Although fetal growth restriction (FGR) is linked to dysregulated immune responses, the immunological profiles of maternal and umbilical cord blood remain poorly understood. The purpose of this study was to compare immune cell subsets in maternal blood (MB) and umbilical cord blood (UCB) between pregnancies that are normal and those that are complicated by FGR. Flow cytometry was used to analyze MB and UCB samples from 33 pregnant women (8 in the FGR group and 25 in the normal control group). In contrast to UCB, which was enriched in naïve T cells, MB from normal pregnancies contained greater

Memory B cells are essential for long-lasting immunity induced by both encountered pathogens and vaccines. Functional diversity within the memory B cell pool is established through interactions between memory B cells and the environment in which they develop and reside. This review will discuss novel insights into the role of cytokines encountered in the microenvironment in modulating memory B cell transcriptomic, epigenetic, and phenotypic identity. Specifically, it will focus on how interferons can prime unique memory B cell subpopulations upregulated in chronic viral infection, autoimmunity

In human blood B cells exist in a variety of forms ranging from antigen-inexperienced naive cells to long-term memory cells, and antibody-producing plasma cells. Historically, the divisions between these types of cells have been defined by a few characteristic surface markers such as IgD, CD27, and CD21, but recent advances in single-cell techniques such as high-parameter cytometry and single-cell sequencing have revealed a greater diversity of populations. An expanding palette of markers has helped to better define B cell subsets but can be a source of confusion when overlapping cell types ar

Radiation therapy induces immunogenic cell death, which can theoretically stimulate T cell priming and induction of tumor-specific memory T cell responses, serving as an in situ vaccine. In practice, this abscopal effect is rarely observed. We use two mouse models of pancreatic cancer to show that a single dose of stereotactic body radiation therapy (SBRT) synergizes with intratumoral injection of agonistic anti-CD40, resulting in regression of non-treated contralateral tumors and formation of long-term immunologic memory. Long-term survival was not observed when mice received multiple fractio

BACKGROUND: Patients with heterotaxy syndrome, commonly associated with complex congenital heart disease (CHD), exhibit a higher risk of severe bacterial infection (SBI). We sought to define the change of a novel immunologic marker, the immunoglobulin M (IgM) memory B-cell percentage, and its association with SBI.
METHODS: We enrolled 46 (M/F 29/17) heterotaxy syndrome patients (42 right atrial isomerism (RAI) and 4 left atrial isomerism (LAI)) aged > 1 y during the period 2010-2012 in a tertiary care center. We analyzed IgM(+)CD27(+) memory B-cell percentages. Patients with simple and comple

The immunologic requirements for generating long-lived protective CD8 T cell memory remain unclear. Memory CD8 populations generated in the absence of CD4 Th cells reportedly have functional defects, and at least a subset of CD8 T cells transiently express CD40 after activation, suggesting that direct CD4-CD8 T cell interactions through CD40 may influence the magnitude and functional quality of memory CD8 populations. To ascertain the role of CD40 in such direct T cell interactions, we investigated CD8 T cell responses in CD40-/- mice after infection with Listeria monocytogenes, an intracellul

Normal immunological memory is thought to be underpinned by T lymphocytes. However, in rheumatoid arthritis there are indications that T-lymphocyte control has been subverted by self-perpetuating B lymphocytes. Potential mechanisms in other autoimmune states are less clear, but a number of observations suggest that misappropriation of immunological memory by B lymphocytes may be a common feature of human autoantibody-associated disease. Put simply, autoantibodies drive their own production. If so, the availability of safe B-lymphocyte-depleting agents provides a potential means for reversal of

Immunological memory to pathogens is associated with clonal expansion and heightened responsiveness of antigen-specific T cells. Recent work emphasizes that, for efficient protective immunity, qualitative changes in T memory cells are vitally important.

