The use of voltage-sensitive dyes to monitor signal-induced changes in membrane potential-ABA triggered membrane depolarization in guard cells.

The plant membrane potential reports on the activity of electrogenic plasma membrane transport processes. The membrane potential is widely used to report for early events associated with changes in light regime, hormone action or pathogen attacks. The membrane potentials of guard cells can be precisely measured with microelectrodes, but this technique is not well suited for rapid screens with large sample numbers. To provide the basis for large-scale membrane potential recordings, we took advantage of voltage-sensitive dyes. Using the fluorescent dyes bis-(1,3-dibutylbarbituric acid)-trimethin

Membrane potential of cells and its regulation during aging. 1. Report: the role of energetic metabolism and plasma membrane phospholipid contents.

Age-dependent changes in the polarization of the plasma membranes (PM) of various cell types and the mechanisms responsible for its regulation were studied in the experiments on the adult (6-8 months) and old (28-32 months) Wistar male rats. The relationship was found between the specificity of cellular function and the pattern of changes in resting potential (RP) and action potential (AP). In senility, changes occur in the ratio of different pathways of energy provision of the ionic transport: the contribution made by enzymic reactions to the mechanism of maintenance of the PM polarization le

Chloride current in mammalian cardiac myocytes. Novel mechanism for autonomic regulation of action potential duration and resting membrane potential.

The properties of the autonomically regulated chloride current (ICl) were studied in isolated guinea pig ventricular myocytes. This current was elicited upon exposure to isoproterenol (ISO) and reversed upon concurrent exposure to acetylcholine (ACh). ICl was time independent and exhibited outward rectification. The responses to ISO and ACh could be blocked by propranolol and atropine, respectively, and ICl was also elicited by forskolin, 8-bromoadenosine 3',5'-cyclic monophosphate, and 3-isobutyl-l-methylxanthine, indicating that the current is regulated through a cAMP-dependent pathway. Light-Triggered Mechanotherapy of Membrane-Mimicking Conjugated Electrolytes Potentiates Antibacterial Efficacy.

The rise of multidrug-resistant (MDR) pathogens has created an urgent need for antibacterial strategies that operate beyond conventional biochemical targets. Here, a light-triggered molecular mechanotherapeutic antibacterial platform based on membrane-mimicking conjugated electrolytes (MMCEs) is reported. Owing to their membrane-compatible architecture, MMCEs can spontaneously intercalate into lipid bilayers while remaining relatively non-disruptive in the dark. To enhance photoinduced mechanical activity, ZBT was developed from the classical BT scaffold by replacing the stilbene-like wing wit

Membrane interfacial potential governs surface condensation and fibrillation of α-Synuclein in neurons.

Biomolecular condensates formed via liquid-liquid phase separation (LLPS) are essential for cellular organization. α-Synuclein, an amyloidogenic protein linked to Parkinson's Disease (PD), undergoes phase separation at high concentrations, but the influence of lipid membranes on this process remains unclear. Here, combining in vitro reconstitution, cell biology, and simulations, we show that membranous interfaces promote α-Synuclein condensation at physiologically relevant sub-critical concentrations ( ~ 10 nM) without crowding agents. Notably, condensation occurs only on membranes with a spec

Targeting the Protein-Membrane Interface Enables Design of Long-Acting CFTR Potentiators.

Many therapeutically relevant membrane proteins possess druggable sites at the lipid-protein interface, but principles guiding ligand design for these sites are not well-defined. CFTR potentiators, a clinically validated drug class for cystic fibrosis, offer a compelling model for exploring membrane-targeted design principles, as their efficacy depends on sustained intramembrane binding to prolong channel opening and chloride conductance. Here, we systematically modified the lipophilic substituent of the CFTR potentiator ABBV-974 and identified an analog that confers markedly increased functio

Prostate-Specific Membrane Antigen (PSMA): A Potential Theranostic Biomarker in Breast Cancer.

