A pair of interneurons influences the choice between feeding and locomotion in Drosophila.

The decision to engage in one behavior often precludes the selection of others, suggesting cross-inhibition between incompatible behaviors. For example, the likelihood to initiate feeding might be influenced by an animal's commitment to other behaviors. Here, we examine the modulation of feeding behavior in the fruit fly, Drosophila melanogaster, and identify a pair of interneurons in the ventral nerve cord that is activated by stimulation of mechanosensory neurons and inhibits feeding initiation, suggesting that these neurons suppress feeding while the fly is walking. Conversely, inhibiting a

The role of interneurons in shaping Purkinje cell responses in the cerebellar cortex.

The well established anatomy of the cerebellar cortex has led to suggestions that cerebellar molecular layer interneurons laterally inhibit Purkinje cells. In support of the anatomical predictions, on-beam excitation and off-beam inhibition of Purkinje cells have been shown to occur when the surface of the cerebellum is electrically excited. Patchy excitation of Purkinje cells with flanking inhibition of sagittally oriented Purkinje cells have also been demonstrated following peripheral stimulation in vivo. To extend these observations, we mapped the functional connectivity between granule cel

[Cytoplasmic syncytial connection--one of three forms of interneuronal connection].

The paper is a review of the current state of the important theoretical problem of neurophysiology about the principal possibility of the existence in the nervous system, apart from the major synaptic and contact connections, also of the cytoplasmic syncytial interneuronal connection. Also analyzed are electron microscopy data and results of observation on behavior of living neurons in tissue culture, which allow suggesting the existence in some loci of the nervous system and under certain conditions of the third form of syncytial interneuronal interactions. Regular signs of syncytial perforat

The fate of the large striatal interneurons expressing calretinin in Huntington's disease.

Huntington's disease (HD) is characterized by the atrophy of the striatum due to losses of projection neurons, while interneurons are relatively spared. However, little is known about the fate of the large interneurons that express calretinin (Cr) in HD. We addressed this issue by applying a double immunofluorescent labeling technique to postmortem striatum from HD patients and controls. We compared the distribution and density of Cr-positive (+) interneurons and their degree of choline acetyltransferase (ChAT) coexpression in normal and HD cases. Large interneurons containing only Cr, ChAT, o

Olfactory interneurons in the brain of the larval sphinx moth Manduca sexta.

1. The physiology and morphology of olfactory interneurons in the brain of larval Manduca sexta were studied using intracellular recording and staining techniques. Antennal olfactory receptors were stimulated with volatile substances from plants and with pure odorants. Neurons responding to the stimuli were investigated further to reveal their response specificities, dose-response characteristics, and morphology. 2. We found no evidence of specific 'labeled-lines' among the odor-responsive interneurons, as none responded exclusively to one plant odor or pure odorant; most olfactory interneuron

Microglial galectin-3 disrupts parvalbumin interneurons and hippocampal synchrony, driving cognitive deficits.

Sepsis-associated encephalopathy (SAE), a devastating neurological complication of systemic inflammation, affects approximately 70% of patients with sepsis. It not only increases mortality but also leaves survivors with persistent cognitive deficits. However, the mechanisms underlying SAE progression remain incompletely understood. Here, using a lipopolysaccharide (LPS)-induced mouse model of SAE, we identify microglial galectin-3 (Gal-3) as a central pathogenic mediator driving systemic inflammation-induced cognitive impairment. Mechanistically, systemic LPS challenge robustly upregulates mic

Exploring Cortical Interneurons in Substance Use Disorder: From Mechanisms to Therapeutic Perspectives.

Interneurons (INs) play a crucial role in the regulation of neural activity within the medial prefrontal cortex (mPFC), a brain region critically involved in executive functions and behavioral control. In recent preclinical studies, dysregulation of INs in the mPFC has been implicated in the pathophysiology of substance use disorder, characterized by vulnerability to chronic drug use. Here, we explore the diversity of mPFC INs and their connectivity and roles in vulnerability to addiction. We also discuss how these INs change over time with drug exposure. Finally, we focus on noninvasive brain

Targeted neuromodulation of spinal interneurons enhances breathing in chronic spinal cord injury.

