Reduced functional connectivity between V1 and inferior frontal cortex associated with visuomotor performance in autism.

Some recent evidence has suggested abnormalities of the dorsal stream and possibly the mirror neuron system in autism, which may be responsible for impairments of joint attention, imitation, and secondarily for language delays. The current study investigates functional connectivity along the dorsal stream in autism, examining interregional blood oxygenation level dependent (BOLD) signal cross-correlation during visuomotor coordination. Eight high-functioning autistic men and eight handedness and age-matched controls were included. Visually prompted button presses were performed with the prefer

The effects of temporal modification of second speech signals on stuttering inhibition at two speech rates in adults.

The recovery of 'gestural' speech information via the engagement of mirror neurons has been suggested to be the key agent in stuttering inhibition during the presentation of exogenous second speech signals. Based on this hypothesis, we expect the amount of stuttering inhibition to depend on the ease of recovery of exogenous speech gestures. To examine this possibility, linguistically non-congruent second speech signals were temporally compressed and expanded in two experiments. In Experiment 1, 12 participants who stutter read passages aloud at normal and fast speech rates while listening to s

In Vivo Expression of an SCA27A-linked FGF14 Mutation Results in Haploinsufficiency and Impaired Firing of Cerebellar Purkinje Neurons.

Autosomal dominant mutations in FGF14 , which encodes intracellular fibroblast growth factor 14 (iFGF14), underlie spinocerebellar ataxia type 27A (SCA27A), a devastating multisystem disorder resulting in progressive deficits in motor coordination and cognitive function. Mice lacking iFGF14 ( Fgf14 -/- ) exhibit similar phenotypes, which have been linked to iFGF14-mediated modulation of the voltage-gated sodium (Nav) channels that control the high frequency repetitive firing of Purkinje neurons, the main output neurons of the cerebellar cortex. To investigate the pathophysiological mechanisms

Antagonism of NMDA receptors but not AMPA/kainate receptors blocks bursting in dopaminergic neurons induced by electrical stimulation of the prefrontal cortex.

Evidence suggests that the prefrontal cortex (PFC) plays an important role in the burst activity of midbrain dopaminergic (DA) neurons. In particular, electrical stimulation of the PFC elicits patterns of activity in DA neurons, closely time-locked to the stimulation, which resemble natural bursts. Given that natural bursts are produced by the activity of excitatory amino acid (EAA)-ergic afferents, if PFC-induced time-locked bursts are homologues of natural bursts, EAA antagonists should attenuate them. Hence, the NMDA (N-methyl-D-aspartate) antagonist CPP (3-((+/-)-2-carboxypiperazin-4-yl)pr

Synchronous cortical oscillatory activity during motor action.

Oscillations of the motor cortex interact with similar activity of the spinal motoneuron pool in the 15-30 Hertz frequency range. Recent observations have demonstrated how this interaction affects the firing of single corticospinal neurons. The interaction, reflected as corticomuscular coherence, occurs for both distal and proximal muscles and it constitutes one connection in a larger web of oscillatory interactions, including several other motor areas in the cortex, thalamus, and cerebellum. New results cast light on the possible functional significance of this interaction. The rhythmic inter

[Sociophysiology: basic processes of empathy].

The aim of this review is to describe sociophysiological and social cognitive processes that underlie the complex phenomenon of human empathy. Automatic reflexive processes such as physiological contagion and action mirroring are mediated by the mirror neuron system. They are a basis for further processing of social signals and a physiological link between two individuals. This link comprises simultaneous activation of shared motor representations. Shared representations lead implicitly via individual associations in the limbic and vegetative system to a shared affective state. These processes

Observing and perceiving: A combined approach to induce plasticity in human motor cortex.

OBJECTIVE: To test whether action observation combined with peripheral nerve electrical stimulation was able to evoke plasticity in the primary motor cortex (M1). METHODS: The stimulation protocol consisted in the observation of a video showing repetitive thumb-index tapping movements (AO) combined with peripheral electrical nerve stimulation (PNS) delivered on the median nerve (AO-PNS). M1 excitability, measured by means of transcranial magnetic stimulation, was compared with that assessed after AO and PNS alone. RESULTS: M1 excitability increased after AO-PNS, whilst no modifications occur

Reduced connectivity between mentalizing and mirror systems in autism spectrum condition.

The mentalizing system and mirror system are thought to play important roles in inferring the internal mental states of others - a process known as mentalizing. Autism spectrum condition (ASC) is associated with difficulties in mentalizing. The aim of this study was to determine whether the behavioural difficulties in mentalizing associated with ASC can be explained by changes in functional connectivity between the mentalizing and mirror system. We recruited 40 adult participants (20 with ASC and 20 typically-developing). Brain activity was monitored using functional magnetic resonance imaging

Loss of Miro1-directed mitochondrial movement results in a novel murine model for neuron disease.

Defective mitochondrial distribution in neurons is proposed to cause ATP depletion and calcium-buffering deficiencies that compromise cell function. However, it is unclear whether aberrant mitochondrial motility and distribution alone are sufficient to cause neurological disease. Calcium-binding mitochondrial Rho (Miro) GTPases attach mitochondria to motor proteins for anterograde and retrograde transport in neurons. Using two new KO mouse models, we demonstrate that Miro1 is essential for development of cranial motor nuclei required for respiratory control and maintenance of upper motor neuro

KCTD10 regulates brain development by destabilizing brain disorder-associated protein KCTD13.

