EDTA-Facilitated Collection of Phloem Exudates for Chemical Analyses.

Phloem exudates can be collected using several methods, including the ethylenediaminetetraacetic acid (EDTA)-facilitated method. Plant parts, such as leaves or inflorescences, are cut, and the cut edges are immediately placed into EDTA solution, which prevents plugging of sieve tubes. After this first inoculation, plant parts are transferred to water, into which phloem exudates are collected for a few hours. Inoculation and collection take place in the dark at high relative humidity. The samples can subsequently be used for different chemical analyses of RNA, proteins and various primary and s

Vegetative and reproductive morphology of Othniophyton elongatum (MacGinitie) gen. et comb. nov., an extinct angiosperm of possible caryophyllalean affinity from the Eocene of Colorado and Utah, USA.

BACKGROUND AND AIMS: Eocene foliage formerly attributed to the extant araliaceous genus Oreopanax was found attached to twigs bearing inflorescences and infructescences unlike those of Araliaceae. Using newly observable characters of phyllotaxy, vegetative and floral buds, infructescences and seeds, we sought to reassess the affinities of this strange angiosperm. METHODS: Fossils were studied from the Parachute Creek Member of the Green River Formation from near Bonanza, Utah and Douglas Pass, Colorado (~47 Mya). Macrofossil impression remains were investigated by low-angle reflected light, a

Diversity of pollination ecology in the Schismatoglottis Calyptrata Complex Clade (Araceae).

Field studies integrating pollination investigations with an assessment of floral scent composition and thermogenesis in tropical aroids are rather few. Thus, this study aimed to investigate the pollination biology of nine species belonging to Schismatoglottis Calyptrata Complex Clade. The flowering mechanism, visiting insect activities, reproductive system, thermogenesis and floral scent composition were examined. Anthesis for all species started at dawn and lasted 25-29 h. Colocasiomyia (Diptera, Drosophilidae) are considered the main pollinators for all the investigated species. Cycreon (Co

Warming in the Maternal Environment Alters Seed Performance and Genetic Diversity of Stylosanthes capitata, a Tropical Legume Forage.

Background/Objectives: Global warming and rising CO2 concentrations pose significant challenges to plant systems. Amid these pressures, this study contributes to understanding how tropical species respond by simultaneously evaluating reproductive and genetic traits. It specifically investigates the effects of maternal exposure to warming and elevated CO2 on progeny physiology, genetic diversity, and population structure in Stylosanthes capitata, a resilient forage legume native to Brazil. Methods: Maternal plants were cultivated under controlled treatments, including ambient conditions (contro

Cinnamomum fasciculatum (Lauraceae), a new species from Guangdong, China.

Cinnamomum fasciculatum, a new species from Guangdong, China, is described and illustrated here. Morphologically, it is most similar to Cinnamomum parthenoxylon in having sericeous terminal buds, glabrous branchlets, glabrous leaves, and fruits of similar shape and size. However, C. fasciculatum can be readily distinguished by its subterminal inflorescences (vs. axillary and lateral) and a later flowering season (June-July vs. March-May). Phylogenetic analysis based on complete chloroplast genomes also supports that it is a new member of C. sect. Camphora and is sister to a clade comprising Ci

A new species of Actinostemon (Euphorbiaceae) from Southern Bahia Atlantic Forest, Brazil.

Actinostemon (Euphorbiaceae, Hippomaneae) is a Neotropical genus, with a total of 16 species. Brazil stands out as its center of diversity with nine species, most from seasonal dry forests of the Atlantic Forest. Through the analysis of herbarium specimens, protologue and field collections, we present here a new species, Actinostemon sessilantherus, from southern Bahia. Actinostemon sessilantherus resembles A. concolor in its alternate, non pseudoverticillate leaves, coriaceous blades and cataphylls, and one pistillate flower per inflorescence, glabrous, without appendages on the carpels. Recessive antimorph alleles reveal novel functions of the Opaque1 myosin XI in maize.

Ideal plant architecture optimizes canopy structure and increases grain yield in maize. However, its underlying genetic mechanisms remain poorly characterized. Two recessive, EMS-induced maize mutants were identified that have reduced stature, opaque kernels, and abnormal subsidiary cell division and are allelic to Opaque1 (o1) which encodes myosin XI-I. These 2 new missense alleles, o1-2995 and o1-tan62, unlike the loss-of-function alleles previously identified, generate O1 protein but paradoxically generate more severe morphological defects. These defects include reduced internode elongation

Loss of Multiple ABCB Auxin Transporters Recapitulates the Major twisted dwarf 1 Phenotypes in Arabidopsis thaliana.

FK506-BINDING PROTEIN 42/TWISTED DWARF 1 (FKBP42/TWD1) directly regulates cellular trafficking and activation of multiple ATP-BINDING CASSETTE (ABC) transporters from the ABCB and ABCC subfamilies. abcb1 abcb19 double mutants exhibit remarkable phenotypic overlap with twd1 including severe dwarfism, stamen elongation defects, and compact circinate leaves; however, twd1 mutants exhibit greater loss of polar auxin transport and additional helical twisting of roots, inflorescences, and siliques. As abcc1 abcc2 mutants do not exhibit any visible phenotypes and TWD1 does not interact with PIN or AU

Assessment of the Cannabinoid Content from Different Varieties of Cannabis sativa L. during the Growth Stages in Three Regions.