Respiratory dysfunction is a debilitating consequence of cervical spinal cord injury (cSCI) with few available treatment options. Restoring function in the chronic phase is challenging due to the limited regenerative capacity of the adult central nervous system. This study investigates the targeted neuromodulation of cervical excitatory interneurons (eINs) to improve functional respiratory recovery in chronic cSCI. Cervical eINs, crucial for acute injury recovery, showed no reduction in numbers at 8- and 12-weeks post injury, highlighting their potential as neuromodulatory targets in the chron

Classification and analysis of retinal interneurons by computational structure under natural scenes.

Inhibitory neurons are diverse across the brain, but for the visual system we lack the ability to functionally classify these neurons under complex natural stimuli. Here we take the approach of classifying retinal amacrine cell responses to natural scenes using optical recording and an interpretable neural network model. We fit mouse amacrine cell responses to a two-layer convolutional neural network model of a class shown previously to accurately capture salamander ganglion cell responses to natural scenes. Using an approach from interpretable machine learning, we determined for each stimulus

Adult trkB Signaling in Parvalbumin Interneurons is Essential to Prefrontal Network Dynamics.

Inhibitory interneurons expressing parvalbumin (PV) are central to cortical network dynamics, generation of γ oscillations, and cognition. Dysfunction of PV interneurons disrupts cortical information processing and cognitive behavior. Brain-derived neurotrophic factor (BDNF)/tyrosine receptor kinase B (trkB) signaling regulates the maturation of cortical PV interneurons but is also implicated in their adult multidimensional functions. Using a novel viral strategy for cell-type-specific and spatially restricted expression of a dominant-negative trkB (trkB.DN), we show that BDNF/trkB signaling i

Presynaptic GABAB receptors functionally uncouple somatostatin interneurons from the active hippocampal network.

Information processing in cortical neuronal networks relies on properly balanced excitatory and inhibitory neurotransmission. A ubiquitous motif for maintaining this balance is the somatostatin interneuron (SOM-IN) feedback microcircuit. Here, we investigated the modulation of this microcircuit by presynaptic GABAB receptors (GABABRs) in the rodent hippocampus. Whole-cell recordings from SOM-INs revealed that both excitatory and inhibitory synaptic inputs are strongly inhibited by GABABRs, while optogenetic activation of the interneurons shows that their inhibitory output is also strongly supp

Cell type-specific expression of acid-sensing ion channels in hippocampal interneurons.

Acid-sensing ion channels (ASICs), a member of the degenerin/epithelial Na+ channel superfamily, are widely expressed in the mammalian CNS. Accumulating evidence suggests that ASIC current density is higher in GABAergic interneurons than that in glutamatergic pyramidal neurons (PNs) in the hippocampus. Such differential expression of ASICs in cortical networks is thought to be a key element for seizure termination. However, GABAergic interneurons are highly diverse; it is unclear whether the functional expression of ASICs differs in distinct GABAergic interneuron subtypes. Moreover, the subuni

Subtypes of substance P receptor immunoreactive interneurons in the rat basolateral amygdala.

Local injections of the neurotoxin SP-saporin into the basolateral amygdala (BLA) are reported to specifically lesion substance P receptor immunoreactive (SPR-IR) interneurons, and to reduce anxiety related behavior. Hence, this technique might provide a means to study how defined interneuron populations regulate neuronal activity in the BLA. However, what interneuron subgroups in the BLA might be targeted by SP-saporin lesions has not been established. This study has used dual-labeling immunofluorescence in the rat BLA to examine SPR-IR neurons for their colocalization with the calcium-bindin

Regulation of excitatory neural input to longitudinal intestinal muscle by myenteric interneurons.

