Glyphosate-based herbicide metabolic profiles in human urine samples through proton nuclear magnetic resonance analysis.

BACKGROUND AND PURPOSE: Glyphosate-based herbicides, extensively utilized worldwide, raise concerns regarding potential human risks due to the detection of glyphosate (GLY) in human body fluids. This study aims to address critical knowledge gaps regarding whether GLY undergoes metabolism in humans, particularly considering the limited information available on human metabolism. EXPERIMENTAL APPROACH: The study investigated GLY and its metabolites in eight amenity horticultural workers using proton nuclear magnetic resonance (1H-NMR) data analysis. Multiple spot urine samples were collected bef

Potential of biogas residue biochar modified by ferric chloride for the enhancement of anaerobic digestion of food waste.

Biogas residue biochar (BRB) and BRB modified by ferric chloride (BRB-FeCl3) were applied to promote anaerobic digestion (AD) of food waste (FW), related mechanisms were also proposed in this study. Results indicated BRB-FeCl3 showed higher specific surface area, more abundant functional groups and impregnate iron than BRB, and they respectively increased 22.50% and 12.79% cumulative methane yields compared with control group because of accelerated volatile fatty acids (VFAs) transformation, which were confirmed by enhanced metabolism of glycolysis, fatty acid degradation and pyruvate. The electronic nature of terminal oxo ligands in transition-metal complexes: ambiphilic reactivity of oxorhenium species.

The synthesis of the Lewis acid-base adducts of B(C6F5)3 and BF3 with [DAAmRe(O)(X)] DAAm = N,N-bis(2-arylaminoethyl)methylamine; aryl = C6F5 (X = Me, 1, COCH3, 2, Cl, 3) as well as their diamidopyridine (DAP) (DAP=(2,6-bis((mesitylamino)methyl)pyridine) analogues, [DAPRe(O)(X)] (X = Me, 4, Cl, 5, I, 6, and COCH3,7), are described. In these complexes the terminal oxo ligands act as nucleophiles. In addition we also show that stoichiometric reactions between 3 and triarylphosphine (PAr3) result in the formation of triarylphosphine oxide (OPAr3). The electronic dependence of this reaction was st

Characterization of serum complement activity of saltwater (Crocodylus porosus) and freshwater (Crocodylus johnstoni) crocodiles.

We employed a spectroscopic assay, based on the hemolysis of sheep red blood cells (SRBCs), to assess the innate immune function of saltwater and freshwater crocodiles in vitro. Incubation of serum from freshwater and saltwater crocodiles with SRBCs resulted in concentration-dependent increases in SRBC hemolysis. The hemolytic activity occurred rapidly, with detectable activity within 2 min and maximum activity at 20 min. These activities, in both crocodilian species, were heat sensitive, unaffected by 20 mM methylamine, and completely inhibited by low concentrations of EDTA, suggesting that t

NMR studies of solvent-assisted proton transfer in a biologically relevant Schiff base: toward a distinction of geometric and equilibrium H-bond isotope effects.

The tautomeric equilibrium in a Schiff base, N-(3,5-dibromosalicylidene)-methylamine 1, a model for the hydrogen bonded structure of the cofactor pyridoxal-5'-phosphate PLP which is located in the active site of the enzyme, was measured by means of 1H and 15N NMR and deuterium isotope effects on 15N chemical shifts at variable temperature and in different organic solvents. The position of the equilibrium was estimated using the one-bond 1J(OHN) and vicinal 3J(H(alpha)CNH) scalar coupling constants. Additionally, DFT calculations of a series of Schiff bases, N-(R1-salicylidene)-alkyl(R2)amines,

Exogenous coreactant-free integrated ECL biosensor based on P5AIn/Fe(II)-MOF for sensitive trimethylamine N-oxide detection.

BACKGROUND: Trimethylamine N-oxide (TMAO) has attracted significant attention as a critical marker for seafood freshness. However, the development of integrated methods combining rapid sample pretreatment with highly sensitive detection for TMAO remains a significant gap. A novel exogenous coreactant-free integrated ECL immunosensor was constructed for sensitive TMAO detection using poly(5-aminoindole)/Iron-based metal-organic framework (P5AIn/Fe-MOF) as a dual-functional platform. RESULTS: In the P5AIn/Fe-MOF composite, the Fe-MOF served as both a co-reaction promoter and an in-situ reductio

The association of trimethylamine N-oxide with diabetic retinopathy Pathology: Insights from network toxicology and molecular docking analysis.

Trimethylamine N-oxide (TMAO), a gut microbiota-derived metabolite, has emerged as a potential contributor to diabetic retinopathy (DR) progression. However, its molecular mechanisms in DR remain unclear. This study integrates network toxicology and multi-omics analyses to elucidate TMAO's role in DR pathogenesis. We identified TMAO-related targets through integration of CTD, SuperPred, and GeneCards databases. Differential expression analysis of DR-related genes was performed using GSE60436 and GSE102485 datasets. We intersected these with TMAO targets to identify key genes. Functional enrich

TMAO converts cytochrome c into a pro-apoptotic peroxidase by destabilizing the heme-Met80 ligation.

Trimethylamine N-oxide (TMAO), a gut microbiota-derived metabolite, has been linked to cardiovascular, renal, and hepatic disorders, but its direct impact on mitochondrial apoptotic machinery remains unclear. Here, we show that TMAO binds cytochrome c (Cyt c), disrupting its structural integrity and converting it into an apoptotically competent species. Spectroscopic analyses revealed that TMAO destabilizes the heme-Met80 axial ligation, shifting Cyt c from its native hexacoordinate to a pentacoordinate state. This conformational change enhances peroxidase activity, exposes hydrophobic cluster

Stroke and Vascular Cognitive Impairment: The Role of Intestinal Microbiota Metabolite TMAO.

