A self-inactivating gamma-retroviral vector reduces manifestations of mucopolysaccharidosis I in mice.

Mucopolysaccharidosis I (MPS I) is a lysosomal storage disease due to deficiency in alpha-L-iduronidase (IDUA) that results in accumulation of glycosaminoglycans (GAGs) throughout the body, causing numerous clinical defects. Intravenous administration of a gamma-retroviral vector (gamma-RV) with an intact long terminal repeat (LTR) reduced the clinical manifestations of MPS I, but could cause insertional mutagenesis. Although self-inactivating (SIN) gamma-RVs in which the enhancer and promoter elements in the viral LTR are absent after transduction reduces this risk, such vectors could be less

The woodchuck model of hepatitis B virus infection.

The woodchuck hepatitis virus (WHV) was the first of the mammalian and avian hepadnaviruses described after discovery of the virus of hepatitis B (HBV). Woodchucks chronically infected with WHV develop progressively severe hepatitis and hepatocellular carcinoma, which present as lesions that are remarkably similar to those associated with HBV infection in humans. The initial virological studies and studies of pathogenesis utilized woodchucks that had been trapped in the wild and had acquired WHV infection naturally. Research with wild woodchucks was complicated by lack of knowledge of their ba

Therapeutic vaccination of woodchucks against chronic woodchuck hepatitis virus infection.

BACKGROUND/AIMS: Therapeutic vaccination is a new approach to treat patients with chronic hepatitis B virus infection. We have used the woodchuck model to examine the efficacy and safety of this approach. METHODS: Seven woodchucks chronically infected with woodchuck hepatitis virus were immunized with surface antigen from this virus, purified from plasma, in conjunction with a peptide named FIS (encompassing amino acids 106-118: FISEAIIHVLHSR from sperm whale myoglobin), which is recognized by T helper lymphocytes. As controls, two woodchucks chronically infected with woodchuck hepatitis viru

Baseline oral microbiome associated with post-living donor liver transplant early complications within 6 months.

Oral dysbiosis is noted in liver cirrhosis. This may play a role in post-liver transplantation (LT) outcomes. This study aimed to assess the baseline oral microbiome and its association with early complications (ECs) within 6 months after LT. This prospective longitudinal study included 94 cirrhotic recipients (CRs) and their living donors. Patients were managed with a standardized immunosuppressive regimen and monitored for 6 months post-LT EC rejection, infection, biliary complications (BCs), and death. Pre-LT and post-LT saliva samples were processed for bacterial sequencing, qPCR validatio

Survey for Babesia spp. in wildlife in the eastern United States.

Babesia is a diverse genus of piroplasms that parasitize the red blood cells of a wide variety of mammals and avian species, including humans. There is a lack of knowledge on the Babesia species of carnivores and mesomammals in the eastern United States and the potential impacts of these species on the health of humans and domestic animals. We surveyed 786 wild mammals in the eastern United States by testing blood, spleen, and heart samples with PCR targeting the 18S rRNA region of apicomplexan parasites. We also performed PCR targeting the cytochrome c oxidase subunit 1 (cox1) region of each

Detection, Prevalence and Phylogenetic Relationships of Demodex spp and further Skin Prostigmata Mites (Acari, Arachnida) in Wild and Domestic Mammals.

This study was conceived to detect skin mites in social mammals through real-time qPCR, and to estimate taxonomic Demodex and further Prostigmata mite relationships in different host species by comparing sequences from two genes: mitochondrial 16S rRNA and nuclear 18S rRNA. We determined the mite prevalence in the hair follicles of marmots (13%) and bats (17%). The high prevalence found in marmots and bats by sampling only one site on the body may indicate that mites are common inhabitants of their skin. Since we found three different mites (Neuchelacheles sp, Myobia sp and Penthaleus sp) in t

Hepadnavirus evolution and molecular strategy of adaptation in a new host.

In order to elucidate the mechanisms of hepadnavirus evolution in vivo and to trace the fate of known quasispecies in a single animal during the acute phase of infection, a woodchuck (Marmota monax) was infected with the hepadnavirus woodchuck hepatitis B virus (WHV). Woodchuck 197 (W197) was injected intravenously with pooled sera collected from a chronic carrier that had been infected originally with a molecular clone of known genome sequence (WHV7). Viral genome variants from both the inoculum and the follow-up sera from W197 were characterized for the presence of quasispecies related to th

Effects of Varied Stimuli on Escape Behavior Diversification of Himalayan Marmots for Different Human Disturbances.

We measured the alert distance (AD), flight-initiation distance (FID), buffer distance (BD), and distance fled (DF) of Himalayan marmots (Marmota himalayana) from four populations experiencing human disturbances of the same persistence but different intensities when subjected to varied stimuli (a running or walking man with or without a leashed dog and a dog alone). We analyzed the effects of different stimuli on the AD, FID, BD, and DF of marmots from each population and the relationship among the AD, FID, and DF to illustrate the escape strategy diversification of the studied marmots for dif

Marmots and Yersinia pestis Strains in Two Plague Endemic Areas of Tien Shan Mountains.

The main purpose of this study was to clarify the role of gray marmots (Marmota baibacina) in the long-term maintenance of highly virulent strains of Yersinia pestis in two plague endemic foci of the Tien Shan Mountains in Kyrgyzstan. We present data from regular observations of populations of M. baibacina and small rodents cohabiting with marmots in the mountainous grasslands of the Sari-Dzhas (east of Issyk-Kul Lake) and the Upper-Naryn (south of Issyk-Kul Lake) natural foci. During 2012-2017, an abundance of marmots and their ectoparasites (fleas and ticks) was significantly higher in Upper

Co-occurrence of Yersinia pestis and a lytic bacteriophage in brain tissue of Eothenomys miletus.

