Crosstalk among hydrogen sulfide (H2S), nitric oxide (NO) and carbon monoxide (CO) in root-system development and its rhizosphere interactions: A gaseous interactome.

Root development in higher plants is achieved by a precise intercellular communication which determines cell fate in the primary embryonic meristem where the gasotransmitters H2S, NO and CO participate dynamically. Furthermore, the rhizosphere interaction of these molecules with microbial and soil metabolism also affects root development. NO regulates root growth and architecture in association with several other biomolecules like auxin indole-3-acetic acid (IAA), ethylene, jasmonic acid (JA), strigolactones, alkamides and melatonin. The CO-mediated signal transduction pathway in roots is clos

NON-IDENTICAL MECHANISMS OF MITOTIC ARREST BY RESPIRATORY INHIBITORS IN PEA ROOT TIPS AND SEA URCHIN EGGS.

The adenosine triphosphate (ATP) content of pea root tips is about 0.40 mmole/kg fresh weight. The effects of partial and complete anaerobiosis, and of carbon monoxide and hydrogen cyanide, on the ATP level are described. The ATP content was shown to correspond closely to the oxygen uptake under these conditions. However, there was no relation between the ATP level and the rate of mitosis, a situation which is in contrast with that in sea urchin eggs. In anaerobic conditions, mitoses in pea root tips could continue at a reduced rate, even though the ATP content had fallen to 1.5 per cent of th

SPATULA regulates CLAVATA1 and GCN5 genetic interaction during gynoecium development in Arabidopsis thaliana.

GENERAL CONTROL NONDEREPRESSIBLE 5 (GCN5) is a histone acetyltransferase in Arabidopsis thaliana that plays a key role in various biological processes, including the development of flowers and inflorescences. We have shown that GCN5 genetically interacts with the CLAVATA (CLV) signalling pathway, potentially affecting gynoecium and inflorescence development by modulating auxin and cytokinin dynamics and regulating WUSCHEL (WUS) expression. The bHLH transcription factor SPATULA (SPT) is also a crucial regulator of gynoecium formation. In clv1gcn5 double mutants, notable phenotypic changes inclu

Metabolic Pathways for Observed Impacts of Crop Load on Floral Induction in Apple.

The triggers of biennial bearing are thought to coincide with embryonic development in apple and occurs within the first 70 days after full bloom (DAFB). Strong evidence suggests hormonal signals are perceived by vegetative apple spur buds to induce flowering. The hormonal response is typically referred to as the floral induction (FI) phase in bud meristem development. To determine the metabolic pathways activated in FI, young trees of the biennial bearing cultivar 'Nicoter' and the less susceptible cultivar 'Rosy Glow' were forced into an alternate cropping cycle over five years and an invers

Plant 22-nt siRNAs mediate translational repression and stress adaptation.

Small interfering RNAs (siRNAs) are essential for proper development and immunity in eukaryotes1. Plants produce siRNAs with lengths of 21, 22 or 24 nucleotides. The 21- and 24-nucleotide species mediate cleavage of messenger RNAs and DNA methylation2,3, respectively, but the biological functions of the 22-nucleotide siRNAs remain unknown. Here we report the identification and characterization of a group of endogenous 22-nucleotide siRNAs that are generated by the DICER-LIKE 2 (DCL2) protein in plants. When cytoplasmic RNA decay and DCL4 are deficient, the resulting massive accumulation of 22-

Machine learning-guided leaf margin classification and discovery that BcPHB promoter selection rewires a BcPHB-BcLMI1 module to shape leaf margins in Brassica campestris.

The morphological diversification of leaf margins reflects the evolutionary reshaping of conserved developmental programs. However, fully deciphering the underlying molecular basis remains challenging due to difficulties in phenotype quantification and the incomplete characterization of complex regulatory networks. In this study, we established a computer-vision framework to decode leaf lobe trait and identified the HD-ZIP III factor PHABULOSA (BcPHB), combining genome-wide association studies (GWAS) and selective sweep analysis in Brassica campestris. We found that promoter variation defines

Metabolic Snorkeling - The Metabolic Interaction Between Plant Organs and Tissues with different Oxygen Availabilities.