BACKGROUND: Hereditary alpha-tryptasemia (HαT) is characterized by elevated basal serum tryptase due to increased copies of the TPSAB1 gene. Individuals with HαT frequently present with multisystem complaints, including anaphylaxis and seemingly functional gastrointestinal (GI) symptoms.
OBJECTIVE: We sought to determine the prevalence of HαT in an irritable bowel syndrome cohort and associated immunologic characteristics that may distinguish patients with HαT from patients without HαT.
METHODS: Tryptase genotyping by droplet digital PCR, flow cytometry, cytometry by time-of-flight, immunohi

BACKGROUND: The intestinal microbiome in early infancy affects immunologic development and thus may affect vaccine memory, though few prospective studies have examined such associations. We examined the association of Bifidobacterium levels in early infancy with memory responses to early vaccination measured at 2 years of age.
METHODS: In this prospective observational study, we examined the association of Bifidobacterium abundance in the stool of healthy infants at 6 to 15 weeks of age, near the time of vaccination, with T-cell and antibody responses measured at 6 weeks, 15 weeks, and 2 year

Invasive trophoblast from Day 34 horse conceptuses survives in extrauterine sites in allogeneic recipients that are immunologically naive to donor major histocompatibility complex class I antigens. The ectopic trophoblast retains its in utero characteristics, including similar lifespan, physiologic effect of its secreted product (equine chorionic gonadotropin) upon the recipient's ovaries, and induction of host immune responses. Immunologic memory has not been considered previously in this experimental system. We hypothesized that primary exposure to ectopic trophoblast would affect the recipi

Neisseria gonorrhoeae, the cause of the sexually transmitted infection gonorrhea, elicits low levels of specific Ig that decline rapidly after the bacteria are cleared. Reinfection with the same serovar can occur, and prior gonococcal infection does not alter the Ig response upon subsequent exposure, suggesting that protective immunity is not induced. The mucosal Ig response apparent during gonorrhea does not correlate with that observed systemically, leading to a suggestion that it is locally generated. In considering whether N. gonorrhoeae directly influences B cells, we observed that gonoco

Innate immunity has long been viewed as lacking memory capabilities. However, an increasing number of studies are challenging this conventional immunological paradigm. In recent years, innate immune memory has been recognized as an essential component of immune defense, allowing organisms to mount either enhanced or diminished responses to previously encountered stimuli. Increasing evidence suggests that innate immune memory may underlie various lung diseases. In this regard, elucidating the mechanisms of innate immune memory in pulmonary diseases not only aids in understanding the underlying

INTRODUCTION: Splenectomy and stem cell transplant have historically provided long-term remissions that seemingly cure a subset of patients with immune thrombocytopenia (ITP). Dozens of modern medical therapies with better safety profiles have revolutionized ITP treatment and several can induce sustained response off treatment (SROT) and delay relapse, providing hope for disease modification and cure.
AREAS COVERED: In this review, we consider how ITP cure might be defined, review key pathophysiologic mechanisms relevant for disease modification, discuss historical approaches, and explore mod

Immune-mediated kidney diseases have a substantial disease burden and frequent disease relapses significantly contribute to the high morbidity. Relapsing disease courses are associated with poor outcome and accelerated progression to end-stage renal disease. The pathomechanisms leading to relapse are diverse and only incompletely understood. Both adaptive and innate immune cells are implicated in the immune memory of these diseases. In addition to antigen-dependent mechanisms of T and B cells, persistent epigenetic and metabolic changes in adaptive and innate immune cells may sustain immunolog

As children age, they become less susceptible to the diverse microbes causing pneumonia. These microbes are pathobionts that infect the respiratory tract multiple times during childhood, generating immunological memory. To elucidate mechanisms of such naturally acquired immune protection against pneumonia, we modeled a relevant immunological history in mice by infecting their airways with mismatched serotypes of Streptococcus pneumoniae (pneumococcus). Previous pneumococcal infections provided protection against a heterotypic, highly virulent pneumococcus, as evidenced by reduced bacterial bur

Tissue resident memory cells (Trm-cells) function as both benign and pathogenic memory cells. Benign Trm-cells play an important role in adaptive immunity by protecting against external pathogens, whereas pathogenic Trm-cells can induce relapses in several chronic skin diseases, including psoriasis. Early and intensive treatment with biologic therapy can lead to effective, long-term disease control by limiting local memory formation in the skin. However, this review finds it important not to compromise the beneficial role of Trm-cells in adaptive immunity when pursuing long-term treatment stra

Immunological memory underpins long-term protection and vaccination. Although early work established the durability and recall capacity of memory T cells, understanding of CD8 T cell memory generation, maintenance, and plasticity has advanced substantially over recent decades. Initial models viewed memory as an outcome of peak effector responses and were defined mainly by persistence. Successive waves of technological innovation, from MHC tetramers and genetic lineage-tracing approaches to single-cell and epigenomic profiling and modern high-throughput gene-perturbation screens, have reshaped
