Green surfactants reveal mechanistic principles governing polymeric microbubble formation and function.

Poly(butyl cyanoacrylate) microbubbles (PBCA MB) are clinically investigated ultrasound-responsive materials whose formation is governed by surfactant-mediated interfacial templating. Here, we used a chemically diverse panel of 14 commercially designated greener non-ionic surfactants to clarify how surfactant chemistry governs PBCA MB formation and performance. By combining physicochemical analysis, dissipative particle dynamics simulations, microscopy, drug-loading experiments, and acoustic characterization, we determined how the architecture of the hydrophobic moiety, PEG chain length, and h

Effect of ultrasound-microbubble exposure on acute myeloid leukemia cancer cell proteome.

The application of ultrasound in combination with microbubbles (USMB) induces a broad spectrum of bioeffects on cells including plasma membrane disruption, cytoskeleton rearrangement, influx/efflux of cytosolic/extracellular molecules, metabolic stress and signalling pathways. This study investigated the effect of USMB on cell proteomics in human acute myeloid leukemia cells. Cells (OCI-AML-5) in suspension were exposed to ultrasound (f = 1 MHz, PD = 20 μs and PRP = 1 ms for 120 s) with and without microbubbles (Definity at 1.7% v/v) at varying acoustic pressures. Following exposure, cells wer

Self-Powered Oxygen Microbubble Generator for Decontamination of Anaerobic Biofilm-Fouled Bioimplants.

Biomedical devices often feature a microgap: confined, minuscule spaces that foster bacterial infiltration and biofilm formation. For instance, peri-implantitis with prevalence rates of 4.7-45% at the patient level is a major complication driven by biofilm infections, characterized by chronic inflammation and implant failure. Anaerobic biofilm residing within the microgap serves as a major source of the peri-implantitis, but tools that remove the biofilm are lacking. Therefore, this study presents a novel preventive strategy employing self-powered microbubbler (SM) for targeted decontamination

Influence of Microbubble Properties and Carrier Frequency on Ultrasound Cavitation-Induced Flow Augmentation.

BACKGROUND: Cavitation of microbubble (MB) contrast agents produces shear-mediated vasodilation and an increase in tissue perfusion. These effects are mediated through endothelial and erythrocyte shear-sensitive pathways. Our aim was to determine whether augmentation of tissue perfusion produced by cavitation is influenced by the deformability of MBs, MB charge-mediated interaction with vascular cells, or ultrasound (US) frequency, which affects MB resonance. METHODS: In mice, highly deformable lipid-stabilized decafluorobutane MBs with neutral or cationic charge, as well as less-deformable M

Laser-induced microbubble as an in vivo valve for optofluidic manipulation in living Mice's microvessels.

Optofluidic regulation of blood microflow in vivo represents a significant method for investigating illnesses linked to abnormal changes in blood circulation. Currently, non-invasive strategies are limited to regulation within capillaries of approximately 10 μm in diameter because the adaption to blood pressure levels in the order of several hundred pascals poses a significant challenge in larger microvessels. In this study, using laser-induced microbubble formation within microvessels of the mouse auricle, we regulate blood microflow in small vessels with diameters in the tens of micrometers.

Microbubble-encapsulated Cobalt Nitrato Complexes for Ultrasound-triggerable Nitric Oxide Delivery.

Cobalt complexes exhibit versatile reactivity with nitric oxide (NO), enabling their utilization in applications ranging from homogeneous catalysis to NO-based modulation of biological processes. However, the coordination geometry around the cobalt center is complex, the therapeutic window of NO is narrow, and controlled NO delivery is difficult. To better understand the complexation of cobalt with NO, we prepared four cobalt nitrato complexes and present a structure-property relationship for ultrasound-triggerable NO release. We hypothesized that modulation of the coordination geometry by lig

Microbubble-based fabrication of resilient porous ionogels for high-sensitivity pressure sensors.

High-sensitivity flexible pressure sensors have obtained extensive attention because of their expanding applications in e-skins and wearable medical devices for various disease diagnoses. As the representative candidate for these sensors, the iontronic microstructure has been widely proven to enhance sensation behaviors such as the sensitivity and limits of detection. However, the fast and tunable fabrication of ionic-porous sensing elastomers remains challenging because of the current template-dissolved or 3D printing methods. Here, we report a microbubble-based fabrication process that enabl

Microbubble-Assisted Ultrasound for Drug Delivery to the Retina in an Ex Vivo Eye Model.

Drug delivery to the retina is one of the major challenges in ophthalmology due to the biological barriers that protect it from harmful substances in the body. Despite the advancement in ocular therapeutics, there are many unmet needs for the treatment of retinal diseases. Ultrasound combined with microbubbles (USMB) was proposed as a minimally invasive method for improving delivery of drugs in the retina from the blood circulation. This study aimed to investigate the applicability of USMB for the delivery of model drugs (molecular weight varying from 600 Da to 20 kDa) in the retina of ex vivo

Microbubble-Assisted Ultrasound for Imaging and Therapy of Melanoma Skin Cancer: A Systematic Review.

