Optical glucose sensor for microfluidic cell culture systems.

Glucose is the primary energy source of human cells. Therefore, monitoring glucose inside microphysiological systems (MPS) provides valuable information on the viability and metabolic state of the cultured cells. However, continuous glucose monitoring inside MPS is challenging due to a lack of suitable miniaturized sensors. Here we present an enzymatic, optical glucose sensor element for measurement inside microfluidic systems. The miniaturized glucose sensor (Ø 1 mm) is fabricated together with a reference oxygen sensor onto biocompatible, pressure-sensitive adhesive tape for easy integration

Learning muscle-health trajectories in ageing: Towards AI-guided organ chips.

Age-related decline in muscle health is commonly defined using cut-offs for strength, muscle quantity or quality, and physical performance. These measures are essential for diagnosis, but prevention and recovery require an understanding of how muscle responds to stress over time. A central question is when recovery remains possible and why older adults with similar clinical measurements follow different trajectories. Clinical cohorts, animal studies and static cell models each address part of this problem. None readily combines human-tissue relevance with controlled perturbation, repeated func

Liver Tissueoid on-a-Chip Modeling Liver Regeneration and Allograft Rejection.

The lack of physiologically relevant in vitro models remains a limitation in liver transplantation research. Progress in organ-on-a-chip technologies enables the generation of clinically translatable data in vitro. A vascularized liver tissueoid-on-a-chip (LToC) model is engineered to replicate human liver tissue's structural and functional features for modeling liver regeneration and allograft rejection. The LToC comprises a microfluidic device containing donor-matched human hepatic progenitor cells and intrahepatic portal vein endothelial cells embedded in a fibrin matrix and maintained in d

Tumor-on-chip and cancer microphysiological platforms: knowledge structure, thematic evolution, and the emergence of organoid-integrated models, 2010-2025.

BACKGROUND: Cancer microphysiological systems (MPS), including tumor-on-chip platforms, combine engineered microenvironments, controlled fluid transport, and tumor-relevant biological models for cancer modeling, drug testing, and translational research. Organoid-integrated chips represent one developing component within this broader field, but the publication growth, knowledge structure, and changing research priorities of cancer MPS and tumor-on-chip research have not been systematically mapped across major bibliographic databases. METHODS: We performed a dual-database bibliometric analysis

Design of a flexing organ-chip to model in situ loading of the intervertebral disc.

The leading cause of disability of all ages worldwide is severe lower back pain. To address this untreated epidemic, further investigation is needed into the leading cause of back pain, intervertebral disc degeneration. In particular, microphysiological systems modeling critical tissues in a degenerative disc, like the annulus fibrosus (AF), are needed to investigate the effects of complex multiaxial strains on AF cells. By replicating these mechanobiological effects unique to the AF that are not yet understood, we can advance therapies for early-stage degeneration at the cellular level. To th

Human liver-kidney model elucidates the mechanisms of aristolochic acid nephrotoxicity.

Environmental exposures pose a significant threat to human health. However, it is often difficult to study toxicological mechanisms in human subjects due to ethical concerns. Plant-derived aristolochic acids are among the most potent nephrotoxins and carcinogens discovered to date, yet the mechanism of bioactivation in humans remains poorly understood. Microphysiological systems (organs-on-chips) provide an approach to examining the complex, species-specific toxicological effects of pharmaceutical and environmental chemicals using human cells. We microfluidically linked a kidney-on-a-chip with

Bioengineering Ovarian Endocrine Function: From Follicle Modeling to Cell-Based Hormone Therapy.

Premature ovarian insufficiency (POI) is a complex endocrine disorder characterized by the loss of ovarian function before age 40, leading to infertility, hypoestrogenism, and systemic complications. Conventional hormone replacement therapies (HRTs) offer only partial relief, failing to replicate the dynamic feedback of endogenous hormone regulation. Advances in tissue engineering, stem cell technology, and microphysiological systems have catalyzed the development of bioengineered platforms that mimic native ovarian endocrine function. This review outlines progress from three-dimensional (3D)

Robust and reusable iridium oxide-modified FTO electrodes for long-term organ-on-a-chip monitoring.

Real-time monitoring in organ-on-a-chip (OoC) systems is critical for capturing dynamic drug responses, yet standard gold (Au) electrodes utilized in electrical cell-substrate impedance sensing (ECIS) suffer from opacity, high cost, and poor reusability. Here, we report a transparent, robust, and reusable sensing interface based on iridium oxide-modified fluorine-doped tin oxide (IrOx@FTO). By electrodepositing nanostructured IrOx onto FTO, we exploited its pseudocapacitive properties to significantly improve interfacial properties, enhancing sensitivity while preserving optical clarity. Engineering human synovial organoids as predictive ecosystems for disease modeling and therapeutic response in rheumatology.

The synovium is a critical site for the initiation and persistence of inflammatory joint diseases, including rheumatoid arthritis (RA), osteoarthritis (OA), and gout. Conventional 2D cultures and animal models fail to recapitulate the multicellular organization, mechanical regulation, and inter-tissue communication that shape human joint pathology. Synovial organoids and related microphysiological systems address these gaps by reconstructing stromal, macrophage, vascular, and immune components within controllable human 3D environments. Here, we review advances in synovial organoid engineering,

Engineered skeletal muscles for disease modeling and drug discovery.