Plants constantly experience partial hypoxia due to varying external oxygen availability and the existence of hypoxic niches, such as the shoot apical meristem, phloem or root nodules. Waterlogging experiments indicate that hypoxic stress at the root does not only lead to a local metabolic response, such as the accumulation of hypoxia-related metabolites, but also causes metabolic alterations in the normoxic shoot. Moreover, hypoxia-related metabolites are exported from the hypoxic root towards the normoxic shoot, where they can be recycled. Maintaining import of glycolytic substrates from the

Gibberellin-mediated internode elongation for grafting and early flowering induction in Brassicaceae crops.

In Brassicaceae crops such as cabbage and turnip, which are harvested for their vegetative organs, floral induction can reduce the eating quality of these organs. Therefore, developing late-flowering varieties that are insensitive to vernalization is a key breeding objective. However, a major challenge in breeding these cultivars lies in the contradiction between the late-flowering trait and the need to induce flowering for crossing. To address this, we developed a flowering induction technique using grafting, in which gibberellin-induced elongated internodes are used as scions and grafted ont

Phosphate regulates malate/citrate-mediated iron uptake and transport in apple.

The accumulation of iron (Fe) in the apical meristem is considered as a critical factor involved in limiting the elongation of roots under low phosphate (Pi) conditions. Furthermore, the antagonism between Fe and Pi largely affects the effective utilization of Fe. Although the lack of Pi serves to increase the effectiveness of Fe in rice under both Fe-sufficient and Fe-deficient conditions, the underlying physiological mechanism governing this phenomenon is still unclear. In this study, we found that low Pi alleviated the Fe-deficiency phenotype in apples. Additionally, low Pi treatments incre

Functional and regulatory analysis of the Arabidopsis thaliana CAX2 cation transporter.

The vacuolar sequestration of metals is an important metal tolerance mechanism in plants. The Arabidopsis thaliana vacuolar transporters CAX1 and CAX2 were originally identified in a Saccharomyces cerevisiae suppression screen as Ca2+/H+ antiporters. CAX2 has a low affinity for Ca2+ but can transport other metals including Mn2+ and Cd2+. Here we demonstrate that unlike cax1 mutants, CAX2 insertional mutants caused no discernable morphological phenotypes or alterations in Ca2+/H+ antiport activity. However, cax2 lines exhibited a reduction in vacuolar Mn2+/H+ antiport and, like cax1 mutants, re

Maize Developmental Biology: Fundamental Discoveries Unlocking Innovation.

Maize (Zea mays) is a cornerstone of agriculture and global genetics research. Every year more than 200 million hectares of maize are grown worldwide for human and animal consumption, and biofuel and energy production. Maize has also been a key research model organism since the 1800s, and over 150 years of research has led to many fundamental discoveries in genetics and developmental biology. These findings include "jumping genes" (transposons) and the first homeobox gene in plants, KNOTTED1, whose discovery overturned the hypothesis that homeobox genes were unique to animals. Research in maiz

Fire adaptation in Neblinaria celiae (Theaceae), a high-elevation rosette shrub endemic to a wet equatorial tepui.

Neblinaria celiae (Theaceae), a rosette shrub endemic to the exceedingly rainy summit of remote Cerro de la Neblina in southern Venezuela, has a previously undescribed set of adaptations to fire. Its growth form entails sparse branching, massive terminal leaf rosettes, and thick bark. It is highly fire-tolerant, with a survival rate of 93% in a stand recently ignited by lightning, vs. 0% in seven co-occurring woody species. Survival increases sharply with rosette height, favoring a sparsely branched habit that would maximize the rate of upward growth through the sparse fuel layer supported by

Cytokinin regulates flowering time in Arabidopsis thaliana through the gibberellin pathway.

Cytokinin and gibberellin are two plant hormones promoting flowering in Arabidopsis thaliana grown under short-day conditions. The activities of both hormones depend on similar signaling components, including several elements of the age pathway. However, to date, the interaction between cytokinin and gibberellin in regulating the transition to flowering remains unknown. In this work, we show that exogenous treatment of late-flowering or non-flowering cytokinin mutants by gibberellin restores their ability to flower. In contrast, the promotion of flowering by cytokinin is suppressed by mutation

Concurrent exposure to water deficit and elevated ambient temperature alleviates root growth defects in Arabidopsis.

