Differential mechanisms of SARS-CoV-2 inactivation by anionic surfactants: a comparative study of fatty acid salts and synthetic surfactants.

Despite advances in COVID-19 vaccines and antivirals, SARS-CoV-2 remains a significant public-health threat. While handwashing helps prevent infection, the anti-SARS-CoV-2 activity of anionic surfactants in hand soaps has not been systematically evaluated. In this study, we compared five surfactants: potassium oleate (C18:1-K), potassium myristate (C14:0-K), potassium laurate (C12:0-K), sodium laureth sulfate (SLES), and sodium dodecyl sulfate (SDS). Viral infectivity assays demonstrated that C18:1-K exhibited remarkable efficacy, reducing SARS-CoV-2 infectivity by more than 10⁵-fold at a conc

Causal relationship between gut microbiota, metabolites, and short stature: a Mendelian randomization study.

BACKGROUND: Previous evidence suggests close relationships between the gut microbiota and short stature, but the causal relationship between them remains unclear. Our study performed Mendelian randomization (MR) analysis to investigate the causal relationships between gut microbiota, blood metabolites, and short stature, and to identify the potential role of blood metabolites as mediators. METHODS: We extracted summary-level data for 119 genera gut microbiota, 309 blood metabolites, and short stature from published genome-wide association studies (GWASs). We applied two-sample MR to infer the

Safety and efficiency of modulating paracellular permeability to enhance airway epithelial gene transfer in vivo.

We evaluated the safety of agents that enhance gene transfer by modulating paracellular permeability. Lactate dehydrogenase (LDH) and cytokine release were measured in polarized primary human airway epithelial (HAE) cells after lumenal application of vehicle, ethyleneglycol-bis-(beta-aminoethyl ether)-N,N'-tetraacetic acid (EGTA), sodium caprate (C10), or sodium laurate (C12). Lung toxicity was assessed after tracheobronchial instillation to murine airways and the relative ability of these agents to enhance in vivo adenoviral gene transfer was evaluated. Lumenal C12 increased LDH release in vi

Bovine tumor necrosis factor-alpha Increases IL-6, IL-8, and PGE2 in bovine fibroblast-like synoviocytes by metabolic reprogramming.

Lameness is a common condition in dairy cattle caused by infectious or noninfectious agents. Joint lesions are the second most common cause of lameness and can be diagnosed in association with the presentation of digit injuries. Fibroblast-like synoviocyte (FLS) are predominant cells of synovia and play a key role in the pathophysiology of joint diseases, thus increasing the expression of proinflammatory mediators. Tumor necrosis factor-alpha (TNF-α) is a potent proinflammatory cytokine involved in cyclooxygenase 2 (COX-2) and proinflammatory cytokine expression in FLS. Previously, TNF-α was d

Differential effect of saturated and unsaturated free fatty acids on the generation of monocyte adhesion and chemotactic factors by adipocytes: dissociation of adipocyte hypertrophy from inflammation.

OBJECTIVE: Obesity is associated with monocyte-macrophage accumulation in adipose tissue. Previously, we showed that glucose-stimulated production by adipocytes of serum amyloid A (SAA), monocyte chemoattractant protein (MCP)-1, and hyaluronan (HA) facilitated monocyte accumulation. The current objective was to determine how the other major nutrient, free fatty acids (FFAs), affects these molecules and monocyte recruitment by adipocytes. RESEARCH DESIGN AND METHODS: Differentiated 3T3-L1, Simpson-Golabi-Behmel syndrome adipocytes, and mouse embryonic fibroblasts were exposed to various FFAs (

Sphingomyelinase D activity in model membranes: structural effects of in situ generation of ceramide-1-phosphate.

The toxicity of Loxosceles spider venom has been attributed to a rare enzyme, sphingomyelinase D, which transforms sphingomyelin to ceramide-1-phosphate. The bases of its inflammatory and dermonecrotic activity, however, remain unclear. In this work the effects of ceramide-1-phosphate on model membranes were studied both by in situ generation of this lipid using a recombinant sphingomyelinase D from the spider Loxosceles laeta and by pre-mixing it with sphingomyelin and cholesterol. The systems of choice were large unilamellar vesicles for bulk studies (enzyme kinetics, fluorescence spectrosco

Molecular docking, Group Interaction Contribution (GIC), and ADMET studies on the role of surfactants in improving the permeability and solubility of mitoxantrone.

CONTEXT: Mitoxantrone (MTX) is an anticancer drug characterized by its hydrophobicity and low bioavailability, which can be increased by adding adjuvants to the formulation, such as surfactants (SFT). They can interact effectively with drugs, improve solubility and permeability, and stabilize them, thanks to unique properties such as amphiphilicity, adsorption, and micellization. METHODS: This novel in silico study explores the intermolecular interactions between 128 surfactants and the chemotherapeutic anticancer agent "Mitoxantrone", using molecular docking and the GIC approach. First, the

Human Serum Albumin (HSA) Suppresses the Effects of Glycerol Monolaurate (GML) on Human T Cell Activation and Function.

Glycerol monolaurate (GML) is a monoglyceride with well characterized anti-microbial properties. Because of these properties, GML is widely used in food, cosmetics, and personal care products and currently being tested as a therapeutic for menstrual associated toxic shock syndrome, superficial wound infections, and HIV transmission. Recently, we have described that GML potently suppresses select T cell receptor (TCR)-induced signaling events, leading to reduced human T cell effector functions. However, how soluble host factors present in the blood and at sites of infection affect GML-mediated

In vitro evaluation of viability, integrity, and inflammation in genital epithelia upon exposure to pharmaceutical excipients and candidate microbicides.

The use of microbicides is a promising approach for the prevention of HIV-1 transmission. Unfortunately, various candidates failed in clinical trials. In some cases, the candidate microbicide even resulted in enhanced virus transmission. Therefore, there is an urgent need to develop more predictive preclinical strategies to anticipate the in vivo efficiency/toxicity rate, including in vitro assays that evaluate effects on epithelial integrity and inflammation. The present study aims to identify potential safety issues concerning the use of microbicides and excipients commonly used in vaginal m