Skeletal muscle is the largest organ of human body with several important roles in everyday movement and metabolic homeostasis. The limited ability of small animal models of muscle disease to accurately predict drug efficacy and toxicity in humans has prompted the development in vitro models of human skeletal muscle that fatefully recapitulate cell and tissue level functions and drug responses. We first review methods for development of three-dimensional engineered muscle tissues and organ-on-a-chip microphysiological systems and discuss their potential utility in drug discovery research and d

Current approaches and advances in placental toxicology.

Despite the crucial role of the placenta in supporting pregnancy and fetal development, research into its susceptibility to environmental exposures has been limited by methodological challenges. We review diverse approaches to studying placental biology and responses to chemical exposures, and provide a comprehensive assessment of traditional and emerging methodologies. Beginning with an overview of placental biology and species differences, we evaluate in vivo and in vitro models, and discuss their strengths and limitations. We examine advances, including placental transfer models, toxicokine

The organ-on-a-chip mirage: Animal replacement, plastic channels, and plastic claims: The False Narrative Behind the War on Animal Research.

Organ-on-a-chip platforms have been widely promoted as imminent replacements for animal research, a theme prominently featured in Science (August 14, 2025, pp. 676-679). This commentary critically examines these claims in light of the current evidence base. While microphysiological systems provide useful adjunctive data, their predictive scope is limited by selective validation, material sorption effects, reliance on nonspecific endpoints, and unverified assumptions about metabolic competence. Economic projections of industry-wide savings rest on optimistic modeling rather than empirical demon

Applications of in vitro new approach methodologies (NAMs) in drug development: A scoping analysis of titles of nonclinical study reports submitted to the FDA.

The impact of New Approach Methodologies (NAMs) in drug development remains incompletely characterized. This scoping analysis used keyword searches of titles of nonclinical study reports submitted to the Center for Drug Evaluation and Research (CDER) from 2011 through 2025, followed by manual curation and classification by platform type and nonclinical study category. Among 574,963 nonclinical study reports submitted during the study period, 1381 in vitro NAM-related reports (0.24%) were identified across 981 applications. Despite this low representation, the annual number of identified in vit

Three-dimensional osteogenic and chondrogenic systems to model osteochondral physiology and degenerative joint diseases.

Tissue engineered constructs have the potential to function as in vitro pre-clinical models of normal tissue function and disease pathogenesis for drug screening and toxicity assessment. Effective high throughput assays demand minimal systems with clearly defined performance parameters. These systems must accurately model the structure and function of the human organs and their physiological response to different stimuli. Musculoskeletal tissues present unique challenges in this respect, as they are load-bearing, matrix-rich tissues whose functionality is intimately connected to the extracellu

Materials for liver-on-a-chip: modules, properties, and future directions.

The liver is the primary hub of our body for metabolism, detoxification, and drug biotransformation. While conventional two-dimensional cell culture and animal models can only partly recapitulate the complex hepatic microenvironment for the study of the liver, Liver-on-a-chip (LiOC) systems can simulate livers by integrating microfluidics, cell biology and engineered microenvironments. It is crucial to apply materials with specific properties in appropriate places because no single material can meet all the requirements for fabrication of LiOC systems. Moreover, improper material selection may

Decoding cellular dynamics and microenvironmental responses in organ-on-chip systems through real-time sensing.

Organ-on-Chip (OoC) systems hold extraordinary promise for predictive drug development and disease modeling. Yet, their translational impact is constrained by several challenges, one of which is the inability to monitor cellular microenvironments and tissue behavior in real time, without destructive sampling. In the absence of real-time sensing capabilities, OoC characterization often relies on intermittent or endpoint measurements, where inputs and endpoints are known, but transient cellular mechanisms driving biological responses remain elusive. This ambiguity limits mechanistic insights and

New approach methodologies complement but cannot replace animal models in critical care medicine research.

Critical care research focuses on life-threatening conditions such as sepsis, trauma, hemorrhage, and burn injury, which account for millions of hospitalizations and hundreds of thousands of deaths annually in the United States alone. Recent policy initiatives by the US Food and Drug Administration and the National Institutes of Health have promoted new approach methodologies, including organoids, organ-on-chip platforms, and computational models, as alternatives to animal research. While new approach methodologies offer valuable tools for mechanistic investigation and screening applications,

Decoding Sebaceous Gland Biology Through Experimental Models: Progress, Limitations, and Future Directions.

The sebaceous gland (SG) is a highly dynamic skin appendage that plays a central role in maintaining skin homeostasis through lipid production, immune modulation, and interactions with the skin microbiome. Dysregulation of SG function is implicated in several dermatological disorders, including acne and other inflammatory skin diseases. Over the past decades, a wide range of experimental models has been developed to investigate SG biology and pathology. These include in vivo animal models, ex vivo human skin explants, primary sebocyte cultures, and immortalized sebocyte cell lines, each offeri
