Effects of Selenium in the Microcirculation of Fructose-Fed Hamsters.

BACKGROUND AND AIMS: Fructose intake is directly related to vascular dysfunction and it is a risk factor for the development of metabolic and cardiovascular diseases, such as obesity, diabetes, and hypertension. Selenium, a component of antioxidant enzymes, improves hyperglycemia and vascular function in diabetic animals. The aim of this study was to evaluate the effects of dietary selenium supplementation on microcirculatory and metabolic parameters of fructose-fed hamsters. METHODS AND RESULTS: Male hamsters (Mesocricetus auratus) had their drinking water substituted or not by 10% fructose

Placenta microstructure and microcirculation imaging with diffusion MRI.

PURPOSE: To assess which microstructural models best explain the diffusion-weighted MRI signal in the human placenta. METHODS: The placentas of nine healthy pregnant subjects were scanned with a multishell, multidirectional diffusion protocol at 3T. A range of multicompartment biophysical models were fit to the data, and ranked using the Bayesian information criterion. RESULTS: Anisotropic extensions to the intravoxel incoherent motion model, which consider the effect of coherent orientation in both microvascular structure and tissue microstructure, consistently had the lowest Bayesian infor

Evaluation of sublingual microcirculation in children with dengue shock.

OBJECTIVE: To report the sublingual microcirculation observed using Sidestream Dark Field imaging in two children with dengue shock. METHOD: Two children, aged 9 and 10 years, were admitted to the pediatric intensive care unit with dengue shock and multiple organ dysfunction. Sublingual microcirculation was assessed in each patient on the first and second days of shock and was assessed a final time when the patients were no longer in shock (on the day prior to extubation) using Sidestream Dark Field technology. The De Backer score and microvascular flow index were used for the analyses. RESU

Microcirculation in the healing of surgical wounds in the oral cavity.

The aim of this research is to evaluate in vivo the characteristics of microcirculation after taking a biopsy sample from the oral mucosa. 20 patients were recruited to the study and all underwent an oral mucosa biopsy for the excision of benign neoformations. The modifications in the oral microcirculation were evaluated in vivo in correspondence to the surgical site through videocapillaroscopy at three different times: 30 min before the biopsy; 48 h after the biopsy; and 7 days after the biopsy. The statistical significance was checked with the Mann-Whitney U-test (P<0.05). The analysis of vi

[Features of systemic microcirculation in patients with chronic dermatoses].

We investigated the parameters of microcirculation of eyes in women of 36-55 years age with atopic dermatitis, microbic eczema and diffuse alopecia. In persons with atopic dermatitis the signs of non-uniformity of capillary caliber and their branching were observed. These abnormalities were accompanied by an increase in the quantities of functioning anastomoses, loops and corners of capillaries. Abnormalities of microcirculation which accompany microbic eczema included alterations in microvessel parallelism, microaneurysm, venous sacculation, pathological forms of branching, vascular new growt

Healing process of venous ulcers: the role of microcirculation.

In order to describe adequately the process of healing in the intermediate degrees, we investigated microcirculatory changes in the venous ulcers at well-defined stages of wound repair. We investigated dynamic changes in microcirculation during the healing process of venous ulcers. Ten venous ulcers were investigated in three consecutive clinical stages of wound healing: non granulation tissue (NGTA), GTA and scar. Subpapillary microcirculation was measured by laser Doppler perfusion (LDP) imaging and expressed using LDP values in arbitrary units. Nutritive perfusion by capillary microscopy an

Hemorheological changes in microcirculation: their mechanism and measurement technique.

Blood fluidity in the capillaries is affected significantly in diseases such as cardiac and brain infarcts, diabetic gangrene and many others. In view of the importance of physiology and pathology of capillary circulation, the hemorheological characteristics of the capillary blood flow are discussed in this article. Also, a new diagnosing technique for blood fluidity disorders is proposed. A computerized system for image analysis and determining blood rheological disorders for clinical and experimental use has also been discussed.

The coronary microcirculation in cyanotic congenital heart disease.

BACKGROUND: Despite an appreciable increase in basal coronary blood flow in cyanotic congenital heart disease, flow reserve remains normal. We hypothesized that preservation of flow reserve resides in remodeling of the coronary microcirculation. Microcirculatory morphometric analyses were performed to test this hypothesis. METHODS AND RESULTS: Necropsy specimens from 4 sources were studied: (1) hearts from patients with Eisenmenger's syndrome (A; n=5), (2) structurally abnormal hearts with ventricular hypertrophy (B; n=8), (3) structurally normal hearts with ventricular hypertrophy (C; n=6),

[Role of microcirculation in multiorgan failure of infectious origin].

The normal host response to infection is a complex process which serves to localize and control bacterial invasion and to initiate repair of injured tissue. This inflammatory process is accompanied by activation of circulating and fixed phagocytic cells and generation of pro-inflammatory and anti-inflammatory mediators. Sepsis results when the prerequisite inflammatory response to infection becomes generalized, and thereby extends to involve otherwise normal tissue which is remote from the initial site of injury or infection. Sepsis is typically associated to cardiovascular alterations that ha

[Morphologic study of the microcirculation in connective tissue diseases].

