Molecular Biomarkers Detected Using Fluorescence in situ Hybridization in a Filipino with Retinoblastoma.

BACKGROUND AND OBJECTIVE: Retinoblastoma is one of the most common intraocular cancers among children usually caused by the loss of retinoblastoma protein function. Despite being a highly heritable disease, conventional diagnostic and prognostic methods depend on clinical examination, with limited consideration of cancer genetics in the standard of care. CD133, KRT19, and MUC1 are commonly explored genes for their utility in liquid biopsies of cancer including lung adenocarcinoma. To date, there are few extensive molecular studies on retinoblastoma in Filipino patients. To this end, the study

Whole-Mount Fluorescence In Situ Hybridization to Study Spermatogenesis in the Anopheles Mosquito.

Spermatogenesis is a complex biological process during which diploid cells undergo successive mitotic and meiotic division followed by large structural changes to form haploid spermatozoa. Besides the biological aspect, studying spermatogenesis is of paramount importance for understanding and developing genetic technologies such as gene drive and synthetic sex ratio distorters, which, by altering Mendelian inheritance and the sperm sex ratio, respectively, could be used to control pest insect populations. These technologies have proven to be very promising in lab settings and could potentially

Exploring X Chromosomal Aberrations in Ovarian Cells by using Fluorescence In Situ Hybridization.

Millions of people worldwide deal with issues concerning fertility. Reduced fertility, or even infertility, may be due to many different causes, including genetic disorders, of which chromosomal abnormalities are the most common. Fluorescence in situ hybridization (FISH) is a well-known and frequently used method to detect chromosomal aberrations in humans. FISH is mainly used for the analysis of chromosomal abnormalities in the spermatozoa of males with numerical or structural chromosomal aberrations. Furthermore, this technique is also frequently applied in females to detect X chromosomal ab

The current and future applications of in situ hybridization technologies in anatomical pathology.

INTRODUCTION: In situ hybridization (ISH) plays an important role in the field of molecular diagnostics, especially in an anatomical pathology laboratory. ISH is a technique that can detect the targeted DNA or RNA sequences in tissue sections from frozen or fixed materials with labeled DNA or RNA probes. Radioactive and non-radioactive probes are the two major probes that can be used to label the targeted nucleic acids. AREAS COVERED: Two decades after the Human Genome Project, ISH has not only simply been applied to identify the chromosomal location of a human gene but has also been extensiv

Myb Immunohistochemical Staining and Fluorescence in situ Hybridization in Salivary Rare Basaloid Lesions.

Objective: Salivary rare basaloid lesions, including cribriform type basal cell adenoma (cBCA), BCA with incomplete capsule (iBCA), sialoblastoma (SB), and intercalated duct hyperplasia (IDH), could easily be misdiagnosed as adenoid cystic carcinoma (AdCC). We aim to identify an approach for differential diagnosis and to establish an optimal workflow concerning the diagnosis of these lesions. Material and methods: A panel of antibodies (MYB, β-catenin, CD117, SOX10, ki67, P63, calponin) and fluorescence in situ hybridization (FISH)-MYB were utilized to distinguish above salivary basaloid disea

HPV RNA in-situ hybridization as a diagnostic aid in papillary laryngeal lesions.

OBJECTIVES: In the larynx, differentiating squamous papillomas from de-novo papillary squamous dysplasias or squamous cell carcinomas (SCC) has significant consequences for management. Overlapping clinical presentations and cytologic changes across the spectrum of papillary lesions presents diagnostic challenges for otolaryngologists and pathologists. In this study, we evaluate whether ribonucleic acid (RNA) in-situ hybridization (ISH) for low-risk and high-risk human papillomavirus (HPV) can help distinguish these lesions. METHODS: We constructed tissue microarrays from 97 papillary laryngea

Detection of Coxiella burnetii in heart valve sections by fluorescence in situ hybridization.

PURPOSE: Infective endocarditis is a severe and potentially fatal disease. Nearly a third of all cases remain culture-negative, making a targeted and effective antibiotic therapy of patients challenging. In the past years, fluorescence in situ hybridization (FISH) has proven its value for the diagnosis of infective endocarditis, particularly when it is caused by fastidious bacteria. To increase the number of infective endocarditis causing agents, which can be identified by FISH, we designed and optimized a FISH-probe for the specific detection of Coxiella burnetii in heart valve tissue. Fluorescence in-situ hybridization and dermoscopy in the assessment of controversial melanocytic tumors.