Plants are often simultaneously subjected to different environmental stresses threatening their very existence. As high temperature events usually coincide with drought conditions, a combination of these stresses is a common occurrence in the field. Recently, global warming has led to an increase in the frequency and intensity of heatwaves and drought across the globe, severely compromising plant performance. However, our understanding of root system responses to combined stress scenarios is limited. Hence, in this study, we investigated how the root system responds under a combined stress env

Synchronisms between bud and cambium phenology in black spruce: early-flushing provenances exhibit early xylem formation.

Bud and cambial phenology represent the adaptation of species to the local environment that allows the growing season to be maximized while minimizing the risk of frost for the developing tissues. The temporal relationship between the apical and radial meristems can help in the understanding of tree growth as a whole process. The aim of this study was to compare cambial phenology in black spruce (Picea mariana (Mill.) B.S.P.) provenances classified as early and late bud flushing. The different phases of cambial phenology were assessed on wood microcores sampled weekly from April to October in

An oncoprotein from the plant pathogen agrobacterium has histone chaperone-like activity.

Protein 6b, encoded by T-DNA from the pathogen Agrobacterium tumefaciens, stimulates the plant hormone-independent division of cells in culture in vitro and induces aberrant cell growth and the ectopic expression of various genes, including genes related to cell division and meristem-related class 1 KNOX homeobox genes, in 6b-expressing transgenic Arabidopsis thaliana and Nicotiana tabacum plants. Protein 6b is found in nuclei and binds to several plant nuclear proteins. Here, we report that 6b binds specifically to histone H3 in vitro but not to other core histones. Comparative Floral Development and Anatomy Reveal Distinct Origins of the Gynophore in Meso-Papilionoideae.

The Meso-Papilionoideae clade comprises most papilionoid legumes and includes small clades with heterogeneous floral morphologies. Some species have a sessile ovary, while in others the gynoecium is elevated by a stalk called a stipe or gynophore. This study provides a qualitative and comparative morphological analysis of meso-papilionoid flowers, focusing on the anatomy, vascularization, and development of the ovary and gynophore. The objective is to unravel the ontogenic origin and anatomical nature of the gynophore in meso-papilionoid flowers. Floral buds at different developmental stages o

The Ethylene Biosynthesis Gene CpACO1A: A New Player in the Regulation of Sex Determination and Female Flower Development in Cucurbita pepo.

A methanesulfonate-generated mutant has been identified in Cucurbita pepo that alters sex determination. The mutation converts female into hermaphrodite flowers and disrupts the growth rate and maturation of petals and carpels, delaying female flower opening, and promoting the growth rate of ovaries and the parthenocarpic development of the fruit. Whole-genome resequencing allowed identification of the causal mutation of the phenotypes as a missense mutation in the coding region of CpACO1A, which encodes for a type I ACO enzyme that shares a high identity with Cucumis sativus CsACO3 and Cucumi

A hypersensitive response-like reaction is involved in hybrid weakness in F1 plants of the cross Capsicum annuum × Capsicum chinense.

Hybrid weakness in Capsicum is characterized by the termination of leaf differentiation after the development of several leaves. F1 plants in some crosses between Capsicum annuum and Capsicum chinense show weakness; this phenomenon has not been investigated in detail since first reported. In the present study, we characterized morphologically and physiologically hybrid weakness in Capsicum. F1 plants did not show weaker growth than their parents 20 days after germination (DAG), but at 40 DAG, the hybrid weakness phenotype was evidenced by almost complete arrest of new leaf formation, delayed i

[Effect of antioxidant-ambiol on the radiation adaptive response].

For studying of the mechanism of adaptive response of plants the seeds of soft wheat Triticum aestivum cultivar Moscovskaya 39 were irradiated in doses 0.25, 50 and 0.25 + 50 Gy and the frequency of cells with aberrations and the mitotic activity in the meristem of seedlings were scored. The pre- and post-treatments of seeds with antioxidant--ambiol were also used. It was found that the exposure of seeds to 0.25 Gy reduce the effects of challenge dose of 50 Gy: the mitotic index increases and the frequency of cells with aberrations decreases--the adaptive response appear.
