds evaluated for atypical antipsychotic activity in apomorphine induced mesh climbing and stereotypic behaviour in mice. The 8-hydroxyquinoline ether derivative 14 has emerged as an important lead compound showing a potential atypical antipsychotic profile. Employing appropriate physicochemical properties, the similarity of the compounds was assessed with respect to some atypical antipsychotic drugs as clozapine, ketanserine, ziprasidone and risperidone.

A new polarographic method is described for the determination of nitrite/nitrogen oxides. The nitrite is first diazotized with p-aminobenzoic acid, followed by coupling to 8-hydroxyquinoline to produce an azo dye. This azo dye is adsorbable on a mercury drop electrode and its reduction is reversible. The linear response range is 6 x 10(-9)-3 x 10(-7) g/ml. The detection limit is 4 x 10(-9) g/ml. This method has been applied to the determination of nitrogen oxides in the air.

A 24-year-old patient treated with hydroxychinoline preparations without cure for acrodermatitis enteropathica since he was nine years old was given zinc oxide orally, at first 50 mg three times, later twice daily. Zinc concentrations in blood plasma, whole blood and sputum were serially determined. At the start of treatment they were significantly below that in four healthy controls. One week after starting zinc therapy the blood level had returned to normal and with it the changes in skin, hair and nails had regressed, as had the emotional disturbances.

We disclose a nickel-catalyzed reaction, which enabled us to difunctionalize unactivated γ,δ-alkenes in ketones with alkenyl triflates and arylboronic esters. Assessing total magnesium (Mg) becomes challenging when cell quantities are minimal. Here, we present a protocol to quantify total intracellular Mg in samples with as few as several thousand cells using a fluorescent plate reader and the fluorescent dye diaza-18-crown-6-hydroxyquinoline-5 (DCHQ5). We describe steps for reagent setup, sample preparation, and assay running. We then detail procedures for sample analysis. This protocol has been validated on different cell lines with data from the literature.
