Discovery, Synthesis, and Activity Evaluation of Novel Small-Molecule Inhibitors Targeting VISTA for Cancer Immunotherapy.

Immune checkpoint inhibitors (ICIs) have been potent therapeutic options for the treatment of multiple types of cancer. However, not all patients experience benefits from ICIs, and discovering inhibitors targeting novel immune checkpoints is necessary. V-domain Ig suppressor of T-cell activation (VISTA) is a novel immune checkpoint. Blockade of the VISTA pathway enhances antitumor immunity in multiple tumor types. Herein, a series of VISTA inhibitors based on the benzimidazole scaffold were discovered. B3 showed the strongest binding affinity to the VISTA protein with a KD value of 0.452 ± 0.1

Design, Synthesis, and Structure-Activity Relationship of Novel Human TLR1/2 Agonists for Potential Immunotherapy.

Capitalizing on the advantages of Toll-like receptor 2 (TLR2) small-molecule agonists combining high potency and low toxicity in cancer therapy, we designed and synthesized a novel series of thiourea-based TLR2 small-molecule agonists and identified a low-toxicity TLR1/2-specific small molecule agonist SMU-C409 (EC50 = 65 ± 3 nM). SMU-C409 incorporates key optimizations: a carbonyl conservation strategy eliminates carboxylesterase-mediated degradation, and a quinoline (N-heterocycle) enables salt formation that boosts solubility by 3.4-fold (free base) and 5.5-fold (hydrochloride) relative to

Self-Propelled In Situ Polymerized Nanoparticles Activating the STING Pathway for Enhanced Bladder Cancer Immunotherapy.

Bladder cancer, a common malignancy of the urinary tract, presents complex therapeutic challenges, thereby necessitating the exploration of innovative treatment strategies. This study introduces a novel, self-propelled nanomedicine delivery system that forms nanoparticles within the bladder lumen by co-infusing dopamine hydrochloride, Mn2+, cGAMP, and urease into the bladder to initiate in situ polymerization. The resulting Mn-cGAMP@PDA-urease (DMCU) nanoparticles possess a urease-modified surface, which acts as an engine to generate propulsive force by breaking down urea. Consequently, this p

Bacteria biohybrids integrating anticancer peptide-loaded nanoparticles for tumor immunotherapy through pyroptosis activation.

Developing bacterial-based biohybrid systems that effectively integrate tumor-targeting, immune modulation, and advanced drug delivery remains a major challenge in cancer therapy. This is primarily due to the complexities of achieving selective tumor colonization, overcoming immune clearance mechanisms, and ensuring controlled drug release within the tumor microenvironment. Here, we introduce P/L@EcN, a bacteria biohybrid system that conjugates ROS-responsive, ruxotemitide (LTX-315)-loaded poly(ethylene glycol)-block-poly(lactide-co-glycolide) (PEG-PLGA) nanoparticles (P/L-NPs) with the tumor-

Targeted immunotherapy of psoriasis: focusing on Janus kinase/signal transducers and activators of transcription signaling pathway.

Psoriasis is a chronic inflammatory skin condition that affects approximately 1%-3% of the global population. The advent of biologic therapies has markedly improved the management of moderate-to-severe cases. While the efficacy and safety of these novel agents are well documented, many patients value the convenience of oral therapies, which not only eliminate the risk of injection site infections and complications but also alleviate the fear and discomfort associated with needles. The Janus kinase (JAK)/signal transducers and activators of transcription (STAT) signaling pathway plays a crucial

Inflammation-Targeting Multienzyme Activity Carbon Dots Loaded with Methotrexate for Synergistic Immunotherapy in Rheumatoid Arthritis.

