Islet transplantation is a promising treatment option for patients with type 1 diabetes (T1D); however, the fate of the graft over time remains difficult to follow, due to the lack of available tools capable of monitoring graft rejection and inflammation prior to islet graft loss. Due to the challenges imposed by the location of the pancreas and the sparsely dispersed beta-cell population within the pancreas, currently, the clinical verification of beta-cell abnormalities can only be obtained indirectly via metabolic studies, which typically is not possible until after a significant deteriorat

Techniques for noninvasive imaging of specific disease-related molecular changes are being developed to enhance diagnosis and therapeutic decision making in the clinical setting, and to facilitate research efforts. Molecular imaging with contrast-enhanced ultrasonography relies on the detection of the acoustic signal produced by microbubble or nanoparticle agents that are targeted to sites of disease. This Review describes the basis for ultrasound molecular imaging, the unique features of contrast agent behavior or detector performance in relation to clinical or research needs, and the progres

BACKGROUND: The diagnosis of Parkinson disease (PD) is based on clinical criteria but misdiagnosis is as high as 25% of cases as confirmed by anatomic-pathologic studies. Since the introduction of in vivo molecular imaging techniques using Single-Photon Emission Computed Tomography and Positron Emission Tomography, the diagnosis of PD became more reliable by assessing dopaminergic and even nondopaminergic systems.
REVIEW SUMMARY: The purpose of this article is to critically review the current data on molecular neuroimaging focusing on the nigrostriatal circuitry and providing useful informati

The ability to visualize molecular processes and cellular regulators of complex pulmonary diseases such as asthma, chronic obstructive pulmonary disease (COPD), or adult respiratory distress syndrome (ARDS), would aid in the diagnosis, differentiation, therapy assessment and in small animal-based drug-discovery processes. Herein we report the application of normalized transillumination and fluorescence molecular tomography (FMT) for the noninvasive quantitative imaging of the mouse lung in vivo. We demonstrate the ability to visualize and quantitate pulmonary response in a murine model of LPS-

X-ray micro-CT is currently used primarily to generate 3D images of micro-architecture (and the function that can be deduced from it) and the regional distribution of administered radiopaque indicators, within intact rodent organs or biopsies from large animals and humans. Current use of X-ray micro-CT can be extended in three ways to increase the quantitative imaging of molecular transport and accumulation within such specimens. (1) By use of heavy elements, other than the usual iodine, attached to molecules of interest or to surrogates for those molecules. The accumulation of the indicator i

Diseases are biological processes, and molecular imaging with positron emission tomography (PET) is sensitive to and informative of these processes. This is illustrated by detection of biological abnormalities in neurological disorders with no computed tomography or MRI anatomic changes, as well as even before symptoms are expressed. PET whole body imaging in cancer provides the means to (i) identify early disease, (ii) differentiate benign from malignant lesions, (iii) examine all organs for metastases, and (iv) determine therapeutic effectiveness. Diagnostic accuracy of PET is 8-43% higher t

BACKGROUND: Intraoperative fluorescence molecular imaging (FMI) is increasingly used to distinguish benign from malignant tumors; however, existing quantitative methods are predominantly data-driven, requiring large datasets and exhibiting limited adaptability in rare diseases or low-incidence tumor surgeries. Furthermore, high false-positive rates and substantial inter-operator variability remain significant challenges, hindering further application of FMI-guided surgery. This study introduces a novel, self-referenced clustering framework that overcomes these limitations by leveraging interna

Diffuse intrinsic pontine glioma (DIPG) is most commonly diagnosed based on imaging criteria, with biopsy often reserved for pontine tumors with imaging features not typical for DIPG (atypical DIPG, 'aDIPG'). The histopathologic and molecular spectra of the clinical entity aDIPG remain to be studied systematically. In this study, thirty-three patients with newly diagnosed pontine-centered tumors with imaging inconsistent with DIPG for whom a pathologic diagnosis was subsequently obtained were included. Neoplasms were characterized by routine histology, immunohistochemistry, interphase fluoresc

PET using established and emerging molecularly targeted radiotracers has become integral to the evaluation and management of brain tumors, providing biologic information that complements conventional MRI to improve tumor characterization, treatment planning, and response assessment across a broad spectrum of neurooncologic diseases. Although [18F]FDG PET remains widely available, the technique is limited by high physiologic grey matter uptake. Somatostatin receptor (SSTR)-targeted PET, including DOTATATE PET, has greatly impacted meningioma evaluation through high tumor-to-background contrast,

Venous thromboembolism (VTE), manifesting as deep vein thrombosis and pulmonary embolism, is a common, recurrent and potentially fatal condition. Anticoagulation remains the cornerstone of therapy, yet determining optimal treatment duration (eg, decision to shorten or extend anticoagulant treatment) is difficult due to the uncertain balance between long-term bleeding risk and recurrence. Future advances in vascular nuclear molecular imaging could critically help support this decision-making process. Current VTE diagnostic imaging relies on structural or perfusion-based techniques to infer thro

Atherosclerosis and abdominal aortic aneurysm (AAA) share the same pathological triad, which includes chronic inflammation, immune cell infiltration, and extracellular matrix (ECM) degradation. Both diseases are characterized by the fact that the disease process may not be diagnosed until a life-threatening event occurs. Current imaging methods diagnose these conditions by assessing the degree of stenosis of the blood vessel, the plaque burden, and the aneurysm diameter. Unfortunately, these methods are not sufficient to measure biological activity, which is important to determine if such pote

Assessment of therapeutic response in oncology is fundamental for evaluating treatment efficacy, guiding clinical decision-making, and determining prognosis. Historically, response evaluation relied primarily on anatomical imaging criteria derived from tumor size changes on computed tomography (CT), most notably the Response Evaluation Criteria in Solid Tumors (RECIST) and its adaptations. However, advances in molecular imaging, particularly positron emission tomography (PET) using [18F]-2-fluoro-2-deoxyglucose ([18F]FDG) and other radiopharmaceuticals, have introduced new biomarkers capable o

Chemiluminescence eliminates interference from real-time excitation light, thereby maximizing the signal-to-noise ratio of imaging. However, the reported chemiluminescent scaffolds remain limited, and the development of novel and efficient chemiluminescent scaffolds is critical and urgent. Herein, we synthesized a series of peroxynitrite (ONOO-)-activated chemiluminescent probes based on a hemicyanine scaffold through molecular engineering, and systematically investigated optical properties as well as their response mechanisms. Upon reaction with ONOO-, the vinylene units in the hemicyanine sc