Recent technological developments in ultrasound (US) imaging and ultrasound contrast agents (UCAs) have improved diagnostic confidence in echography. In the clinical management of melanoma, contrast-enhanced ultrasound (CEUS) imaging complements conventional US imaging (i.e., high-resolution US and Doppler imaging) for clinical examination and therapeutic follow-up. These developments have set into motion the combined use of ultrasound and UCAs as a new modality for drug delivery. This modality, called sonoporation, has emerged as a non-invasive, targeted and safe method for the delivery of th

Use of Nitric Oxide Donor-Loaded Microbubble Destruction by Ultrasound in Thrombus Treatment.

In the presence of a vascular thrombus, the recovery of blood flow and vascular recanalization are very important to prevent tissue damage. An alternative procedure to thrombolysis is required for patients who are unable to receive surgery or thrombolytic drugs due to other physical conditions. Recently, the performance of thrombolysis combined with microbubbles has become an attractive and effective therapeutic procedure. Indeed, in a recent study, we demonstrated that, upon exposure to ultrasound, liposomes loaded with nitric oxide release agonists conjugated to microbubbles; therefore, ther

Quantification of microbubble-induced sonothrombolysis in an ex vivo non-human primate model.

BACKGROUND: In vitro studies with ultrasound (US) and microbubbles (MB) have reported that sono-thrombolysis can be achieved at high peak rarefactional acoustic pressure amplitudes (PRAPAs) using 0.25 and 1.05 MHz US frequencies. OBJECTIVE: The aim of the current study was to determine if these parameters work on an ex vivo physiological model of thrombosis. METHODS: A thrombogenic device was placed in an ex vivo chronic arteriovenous shunt in juvenile baboons. Platelet accumulation was measured by dynamic imaging of the device and the 10 cm thrombus tail with 111 In-labeled platelets. After

A New Mechanism of Light-Induced Bubble Growth to Propel Microbubble Piston Engine.

Radiation pressure refers to the momentum transfer of photons during light "particles" impacting a surface. The force is too small to drive microengines. Different from the classical radiation pressure, the indirect radiation pressure (Fm ) is introduced, coming from the momentum change of light-induced bubble expansion. Fm is shown to obey Fm ∼ (I·rb )2 , behaving faster growth of indirect radiation pressure versus light intensity I and bubble radius rb . An effective bubble size range is identified for Fm to suppress other forces for bubble in liquid. The top laser irradiation on nanofluid i

Optimization of microbubble enhancement of hyperthermia for cancer therapy in an in vivo breast tumour model.

We have demonstrated that exposing human breast tumour xenografts to ultrasound-stimulated microbubbles enhances tumour cell death and vascular disruption resulting from hyperthermia treatment. The aim of this study was to investigate the effect of varying the hyperthermia and ultrasound-stimulated microbubbles treatment parameters in order to optimize treatment bioeffects. Human breast cancer (MDA-MB-231) tumour xenografts in severe combined immunodeficiency (SCID) mice were exposed to varying microbubble concentrations (0%, 0.1%, 1% or 3% v/v) and ultrasound sonication durations (0, 1, 3 or

Magnetic Nanoliposomes as in Situ Microbubble Bombers for Multimodality Image-Guided Cancer Theranostics.

Nanosized drug delivery systems have offered promising approaches for cancer theranostics. However, few are effective to simultaneously maximize tumor-specific uptake, imaging, and therapy in a single nanoplatform. Here, we report a simple yet stimuli-responsive anethole dithiolethione (ADT)-loaded magnetic nanoliposome (AML) delivery system, which consists of ADT, hydrogen sulfide (H2S) pro-drug, doped in the lipid bilayer, and superparamagnetic nanoparticles encapsulated inside. HepG2 cells could be effectively bombed after 6 h co-incubation with AMLs. For in vivo applications, after prefere

Hypospadias is a congenital anomaly of the penis with an occurrence of approximately 1 in 200 boys, but the etiology of the majority of hypospadias has remained unknown. Numerous genes have been reported as having variants in hypospadias patients, and many studies on genetic deletion of key genes in mouse genital development have also been published. Until now, no comparative analysis in the genes related literature has been reported. The basic knowledge of penile development and hypospadias is mainly obtained from animal model studies. Heterogeneous staining in the liver parenchyma after the injection of perflubutane microbubble contrast agent.

This study aimed to characterize the features of heterogeneous staining in the liver after injection of perflubutane microbubble agent (Sonazoid(TM), 0.0075 mL/kg). Digitized hepatic contrast sonograms from 906 subjects were reviewed to assess time-related changes in heterogeneous staining and the possible association between this effect and the clinical backgrounds was analyzed. Heterogeneous staining was found in seven subjects (0.77%) on 15-min phase sonograms. The staining initially appeared as hyper-enhanced circular spots in the liver 10 min or later after the agent injection. The number
