Cold Tolerance and Differential Expression of Cuticular Protein Genes in Sungaya inexpectata Zompro, 1996 (Insecta: Phasmatodea).

Sungaya inexpectata is a tropical stick insect endemic to the Philippines, providing a useful system for investigating cold responses in tropical ectotherms. In this study, we exposed individuals to low temperature (8 °C) and normal temperature (25 °C) and characterized their transcriptomic responses. A total of 1656 differentially expressed genes were identified, including those involved in energy metabolism, cuticular proteins (CPs), and heat shock proteins. Since CP-related genes were notably enriched, we focused on this family. qPCR assessment provided preliminary expression profiles for s

The jewel wasp Nasonia vitripennis utilizes two single-copy, protamine-like sperm nuclear basic proteins.

The DNA of sperm is uniquely packaged into an exceptionally condensed chromatin state with sperm nuclear basic proteins (SNBPs). These proteins are diverse, including protamines, protamine-like (PL) proteins, testis-specific (TS) histones, and high mobility group (HMG)-box proteins. Usage of SNBP type varies widely among and within phyla. Moreover, SNBPs can be evolutionarily dynamic in copy number and amino acid composition, even among closely related species. Despite the diversity of insects, little is known about SNBP usage across members of this taxonomic group. A previous study biochemica

Entomo-venomics: The evolution, biology and biochemistry of insect venoms.

The insects are a hyperdiverse class containing more species than all other animal groups combined-and many employ venom to capture prey, deter predators and micro-organisms, or facilitate parasitism or extra-oral digestion. However, with the exception of those made by Hymenoptera (wasps, ants and bees), little is known about insect venoms. Here, we review the current literature on insects that use venom for prey capture and predator deterrence, finding evidence for fourteen independent origins of venom usage among insects, mostly among the hyperdiverse holometabolan orders. Many lineages, inc

Edible Insect-Based Beverages: A Narrative Review of Functional, Technological, and Experimental Dimensions.

Edible insects are increasingly recognized as sustainable and nutrient-dense ingredients with potential applications across diverse food systems. While their use in solid foods has been widely explored, the incorporation of insect-derived ingredients into beverages remains fragmented and insufficiently conceptualized. This narrative review critically examines the current state of insect-based beverages, integrating technological, nutritional, cultural, and market-oriented perspectives. Rather than adopting a fully systematic review methodology, this article synthesizes representative scientifi

Identification of carmine allergens among three carmine allergy patients.

BACKGROUND: There have been several reports of carmine allergy; however, identification of the responsible carmine allergens has not been widely documented. METHODS: Three female patients presented with a history of anaphylaxis and/or urticaria/angioedema after ingestion of carmine-containing foods. All three patients had 4+ skin prick tests to carmine. Among them, two patients were confirmed to have carmine allergy by blinded, placebo-controlled food challenges to carmine. SDS-PAGE of cochineal insects and carmine, immunoblotting for IgE antibody with sera from all three patients, and immuno

Identification of AcMNPV GP64-binding proteins through a combinational use of a self-biotinylated virus and the cross-linking method.

Baculoviruses are potential vectors of gene therapy for the ability to transfer gene high efficiently into mammalian cells. However, cell membrane proteins which interact with baculoviral glycoproteins have not been identified. In this study, we developed a self-biotinylated AcMNPV bearing biotinylated GP64 glycoproteins. This recombinant virus demonstrated the capability to infect insect cells and to transduct mammalian cells. Using this biotinylated virus, a protein >170Kda which could specifically interact with GP64 proteins was identified from virus transducted BHK-21 cells through cross-l

Specific antisera produced by direct immunization with slices of polyacrylamide gel containing small amounts of protein.

Rabbits were injected with slices of polyacrylamide gels containing entrapped insect proteins after separation by electrophoresis. Specific antibodies were produced independently of the nature of the gel (with or without sodium dodecyl sulphate) and of the staining technique (amido black or Coomassie Blue). The procedure appears to be a rapid and simple method for production of antibodies specific to proteins separated in minute quantities from a complex mixture.