Although the 'gold standard' for melanoma diagnosis remains histopathological analysis, presently dermoscopists play a significant role in the diagnostic process. However, even a combined approach may not allow a clear-cut judgment on equivocal melanocytic lesions. Fluorescence in-situ hybridization (FISH) can offer assistance in the evaluation of chromosome abnormalities associated with malignancies, and its role is emerging in melanoma diagnosis. The aim of this study was to evaluate the diagnostic role of the FISH in the assessment of controversial lesions, defined as those lesions showing

Detection of HPV by in situ hybridization in thin-layer (ThinPrep) cervicovaginal samples.

We have studied an automated in situ hybridization (ISH) method as a possible alternative approach for detecting high-risk human papillomavirus (HPV) in monolayer (ThinPrep) cervico-vaginal samples, comparing the results with those obtained by polymerase chain reaction (PCR) using consensus primers and studying the relationship between the ISH staining pattern and the viral integration in HPV 16-positive cases. Eighty atypical squamous cells of undetermined significance (ASCUS) and low-grade squamous intraepithelial lesion (LSIL) cases were used for our purposes. The patients were monitored th

The evidence of maternal microchimerism in biliary atresia using fluorescent in situ hybridization.

BACKGROUND: Biliary atresia (BA) is a cholestatic disease of unknown etiology. It has recently been suggested that graft-vs-host disease caused by microchimerism is an etiology in the development of autoimmune disease. Moreover, the liver is a frequent target organ of graft-vs-host disease. The aim of this study is to identify the presence and extent of maternal microchimerism and to determine whether it plays a role in the etiology of BA. METHODS: The liver biopsy specimens of 6 male patients with BA (BA group) and 6 males with other liver diseases (non-BA group) were assayed for X- and Y-ch

[Detection of mosaicism in women with Turner's syndrome using fluorescence in situ hybridization].

BACKGROUND: According to the literature approximately 50% of patients with Turner's syndrome have karyotype 45,X in every cell, the rest have two or more cell lines with mosaics of sex chromosomes. New methods have shown that the mosaicism is probably more frequent than expected from classical cytogenetics examinations. The aim of this study was to detect numerical changes of sex chromosomes in mitoses and interphase nuclei of cultivated peripheral lymphocytes of patients with Turner's syndrome by means of FISH, to compare the sensitivity of classical and molecular-cytogenetic methods and to e

Fluorescent in situ hybridization for identifying cytogenetic abnormalities in inadequate and suboptimal specimens.

Conventional cytogenetic analysis is often hampered by its dependency on the evaluation of dividing cells. When the mitotic index is low, or the cytogenetic preparation suboptimal, an accurate diagnosis often cannot be achieved using standard GTG banding. Molecular cytogenetics using fluorescent in situ hybridization (FISH) can be extremely useful in such a situation because it can be performed on nondividing interphase nuclei, and can usually be carried out on existing slides without requiring an additional specimen. The present paper presents two case histories to illustrate the utility of F

[Fluorescent in situ hybridization of enriched SFA-DNA on pachytene chromosomes in mouse].

In the present study, the primary constrictions of mouse pachytene chromosomes stained with Hoechst 33258, and those stained by indirect immunofluorescence method with CREST antiserum were compared with those hybridized by the probe DNA isolated from the enriched mouse spindle fiber attachments. It was found that the DNA probe hybridized not only with the centromeric regions on synaptonema complexes but also with the DNA of pericentromeric heterochromatin of all autosomes. Moreover, the DNA probe hybridized also with the centromeric regions of both X and Y chromosomes. It was concluded that th

Detection of chromosome aberrations in interphase nuclei using fluorescence in situ hybridization technique.

We report here several experiences of interphase cytogenetics, using fluorescence in situ hybridization (FISH) technique, for the detection of chromosome aberrations. FISH, using alpha satellite specific probes of 18, X, Y chromosomes, was done in interphase nuclei from peripheral blood of patients with Edwards' syndrome, Klinefelter's syndrome and Turner's syndrome with healthy male and female controls, respectively. The distributions of fluorescent signals in 100 interphase nuclei were well correlated with metaphase findings. Nowadays FISH plays an increasingly important role in a variety of