Background: Subtype classification for breast cancer (BC) patients is important for risk-stratification. Unfortunately, this parameter is not always able to discriminate between high- and low-risk diseases. Glutamate Carboxypeptidase-II (GCPII), also known as prostate-specific membrane antigen (PSMA), could be an important biomarker of aggressiveness, given that it has been reported to be expressed in BC tumor cells and even more in endothelial cells of tumor vessels. Methods: We analyzed 22 Luminal A, 47 Luminal B, 9 HER2-positive (HER2+), and 23 triple-negative (TN) BC to assess whether PSMA

Cell membrane-based nanovesicles as potential drug delivery systems for atherosclerosis therapy.

Atherosclerosis is the main underlying cause of death, being originated by endothelial dysfunction, which favor the recruitment of different cell types and expression of several factors contributing to plaque development and growth. Current standard treatments are focused on the systemic administration of statins, which reduce the blood cholesterol levels and provide additional anti-inflammatory and plaque-stabilizing effects but also present important adverse side effects such as myopathy and liver damage, among others. To overcome such drawbacks, drug delivery systems have emerged to directl

Identifying Potential Drug Targets for Membranous Nephropathy Through an Analysis of Mendelian Randomization.

Membranous nephropathy (MN) is the most common cause of adult nephrotic syndrome. Current treatments rely heavily on immunosuppressants; however, some patients do not achieve the desired therapeutic effect. Therefore, the identification of new drug targets and the development of novel medications are of urgent importance. In this study, we collected protein quantitative trait loci (pQTLs) for 734 circulating plasma proteins (CPPs) from previous research. Using principles of Mendelian genetics, we conducted a Mendelian randomization (MR) analysis using three inferential methods: the Wald ratio,

Optical Redox Imaging Predicts Post-Loading Cartilage Mitochondrial Membrane Potential.

PURPOSE: Disrupted cellular redox balance is associated with various diseases, including osteoarthritis. Although mitochondrial (MT) membrane potential is a proxy for redox balance, the translational potential of this method is limited by exogenous dye. Therefore, the objective of this study was to predict changes in MT membrane potential in response to mechanical loading using dye-free optical redox imaging (ORI). A secondary objective was to determine the effect of loading on ORI metrics. METHODS: Full-thickness porcine cartilage strips were subjected to tensile loading at one of two strain

Efflux pumps mediate changes to fundamental bacterial physiology via membrane potential.

Efflux pumps are well known to be an important mechanism for removing noxious substances such as antibiotics from bacteria. Given that many antibiotics function by accumulating inside bacteria, efflux pumps contribute to resistance. Efflux pump inactivation is a potential strategy to combat antimicrobial resistance, as bacteria would not be able to pump out antibiotics. We recently discovered that the impact of loss of efflux function is only apparent in actively growing cells. We demonstrated that the global transcriptome of Salmonella Typhimurium is drastically altered during slower growth l

Venturicidin A affects the mitochondrial membrane potential and induces kDNA loss in Trypanosoma brucei.

Neglected tropical diseases caused by trypanosomatid parasites have devastating health and economic consequences, especially in tropical areas. New drugs or new combination therapies to fight these parasites are urgently needed. Venturicidin A, a macrolide extracted from Streptomyces, inhibits the ATP synthase complex of fungi and bacteria. However, its effect on trypanosomatids is not fully understood. In this study, we tested venturicidin A on a panel of trypanosomatid parasites using Alamar Blue assays and found it to be highly active against Trypanosoma brucei and Leishmania donovani, but

Effect of the mitochondrial membrane potential on the absorbance of the reduced form of cytochrome c oxidase.

The effect of mitochondrial membrane potential (ΔΨm) on the absorbance of the reduced cytochrome c oxidase (COX) was evaluated in isolated rabbit heart mitochondria using integrating sphere optical spectroscopy. Maximal reduction of the mitochondrial cytochromes was achieved by either blowing nitrogen to remove oxygen, or by adding cyanide. Gradual depolarization of ΔΨm by adding increasing concentrations of uncoupler resulted in an increase of up to 50 % in the absorbance of cytochrome aa3 under nitrogen saturation, and of 25 % with cyanide. Cytochrome aa3 absorbance increases were also obser

Sterilized Polyhexanide-Releasing Chitosan Membranes with Potential for Use in Antimicrobial Wound Dressings.