Chemosensory proteins, major salivary factors in caterpillar mandibular glands.

Research in the field of insect-host plant interactions has indicated that constituents of insect saliva play an important role in digestion and affect host chemical defense responses. However, most efforts have focused on studying the composition and function of regurgitant or saliva produced in the labial glands. Acknowledging the need for understanding the role of the mandibular glands in herbivory, we sought to make a qualitative and semi-quantitative comparison of soluble luminal protein fractions between mandibular and labial glands of Vanessa gonerilla butterfly larvae. From Molecular Recognition to Behavioral Control: Dual-Target Aphid Repellent Discovery of Novel Ionone Derivatives.

Odorant-binding proteins (OBPs) are promising targets for insect behavior regulators. We established an original target-to-behavior strategy for developing dual-target aphid repellents. Structure-based virtual screening initially identified β-ionone as a lead compound, from which 24 novel ionone derivatives were designed and synthesized targeting Acyrthosiphon pisum OBP3 and OBP7. Most ionone derivatives exhibited enhanced binding affinities to both ApisOBP3 (5.14-20.70 μM) and ApisOBP7 (3.64-28.68 μM). Molecular dynamics simulations and alanine site-directed mutagenesis revealed that mutation

Mode of action of nanoparticles against insects.

The employment of nanoparticles obtained through various synthesis routes as novel pesticides recently attracted high research attention. An impressive number of studies have been conducted to test their toxic potential against a wide number of arthropod pests and vectors, with major emphasis on mosquitoes and ticks. However, precise information on the mechanisms of action of nanoparticles against insects and mites are limited, with the noteworthy exception of silica, alumina, silver, and graphene oxide nanoparticles on insects, while no information is available for mites. Here, I summarize cu

NlAgo3 and NlBiwi are essential for piRNAs biogenesis and female reproduction in Nilaparvata lugens.

P-element-induced wimpy testis (PIWI) -interacting RNAs (piRNAs) are short non-coding RNAs crucial for silencing transposable elements (TEs) and maintaining genome stability, particularly in germ cells. While most research on piRNAs and PIWI proteins has traditionally focused on model organisms such as Drosophila, emerging studies are now extending to other insect orders. However, little is known about piRNAs in insects of the Hemiptera order. Here, we investigated the role of piRNAs in the brown planthopper (BPH), Nilaparvata lugens, a significant pest of rice that belongs to the Hemiptera or

Planthopper salivary sheath protein LsSP1 contributes to manipulation of rice plant defenses.

Salivary elicitors secreted by herbivorous insects can be perceived by host plants to trigger plant immunity. However, how insects secrete other salivary components to subsequently attenuate the elicitor-induced plant immunity remains poorly understood. Here, we study the small brown planthopper, Laodelphax striatellus salivary sheath protein LsSP1. Using Y2H, BiFC and LUC assays, we show that LsSP1 is secreted into host plants and binds to salivary sheath via mucin-like protein (LsMLP). Rice plants pre-infested with dsLsSP1-treated L. striatellus are less attractive to L. striatellus nymphs t

Development of real-time RT-PCR for evaluation of JEV clearance during purification of HPV type 16 L1 virus-like particles.

Insect cell culture has greatly increased in part due to the widespread use of insect virus-based vectors for efficient expression of foreign proteins. Insect cells such as Sf9 cells are susceptible to arboviruses which may pose a safety concern by adventitious introduction during the production process. The objective of this study was to establish techniques for viral clearance validation of insect cell-derived biotechnological products using Japanese encephalitis virus (JEV) as a model, since JEV is a member of arthropod-borne flaviviruses that are known to be infectious in insect cells. Manure to insect farming for feed mitigates nitrogen and greenhouse gas emissions.

Livestock manure is nutrient-rich and represents a promising feedstock for insect farming, with insect protein offering a high-quality alternative to conventional crop-based livestock feed. This pathway has substantial potential to mitigate nitrogen (N) and greenhouse gas (GHG) emissions from livestock systems. Here, we integrate global livestock models with trade, land-use, and cost-benefit analysis to quantify the environmental and economic implications of manure-to-insect conversion while ensuring the recycling of manure to croplands to maintain soil fertility across 166 countries. We show

Simulation of insect enriched bread consequences on gut microbiota.

