Texture Analysis of Fibrous Meningioma Using Label-Free Multiphoton Microscopy.

Fibrous meningiomas, a common type of brain tumor, present surgical challenges due to their variable hardness, which is crucial for complete resection and patient prognosis. This study explores the use of label-free multiphoton microscopy (MPM) for the objective assessment of the texture of fibrous meningiomas. Fresh tumor samples from 20 patients were analyzed using both multichannel and lambda mode MPM, with quantitative image analysis algorithms determining collagen content and multi-peak spectral fitting providing additional optical collagen metrics. The study compared medium and hard fibr

Cortical ensembles selective for context.

Neural processing of sensory information is strongly influenced by context. For instance, cortical responses are reduced to predictable stimuli, while responses are increased to novel stimuli that deviate from contextual regularities. Such bidirectional modulation based on preceding sensory context is likely a critical component or manifestation of attention, learning, and behavior, yet how it arises in cortical circuits remains unclear. Using volumetric two-photon calcium imaging and local field potentials in primary visual cortex (V1) from awake mice presented with visual "oddball" paradigms

Intravital multiphoton microscopic imaging platform for ocular surface imaging.

The purpose of this study is to provide an intravital noninvasive multiphoton microscopic platform for long-term ocular imaging in transgenic fluorescent mice with subcellular resolution. A multiphoton microscopic system with tunable laser output was employed. We designed a mouse holder incorporated with stereotaxic motorized stage for in vivo three-dimensional imaging of ocular surface in 3 transgenic mouse line with fluorescent protein (FP) expression to visualize distinct structures. With our imaging platform and the expression of FPs, we obtained the three-dimensional images across the who

Two-photon imaging of cellular activities in oxygen sensing tissues.

The cellular oxygen sensing system of the body ensures appropriate adaptation of cellular functions toward hypoxia by regulating gene expression and ion channel activity. Two-photon laser microscopy is an ideal tool to study and prove the relevance of the molecular mechanisms within oxygen sensing pathways on the cellular and complex tissue or organ level. Images of hypoxia inducible factor 1 (HIF-1) subunit nuclear mobility and protein-protein interaction in living cells, of hypoxia-induced changes in membrane potential and intracellular calcium of live ex vivo carotid bodies as well as of ra

Intravital two-photon microscopy for studying the uptake and trafficking of fluorescently conjugated molecules in live rodents.

In this study, we describe an experimental system based on intravital two-photon microscopy for studying endocytosis in live animals. The rodent submandibular glands were chosen as model organs because they can be exposed easily, imaged without compromising their function and, furthermore, they are amenable to pharmacological and genetic manipulations. We show that the fibroblasts within the stroma of the glands readily internalize systemically injected molecules such as fluorescently conjugated dextran and BSA, providing a robust model to study endocytosis. We dynamically image the traffickin

Zonal hepatocellular responses to acute ethanol consumption: impacts on mitochondrial function and liver metabolism.

Hepatocellular mitochondrial depolarization (mtDepo) after ethanol (EtOH) increases respiration to stimulate EtOH detoxification. mtDepo also triggers mitophagy, which may contribute to alcohol-associated liver disease. This study characterized sublobular respiration and distribution of mtDepo and mitophagy after acute EtOH. C57BL/6J and GFP-LC3 transgenic mice were gavaged with 6 g/kg EtOH or vehicle and administered MitoTracker Red (MTR). Hepatocytes were zonally sorted by MTR fluorescence for assessment of oxygen consumption rates (OCRs). Cytochrome P4502E1 (CYP2E1) immunolabeling identifie

Replication and distribution of Toxoplasma gondii in the small intestine after oral infection with tissue cysts.

Natural infection by Toxoplasma gondii occurs via oral ingestion of tissue cysts that rupture in the small intestine, releasing zoites that infect locally before disseminating throughout the host. The studies presented here used fluorescent parasites combined with flow cytometry and multiphoton microscopy techniques to understand the events associated with parasite replication in the mucosa. At 3 days postinfection with tissue cysts, parasites were localized in small foci and flow cytometry revealed parasites present in macrophages, neutrophils, and monocytes in the lamina propria. By day 6 po

Label-free two-photon fluorescence lifetime imaging of FADH2 for rapid diagnosis and grading of glioma.

SIGNIFICANCE: Glioma diagnosis currently relies on histopathology, which provides essential morphological information but fails to capture the dynamic metabolic heterogeneity underlying tumor progression. AIM: This study seeks to establish a label-free fluorescence lifetime imaging strategy to characterize the metabolic microenvironment of gliomas, with the goal of achieving accurate tumor grading and real-time malignancy assessment. APPROACH: We developed a phasor plot-based two-photon fluorescence lifetime imaging microscopy (TP-FLIM) system. This imaging approach was utilized to evaluate

Dysregulated Epithelial Shedding Promotes Early Intestinal Hyperpermeability Following Major Burn Injury.

A major burn injury triggers systemic inflammation and metabolic responses beyond the skin. Early intestinal barrier failure can amplify postburn inflammation and organ dysfunction, yet the dynamic epithelial lesions that generate focal leakage in vivo remain unclear. We characterized villus-tip epithelial shedding, epithelial gap formation, and tracer leak sites early after burn injuries using intravital multiphoton fluorescence microscopy. Male C57BL/6 mice were assigned to control or burn groups (30%-35% total body surface area full-thickness burn; n = 6/group). Intestinal permeability was

AI-based digital pathology provides newer insights into lifestyle intervention-induced fibrosis regression in MASLD: An exploratory study.