Hemp (Cannabis sativa L.) belongs to the Cannabaceae family. It is very rich in chemical constituents, especially the cannabinoids which has not been reported in any other plant, and has broad pharmacological properties. Hemp as a multi-purpose crop is a good source of fibers, seed, fixed and volatile oil. It is known that the cannabinoid content of hemp is related to genetic factors, as well as plant's growth stages and environmental factors such as latitude, altitude, weather, particularly moisture availability and nutrient supply during the growing season. The present study was designed to

First Report of Head Rot of Brassica oleracea convar. botrytis var. italica Caused by Pseudomonas fluorescens in Southern Italy.

During the winter of 2004, symptoms were observed in commercial cauliflower (Brassica oleracea L. convar. botrytis (L.) Alef. var. italica) fields of "romanesco" type (cv. Navona) in Apulia, southern Italy. These symptoms were noted on inflorescences that were almost ready for harvest, and a bacterial etiology was suspected. In particular, the corymbs showed water-soaked and brown discolored areas which then rotted. The above alterations involved the whole inflorescences, or in some cases, only a few florets. Longitudinal sections of the symptomatic inflorescences or the single floret showed b

Ethnobotanical study on edible flowers in Xishuangbanna, China.

BACKGROUND: Edible flowers (EFs) represent valuable sources of both food and medicinal resources, holding the promise to enhance human well-being. Unfortunately, their significance is often overlooked. Ethnobotanical studies on the EFs are lacking in comparison with their botanical and phytochemical research. The practice of consuming flowers as food has a rich culture and long history in China, especially among different linguistic groups in Xishuangbanna, Yunnan. However, economic activities have led to a decline of this tradition. Consequently, preserving the traditional knowledge and cultu

The Effect of Biogeographic Origin on the Adaptive Trajectories of Plants in Urban Environments.

Parallel adaptation is a hallmark of urban evolution, yet whether plant responses to urbanization are globally consistent or influenced by biogeographic origin remains unclear. We investigated pollinator communities, floral display and reproductive allocation in Trifolium repens along urbanization gradients in 11 European urban areas spanning boreal, continental, and Mediterranean biogeographic regions. We found strong differences, with boreal and continental gradients showing no consistent floral trait shifts, whereas Mediterranean gradients showed parallel reductions in flower and infloresce

Incomplete functional divergence drives MADS-box gene synergy during floral development in tomato.

Genetic synergy arises from interactions between functionally related genes that control complex traits in plants and animals. Synergy occurs when the combined effect of multiple genes exceeds the additive contribution from each individual gene. However, genetic mechanisms that drive and maintain synergy remain underexplored. Here, we investigated synergistic interactions among SEPALLATA (SEP) MADS-box genes during floral development in tomato. We discovered that SEP gene synergy emerges from duplicated genes that partitioned functions to regulate inflorescence architecture and floral organ id

Structural analysis of phosphatidylcholine of plant tissue.

Pure preparations of phosphatidylcholine were isolated from spinach leaf chloroplasts, spinach leaf microsomes, and cauliflower inflorescence. The isolated phosphatidylcholine was treated with snake venom phospholipase A, and the fatty acid distribution and composition of the fatty acid methyl esters prepared from the lysophosphatidylcholine and the freed fatty acid were determined by gas-liquid chromatography. The results showed that saturated fatty acids were preferentially esterified at position 1 and unsaturated fatty acids at position 2. The phosphatidylcholine from cauliflower was also t

Characterization and antimicrobial activity of essential oils of industrial hemp varieties (Cannabis sativa L.).

The present study focused on inhibitory activity of freshly extracted essential oils from three legal (THC<0.2% w/v) hemp varieties (Carmagnola, Fibranova and Futura) on microbial growth. The effect of different sowing times on oil composition and biological activity was also evaluated. Essential oils were distilled and then characterized through the gas chromatography and gas chromatography-mass spectrometry. Thereafter, the oils were compared to standard reagents on a broad range inhibition of microbial growth via minimum inhibitory concentration (MIC) assay. Microbial strains were divided i

A FILAMENTOUS FLOWER orthologue plays a key role in leaf patterning in opium poppy.

The plant-specific YABBY genes were initially defined by their roles in determining abaxial/adaxial cell fate in lateral organs of eudicots, and repressing meristematic genes in differentiating tissues such as leaves. In Arabidopsis thaliana FILAMENTOUS FLOWER (FIL) is also required for inflorescence and floral meristem establishment and flower development in a pathway involving the floral transition and identity genes. Here we describe the characterization of a FIL orthologue from the basal eudicot, Papaver somniferum (the opium poppy), and demonstrate a role for the gene in patterning the hi