In rheumatoid arthritis (RA), excessive reactive oxygen species (ROS) and chronic inflammation drive damage to the synovium, cartilage, and bone. Developing precise and synergistic therapy for RA is crucial for improving remission rates. Here, carbon dots (CDs) with multienzyme activity and inflammation-targeting capabilities are designed to deliver methotrexate (MTX) for synergistic RA treatment. Specifically, positively charged CDs with porphyrin iron cores and amino-functionalized surfaces are synthesized to simultaneously scavenge hydrogen peroxide, superoxide anions, and hydroxyl radicals

Light-Controlled Bioorthogonal Chemistry Altered Natural Killer Cell Activity for Boosted Adoptive Immunotherapy.

Natural killer (NK) cell-based immunotherapy has received much attention in recent years. However, its practical application is still suffering from the decreased function and inadequate infiltration of NK cells in the immunosuppressive microenvironment of solid tumors. Herein, we construct light-responsive porphyrin Fe array-armed NK cells (denoted as NK@p-Fe) for cell behavior modulation via bioorthogonal catalysis. By installing cholesterol-modified porphyrin Fe molecules on the NK cell surface, a catalytic array with light-harvesting capabilities is formed. This functionality transforms NK

Recently activated CD4 T cells in tuberculosis express OX40 as a target for host-directed immunotherapy.

After Mycobacterium tuberculosis (Mtb) infection, many effector T cells traffic to the lungs, but few become activated. Here we use an antigen receptor reporter mouse (Nur77-GFP) to identify recently activated CD4 T cells in the lungs. These Nur77-GFPHI cells contain expanded TCR clonotypes, have elevated expression of co-stimulatory genes such as Tnfrsf4/OX40, and are functionally more protective than Nur77-GFPLO cells. By contrast, Nur77-GFPLO cells express markers of terminal exhaustion and cytotoxicity, and the trafficking receptor S1pr5, associated with vascular localization. A short cour

Enhancing anti-tumor immunity through liposomal oxaliplatin and localized immunotherapy via STING activation.

The cyclic GMP-AMP synthase-stimulator of interferon genes (cGAS-STING) pathway is a promising approach for anti-cancer immunotherapy by bridging innate and adaptive immunity. Recent evidence suggests that chemotherapy-induced DNA damage can directly induce dendritic cell (DC) maturation and recruitment, which synergizes with STING activation to enhance anti-tumor effects. As an immunogenic cell death (ICD) inducer, oxaliplatin generates massive double-stranded DNA (dsDNA) crosslinks, release of tumor-associated antigens and promoting the "eat me" signal. STING activation improves antigen immu

A Nanobody-Bioorthogonal Catalyst Conjugate Triggers Spatially Confined Prodrug Activation for Combinational Chemo-immunotherapy.

Checkpoint inhibitors have been used with chemotherapy to improve antitumor efficacy. However, overcoming the immunosuppressive effect of chemotherapeutics remains a challenge. We report a nanobody-catalyst conjugate Ru-PD-L1 by fusing a ruthenium catalyst to an anti-PD-L1 nanobody. After administration of Ru-PD-L1 and a doxorubicin (DOX) prodrug, Ru-PD-L1 disrupts the PD-L1/PD-1 interaction and catalyzes the uncaging of the DOX prodrug. The spatially confined release of DOX reduces its systemic toxicity and leads to immunogenic cell death (ICD). The induced ICD triggers antitumor immune respo

Secondary resistance to immunotherapy associated with β-catenin pathway activation or PTEN loss in metastatic melanoma.

BACKGROUND: While cancer immunotherapies including checkpoint blockade antibodies, adoptive T cell therapy, and even some vaccines have given rise to major clinical responses with durability in many cases, a subset of patients who initially respond subsequently develop secondary resistance to therapy. Tumor-intrinsic mechanisms of acquired immunotherapy resistance are incompletely understood. METHODS: Baseline and treatment-resistant tumors underwent molecular analysis via transcriptional profiling or genomic sequencing for oncogenic alterations and histologic analysis for T cell infiltration

No need for constant help: human IgG2 antibodies have an autonomous agonistic activity for immunotherapy of cancer.

