One-step Access to Resorcinsalens-Solvent-Dependent Synthesis, Tautomerism, Self-sorting and Supramolecular Architectures of Chiral Polyimine Analogues of Resorcinarene.

Substituted 2,4- and 4,6-dihydroxyisophthalaldehydes were condensed with optically pure and racemic trans-1,2-diaminocyclohexane to form resorcinarene-like polyimine macrocycles (resorcinsalens), the structure and stoichiometry of which were controlled by the choice of the reaction medium. Particularly, the cyclocondensation reactions were driven by the solubility, tautomerization, or by social self-sorting. The resorcinsalens crystallized as inclusion compounds, in which the guest molecules were situated either in channels or in voids. In the highly hydrated crystals of one of the [2+2] macro

Electrophysiological identification of 4 macrocyclic lactones as female-specific volatiles of the agarwood tree defoliator Heortia vitessoides (Lepidoptera: Crambidae).

Agarwood trees (Aquilaria spp.) are widely cultivated in tropical Asia for their valuable resin. The defoliator moth Heortia vitessoides Moore (Lepidoptera: Crambidae) is a devastating pest that significantly limits the productivity of agarwood plantations. Sex pheromones offer a sustainable and efficient approach for monitoring and controlling this pest. In this study, we report the identification of female-specific volatiles (FVs) as candidate sex pheromones produced by the female of H. vitessoides. The FVs were identified and extracted from the 3rd to 5th abdominal segments of the calling f

A New Dy(III) Complex: Fluorescence Performances, Loading with Resveratrol-Hydrogels Against Skin Aging and Molecular Docking.

In the present research, novel lanthanide coordination compounds [DyL(PhCOO)(CH3OH)](ClO4)2·(CH3OH)2 (1) were characterized by the compression of 2,6-diformyl-4-methyl-phenol (dmp) and 1,3-diamino-2-propanol using benzoate as the secondary ligand, where L indicates the deprotonated macrocyclic ligand. Through the high structural rigidity driven by the coordination of the macrocyclic ligand formed by condensation in methanol solution and sodium benzoate with Dy(ClO4)3·6H2O, compound 1 exhibits outstanding cyan-emitting fluorescence performance and potential applications as a fluorescent materia

Study on enantiomeric separation of basic drugs by NACE in methanol-based medium using erythromycin lactobionate as a chiral selector.

A wide variety of chiral selectors have been employed in CZE, and among them macrocyclic antibiotics including glycopeptides, ansamycins, aminoglycosides and polypeptides exhibited prominent enantioselective properties toward abundant racemic compounds. Compared with CZE, the use of macrocyclic antibiotics as chiral selectors in NACE has not been reported previously. In this study, an approach to the enantioseparation of basic drugs by means of NACE with erythromycin lactobionate (EL) belonging to the group of macrolide antibiotics has been investigated. Especially different from the above fou

Dual templated synthesis of silver acetylide cluster-encapsulated supramolecular boxes.

A dual-templated approach for the controllable synthesis of metal cluster complexes is described. By using an acetylide-containing anion with specific geometry as a central template and a macrocyclic coordinative compound as a peripheral one, two multinuclear silver-acetylide cluster-encapsulated supramolecular boxes were synthesized.

Nitrene-Transfer Chemistry to Olefins Mediated by Co(III) Catalysts Supported by a Tetraamido Macrocycle.

A tetraamido macrocyclic ligand scaffold (L1H4) has been synthesized and metalated with Co(III) to afford anionic metal complexes [CoIII(L1)]- (cations: K+, PPh4+) that are paramagnetic (S = 1) in solution and display semi-reversible reduction waves (CoII/III) and irreversible anodic events in cyclic voltammograms. These macrocyclic Co(III) compounds have been examined as nitrene-transfer catalysts, involving iminoiodinane precursors (PhI=NR) and olefins, to reveal efficient aziridinations and/or allylic aminations, especially with sterically unencumbered styrenes. Mechanistic studies indicate

A unified deep generative framework for surrogate-guided molecular discovery across diverse molecular spaces.

A unified deep generative framework for surrogate-guided molecular discovery across diverse molecular spaces is presented to connect forward property prediction with property-oriented candidate generation. A high-accuracy surrogate predictor based on message-passing neural networks is decoupled from heterogeneous generators, including sequence models and diffusion models, and integrated into a closed-loop strategy of generate, score, select, and regenerate to progressively enrich high-performing candidates without modifying the underlying model architectures. The framework is evaluated on thre

De Novo Boronic Acid-Containing Macrocyclic Peptides That Selectively Bind a Sialylated N-Glycan.

Glycan-mediated molecular recognition is central to many diseases, yet designing selective synthetic glycan binders is difficult due to the limited functional group diversity of carbohydrates. Boronic acids can form reversible covalent bonds with cis-diols but typically lack selectivity. To address this, we developed conformationally constrained macrocyclic peptides incorporating boronic acid-containing amino acids using genetic code reprogramming and the RaPID system. Screening against the N-linked biantennary sialylated glycan N-A2G2S2 yielded de novo macrocycles with single-digit micromolar

Single-Molecule Macrocyclic Carrier Overcomes the Direct Transmembrane of Medium-Sized Cargoes.

