Deciphering Cis-Regulatory Element Mediated Combinatorial Regulation in Rice under Blast Infected Condition.

Combinations of cis-regulatory elements (CREs) present at the promoters facilitate the binding of several transcription factors (TFs), thereby altering the consequent gene expressions. Due to the eminent complexity of the regulatory mechanism, the combinatorics of CRE-mediated transcriptional regulation has been elusive. In this work, we have developed a new methodology that quantifies the co-occurrence tendencies of CREs present in a set of promoter sequences; these co-occurrence scores are filtered in three consecutive steps to test their statistical significance; and the significantly co-oc

Phase-contrast microtomography unveils mechanisms of root colonization by a vascular fungal pathogen.

Soil-borne vascular pathogens pose serious threats to agriculture with complex invasion strategies that remain poorly characterized compared to foliar pathogens. While foliar pathogens like Magnaporthe oryzae employ specialized appressoria to penetrate plant surfaces through a combination of mechanical force and enzymatic degradation, the invasion mechanisms of vascular pathogens that lack classical appressoria have remained largely theoretical. The nanoscale processes governing root penetration and colonization by these pathogens are particularly challenging to visualize due to technical limi

Evolutionary Systems Biology Identifies Genetic Trade-offs in Rice Defense against Aboveground and Belowground Attackers.

Like other plants, wild and domesticated rice species (Oryza nivara, O. rufipogon, and O. sativa) evolve in environments with various biotic and abiotic stresses that fluctuate in intensity through space and time. Microbial pathogens and invertebrate herbivores such as plant-parasitic nematodes and caterpillars show geographical and temporal variation in activity patterns and may respond differently to certain plant-defensive mechanisms. As such, plant interactions with multiple community members may result in conflicting selection pressures on genetic polymorphisms. Here, through assays with

Discovery of Acaricidal, Insecticidal, and Fungicidal Candidates Inspired by Natural Ethyl Cinnamate Compounds Isolated from Polygonum orientale L.

As a continuation of our research on the pesticide development of Polygonum orientale L., the chemical constituents of the seeds of P. orientale were systematically investigated. Eleven natural compounds (PO-1 to PO-11) were isolated from the EtOAc extract of P. orientale. Notably, compound PO-9 and its dimeric compound PO-10 were first isolated from P. orientale and possessed excellent acaricidal activity against Tetranychus cinnabarinus. With PO-9 and PO-10 as the lead compounds, two series of cinnamate derivatives were further synthesized, and their acaricidal, insecticidal, and fungicidal

SPOTTED-LEAF7 targets the gene encoding β-galactosidase9, which functions in rice growth and stress responses.

β-Galactosidases (Bgals) remove terminal β-D-galactosyl residues from the nonreducing ends of β-D-galactosidases and oligosaccharides. Bgals are present in bacteria, fungi, animals, and plants and have various functions. Despite the many studies on the evolution of BGALs in plants, their functions remain obscure. Here, we identified rice (Oryza sativa) β-galactosidase9 (OsBGAL9) as a direct target of the heat stress-induced transcription factor SPOTTED-LEAF7 (OsSPL7), as demonstrated by protoplast transactivation analysis and yeast 1-hybrid and electrophoretic mobility shift assays. Engineering Climate-Resilient Rice Using a Nanobiostimulant-Based "Stress Training" Strategy.

Under a changing climate, cultivating climate-resilient crops will be critical to maintaining food security. Here, we propose the application of reactive oxygen species (ROS)-generating nanoparticles as nanobiostimulants to trigger stress/immune responses and subsequently increase the stress resilience of plants. We established three regimens of silver nanoparticles (AgNPs)-based "stress training": seed training (ST), leaf training (LT), and combined seed and leaf training (SLT). Trained rice seedlings were then exposed to either rice blast fungus (Magnaporthe oryzae) or chilling stress (10 °C

Expression and characterization of recombinant fungal acetyl-CoA carboxylase and isolation of a soraphen-binding domain.

Acetyl-CoA carboxylase (ACC) catalyses the first step in fatty-acid biosynthesis. Owing to its role in primary metabolism, ACC has been exploited as a commercial herbicide target and identified as a chemically validated fungicide target. In animals, ACC is also a key regulator of fat metabolism. This function has made ACC a prime target for the development of anti-obesity and anti-Type II diabetes therapeutics. Despite its economic importance, there is a lack of published information on recombinant expression of ACC. We report here the expression of enzymically active fungal (Ustilago maydis )

Brachypodium distachyon. A new model system for functional genomics in grasses.

A new model for grass functional genomics is described based on Brachypodium distachyon, which in the evolution of the Pooideae diverged just prior to the clade of "core pooid" genera that contain the majority of important temperate cereals and forage grasses. Diploid ecotypes of B. distachyon (2n = 10) have five easily distinguishable chromosomes that display high levels of chiasma formation at meiosis. The B. distachyon nuclear genome was indistinguishable in size from that of Arabidopsis, making it the simplest genome described in grasses to date. B. distachyon is a self-fertile, inbreeding

Eucalyptus camaldulensis properties for use in the eradication of infections.

Eucalyptus camaldulensis (E. camaldulensis), called the eucalyptus has so many characteristics such as antimicrobial features. Common names include red gum, red chewing gum, river chewing gum, red chewing gum. Its class is Eucalyptus, which is comprised of 800 species worldwide, but three or four species are found in Australia. This tree generally grows on the edge of rivers with continuous or seasonal water. Most of the gray clay soils run along the riverside and are exposed to frequent floods, and clay content can save more water into the tree. Pharmacy departments and research groups have f

The basic helix-loop-helix transcription factor Crf1 is required for development and pathogenicity of the rice blast fungus by regulating carbohydrate and lipid metabolism.

Pyricularia oryzae is a plant pathogen causing rice blast, a serious disease spreading in cultivated rice globally. Transcription factors play important regulatory roles in fungal development and pathogenicity. Here, we characterized the biological functions of Crf1, a basic helix-loop-helix (bHLH) transcription factor, in the development and pathogenicity of P. oryzae with functional genetics, molecular and biochemical approaches. We found that CRF1 is necessary for virulence and plays an indispensable role in the regulation of carbohydrate and lipid metabolism in P. oryzae. Deletion of CRF1

A novel jasmonic acid-inducible rice myb gene associates with fungal infection and host cell death.

Endogenous signal molecules such as jasmonic acid (JA) and salicylic acid (SA) play an important role in induced resistance against pathogen infection and insect herbivory. In rice seedlings, JA is an effective inducer of systemic acquired resistance (SAR) against infection of blast fungus (Pyricularia grisea). To gain further insights into JA-mediated defense signaling pathways, we isolated and characterized a pathogen- and JA-induced rice gene (JAmyb) that encodes a Myb transcription factor. The JAmyb gene was induced within 1 day after fungal infection in resistant and susceptible interacti

An enzymatic glucose/O2 biofuel cell operating in human blood.

Enzymatic biofuel cells (BFCs) may power implanted medical devices and will rely on the use of glucose and O2 available in human bodily fluids. Other than well-established experiments in aqueous buffer, little work has been performed in whole human blood because it contains numerous inhibiting molecules. Here, we tested our BFCs in 30 anonymized, random and disease-free whole human blood samples. We show that by designing our anodic and cathodic bioelectrocatalysts with osmium based redox polymers and home-made enzymes we could reach a high selectivity and biofunctionnality.
