Trophoblast metabolic reprogramming triggers adverse pregnancy outcomes during DENV-2 infection in mice.

Adverse pregnancy outcomes associated with dengue virus (DENV) infection are a severe manifestation of dengue fever; however, we still poorly understand the underlying mechanisms. Trophoblast dysfunction in the placenta contributes centrally to various pregnancy complications. As the functional unit of the placenta, the syncytiotrophoblast (STB) requires continuous fusion of precursor trophoblasts and adequate energy supply. In this study, we established a model of adverse pregnancy by infecting Ifnar1-/- pregnant mice with DENV-2. We did not detect viral replication within the placental troph

Risk factors and rechallenge outcomes in severe immune-mediated hepatotoxicity: Real-world evidence from two high-volume centers in China.

BACKGROUND AND AIMS: Immune checkpoint inhibitors (ICIs) improve cancer outcomes but can lead to immune-mediated hepatotoxicity (IMH). Real-world data on severe IMH remain limited. This study aimed to identify risk factors and evaluate clinical outcomes, including the safety of ICIs rechallenge. METHODS: This retrospective dual-center study included patients who developed IMH after ICIs therapy at two high-volume hospitals in China. Univariable and multivariable logistic regression were used to identify independent risk factors. The timing of onset was assessed by treatment cycles, and outcom

Subphenotypes of SARS-CoV-2-Associated ARDS Overlap Each Other: A Retrospective Analysis.

Background  Severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) virus-associated pneumonia and acute respiratory distress syndrome (ARDS) were often associated with hyperinflammation and elevation of several serum inflammatory markers but usually less than what is observed in non-coronavirus disease (COVID) ARDS. Elevated inflammatory markers such as C-reactive protein, interleukin (IL)-6, etc., are associated with severe infection. This study identified subphenotypes of COVID-19 ARDS patients by latent profile analysis in a cohort of Indian patients. Methods  Data of n  = 233 adult I

Streptococcus pyogenes and EBV coinfection in severe adult meningoencephalitis: a rare diagnosis in a diabetic patient.

BACKGROUND: Intracranial infection caused by Streptococcus pyogenes is extremely rare in adults. We report a case of Streptococcus pyogenes combined with Epstein-Barr virus (EBV) infection in cerebrospinal fluid (CSF), which was diagnosed by targeted next-generation sequencing (tNGS). The findings were subsequently confirmed by capillary electrophoresis (CE) and quantitative real-time PCR (qPCR), respectively. Meanwhile, digital PCR (dPCR) detection indicated the presence of Streptococcus species DNA in the blood. Brain magnetic resonance imaging (MRI) suggested the possibility of meningitis.

Clinical characteristics and laboratory findings in Danish children hospitalized with primary Epstein-Barr virus infection.

BACKGROUND: Epstein-Barr virus (EBV) positive infectious mononucleosis (IM) is a common disease in adolescents. However, IM is often considered a rare disease in early childhood. We aimed to describe the classical presentation of adolescent EBV-associated IM compared to EBV infection at younger age. METHODS: All immunocompetent children hospitalized at Hvidovre University Hospital, Copenhagen between 2002 and 2013, who presented with clinical features that prompted a laboratory test for EBV, and who tested positive by presence of EBV-specific antibodies, heterophile antibodies or a positive E

Serum levels of soluble vascular cell adhesion molecules may correlate with the severity of dengue virus-1 infection in adults.

The damage of vascular endothelial cells has been speculated to be involved in the pathogenesis of dengue virus (DENV) infection. However, little is known about the role of soluble vascular cell adhesion molecule-1 (sVCAM-1) in predicting the severity of dengue infection in adults. In this study, 51 adults with DENV-1 infection (21 with severe dengue and 30 with dengue fever (DF) were included, and their serum levels of sVCAM-1 and other parameters were determined. The results indicated that the levels of sVCAM-1 were elevated on days 1-3 to 16.75 (11.55-34.74) ng/mL in the severe dengue patie

Facial herpes zoster complicated by varicella zoster virus (VZV) encephalitis: The diagnostic significance of atypical lymphocytes in cerebrospinal fluid (CSF).

BACKGROUND: In general, viral infections of the central nervous system (CNS) manifest as encephalitis and, less commonly, as meningoencephalitis or aseptic meningitis. Varicella zoster virus (VZV) is an uncommon cause of encephalitis. METHODS: Herpes zoster (shingles) is a cutaneous reactivation of previous chickenpox infection due to VZV. Herpes zoster may be dermatomal (ie, <3 dermatomes) or disseminated (ie, >3 dermatomes). Decreased cell-mediated immunity from stress, steroids, or immunosuppressive drugs often precede dermatomal/disseminated herpes zoster. With herpes zoster, the closer t

Reduction of serum interferon (IFN)-gamma concentration and lupus development in NZBxNZWF(1)mice by lactic dehydrogenase virus infection.

The complexity of cytokine regulation and the imbalance of helper T (Th)1 and Th2 subsets in systemic lupus erythematosus (SLE) animal models and human SLE are well recognized. In this study in NZBxNZWF(1)mice, the effects of lactic dehydrogenase virus (LDV) infection on the production of interferon (IFN)-gamma in the serum and the development of autoimmune disease were examined. The progress of the disease (the development of glomerulonephritis, formation of glomerular IgG and C3 deposits, increase in the blood urea nitrogen values, and mortality) was parallel with an increase in serum IFN-ga

In Vitro Human Placental Studies to Support Adenovirus-Mediated VEGF-DΔNΔC Maternal Gene Therapy for the Treatment of Severe Early-Onset Fetal Growth Restriction.

Severe fetal growth restriction (FGR) affects 1 in 500 pregnancies, is untreatable, and causes serious neonatal morbidity and death. Reduced uterine blood flow (UBF) is one cause. Transduction of uterine arteries in normal and FGR animal models using an adenovirus (Ad) encoding VEGF isoforms increases UBF and improves fetal growth in utero. Understanding potential adverse consequences of this therapy before first-in-woman clinical application is essential. The aims of this study were to determine whether Ad.VEGF-DΔNΔC (1) transfers across the human placental barrier and (2) affects human place

Circulating Exosomes Drive Persistent Neuronal Dysfunction in Post-Herpetic Neuralgia Patients.

OBJECTIVE: Post-herpetic neuralgia (PHN) is a debilitating chronic pain condition persisting beyond 3 months after herpes zoster (HZ), yet the mechanisms driving the transition from acute viral injury to chronic neuropathic pain remain poorly characterized. We tested whether circulating exosomes are sufficient to drive sensory neuron dysfunction independent of direct varicella zoster virus infection. METHODS: Human nociceptive neurons were either directly infected with varicella zoster virus or exposed to exosomes isolated from sera of control individuals, patients with acute HZ, or patients