BACKGROUND AND AIMS: Lifestyle intervention is the mainstay of therapy for metabolic dysfunction-associated steatohepatitis (MASH), and liver fibrosis is a key consequence of MASH that predicts adverse clinical outcomes. The placebo response plays a pivotal role in the outcome of MASH clinical trials. Second harmonic generation/two-photon excitation fluorescence (SHG/TPEF) microscopy with artificial intelligence analyses can provide an automated quantitative assessment of fibrosis features on a continuous scale called qFibrosis. In this exploratory study, we used this approach to gain insight

In Vivo Imaging of Inflamed Glomeruli Reveals Dynamics of Neutrophil Extracellular Trap Formation in Glomerular Capillaries.

Neutrophil extracellular traps (NETs) have been documented in glomeruli of patients with glomerulonephritis. However, the dynamics of NET formation in the glomerulus and their functional contribution to acute glomerular injury are poorly understood. Herein, we used in vivo multiphoton microscopy to investigate NET formation in the acutely inflamed glomerulus. Glomerular inflammation was induced using an antibody against the glomerular basement membrane. After induction of inflammation, multiphoton microscopy revealed that approximately 20% of glomeruli contained structures composed of extracel

A genetically encoded calcium indicator for chronic in vivo two-photon imaging.

Neurons in the nervous system can change their functional properties over time. At present, there are no techniques that allow reliable monitoring of changes within identified neurons over repeated experimental sessions. We increased the signal strength of troponin C-based calcium biosensors in the low-calcium regime by mutagenesis and domain rearrangement within the troponin C calcium binding moiety to generate the indicator TN-XXL. Using in vivo two-photon ratiometric imaging, we show that TN-XXL exhibits enhanced fluorescence changes in neurons of flies and mice. TN-XXL could be used to obt

Automatic and label-free identification of blood vessels in gliomas using the combination of multiphoton microscopy and image analysis.

Currently, the targeted treatment of tumor based on the tumor microenvironment is newly developed. Blood vessels are the key parts in the tumor microenvironment, which is taken as a new visible target for tumor therapy. Multiphoton microscopy (MPM), based on the second harmonic generation and two-photon excited fluorescence, is available to make the label-free analysis on the blood vessels in human gliomas. MPM can reveal the vascular morphological characteristics in gliomas, including vascular malformation, intense vascular proliferation, perivascular collagen deposition, perivascular lymphoc

Macroscopic and microscopic imaging modalities for diagnosis and monitoring of urogenital schistosomiasis.

Urogenital schistosomiasis remains a major global challenge. Optimal management of this infection depends upon imaging-based assessment of sequelae. Although established imaging modalities such as ultrasonography, plain radiography, magnetic resonance imaging (MRI), narrow band imaging, and computerized tomography (CT) have been used to determine tissue involvement by urogenital schistosomiasis, newer refinements in associated technologies may lead to improvements in patient care. Moreover, application of investigational imaging methods such as confocal laser endomicroscopy and two-photon micr

CdSe/CdS-quantum rods: fluorescent probes for in vivo two-photon laser scanning microscopy.

CdSe/CdS-Quantum-dots-quantum-rods (QDQRs) with an aspect ratio of ∼ 6 are prepared via the seeded growth method, encapsulated within a shell of crosslinked poly(isoprene)-block-poly(ethylene glycol) (PI-b-PEG) diblock copolymer, and transferred from the organic phase into aqueous media. Their photoluminescence quantum yield (PLQY) of 78% is not compromised by the phase transfer. Within a period of two months the PLQY of QDQRs in aqueous solution at neutral pH decreases only slightly (to ∼ 65%). The two-photon (TP) action cross sections of QDQRs (∼ 10(5) GM) are two orders of magnitude higher

Digital quantitation of bridging fibrosis and septa reveals changes in natural history and treatment not seen with conventional histology.

BACKGROUND AND AIMS: Metabolic dysfunction-associated steatohepatitis (MASH) with bridging fibrosis is a critical stage in the evolution of fatty liver disease. Second harmonic generation/two-photon excitation fluorescence (SHG/TPEF) microscopy with artificial intelligence (AI) provides sensitive and reproducible quantitation of liver fibrosis. This methodology was applied to gain an in-depth understanding of intra-stage fibrosis changes and septa analyses in a homogenous, well-characterised group with MASH F3 fibrosis. METHODS: Paired liver biopsies (baseline [BL] and end of treatment [EOT])

Quantitative evaluation of the human vocal fold extracellular matrix using multiphoton microscopy and optical coherence tomography.

Identifying distinct normal extracellular matrix (ECM) features from pathology is of the upmost clinical importance for laryngeal diagnostics and therapy. Despite remarkable histological contributions, our understanding of the vocal fold (VF) physiology remains murky. The emerging field of non-invasive 3D optical imaging may be well-suited to unravel the complexity of the VF microanatomy. This study focused on characterizing the entire VF ECM in length and depth with optical imaging.
