Membrane-associated human tyrosinase is an enzymatically active monomeric glycoprotein.

Human tyrosinase (hTyr) is a Type 1 membrane bound glycoenzyme that catalyzes the initial and rate-limiting steps of melanin production in the melanosome. Mutations in the Tyr gene are linked to oculocutaneous albinism type 1 (OCA1), an autosomal recessive disorder. Currently, the application of enzyme replacement therapy for a treatment of OCA1 is hampered by the absence of pure hTyr. Here, full-length hTyr (residues 1-529) was overexpressed in Trichoplusia ni larvae infected with a baculovirus, solubilized with detergent and purified using chromatography. Michaelis-Menten kinetics, enzymatic

Ocular findings in patients with oculocutaneous albinism type ia with G47D tyrosinase gene mutation in Puerto Rico: a case report.

UNLABELLED: Previous studies have suggested that the G47D mutation leads patients to develop Oculocutaneous albinism (OCA) type IA. This mutation has been described in the Canary Islands. Historically, there has been a migration from the Canary Islands to some regions of Puerto Rico. OBJECTIVE: To report on the ocular findings of two Puerto Rican patients with OCA IA due to the G47D Tyrosinase gene mutation. PATIENT AND FINDINGS: Two unrelated patients with OCA underwent a comprehensive eye examination and were referred for genetic analysis. Patients had almost total iris transillumination, c

The melanogenesis and mechanisms of skin-lightening agents--existing and new approaches.

Skin-lightening products are commercially available for cosmetic purposes to obtain lighter skin complexion. Clinically, they are also used for treatment of hyperpigmentary disorders such as melasma, café au lait spot and solar lentigo. All of these target naturally melanin production, and many of the commonly used agents are known as competitive inhibitors of tyrosinase, one of the key enzymes in melanogenesis. In this review, we present an overview of commonly used skin-whitening ingredients that are commercialized, but we also hypothesize on other mechanisms that could be important targets

Molecular basis of type I (tyrosinase-related) oculocutaneous albinism: mutations and polymorphisms of the human tyrosinase gene.

Type I (tyrosinase related) oculocutaneous albinism (OCA) results from mutations of the tyrosinase gene on chromosome 11q that lead to reduced or absent melanin pigment synthesis. The phenotype of Type I OCA is broad, ranging from a total lack to only a moderate reduction of melanin, and the phenotypic variation is associated with different mutant alleles at the tyrosinase locus. A total of 36 mutations have been identified in Type I OCA including 24 missense, 4 nonsense, and 8 frameshift mutations. The majority of affected individuals have been compound heterozygotes with different maternal a

Relationship between the biodegradative capability of soil micromycetes for pentachlorophenol and for pentachloronitrobenzene.

A collection of 1056 strains in our laboratory were incubated with various xenobiotics among which were two potent fungicides: pentachlorophenol (PCP) and pentachloronitrobenzene (PCNB). The production of extracellular phenoloxidases were examined, using a series of ten different reagents. On the whole, PCNB is less accessible to fungal degradation than PCP. Although no correlation was found between the biodegradative capability of individual fungal strains for PCP or for PCNB, when taxonomic groups were considered as a whole, the same activity profiles were found. Zygomycetes were the most ef

Characterization of human hairbulb tyrosinase: properties of normal and albino enzyme.

Human hairbulb tyrosinase from normally pigmented and tyrosinase-positive oculocutaneous albino (TPA) hairbulbs was studied by single hairbulb and by pooled hairbulb assay procedures. The response to temperature and pH was the same for TPA and normal enzyme. The Km for tyrosine as substrate and the Km or dopa as cofactor was the same for TPA and normal enzyme. These studies show that TPA tyrosinase is kinetically normal and that the defect with this form of albinism must be elsewhere in the melanin pathway.

A new amino acid, 3-(2,5-SS-dicysteinyl-3,4-dihydroxyphenyl)alanine, from the tapetum lucidum of the gar (Lepisosteidae) and its enzymic synthesis.

The tapetum lucidum of the alligator gar Lepisosteus was shown by t.l.c. to contain a new phenolic amino acid, which is apparently a major constituent of the reflecting material. It was isolated in a yield of 0.5 mg/eye and its physical and chemical characteristics, especially reductive hydrolysis with hydriodic acid giving dopa (3,4-dihydroxyphenylalanine) and cysteine, suggested that it might to SS-dicysteinyldopa. Tyrosinase oxidation of L-dopa in the presence of an excess of L-cysteine yielded, in addition to known 5- and 2-S-cysteinyldopa, the same amino acid as that isolated from the eye

Effects of neuromelanin buildup in rodent nigral dopamine neurons: implications for sex-biased vulnerability in Parkinson's disease.

Neuromelanin (NM) is a dark pigment that accumulates with age in human substantia nigra pars compacta (SNpc) dopamine (DA) neurons, conferring the dark look that inspired nigral area's name. Although NM has long been associated with Parkinson's disease (PD), as melanized neurons favorably degenerate during disease development, NM functions within SNpc DA neurons are still mostly elusive. Here, using an NM-producing rat model generated by viral vector-induced expression of human tyrosinase (hTyr), we inspected NM impact on survival and activity of nigral DA neurons, nigral mitochondrial functio

A click-compatible BmTyr platform for biotin-free proximity labeling and subcellular proteome profiling in primary T cells.

