Immunoinformatics-based design and in silico evaluation of a multi-epitope vaccine candidate against Naegleria fowleri.

Primary amoebic meningoencephalitis (PAM) is caused by Naegleria fowleri, a free-living amoeba. It is a fatal infection with more than 95% mortality rate. Even though several combinations of antimicrobial agents were used to treat the condition, the overall survival remains poor and no effective vaccine is currently available. In this study, an immunoinformatic approach was utilized for designing a multi-epitope vaccine against N. fowleri. The sequences of antigenic proteins were retrieved for epitope prediction. The computational prediction identified 400 CTL, 590 HTL, and 65 B-cell high-bind

Reverse vaccinology-based design of a universal multiepitope vaccine against chikungunya virus: Phylogenetic and immunoinformatics approaches.

Chikungunya virus (CHIKV) infection is a re-emerging arboviral disease in tropical and subtropical regions. In addition to acute febrile syndrome, CHIKV infection may lead to chronic articular manifestations that significantly affect a long-term quality of life. This study aimed to design a universal vaccine candidate covering all circulating genotypes of CHIKV based on conserved multiepitope platform. We employed a large scale phylogenetic and immunoinformatic approach to identify conserved regions of the open reading frames (ORF2) region encoding viral structural proteins. This study ultimat

Epitope-based vaccine of NiV targeting glycoprotein and fusion protein: an integrated immunoinformatics and bioinformatics approach.

NiV is an ongoing infection that causes severe complications, and its frequent outbreaks make its treatment more complicated in terms of the proper therapeutic design. No viable vaccine candidate exists to combat NiV; however, efforts are underway, and some vaccines are in clinical trials. Therefore, an integrated immunoinformatics and bioinformatics approach was employed to design a promising epitope-based vaccine targeting glycoprotein and fusion protein, which are critical for host-pathogen interactions. The two targets, i.e., glycoprotein and fusion protein, were examined to identify B- an

In silico design of a citrullinated protein-based multi-epitope vaccine for rheumatoid arthritis using immunoinformatics.

Rheumatoid arthritis (RA) is a chronic autoimmune disorder marked by synovial inflammation and joint deformity. Anti-citrullinated protein antibodies is one of the key biomarkers for RA, produced against citrullinated peptides and contribute to inflammation by forming immune complexes. While current RA treatments may help in managing the symptoms, they may compromise the immune system by increasing vulnerability to infections. In this study, an immunoinformatics approach was used to design a multi-epitope vaccine targeting citrullinated peptides associated with RA. Citrullinated peptides were

Design of a multi-epitope vaccine against intestinal parasites associated with colorectal cancer using immunoinformatics approaches.

The increasing prevalence of gastrointestinal infections caused by Cryptosporidium parvum and Schistosoma mansoni, coupled with the lack of definitive treatments, underscores the need for vaccine development to prevent infection-related cancers. Using experimentally validated epitopes for vaccine development enhances confidence in inducing strong and durable immune responses compared to predicted epitopes, which require lab validation. In this study, we employed a unique computational strategy using 197 experimentally validated epitopes of C. parvum and S. mansoni obtained from the IEDB databa

Designing a Multi-epitope Vaccine against the SARS-CoV-2 Variant based on an Immunoinformatics Approach.

BACKGROUND: SARS-CoV-2 is a life-threatening virus in the world. Scientific evidence indicates that this pathogen will emerge again in the future. Although the current vaccines have a pivotal role in the control of this pathogen, the emergence of new variants has a negative impact on their effectiveness. OBJECTIVES: Therefore, it is urgent to consider the protective and safe vaccine against all subcoronavirus species and variants based on the conserved region of the virus. Multi-epitope peptide vaccine (MEV), comprised of immune-dominant epitopes, is designed by immunoinformatic tools and it

Chimeric vaccine design against the epidemic Langya Henipavirus using immunoinformatics and validation via immune simulation approaches.

In July 2022, a new virus called Langya virus (LayV) was discovered in China in patients who had a fever. This virus is a type of Henipavirus (HNV) and is considered a potential threat as it could spread from animals to humans. It causes respiratory disease with symptoms including fever, coughing, and fatigue and is closely linked to two other henipaviruses that are known to infect humans, namely Hendra and Nipah viruses. These viruses may cause fatal respiratory illnesses. Investigators believe that the LayV is spread by shrews, and may have infected humans directly or via an intermediary spe

Integrated immunoinformatics and subtractive proteomics approach for multi-epitope vaccine designing to combat S. pneumoniae TIGR4.