Wound infection is a common complication of chronic wounds. It can impair healing, which may not occur without external help. Antimicrobial dressings (AMDs) are a type of external help to infected chronic wounds. In this study, highly porous membranes made of only chitosan and containing the antiseptic polyhexanide (poly(hexamethylene biguanide); PHMB) were prepared by cryogelation, aiming to be used in AMDs. These membranes exhibited a water swelling capacity of 748%, a water drop penetration time of 11 s in a dry membrane and a water vapor transmission rate of 34,400 g H2O/m2/24 h when in co

Contactin 1, a Potential New Antigen Target in Membranous Nephropathy: A Case Report.

Several novel antigens have recently been characterized in membranous nephropathy (MN), but those involved in the rare cases of MN associated with inflammatory neuropathies remain elusive. Although several antibodies have been identified in the serum, there is no evidence so far for their deposition in glomeruli. We report the case of a 73-year-old woman who was referred because of subacute onset of proximal asymmetric lower limb weakness together with ataxic gait. She was diagnosed with inflammatory neuropathy. Testing showed an estimated glomerular filtration rate of 73mL/min/1.73m2, hypoalb

Relapse of pathological angiogenesis: functional role of the basement membrane and potential treatment strategies.

Blinding eye diseases such as corneal neovascularization, proliferative diabetic retinopathy, and age-related macular degeneration are driven by pathological angiogenesis. In cancer, angiogenesis is key for tumor growth and metastasis. Current antiangiogenic treatments applied clinically interfere with the VEGF signaling pathway-the main angiogenic pathway-to inhibit angiogenesis. These treatments are, however, only partially effective in regressing new pathologic vessels, and the disease relapses following cessation of treatment. Moreover, the relapse of pathological angiogenesis can be rapid

Small Molecule Channels Harness Membrane Potential to Concentrate Potassium in trk1Δtrk2Δ Yeast.

Many protein ion channels harness membrane potential to move ions in opposition to their chemical gradient. Deficiencies of such proteins cause several human diseases, including cystic fibrosis, Bartter Syndrome, and proximal renal tubular acidosis. Using yeast as a eukaryotic model system, we asked whether, in the context of a protein ion channel deficiency in vivo, small molecule channels could similarly harness membrane potential to concentrate ions. Trk potassium transporters use membrane potential to move potassium from a relatively low concentration outside cells (∼15 mM) to one of >10×

Roles of VMP1 in Autophagy and ER-Membrane Contact: Potential Implications in Neurodegenerative Disorders.

Cellular communication processes are highly dynamic and mediated, at least in part, by contacts between various membrane structures. The endoplasmic reticulum (ER), the major biosynthetic organelle of the cell, establishes an extensive network with other membrane structures to regulate the transport of intracellular molecules. Vacuole membrane protein 1 (VMP1), an ER-localized metazoan-specific protein, plays important roles in the formation of autophagosomes and communication between the ER and other organelles, including mitochondria, autophagosome precursor membranes, Golgi, lipid droplets,

D-Chiro-Inositol Improves Sperm Mitochondrial Membrane Potential: In Vitro Evidence.

The use of inositols in endocrinological clinical practice is increasingly widespread. Most of the existing evidence concerns myoinositol (MYO), the most abundant form in nature, especially in women with polycystic ovarian syndrome. We have previously shown that MYO increases sperm motility in patients with asthenozoospermia by the increase of sperm mitochondrial membrane potential (MMP), a biofunctional sperm parameter closely associated to sperm motility. The aim of this study was to evaluate the effects of D-chiro-inositol (DCI), another biologically active isoform of inositols, on sperm MM
