Potential Application and Bactericidal Mechanism of Lactic Acid-Hydrogen Peroxide Consortium.

It has been found that lactic acid and hydrogen peroxide (H2O2) displayed co-operatively enhanced killing activity to pathogens. The synergistic effect was investigated with using several microbe species, suggesting that low concentration of lactic acid and H2O2 could kill both Gram-negative and Gram-positive bacteria or even fungal pathogens. To explore the mechanism of synergistic sterilization of lactic acid and H2O2, Escherichia coli DH5α was used as the indicator bacteria. Lactic acid and H2O2 could generate hydroxyl radicals depending on the intracellular iron ions. The genomic DNA of tr

A Lactic Acid Bacteria (LAB)-Based Vaccine Candidate for Human Norovirus.

Human noroviruses (HuNoVs) are responsible for more than 95% of the non-bacterial acute gastroenteritis epidemics in the world. The CDC estimates that every year 21 million individuals suffer from HuNoV-induced gastroenteritis in the United States. Currently, there is no FDA-approved vaccine for HuNoVs. Development of an effective vaccine has been hampered by the lack of an efficient cell culture system for HuNoVs and a suitable small animal model for pathogenesis study. In this study, we developed lactic acid bacteria (LAB) as a vector to deliver HuNoV antigen. A LAB strain (Lactococcus lacti

Hydrogenation of Lactic Acid to 1,2-Propanediol over Ru-Based Catalysts.

The catalytic hydrogenation of lactic acid to 1,2-propanediol with supported Ru catalysts in water was investigated. The influence of catalyst support (activated carbon, γ-Al2O3, SiO2, TiO2, and CeO2) and promoters (Pd, Au, Mo, Re, Sn) on the catalytic performance was evaluated. Catalytic tests revealed that TiO2 yields the best Ru catalysts. With a monometallic Ru/TiO2 catalyst, a 1,2-propanediol yield of 70 % at 79 % lactic acid conversion was achieved at 130 °C after 20 h reaction. Minor byproducts of the hydrogenation reaction were propionic acid, ethanol, 1-propanol, and 2-propanol. For t

Metabolic acidosis in short bowel syndrome: think D-lactic acid acidosis.

Short bowel syndrome (SBS) is a condition when a person's gastrointestinal function is insufficient to supply the body with essential nutrients and hydration. Patients with SBS suffer from diarrhoea and symptoms of malabsorption such as weight loss, electrolyte disturbances and vitamin deficiencies. Long-term management of this condition can be complicated by the underlying disease, the abnormal bowel function and issues related to treatment like administration of parenteral nutrition and the use of a central venous catheter. Here, we describe a case of D-lactic acid acidosis, a rarer complica

Metabolic engineering of Bacillus subtilis for production of D-lactic acid.

Poly lactic acid (PLA) based plastics is renewable, bio-based, and biodegradable. Although present day PLA is composed of mainly L-LA, an L- and D- LA copolymer is expected to improve the quality of PLA and expand its use. To increase the number of thermotolerant microbial biocatalysts that produce D-LA, a derivative of Bacillus subtilis strain 168 that grows at 50°C was metabolically engineered. Since B. subtilis lacks a gene encoding D-lactate dehydrogenase (ldhA), five heterologous ldhA genes (B. coagulans ldhA and gldA101, and ldhA from three Lactobacillus delbrueckii) were evaluated. Lactic acid bacterial extracts as anti-Helicobacter pylori: a molecular approach.

BACKGROUND: Helicobacter pylori (H. pylori) infection, the main cause of chronic gastritis, increases gastric cancer risk. Antibiotics-based H. pylori eradication treatment is 90% effective. However, it is expensive and causes side effects and antibiotic resistance. Lactic acid bacteria (LAB) could present a low-cost, large-scale alternative solution to prevent or decrease H. pylori colonization. AIM: This work aimed to study the inhibitory effects of LAB strains on the growth and pathogenic activity of H. pylori stains. To this end, we have selected the most virulent H. pylori strains (out o

Potential of wine-associated lactic acid bacteria to degrade biogenic amines.

Some lactic acid bacteria (LAB) isolated from fermented foods have been proven to degrade biogenic amines through the production of amine oxidase enzymes. Since little is known about this in relation to wine micro-organisms, this work examined the ability of LAB strains (n=85) isolated from wines and other related enological sources, as well as commercial malolactic starter cultures (n=3) and type strains (n=2), to degrade histamine, tyramine and putrescine. The biogenic amine-degrading ability of the strains was evaluated by RP-HPLC in culture media and wine malolactic fermentation laboratory

[Evaluation of group B streptococcus susceptibility to lactic acid bacteria strains].

Streptococcus agalactiae (Group B streptococci--GBS) is one of the coexistent components of vaginal microflora. Lactic Acid Bacteria (LAB) are the predominant bacteria in the physiological flora of vagina. The production of antagonistic substances like short chain fatty acids, hydrogen peroxide, bacteriocin-like substances protects against multiplication of pathogenic microorganisms. However, during pregnancy there is a disturbance of vaginal microflora and there are optimal conditions for bacterial or candidal vaginitis. Despite sufficient protection of LAB during pregnancy, Streptococcus aga

Lactic acid bacteria-enclosing poly(epsilon-caprolactone) microcapsules as soil bioamendment.