UNLABELLED: We surveyed plague in the Apodemus chevrieri-Eothenomys miletus natural plague focus on the Yunnan Plateau, China. From 2023 to 2025, plague-positive E. miletus were detected in Gaomei, Xiaomachang, and Tangshang. In the brain tissue of an E. miletus carcass, both Yersinia pestis and a Yersinia pestis-specific bacteriophage (YN2024) were simultaneously isolated, and their simultaneous presence was observed via transmission electron microscopy (TEM). This observation was supported by previous analysis of a similar co-occurrence of Y. pestis and bacteriophage from the Marmota himalay

Innate and adaptive immunity associated with resolution of acute woodchuck hepatitis virus infection in adult woodchucks.

Viral and/or host factors that are directly responsible for the acute versus chronic outcome of hepatitis B virus (HBV) infection have not been identified yet. Information on immune response during the early stages of HBV infection in humans is mainly derived from blood samples of patients with acute hepatitis B (AHB), which are usually obtained after the onset of clinical symptoms. Features of intrahepatic immune response in these patients are less studied due to the difficulty of obtaining multiple liver biopsies. Woodchuck hepatitis virus (WHV) infection in woodchucks is a model for HBV inf

Treatment of woodchuck hepatitis virus infection in vivo with 2', -3'-dideoxycytidine (ddC) and 2',-3'-dideoxycytidine monophosphate coupled to lactosaminated human serum albumin (L-HSA ddCMP).

Dideoxycytidine (ddC) is a nucleoside analogue active against human immunodeficiency virus and with in vitro activity against human hepatitis B virus. We investigated the ability of ddC to inhibit one of the Hepadnaviridae, the woodchuck hepatitis virus and compared the results with the effect obtained by a conjugate of lactosaminated human serum albumin 2',-3'-dideoxycytidine monophosphate (L-HSA ddCMP). This compound specifically enters the hepatocyte via the asialoglycoprotein receptor. We treated five chronic woodchuck hepatitis virus carriers with intravenous injections of 0.5 mg/kg body

A single-cell atlas of the woodchuck liver reveals cellular programs conserved in human HBV infection.

BACKGROUND & AIMS: The eastern woodchuck (Marmota monax), which can be naturally infected with woodchuck hepatitis virus (WHV), has served as a model for studying aspects of human HBV infection, including the establishment of chronic infection and progression from chronic hepatitis to liver cancer. However, the cellular landscape of the woodchuck liver and its parallels to HBV infection remain uncharacterized. METHODS: We generated a single-cell and spatial transcriptomic atlas of the woodchuck liver in health and chronic WHV infection, characterizing cell types and infection-associated proce

The impact of HBV infection on gut microbiota using Chinese woodchuck model with woodchuck hepatitis virus (WHV) infection.

Hepatitis B virus (HBV) infection seems to be related to gut microbiota. This study aims to explore the effects of HBV infection on gut microbiota and possible immunological mechanisms using the Chinese woodchuck model. Nine adult woodchucks were randomly divided into Cyclosporine A (CsA) or Control group. Animals were orally treated with CsA and saline for 2 weeks before WHV inoculation, and continued until 8 weeks after that. Blood CsA concentrations were tested at 2 weeks after administration and before discontinuation. Quantitative PCR was used to detect serum WHV DNA. Flow cytometry was u

Identification of an intrahepatic transcriptional signature associated with self-limiting infection in the woodchuck model of hepatitis B.

UNLABELLED: The woodchuck model of hepatitis B virus (HBV) infection displays many characteristics of human infection and has particular value for characterizing the host immune responses during the development of chronic infection. Using the newly developed custom woodchuck microarray platform, we compared the intrahepatic transcriptional profiles of neonatal woodchucks with self-limiting woodchuck hepatitis virus (WHV) infection to those woodchucks progressing to persistent WHV infection. This revealed that WHV does not induce significant intrahepatic gene expression changes during the early

Persistence of isolated antibodies to woodchuck hepatitis virus core antigen is indicative of occult infection.

Antibodies against virus nucleocapsid (anticore) normally accompany hepadnaviral hepatitis but they may also occur in the absence of symptoms and other serological indicators of the infection. This situation can be encountered following a clinically and serologically unapparent exposure to hepatitis B virus (HBV) or after recovery from hepatitis B. In this study, woodchucks inoculated with woodchuck hepatitis virus (WHV) were investigated to determine the relationship between anticore detection and the molecular status of virus replication in a primary WHV surface antigen (WHsAg)-negative infe

Efficiency of oxetanocin-G, a novel nucleoside against the woodchuck hepatitis virus.

Oxetanocin-G (OXT-G), a potent antiviral agent, is a novel nucleoside isolated from the culture filtrate of Bacillus megaterium. We investigated the antiviral effect of oral administration of OXT-G for five days on woodchuck hepatitis virus (WHV), in vivo, using 12 woodchucks. Woodchucks were randomized into each of four treatment groups according to the dose of OXT-G. Two out of six woodchucks treated with 1.0 or 2.0 mg/kg/day of OXT-G died. After treatment with OXT-G, serum levels of WHV-DNA significantly decreased in all woodchucks. However, the antiviral effect was only partial and levels

Therapeutic vaccines in HBV: lessons from HCV.

Currently, millions of people infected with hepatitis B virus (HBV) are committed to decades of treatment with anti-viral therapy to control viral replication. However, new tools for immunotherapy that include both viral vectors and molecular checkpoint inhibitors are now available. This has led to a resurgence of interest in new strategies to develop immunotherapeutic strategies with the aim of inducing HBeAg seroconversion--an end-point that has been associated with a decrease in the rates of disease progression. Ultimately, a true cure will involve the elimination of covalently closed circu