Direct transmembrane transport plays an important role in the biomedical field. Although well-established methods exist for the direct membrane transport of small-sized cargoes (e.g., water and ions) and large-sized cargoes (e.g., proteins, DNAs, polysaccharides), they are challenging to directly apply to medium-sized cargoes (e.g., drugs, probes, oligopeptides, and growth hormones). Here, we present a negatively charged single molecular carrier, carboxyl-modified azocalix[6]arene (CAC6A), as a direct transmembrane tool that facilitates the transport of medium-sized cargoes. The cellular uptak

Macrocyclic skeletal modification approach to the anti-trypanosomal macrolides, actinoallolides.

Herein, we report the construction of 16-membered macrocycles that were designed as intermediates toward the unified synthesis of actinoallolides. Key to our synthesis was the use of Mitsunobu macrocyclization, followed by a sequence of Birch reduction and oxidative C-C cleavage to edit the macrocycle.

Syntheses and investigation of metal complexes with macrocyclic polythioether ligands.

Copper(I) complexes with the macrocyclic thioether ligands 1,4,8,11-tetrathiacyclotetradecane (tetrathiacyclam, 14-S4) and 1,8-dithia-4,11-diazacyclotetradecane (dithiacyclam, 14-N2S2) were synthesised and structurally characterised. While the copper(I) complexes showed no reactivity towards dioxygen, the formation of "dioxygen adduct complexes" could be spectroscopically detected with ozone using low temperature stopped-flow techniques. Furthermore, it was possible to synthesise and characterise iron(II) and cobalt(II) complexes with the tetrathiacyclam ligand. No "dioxygen adduct" intermedia

Size-selective recognition by a tubular assembly of phenylene-pyrimidinylene alternated macrocycle through hydrogen-bonding interactions.

Study of artificial tubular assemblies as a useful host scaffold for size-selective recognition and release of guest molecules is an important subject in host-guest chemistry. We describe well-defined self-assembled nanotubes (NT6mer) formed from π-conjugated m-phenylene-pyrimidinylene alternated macrocycle 16mer that exhibit size-selective recognition toward a specific aromatic acid. In a series of guest molecules, a size-matched trimesic acid (G3) gives inclusion complexes (NT6mer⊃G3) in dichloromethane resulting in an enhanced and red-shifted fluorescence. Macrocycle-mediated modulation and repurposing of antibacterial and non-antibacterial drugs.

The rapid emergence of antibiotic resistance has outpaced the discovery of new antibiotics, motivating alternative strategies to enhance existing therapeutics. Supramolecular macrocycles, characterized by defined cavities and host-guest complexation, provide a versatile platform for drug modulation. This review summarizes recent advances in macrocycle-based systems, including cyclodextrins, cucurbiturils, calixarenes, pillararenes, and crown ethers, with emphasis on their ability to modulate both antibacterial and non-antibacterial drugs. Unlike previous reviews focused on new antibacterial ma

Ugi Multicomponent Reaction Based Synthesis of Medium-Sized Rings.

An Ugi multicomponent reaction based two-step strategy was applied to generate medium-sized rings. In the first linear expansion phase, a series of diamines reacted with cyclic anhydrides to produce different lengths of terminal synthetic amino acids as the starting material for the second phase. The Ugi-4-center 3-component reaction was utilized to construct complex medium-sized rings (8-11) by the addition of isocyanides and oxo components. This method features mild conditions and a broad substrate scope.

Fragmentation of nitrone triflates to 9-membered rings.

A new fragmentative rearrangement of nitrone derivatives to form 9-membered rings is reported. The fragmentations are triggered when nitrones are treated with triflic anhydride; a C-C bond antiperiplanar to the cleaving N-O bond is activated either by an oxygen lone pair or by an electron-rich aromatic ring. In the former case, further cyclization of the 9-membered intermediate leads to a rearranged condensed ring system, but when triggered by arenes, 9-membered ring amides are isolated.

Discovery of Hirsutellones as Nucleolin-Targeting Natural Macrocycles for Triple-Negative Breast Cancer Treatment.

Triple-negative breast cancer (TNBC) remains a major clinical challenge due to its aggressive behavior, high metastatic potential, and limited therapeutic options, as current treatments often constrained by resistance and toxicity. Here, ten structurally distinct macrocyclic alkaloids were discovered from Didymella sp. Compounds 1 and 2 possessed the 13-membered macrocyclic ring alongside the novel spiro junction at C-3/C-18. Among them, phomapyrrolidone A (Ppd A) was the most active compound in the series, showing the lowest IC50 values and favorable selectivity toward TNBC cells. Integrated