Free plant growth-promoting bacteria in soil bioamendments (SBA) are easily outnumbered by competitors and predators in agricultural soils. Microencapsulation of the bacteria is an effective technique that provides a suitable microenvironment for their survival. In this study, we attempted to prepare poly(epsilon-caprolactone) (PCL) microcapsules enclosing lactic acid bacteria (LAB), a plant growth-promoting bacteria, using the solvent-evaporation method via water-in-oil-in-water (W/O/W) emulsion. Three preparation parameters in the emulsion system were optimized based on the lactic acid produ

Poly-L-lactic acid: a new dimension in soft tissue augmentation.

Poly-L-lactic acid (PLLA) is a biodegradable, synthetic polymer of L-lactic acid that has been used in a variety of human medical applications for over 40 years. It has recently been approved in the United States for the treatment of HIV-associated facial lipoatrophy. This indication, combined with a European experience that indicates a clinical benefit persisting for up to 2 years, has made PLLA an increasingly popular injectable soft tissue filler in healthy patients. However, controlled clinical studies are necessary to determine whether PLLA benefits healthy patients seeking contour restor

Poly-L-lactic acid: a temporary filler for soft tissue augmentation.

A number of soft tissue filling agents are available in the U.S. and Europe. In Europe, poly-L-lactic acid was approved for soft tissue augmentation and correction of wrinkles in 1999. The clinical results from this synthetic implant may last up to two years. In this article, we report our experience using poly-L-lactic acid for correction of facial lipoatrophy and wrinkles in 300 patients since 1999. Poly-L-lactic acid is an ideal, resorbable filler material for soft tissue augmentation, especially in the lower one-half of the face. The aesthetic results are excellent and long-lasting. Clinic

Encapsulated lactic acid bacteria for control of malolactic fermentation in wine.

The kinetics of both malolactic fermentation in Chardonnay wine by encapsulating Lactobacillus casei cells in pectate gel and lyophilized Oenococcus oeni culture has been carried out. The influence of acidity, sulfur dioxide content, and organic acid content on the malolactic activity of the bacteria has been controlled. Encapsulated bacteria degraded 30%, of malic acid in white wine, deacidifying it from pH 3.15 to 3.40, whereas the lyophilized culture degraded 48% of malic acid, deacidifying from pH 3.15 to 3.60. The degree of conversion of malic acid in wine by the encapsulated cells was tw

Continuous lactic acid fermentation using a plastic composite support biofilm reactor.

An immobilized-cell biofilm reactor was used for the continuous production of lactic acid by Lactobacillus casei subsp. rhamnosus (ATCC 11443). At Iowa State University, a unique plastic composite support (PCS) that stimulates biofilm formation has been developed. The optimized PCS blend for Lactobacillus contains 50% (wt/wt) agricultural products [35% (wt/wt) ground soy hulls, 5% (wt/wt) soy flour, 5% (wt/wt) yeast extract, 5% (wt/wt) dried bovine albumin, and mineral salts] and 50% (wt/wt) polypropylene (PP) produced by high-temperature extrusion. The PCS tubes have a wall thickness of 3.5 m

Peritoneal fluid lactic acid and diagnostic dilemmas in acute abdominal disease.

Our previous canine research suggested that the determination of peritoneal fluid lactic acid levels may be helpful in the evaluation of potential acute abdomen cases. To investigate the clinical significance of those findings, we obtained simultaneous peritoneal and plasma lactic acid values from patients undergoing emergency celiotomy or in whom surgical consultation was sought to rule out an acute abdomen. The lactic acid value was significantly higher in peritoneal fluid than in plasma in patients who were found to have hollow viscus perforation, gangrenous intestine, peritonitis, or intra

In-vivo degradation of poly(lactic acid) of different molecular weights.

Lactic acid was polymerized using tetraphenyl tin as a catalyst. The molecular weight of the resultant poly(lactic acid) varied between 0.89 X 10(6) and 2.94 X 10(6) depending upon the concentration of the catalyst used. In-vivo degradation of the poly(lactic acid) samples having 4 different molecular weights were studies by implanting these in Wistar rats. It was found that at the end of a 48-week implantation period lower molecular weight poly(lactic acid) samples were degraded at a faster rate than the higher molecular weight samples.

Lactic acid levels in pleural fluid from patients with bacterial pleuritis.

Pleural fluid lactic acid estimations were carried out on 60 samples by gas-liquid chromatography. Lactic acid levels in 12 patients with bacterial pleural infection were statistically significantly higher (mean, 287 mg/dl; range, 135 to 482 mg/dl) than in 18 patients with malignancy (mean, 71 mg/dl; range, 24 to 157 mg/dl) and 30 other patients with pleural effusions (mean, 19 mg/dl; range, 10 to 57 mg/dl). Effect of ethanol on lactic acid production by exercised normal muscle.

Several studies have suggested that muscle lactic acid production is defective in alcoholic myopathy. However, normal controls have not been evaluated. To study the effects of ethanol on the production of lactic acid in normal exercised muscle, oral ethanol (1 g per kg body weight, 20% v/v) was given to eight nonalcoholic subjects. Forearm ischemic exercise with a total work load of 7.2 kg-m/min was performed just before and 90 min after ingestion of ethanol. At the time of exercise, the serum ethanol level was 95.7 +/- 15.1 mg% (mean +/- SD). Resting serum lactic acid was significantly higher
