Effects of trichloroethylene and its metabolites on rodent hepatocyte intercellular communication.

Chronic exposure to trichloroethylene (TCE) results in hepatocellular cancer in mice but not rats. The induction of hepatic tumors by TCE appears to be mediated through nongenotoxic or tumor promotion mechanisms. One cellular effect exhibited by a number of nongenotoxic carcinogens and tumor promoters is the inhibition of gap junction mediated intercellular communication. In the present study, the effects of trichloroethylene (TCE) and its metabolites, trichloracetic acid (TCA), trichloroethanol (TCEth), and chloral hydrate (CH) on gap junction mediated intercellular communication in cultured

Re-examination of the carotid body ultrastructure with special attention to intercellular membrane appositions.

Ultrathin sections of the carotid body of adult rats, processed through freeze-substitution after aldehyde-prefixation, showed a substantial number of presumed gap junctions between two adjacent chief cells, between chief and sustentacular cells, and between chief cells and carotid nerve terminals. The junctions showed a very narrow intercellular space of 2 nm and ranged in length from 200 nm to 1 micron. They may form the morphological substrate for electrical coupling between cells, the occurrence of which has been demonstrated by electrophysiology. Further studies using freeze-fracture and

The role of extracellular matrix in the formation of the sclerotome.

The development of the sclerotome is considered as a model for the formation of mesenchyme from an epithelium. In early epithelial somites, transmission and scanning electron microscopy indicate considerable ultrastructural similarity between the future sclerotome and dermamyotomal regions. Subsequently, these two regions diverge in their development. In the forming dermamyotome, junctional complexes become more extensive and the cells become elongated, closely applied to each other, and have angular surface contours. In the forming sclerotome, there is an early reduction in apical junctions.

Role of aPKC isoforms and their binding partners Par3 and Par6 in epidermal barrier formation.

The skin water barrier, essential for terrestrial life, is formed by a multilayered stratifying epithelium, which shows a polarized distribution of both differentiation and intercellular junction markers. Recently, several reports showed the crucial importance of tight junctions for the in vivo water barrier function of the skin. In simple epithelial cells, intercellular junction formation is closely coupled to the establishment of polarity. However, if and how polarity proteins contribute to epidermal differentiation and junction formation is not yet known. Here, we have characterized the loc

Expression of claudins, occludin, junction adhesion molecule A and zona occludens 1 in canine organs.

Tight junctions are the outermost structures of intercellular junctions and are classified as transmembrane proteins. These factors form selective permeability barriers between cells, act as paracellular transporters and regulate structural and functional polarity of cells. Although tight junctions have been previously studied, comparison of the transcriptional‑translational levels of these molecules in canine organs remains to be investigated. In the present study, organ‑specific expression of the tight junction proteins, claudin, occludin, junction adhesion molecule A and zona occludens 1 wa

Desmosomes and gap-junctions in various epidermoid preneoplastic and neoplastic lesions of the cervix uteri.

The distribution of desmosomes and of gap-junctions was studied by morphometric means, at the electron microscopic level, in the following lesions of the uterine cervix: metaplasia, moderate and severe dysplasia, carcinoma in situ and invasive epidermoid carcinoma. The results were compared with normal exocervical epithelium. The proportion of the cell surface occupied by gap-junctions was decreased in all lesions; gap-junctions were statistically absent from moderate dysplasia and more advanced lesions. The number of desmosomes decreased gradually from metaplasia to invasive carcinoma, with e

Gap junction protein connexin43 exacerbates lung vascular permeability.

Increased vascular permeability causes pulmonary edema that impairs arterial oxygenation and thus contributes to morbidity and mortality associated with Acute Respiratory Distress Syndrome and sepsis. Although components of intercellular adhesive and tight junctions are critical for maintaining the endothelial barrier, there has been limited study of the roles of gap junctions and their component proteins (connexins). Since connexins can modulate inflammatory signaling in other systems, we hypothesized that connexins may also regulate pulmonary endothelial permeability. The relationships betwe

Shigella flexneri enters human colonic Caco-2 epithelial cells through the basolateral pole.

The commonly accepted view that enteroinvasive bacteria enter cells of the intestinal epithelial lining through the apical surface can be challenged in the case of shigellosis. This study is based on in vitro experiments that showed that the invasion of human colonic Caco-2 cells by Shigella flexneri occurred through the basolateral pole of these cells. In these experiments, the few bacteria that interacted with the apical surface either bound to microvilli of the cell dome without causing detectable alteration or bound at the level of intercellular junctions at which they demonstrated a limit

Gap junctions in the eye: evidence for heteromeric, heterotypic and mixed-homotypic interactions.

Some of the best evidence that different types of gap junction proteins (connexins) interact with each other in vivo has been found in the eye. This review focuses on three diverse ocular tissues that may contain heterotypic or heteromeric gap junction channels. Each of the tissues uses gap junctions in a superlative fashion: The crystalline lens has an exceptionally high density of gap junctions; the ciliary body expresses a surprising variety of connexins; the neural retina shows remarkable specificity in the patterns of intercellular coupling.