Streptococcus pneumoniae is one of the major precarious pathogens accountable for over 1.2 million fatalities annually. The key drivers for pneumococcal vaccine development involve high morbidity and mortality in over one million cases, especially in very young children and the elderly. In this study, immunoinformatics was integrated with subtractive proteomics to find antigenic proteins for designing a multi-epitope vaccine against S. pneumoniae. As prospective vaccine targets, the developed pipeline identified two antigenic proteins, i.e., penicillin-binding protein and ATP synthase subunit.

Designing a novel E2-IFN-γ fusion protein against CSFV by immunoinformatics and structural vaccinology approaches.

Subunit vaccines with high purity and safety are gradually becoming a main trend in vaccinology. However, adjuvants such as interferon-gamma (IFN-γ) are required to enhance immune responses of subunit vaccines due to their poor immunogenicity. The conjugation of antigen with adjuvant can induce more potent immune responses compared to the mixture of antigen and adjuvant. At the same time, the selection of linker, indispensable in the construction of the stable and bioactive fusion proteins, is complicated and time-consuming. The development of immunoinformatics and structural vaccinology appro

A cutting-edge immunoinformatics approach for design of multi-epitope oral vaccine against dreadful human malaria.

Human malaria is a pathogenic disease mainly caused by Plasmodium falciparum, which was responsible for about 405,000 deaths globally in the year 2018. To date, several vaccine candidates have been evaluated for prevention, which failed to produce optimal output at various preclinical/clinical stages. This study is based on designing of polypeptide vaccines (PVs) against human malaria that cover almost all stages of life-cycle of Plasmodium and for the same 5 genome derived predicted antigenic proteins (GDPAP) have been used. For the development of a multi-immune inducer, 15 PVs were initially

Vaccine Design from the Ensemble of Surface Glycoprotein Epitopes of SARS-CoV-2: An Immunoinformatics Approach.

The present study aimed to work out a peptide-based multi-epitope vaccine against the severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2). We predicted different B-cell and T-cell epitopes by using the Immune Epitopes Database (IEDB). Homology modeling of the construct was done using SWISS-MODEL and then docked with different toll-like-receptors (TLR4, TLR7, and TLR8) using PatchDock, HADDOCK, and FireDock, respectively. From the overlapped epitopes, we designed five vaccine constructs C1-C5. Based on antigenicity, allergenicity, solubility, different physiochemical properties, and mo

Exploring Leptospiral proteomes to identify potential candidates for vaccine design against Leptospirosis using an immunoinformatics approach.

Leptospirosis is the most widespread zoonotic disease, estimated to cause severe infection in more than one million people each year, particularly in developing countries of tropical areas. Several factors such as variable and nonspecific clinical manifestation, existence of large number of serovars and asymptomatic hosts spreading infection, poor sanitation and lack of an effective vaccine make prophylaxis difficult. Consequently, there is an urgent need to develop an effective vaccine to halt its spread all over the world. In this study, an immunoinformatics approach was employed to identify

An immunoinformatics-driven multiepitope subunit vaccine targeting Oropouche virus: Molecular docking, immune and dynamics simulation approaches.

The Oropouche virus (OROV) is an emerging zoonotic orthobunyavirus responsible for recurring outbreaks of febrile illness and neurological disorders in South and Central America. Despite its growing public health issues, no FDA-approved vaccines or antiviral therapies currently exist. To address this gap, a multiepitope subunit vaccine was designed to target OROV glycoproteins G1 (Gn), G2 (Gc), and the nucleocapsid (N) protein using a reverse-vaccinology (RV) approach. For vaccine construction, suitable epitopes (MHC-I, MHC-II, and linear B-cell) were selected based on percentile rank, antigen

Immunoinformatics Design of a Broad-Spectrum Multi-Epitope Vaccine Targeting HA2 and M1 of H9N2 AIV.

H9N2 avian influenza virus (AIV) continues to mutate, leading to immunosuppression and secondary infections in poultry. Traditional inactivated vaccines mainly induce humoral immunity and have limited cross-protection efficacy against various subtypes of virus strains. In this study, we targeted the HA2 and M1 proteins of H9N2 as antigens and used immunoinformatics methods to design a broad-spectrum multi-epitope vaccine (MEV) that can simultaneously activate humoral and cellular immunity. Firstly, through systematic evolutionary analysis and sequence comparison, highly conserved amino acid se