Agonistic antibodies specific for members of the tumor necrosis factor receptor protein family hold great promise for immunotherapy of cancer. In this issue of Cancer Cell, White and colleagues provide evidence that the human IgG2 subclass may represent a superior backbone for the use of these antibodies in human therapy.

Active comparator-controlled, rater-blinded study of corticotropin-based immunotherapies for opsoclonus-myoclonus syndrome.

To test the efficacy and safety of corticotropin-based immunotherapies in pediatric opsoclonus-myoclonus syndrome, 74 children received corticotropin alone or with intravenous immunoglobulin (groups 1 and 2, active controls); or both with rituximab (group 3) or cyclophosphamide (group 4); or with rituximab plus chemotherapy (group 5) or steroid sparers (group 6). There was 65% improvement in motor severity score across groups (P < .0001), but treatment combinations were more effective than corticotropin alone (P = .0009). Groups 3, 4, and 5 responded better than group 1; groups 3 and 5 respond

Grass pollen immunotherapy induces mucosal and peripheral IL-10 responses and blocking IgG activity.

T regulatory cells and IL-10 have been implicated in the mechanism of immunotherapy in patients with systemic anaphylaxis following bee stings. We studied the role of IL-10 in the induction of clinical, cellular, and humoral tolerance during immunotherapy for local mucosal allergy in subjects with seasonal pollinosis. Local and systemic IL-10 responses and serum Ab concentrations were measured before/after a double-blind trial of grass pollen (Phleum pratense, Phl P) immunotherapy. We observed local increases in IL-10 mRNA-positive cells in the nasal mucosa after 2 years of immunotherapy, but

Anti-Angiogenic Agent Combined with Anti-PD-1 Immunotherapy Showed Activity in Patients With Classical Hodgkin Lymphoma Who Have Failed Immunotherapy: A Retrospective Case Report Study.

BACKGROUND: PD-1/PD-L1 inhibitor immunotherapy has showed impressive activity in various cancers, especially relapsed/refractory (r/r) classical Hodgkin lymphoma (cHL). However, acquired resistance is inevitable for most patients. Sometimes severe side effects also lead to treatment termination. When immunotherapy failed, alternative treatment options are limited. In the past few years, we have used the anti-angiogenic agent apatinib and PD-1 inhibitor camrelizumab to treat cHL patients who failed prior immunotherapy. In this study, we analyzed the data of these patients. PATIENTS AND METHODS

Identification of Active Compounds of Centipeda minima for Potentiating Tumor Immunotherapy in Non-Small Cell Lung Cancer via FANCD2-cGAS/STING/IFN-I Signaling Axis.

Activation of cGAS/STING/IFN-I signaling pathway serves as a potential therapeutic strategy to enhance CD8+ T cells infiltration and augment antitumor immunotherapy. The medicinal herb Centipeda minima has been traditionally employed to treat respiratory diseases and cancer; however, whether C. minima and its bioactive constituents can activate CD8+ T cell-mediated antitumor immunity in non-small cell lung cancer (NSCLC) remains elusive. In this study, the ethanol extract of C. minima (ECM) significantly enhanced the cytotoxic activity of human peripheral blood mononuclear cell-derived activat

Case report: Primary hepatocellular carcinoma with portal vein tumor thrombus characterized by active tumor immune microenvironment achieving a complete response following treatment of combined immunotherapy.

Portal vein tumor thrombus (PVTT) is a frequent complication in hepatocellular carcinoma (HCC). HCC patients with PVTT have the characteristics of less treatment tolerance and poor prognosis. Immunotherapy, especially combined immunotherapy, has been successfully used in advanced HCC. However, there are no recognized universally indicators that can predict response or resistance to immunotherapy for HCC. Herein, we reported a 58-year-old HCC patient with PVTT, cirrhosis and chronic viral hepatitis, who achieved complete response (CR) after combined immunotherapy (camrelizumab combined with sor