KCTD10 belongs to the KCTD (potassiumchannel tetramerization domain) family, many members of which are associated with neuropsychiatric disorders. However, the biological function underlying the association with brain disorders remains to be explored. Here, we reveal that Kctd10 is highly expressed in neuronal progenitors and layer V neurons throughout brain development. Kctd10 deficiency triggers abnormal proliferation and differentiation of neuronal progenitors, reduced deep-layer (especially layer V) neurons, increased upper-layer neurons, and lowered brain size. Mechanistically, we screene

Compressive streak microscopy for fast sampling of fluorescent reporters of neural activity.

SIGNIFICANCE: In vivo one-photon fluorescence imaging of calcium and voltage indicators expressed in neurons enables noninvasive recordings of neural activity with submillisecond precision. However, data acquisition speed is limited by the frame rate of cameras. AIM: We developed a compressive streak fluorescence microscope to record fluorescence in individual neurons at high speeds ( ≥ 200 frames per second) exceeding the nominal frame rate of the camera by trading off spatial pixels for temporal resolution. APPROACH: Our microscope leverages a digital micromirror device for targeted illum

NOCICEPTRA: Gene and microRNA Signatures and Their Trajectories Characterizing Human iPSC-Derived Nociceptor Maturation.

Nociceptors are primary afferent neurons serving the reception of acute pain but also the transit into maladaptive pain disorders. Since native human nociceptors are hardly available for mechanistic functional research, and rodent models do not necessarily mirror human pathologies in all aspects, human induced pluripotent stem cell-derived nociceptors (iDN) offer superior advantages as a human model system. Unbiased mRNA::microRNA co-sequencing, immunofluorescence staining, and qPCR validations, reveal expression trajectories as well as miRNA target spaces throughout the transition of pluripot

Phenotypes and peripheral mechanisms underlying inflammatory pain-related behaviors induced by BmK I, a modulator of sodium channels.

The integrated mechanisms of dynamic signaling of sodium channels involved in clinical pain are still not yet clear. In this study, a new rat inflammatory pain model was developed by using the unilateral intraplantar injection of BmK I, a receptor site 3-specific modulator of sodium channels from the venom of scorpion Buthus martensi Karsch (BmK). It was found that BmK I could induce several kinds of inflammatory pain-related behaviors including spontaneous pain companied with unique episodic paroxysms, primary thermal hypersensitivity, and mirror-image mechanical hypersensitivity with differe

EEG correlates of action observation in humans.

To investigate electrophysiological correlates of action observation electroencephalogram (EEG) was recorded while participants observed repetitive biological (human) or non-biological movements (at a rate of 2 Hz). Steady-state evoked potentials were analyzed and their neural sources were investigated using low resolution electromagnetic tomography analysis (LORETA). Results revealed significantly higher activation in the primary motor and premotor cortex, supplementary motor area as well as the posterior parietal cortices during observation of biological movements, supporting mirror properti

Central clocking.

The main questions in circadian neurobiology are: how many oscillators are involved; how are their daily oscillations generated and synchronized to the external world; and how do they signal time of day to the organism. The suprachiasmatic nuclei of the hypothalamus (SCN) are well established as the principal circadian oscillator of mammals. Their 10,000 or so 'clock' neurones drive our overt rhythms-the daily patterning we observe in our physiology and behaviour being mirrored perfectly by their spontaneous cycle of neuronal activity. However, they are not our only circadian oscillator, their

"Schrödinger's Rat": Testing Consciousness Under Anesthesia Alters Brain State and Neuronal Responses in Auditory Cortex.

PURPOSE: In anesthesia research and clinical practice, the level of consciousness is often assessed by stimulating subjects and observing their responses. It is possible that such stimulation alters the very state of consciousness being assessed. Whether this phenomenon is accompanied by specific cortical activities remains unknown. Here, we tested the effects of behavioral assessments on neuronal activity in the rat primary auditory cortex (A1) and perirhinal cortex (PrC). METHODS: Neuronal spiking activity was recorded from microwire arrays targeting A1 and/or PrC in rats (n = 9), whilst pr

Is emotional contagion special? An fMRI study on neural systems for affective and cognitive empathy.

Empathy allows us to simulate others' affective and cognitive mental states internally, and it has been proposed that the mirroring or motor representation systems play a key role in such simulation. As emotions are related to important adaptive events linked with benefit or danger, simulating others' emotional states might constitute of a special case of empathy. In this functional magnetic resonance imaging (fMRI) study we tested if emotional versus cognitive empathy would facilitate the recruitment of brain networks involved in motor representation and imitation in healthy volunteers. Decoding state-dependent cortical-cerebellar cellular functional connectivity in the mouse brain.

The cortex and cerebellum form multi-synaptic reciprocal connections. We investigate the functional connectivity between single spiking cerebellar neurons and the population activity of the mouse dorsal cortex using mesoscale imaging. Cortical representations of individual cerebellar neurons vary significantly across different brain states but are drawn from a common set of cortical networks. These cortical-cerebellar connectivity features are observed in mossy fibers and Purkinje cells as well as neurons in different cerebellar lobules, albeit with variations across cell types and regions. Brain activations during execution and observation of visually guided sequential manual movements in autism and in typical development: A study protocol.

Motor issues are frequently observed accompanying core deficits in autism spectrum disorder (ASD). Impaired motor behavior has also been linked to cognitive and social abnormalities, and problems with predictive ability have been suggested to play an important, possibly shared, part across all these domains. Brain imaging of sensory-motor behavior is a promising method for characterizing the neurobiological foundation for this proposed key trait. The present functional magnetic resonance imaging (fMRI) developmental study, involving children/youth with ASD, typically developing (TD) children/y