Insect powders are newly regulated in Europe and can be used to fortify protein-enriched bakery products. Because protein sources influence gut microbial balance, assessing their impact on the human microbiome is essential. This study investigated how cricket (Acheta domesticus) powder affects colon microbiota and whether sourdough fermentation improves protein-enriched breads. Using an in vitro gut model combining Infogest® digestion with MICODE© colon microbiota fermentation, four bread prototypes were examined. Metabolomic and microbiomic analyses showed that sourdough breads with insect po

Cooking Has Variable Effects on the Fermentability in the Large Intestine of the Fraction of Meats, Grain Legumes, and Insects That Is Resistant to Digestion in the Small Intestine in an in Vitro Model of the Pig's Gastrointestinal Tract.

This study aimed to evaluate the fermentation in the large intestine of indigestible dietary protein sources from animal, insect, and plant origin using an in vitro model of the pig's gastrointestinal tract. Protein sources were used raw and after a cooking treatment. Results showed that the category of the ingredient (meats, insects, or grain legumes) exerts a stronger impact on enzymatic digestibility, fermentation patterns, and bacterial metabolites such as short-chain fatty acids (SCFA) and hydrogen sulfide (H2S) than the cooking treatment. The digestibility and the fermentation characteri

Proteomic and transcriptomic analyses of saliva and salivary glands from the Asian citrus psyllid, Diaphorina citri.

Salivary secretions play critical roles in interactions among insects, insect-vectored pathogens, and host plants. The Asian citrus psyllid Diaphorina citri is a sap-sucking Hemipteran that serves as a vector for Candidatus Liberibacter asiaticus, the causal agent of citrus greening disease ("Huanglongbing" or HLB). D. citri continuously injects saliva into host plants using specialized stylets so as to feed and transmit the HLB pathogen. Knowledge on the composition and function of salivary proteins of this pest is very limited. In this study, proteomic and transcriptomic approaches were adop

Generation of expression clone set for functional proteomics of human gastric and liver cancers.

Two thousand sixty-eight multi-purpose expression clones for the 326 candidate genes related to gastric or liver cancers were constructed using the Gateway system. These clones can be expressed as His, Glutathione-S-transferase (GST) or Enhanced version of the green fluorescent protein (EGFP) fusion proteins in E. coli, insect cells or mammalian cells. For the 246 E. coli expression clones, the GST fusion proteins had greater expression efficiency and solubility than the His fusion proteins. Approximately 20% of the expressed proteins had unexpected molecular weights. A detailed sequence analy

Global Gene Expression Profiling through the Complete Life Cycle of Trypanosoma vivax.

The parasitic flagellate Trypanosoma vivax is a cause of animal trypanosomiasis across Africa and South America. The parasite has a digenetic life cycle, passing between mammalian hosts and insect vectors, and a series of developmental forms adapted to each life cycle stage. Each point in the life cycle presents radically different challenges to parasite metabolism and physiology and distinct host interactions requiring remodeling of the parasite cell surface. Transcriptomic and proteomic studies of the related parasites T. brucei and T. congolense have shown how gene expression is regulated d

A potential role for the interaction of Wolbachia surface proteins with the Drosophila microtubulin in maintenance of endosymbiosis and affecting spermiogenesis.

Wolbachia, as a widely infected intracellular symbiotic bacterium in Arthropoda, is able to manipulate the reproduction of insect hosts for facilitating their own transmission. Cytoplasmic incompatibility (CI) is the most common phenotype that Wolbachia induced in insect hosts where they resulted in the failure of uninfected egg hatch when fertilized with the sperm derived from Wolbachia-infected males, suggesting that the sperm are modified by Wolbachia during spermatogenesis. Although the molecular mechanisms of CI are beginning to be understood, the effects of Wolbachia on the symbiotic rel