Proximity labeling has revolutionized the study of dynamic subcellular proteomes by enabling the capture of transient protein interactions within living cells, yet the application of existing platforms to hard-to-transfect primary cells remains challenging. Here, we leverage a bioorthogonal proximity labeling platform based on the copper-dependent tyrosinase BmTyr to profile subcellular proteomes in primary T cells. This system catalyzes the subcellular incorporation of an alkyne-phenol probe, enabling subsequent click-compatible conjugation to versatile azide-bearing tags for fluorescence ima

Discovery of novel tyrosinase-inhibitory peptides FW and LAW from bullfrog (Lithobates catesbeianus) muscle protein: In silico screening, inhibition kinetics, gastrointestinal stability, and anti-melanogenic efficacy in zebrafish.

Bullfrog (Lithobates catesbeianus) muscle protein remains an underexplored source of bioactive peptides. This study applied an integrated in silico-in vitro-in vivo workflow to identify tyrosinase (TYR)-inhibitory peptides from bullfrog muscle protein hydrolysates. Among five proteases evaluated, acid protease hydrolysis for 3 h produced the highest TYR-inhibitory activity. Following ultrafiltration and Sephadex G-15 gel filtration, the most active fraction (<1 kDa) was analyzed by LC-MS/MS, yielding 71 candidate peptide sequences. The dipeptide FW and tripeptide LAW were prioritized through P

An upside to chronic inflammation? Starvation reconfigures immune pathways in Manduca sexta, producing chronic inflammation that may help defend against bacterial infection.

Most animals face periods of food scarcity, and pathogens are ever-present. Therefore, selection will favour animals that can maintain immune defense during starvation, despite declining resources. Using the caterpillar Manduca sexta as a model system, we show how two days of starvation reconfigures the immune system. Metabolomic and proteomic analysis of the plasma showed that starvation shifted the immune system towards a more 'pro-inflammatory' state, with increased activity in the phenoloxidase/eumelanin pathways relative to controls. Melanization occurred more rapidly in starved caterpill

Selective Electrochemical Biosensors for Tyrosinase and Tyramine via Enzyme-Mediated Oxidation Reaction.

Tyrosinase, a copper-containing oxidase, holds promise as a critical biomarker for diverse skin-related infections and vitiligo and melanoma tumors. Beyond its role as a critical biomarker and in biological pigmentation, tyrosinase is also involved in the oxidation of biogenic amines such as tyramine. Tyramine as a tyrosinase substrate serves as a precursor in the synthesis of various neurotransmitters such as dopamine and norepinephrine and contributes to significant neurological processes. It is well-stated that tyrosinase could oxidize tyramine in the presence of oxygen to give dopamine as

Signal-On Detection of Dopamine and Tyrosinase Using Tris(hydroxypropyl)phosphine as a New Lucigenin Chemiluminescence Coreactant.

Dopamine (DA) is a very imperative neurotransmitter in our body, since it contributes to several physiological processes in our body, for example, memory, feeling, cognition, cardiovascular diseases, and hormone secretion. Meanwhile, tyrosinase is a critical biomarker for several dangerous skin diseases, including vitiligo and melanoma cancer. Most of the reported chemiluminescent (CL) methods for monitoring DA and tyrosinase are signal-off biosensors. Herein, we introduce a new chemiluminescent "signal-on" system, lucigenin-tris(hydroxypropyl)phosphine (THPP), for the selective determination

Design, Synthesis, and Biological Evaluation of Novel Hybrids Containing Dihydrochalcone as Tyrosinase Inhibitors to Treat Skin Hyperpigmentation.

Excessive melanin deposition may lead to a series of skin disorders. The production of melanin is carried out by melanocytes, in which the enzyme tyrosinase performs a key role. In this work, we identified a series of novel tyrosinase inhibitor hybrids with a dihydrochalcone skeleton and resorcinol structure, which can inhibit tyrosinase activity and reduce the melanin content in the skin. Compound 11c possessed the most potent activity against tyrosinase, showing IC50 values at nanomolar concentration ranges, along with significant antioxidant activity and low cytotoxicity. Furthermore, in vi

The anti-melanogenic properties of Swietenia macrophylla king.

Fair flawless skin is the goal for some cultures and the development of irregular skin pigmentation is considered an indication of premature skin aging. Hence, there is a rising demand for skin whitening cosmetics. Thus, this research will be focusing on discovering the anti-pigmentation properties of Swietenia macrophylla seeds. Firstly, the seeds were extracted with ethanol and further fractionate based on their polarity before testing them on zebrafish embryos. The ethanolic extract of the seed demonstrated significant inhibition of both tyrosinase activity and melanin production in the emb

Antamanide Analogs as Potential Inhibitors of Tyrosinase.

The tyrosinase enzyme, which catalyzes the hydroxylation of monophenols and the oxidation of o-diphenols, is typically involved in the synthesis of the dark product melanin starting from the amino acid tyrosine. Contributing to the browning of plant and fruit tissues and to the hyperpigmentation of the skin, leading to melasma or age spots, the research of possible tyrosinase inhibitors has attracted much interest in agri-food, cosmetic, and medicinal industries. In this study, we analyzed the capability of antamanide, a mushroom bioactive cyclic decapeptide, and some of its glycine derivative