Upregulation of the expression of tight and adherens junction-associated proteins during maturation of neonatal pancreatic islets in vitro.

Cell-cell contacts mediated by intercellular junctions are crucial for proper insulin secretion in the endocrine pancreas. The biochemical composition of the intercellular junctions in this organ and the role of junctional proteins in endocrine pancreatic dysfunctions are still unclear. In this study, we investigated the expression and cellular location of junctional and cytoskeletal proteins in cultured neonatal rat pancreatic islets. Neonatal B-cells had an impaired insulin secretion compared to adult cells. Cultured neonatal islets showed a time-dependent increase in the glucose-induced sec

Expression of connexin 43 in breast cancer in comparison with mammary dysplasia and the normal mammary gland.

Gap junctional intercellular communication (GJIC) plays a critical role in tissue development and differentiation and probably in carcinogenesis. The purpose of the study was to evaluate the expression and localisation of Cx43 in 40 cases of mammary dysplasia and 29 cases of breast cancer (without primary chemotherapy). The tissue sections were investigated for Cx43 expression by immunohistochemistry. In the normal mammary gland there was an intercellular, punctate staining pattern, mainly between myoepithelial cells, characteristic of functional gap junctions. In dysplasias there was mainly m

A disease in dogs resembling human pemphigus vulgaris: case reports.

A pemphigus-like disease occurred in 5 dogs. The canine disease closely resembled pemphigus vulgaris in man and was characterized by ulcerative lesions in the oral cavity, on the skin, and at mucocutaneous junctions. Histologically, the first sign of the disease was disappearance of the intercellular bridges in the lower epidermis. The resulting acantholysis was diagnostic for pemphigus. Antibodies to the intercellular space substance were detected by immunofluorescent staining in serum of 4 of the dogs. High doses of corticosteroids were effective in controlling the disease.

Expression of connexin 43 protein in testes, epididymides and prostates of stallions.

REASONS FOR PERFORMING STUDY: Connexin 43 (Cx43) is a ubiquitously distributed gap junction protein in testes and other reproductive tissues. Adjacent cells share ions and small metabolites through intercellular channels, which are present in gap junctions. Previously, Cx43 has not been reported in testes, epididymides and prostates either in healthy stallions or cryptorchid horses. OBJECTIVES: To demonstrate the expression pattern of Cx43 in the reproductive tissues of stallions and examine whether naturally occurring bilateral cryptorchidism has any influence on distribution and expression

Viral Infection and Connexin Dysfunction in the Heart.

PURPOSE OF REVIEW: Gap junctions, comprising connexin proteins, enable the direct intercellular electrical coupling of cardiomyocytes, and disruption of this process is arrhythmogenic. In addition, gap junctions effect metabolic coupling and of relevance to this review, propagate host antiviral immune responses. Accordingly, connexins have emerged as viral targets during infection. This review summarizes current knowledge regarding contributions of inflammation vs virally encoded factors in driving alterations to cardiac gap junction function. RECENT FINDINGS: In addition to host immune-media

Epidermal tight junctions in health and disease.

The skin, the largest organ of the body, is an essential barrier that under homeostatic conditions efficiently protects and/or minimizes damage from both environmental (e.g. microorganisms, physical trauma, ultraviolet radiation) and endogenous (e.g., cancers, inflammation) factors. This formidable barrier function resides mainly in the epidermis, a dynamic, highly-stratified epithelium. The epidermis has 2 major barrier structures: stratum corneum, the outmost layer and tight junctions, intercellular junctions that seal adjacent keratinocytes in the stratum granulosum, found below the stratum

Chorda-evoked opening of tight junctions in rat submandibular salivary acini demonstrated by microperoxidase.

The permeability of these junctions from the interstitium to the lumen was examined by using an ultrastructural tracer, microperoxidase, in conjunction with electron microscopy. In the resting gland, the reaction product of microperoxidase was seen in the interstitial and intercellular spaces, but not within acinar lumina; thus the tight junction was impermeable to microperoxidase (junction closed). Intraductal injection of hypertonic sucrose solution (1000 mOsm; 30 microliters) caused a sustained elevation of the luminal pressure, indicating osmotic water flow into the lumen due to the presen

Do Vascular and Extracellular Measurements Consistently Reflect Intracellular pO2?

Oxygen measurements are routinely made either in the vasculature or in the extracellular fluid surrounding the cells of tissues. Yet, metabolic oxygen availability depends on the pO2 within the cells, as does the enhancing effect of oxygen on radiotherapy outcomes. This article reports quantitative modeling work examining the effect of cellular plasma membrane composition on tissue permeability, as a window into tissue oxygen gradients. Previous application of the model indicates that lipid-mediated diffusion pathways accelerate oxygen transfer from capillaries to intracellular compartments an