Capillaroscopy is a non-invasive diagnostic test used to study microvascular abnormalities which are present in many disorders, particularly some rheumatic pathologies, such as connective tissue diseases. In systemic sclerosis, capillaroscopy allows detection of pathognomonic microvascular alterations. In other connective tissue diseases, including systemic lupus erythematosus, dermatopolymyositis, undifferentiated connective tissue diseases, and mixed connective tissue disease, the capillaroscopic patterns, although non-specific, can provide a valid support for the diagnosis. In Raynaud's phe

Skin microcirculation in healthy subjects and patients with arteriosclerosis.

The concept to use the human skin microcirculation as a pharmacological in-vivo test system is old; however, methods developed in the 50s have been abandoned because of side effects and/or use of radioactive substances. We describe a newly developed minimally invasive method that allows in-vivo pharmacology in the human skin microcirculation injecting very low doses of a substance of drug without any systemic effects. The double injection technique (DIT) bears the potential to predict the effects of a drug and/or the vascular reactivity or dysfunction of other less accessible areas of the circ

Alcohol and endogenous nitric oxide in hepatic microcirculation.

This study investigated the role of endogenous nitric oxide in the regulation of hepatic vascular tone in the presence of ethanol. In the perfused rat liver, upon the initiation of ethanol infusion into the liver, portal pressure was increased in a dose-dependent manner, reaching maximal levels in 2-5 min, then decreasing gradually. Simultaneous infusion of N(G)-monomethy 1-L-arginine, a nitric oxide synthesis inhibitor, enhanced this ethanol-induced increase in portal pressure. This enhancement was reversed by simultaneous infusion of a precursor of nitric oxide, L-arginine. These results sug

Reactive hyperemia of skin microcirculation in septic neonates.

Reactive hyperemia after 1 min of arterial occlusion was studied in back, thigh and heel skin of 40 preterm and full-term neonates using laser Doppler flowmetry. Twelve infants had clinical signs of septicemia, but normal laboratory tests at the time of fluxmetry. However, CRP, leukocyte count and the ratio of immature to total neutrophils increased during the following days and septicemia was confirmed by positive blood cultures (septic group). Seven neonates with clinical signs of septicemia had developed neither positive blood cultures nor laboratory signs (non-septic group). Fifteen were h

[Microcirculation of skeletal muscle after ischemia and reperfusion].

The postischemic reperfusion-damage in the microcirculation of skeletal muscle is characterized by 1) heterogenous capillary perfusion with capillary perfusion deficiency ("no reflow"), 2) microvascular accumulation of leukocytes, which adhere to the endothelium of postcapillary venules, and 3) increase of vessel permeability with swelling of endothelial cells and with interstitial edema. Two models of pathomechanism may be differentiated: The capillary perfusion-deficiency of the no-reflow phenomenon prolongs the duration of ischemia extending the period of postischemic reperfusion and induce

Dynamics of leukocyte-endothelium interactions in the splanchnic microcirculation.

In vivo dynamics of the interaction between leukocytes and the endothelium following direct activation of the white blood cells (WBCs), apart from possible endothelial cell activation, were studied in arterioles, capillaries, and venules of splanchnic tissue (rabbit omentum). WBCs were isolated using either density gradient or centrifugation techniques, labeled with fluorescent dyes, and exposed to physiological solutions with or without the chemoactivator N-formyl-methionyl-leucyl-phenlyalanine (FMLP). WBCs isolated using standard density gradient separation techniques rapidly disappeared fro

Human immunodeficiency virus (HIV) leukoencephalopathy and the microcirculation.

We studied the brains of three patients with acquired immune deficiency syndrome (AIDS), all of whom developed subacutely progressive dementia unassociated with opportunistic infection or neoplasm in the central nervous system. Computed tomographic (CT) scans of the head revealed cortical atrophy, ventricular dilation, and diffuse hypodensity of the centrum semiovale. On microscopic examination, the cerebral and cerebellar white matter in all cases showed diffuse and focal, angiocentric regions of myelin pallor, focal vacuolization, and extensive gliosis. The rat liver microcirculation in alcohol-induced hepatomegaly.

It has been suggested that hepatocyte enlargement can lead to compression of the extracellular space (sinusoidal and interstitial) and induce portal hypertension. However, this hypothesis has never been tested by measuring the vascular and extravascular spaces in the intact liver. The aim of the present study was to investigate the effects of chronic alcohol intake on the hepatic microcirculation using Goresky's multiple-indicator dilution technique in the isolated perfused rat liver. Female rat littermates were pair-fed either ethanol (n = 7) or an isocaloric carbohydrate diet (n = 7) for 21

Capillary plugging by granulocytes and the no-reflow phenomenon in the microcirculation.

Granulocytes are large, stiff viscoelastic cells that adhere naturally to the vascular endothelium. On their passage through the capillary network they have to be deformed, and recent evidence indicates that they may impose a significant hemodynamic resistance. The entry time of granulocytes into capillaries is about three orders of magnitude longer than that for red cells. Inside the capillary the granulocytes move with a lower velocity than red cells. Under conditions when the capillary perfusion pressure is reduced and/or elevated levels of inflammatory products are present that increase th

Increased permeability of hamster microcirculation to glycosylated albumin.

The permeability of the microcirculation to native and glycosylated albumin was tested in 25 non-diabetic Syrian hamsters. The microvasculature of the cheek pouch was studied by a fluorescent video-microscopy technique after the animals had been injected with fluorescent native or glycosylated albumin or with both. Native albumin remained in the cheek pouch microvasculature, whereas glycosylated albumin leaked out of the microvascular bed along the postcapillary and collecting venules. The extravascular leakage of glycosylated albumin, probably due to its electrical or conformational change, m