The circuit of myenteric interneurons that regulate excitatory input to longitudinal colonic muscle was identified using dispersed ganglia and longitudinal muscle strips with adherent myenteric plexus from rat distal colon. The preparations enabled measurement of neurotransmitter release from interneurons and/or excitatory motoneurons innervating longitudinal muscle. 1, 1-Dimethyl-4-phenylpiperizinium (DMPP) and somatostatin were used to activate myenteric neurons in dispersed ganglia and muscle strips, respectively. DMPP-stimulated vasoactive intestinal peptide (VIP) release in dispersed gang

Differential Cortical and Thalamic Engagement of Cholinergic Interneurons in the Nucleus Accumbens Core.

Cholinergic interneurons (CINs) in the nucleus accumbens (NAc) play a key role in regulating motivated behaviors. Here we examine the connectivity and functional impact of cortical and thalamic inputs onto CINs in the NAc core of mice of either sex. We first use cell type-specific retrograde anatomy to identify the prefrontal cortex and thalamus as putative afferents. We then combine ex vivo slice physiology and optogenetics to characterize the properties of synapses onto CINs. We demonstrate that thalamic inputs strongly facilitate, whereas cortical inputs exhibit marked depression. We also s

Rapid and Robust Generation of Human Cortical Interneurons from Induced Neural Stem Cells.

Current protocols for generating cortical interneurons from human pluripotent stem cells are hindered by slow differentiation kinetics and poor reproducibility across cell lines. Here, we present a defined small-molecule-based strategy that efficiently directs human-induced neural stem cells (hiNSCs) toward cortical GABAergic interneurons within 14-18 days, which is substantially faster than conventional methods. Short-term dual-SMAD and WNT inhibition rapidly commits hiNSCs to an interneuron progenitor fate, reaching transcriptional states equivalent to those obtained with prolonged protocols

Electroacupuncture Attenuates Cyclophosphamide-Induced Bladder Pain by Restoring Autophagy in Spinal Inhibitory Interneurons.

Interstitial cystitis/bladder pain syndrome is characterized by chronic pelvic pain and urinary symptoms. Autophagy and oxidative stress are essential to neuronal homeostasis, and their dysregulation in spinal circuits may contribute to persistent pain. While electroacupuncture (EA) has been reported to alleviate cystitis-related pain, the associated cellular changes in the spinal dorsal horn remain incompletely understood. In this study, we used the cyclophosphamide (CYP)-induced cystitis rat model to investigate the cellular and molecular mechanisms underlying the analgesic effects of EA. Be

T Cell-Mediated Targeting of Interneurons in Mice Shapes Hippocampal Remodeling and Epilepsy.

OBJECTIVE: Autoimmune encephalitis (AE) is associated with autoantibodies targeting distinct neuronal populations. In AE, antibodies against glutamate decarboxylase 65 (GAD65), expressed in GABAergic interneurons, are frequently detected. In GAD65-AE, hippocampal biopsies often show infiltrates of CD8+ cytotoxic T cells (CTLs), suggesting a prominent T cell-associated pathology. This has led to the hypothesis that CTLs contribute to neuronal injury in GABAergic circuits. We investigated whether selective CTL-mediated dysfunction of hippocampal interneurons may contribute to temporal lobe epile

Reduced inhibition of hippocampal adult-born granule cells by parvalbumin interneurons after TBI.

Traumatic brain injury (TBI) is one of the leading causes of acquired temporal lobe epilepsy. TBI drives hippocampal circuit rearrangements that may contribute to increased seizure risk, such as altered inhibitory circuit function and aberrant post-traumatic neurogenesis. In the hippocampal dentate gyrus, adult-born dentate granule cells (DGCs) acquire inhibitory synaptic inputs from parvalbumin-expressing (PV) interneurons early in their maturation. These inputs are important for circuit integration and feedforward inhibition of these neurons. To test whether DGCs born after TBI have function