The gut microbiome interacts with the brain bidirectionally through the microbiome-gutbrain axis, which plays a key role in regulating various nervous system pathophysiological processes. Trimethylamine N-oxide (TMAO) is produced by choline metabolism through intestinal microorganisms, which can cross the blood-brain barrier to act on the central nervous system. Previous studies have shown that elevated plasma TMAO concentrations increase the risk of major adverse cardiovascular events, but there are few studies on TMAO in cerebrovascular disease and vascular cognitive impairment. [Effect of traditional Chinese medicine in attenuating chronic kidney disease and its complications by regulating gut microbiota-derived metabolite trimethylamine N-oxide: a review].

Trimethylamine N-oxide(TMAO), a metabolite of gut microbiota, is closely associated with chronic kidney disease(CKD). It can aggravate the kidney injury and promote the occurrence of complications of CKD mainly by inducing renal fibroblast activation, vascular endothelial inflammation, macrophage foaming, platelet hyperreactivity, and inhibition of reverse cholesterol transport. Thus it is of great significance for clinical treatment of CKD to regulate circulating TMAO and alleviate its induced body damage. Currently, therapeutic strategies for TMAO regulation include dietary structure adjustm

[Gut microbiome in heart failure and aortic stenosis].

The gut microbiome is linked to the development of individual diseases. Patients with congestive heart failure (HF) develop intestinal wall edema due to venous congestion, which impairs absorption function and allows bacterial overgrowth. Consequently, the pathogenous bacterial strains produce many harmful substances, including trimethylamine N-oxide (TMAO) and endotoxin (LPS - lipopolysaccharide), which lead to deterioration of HF. These discoveries led to hypothesis about the heart-bowel axis. High levels of TMAO present in patients with HF predispose to higher long-term mortality, even afte

Retrospective identification of new psychoactive substances in patient samples submitted for clinical drug analysis.

New psychoactive substances (NPS) are life threatening through unpredictable toxicity and limited analytical options for clinicians. We present the retrospective identification of NPS in raw data from a liquid chromatography-high resolution mass spectrometry (LC-HRMS)-based multidrug panel analysis on 14 367 clinical oral fluid samples requested during 2019 mainly by psychiatric and addiction care clinics. Retrospectively analysed NPS included 48 notified originally in 2019 by the European Union Early Warning System (EU EWS) and 28 frequently reported in Sweden. Of 88 included NPS, 34 (mitragy

Proteoform-Resolved Cross-Linking Reveals Environment-Dependent Structural Effects of α-Synuclein S129 Phosphorylation.

Intrinsically disordered proteins (IDPs) drive many neurodegenerative disorders, but their structures remain difficult to define because they populate dynamic ensembles that change with the chemical environment. This problem is central for α-synuclein (aSyn), a Parkinson's disease-linked IDP in which S129 phosphorylation is highly enriched in disease-associated aggregates. Although S129 phosphorylation stabilizes a more compact aSyn ensemble in dilute solution, whether this structural effect persists across other biochemical environments has remained unknown. Here, we developed a quantitative

Signed, sealed, delivered: a generalizable model for living biotherapeutic dosing and metabolism.

Living Biotherapeutic Products (LBPs) offer a promising therapeutic strategy for metabolic disorders rooted in gut microbiome dysfunction, yet quantitative frameworks for predicting their efficacy remain underdeveloped. We introduce the Bacterial Compartment Absorption and Transit (BCAT) model, a pharmacokinetic-pharmacodynamic framework that couples probiotic transit, endogenous microbiome metabolism, and enzymatic transformation within a unified dose-optimization setting. Building on the classical CAT model, BCAT incorporates mechanistically-derived colon compartments and treats dosing time

Gut Microbial Choline TMA-Lyase CutC: From Metabolic Mechanism to a Novel Therapeutic Target for Diseases.

In recent years, the pivotal role of the gut microbiota and its metabolites in host health and disease has garnered increasing attention. Dietary phosphatidylcholine and choline are metabolized by gut bacteria to generate trimethylamine (TMA). Upon entering the bloodstream, TMA is oxidized by host liver enzymes to trimethylamine N-oxide (TMAO), a known independent risk factor for various systemic diseases, including atherosclerosis, thrombosis, and chronic kidney disease. Within this complex "diet-gut-host" metabolic axis, the microbial choline TMA-lyase (CutC) acts as the key rate-limiting en

Electric-Field Pixel Array Biosensor for Catalytic Ion Detection in Diabetic Nephropathy.

Trimethylamine N-oxide (TMAO) has recently emerged as a clinically relevant biomarker for the early diagnosis and risk stratification of chronic diabetic nephropathy and cardiovascular disease. Here, we report an electric-field-driven pixel array urine biosensor for the assessment of diabetic nephropathy and for elucidating the relationship among urine TMAO, glucose, and urine microalbumin-to-creatinine ratio (UACR). The purified trimethylamine N-oxide reductase (torA) enzyme catalyzes the reduction of TMAO to trimethylamine (TMA) and H2O, during which generated H+ ions modulate the local ioni

Dietary Nutrients, Gut Microbiota, and Cardiac Function: From Metabolic Mechanisms to Clinical Applications.

BACKGROUND: The heart depends on a continuous and flexible energy supply from fatty acids, glucose, and other substrates. Emerging evidence shows that gut microbiota-derived metabolites-such as trimethylamine-N-oxide (TMAO), short-chain fatty acids (SCFAs), secondary bile acids, indoles, phenylacetylglutamine (PAGln), and branched-chain amino acids-modulate cardiac metabolism and function. Although clinical evidence linking these metabolites to cardiovascular outcomes is expanding, most data remain associative, with limited causal or interventional proof. METHODS: A comprehensive narrative re
