Neisseria meningitidis, Neisseria lactamica and Moraxella catarrhalis share cross-reactive carbohydrate antigens.

Carriage of commensal bacteria species is associated with the development of natural immunity to meningococcal disease, with lipo-oligosaccharides (LOS) of meningococci being one of the main virulence factors associated with severity of meningococcal disease. Meningococcal reference strains and isolates from the commensal species Neisseria lactamica and Moraxella catarrhalis were assessed for the presence of cross-reactive glycoconjugate antigens. Binding of human blood group antibodies of the P and Ii system to meningococcal immunotype reference strains were in accordance with the presence of

Biochemical and immunological properties of lactoferrin binding proteins from Moraxella (Branhamella) catarrhalis.

The Neisseriaceae can acquire iron (Fe) from lactoferrin (Lf) using host-Lf receptors on the bacterial surface. The binding proteins that are proposed to constitute the receptor have been identified by isolation with immobilized Lf. Using CopB-specific monoclonal antibodies and isogenic CopB mutants, we demonstrate that the 84 kDa protein isolated with immobilized human Lf from Moraxella catarrhalis using low stringency conditions is CopB, an 84 kDa membrane-spanning protein with similarities to other TonB-dependent outer membrane proteins. Affinity isolation of Lf receptors from a variety of

Genetic diversity, virulence genes, and antimicrobial resistance of Moraxella bovoculi and Moraxella bovis from infectious bovine keratoconjunctivitis: insights from MALDI-TOF-MS, 16S rRNA analyses, and other ocular bacteria.

BACKGROUND: Moraxella bovis is the main etiological agent of infectious bovine keratoconjunctivitis (IBK). However, Moraxella bovoculi and other bacteria are also frequently isolated from IBK. The recurrent detection of M. bovoculi suggests a role in IBK, though its contribution remains unclear. This study aimed to characterize M. bovoculi and M. bovis isolates from IBK cases and evaluate the role of other ocular bacteria. METHODS: Eye swabs from cattle with keratoconjunctivitis were cultured and identified using MALDI-TOF MS, with 16S rRNA sequencing for confirmation. Virulence gene analysis

Moraxella catarrhalis Restriction-Modification Systems Are Associated with Phylogenetic Lineage and Disease.

Moraxella catarrhalis is a human-adapted pathogen, and a major cause of otitis media (OM) and exacerbations of chronic obstructive pulmonary disease. The species is comprised of two main phylogenetic lineages, RB1 and RB2/3. Restriction-modification (R-M) systems are among the few lineage-associated genes identified in other bacterial genera and have multiple functions including defense against foreign invading DNA, maintenance of speciation, and epigenetic regulation of gene expression. Here, we define the repertoire of R-M systems in 51 publicly available M. catarrhalis genomes and report th

Moraxella catarrhalis: from interactions with the host immune system to vaccine development.

Moraxella catarrhalis is a human-restricted commensal that over the last two decades has developed into an emerging respiratory tract pathogen. The bacterial species is equipped with various adhesins to facilitate its colonization. Successful evasion of the human immune system is a prerequisite for Moraxella infection. This strategy involves induction of an excessive proinflammatory response, intervention of granulocyte recruitment to the infection site, activation of selected pattern recognition receptors and cellular adhesion molecules to counteract the host bacteriolytic attack, as well as,

The Moraxella catarrhalis nitric oxide reductase is essential for nitric oxide detoxification.

Moraxella catarrhalis is a Gram-negative obligate aerobe that is an important cause of human respiratory tract infections. The M. catarrhalis genome encodes a predicted truncated denitrification pathway that reduces nitrate to nitrous oxide. We have previously shown that expression of both the M. catarrhalis aniA (encoding a nitrite reductase) and norB (encoding a putative nitric oxide reductase) genes is repressed by the transcriptional regulator NsrR under aerobic conditions and that M. catarrhalis O35E nsrR mutants are unable to grow in the presence of low concentrations of nitrite (W. Outbreaks of keratoconjunctivitis in a camel herd caused by a specific biovar of Moraxella canis.

Two tributyrin hydrolysis-negative Moraxella isolates obtained in cases of keratoconjunctivitis in Camelus dromedarius in the Canary Islands showed highest degrees of 16S rRNA gene sequence similarity to Moraxella canis. A level of DNA relatedness to the M. canis type strain of 79% confirmed the identity of the isolates as a tributyrin hydrolysis-negative biovar of M. canis.

The other siblings: respiratory infections caused by Moraxella catarrhalis and Haemophilus influenzae.

Respiratory infections remain substantial causes of morbidity and mortality globally. In this paper, two substantial players in bacterial-associated respiratory disease are assessed as to their respective roles in children and adults and in the developed and developing world. Moraxella catarrhalis, although initially thought to be a nonpathogen, continues to emerge as a cause of upper respiratory disease in children and pneumonia in adults. No vaccine is currently available to prevent M. catarrhalis infection. Haemophilus influenzae type b, originally thought to be the cause of influenza, has

Characterization of hemolysin of Moraxella bovis using a hemolysis-neutralizing monoclonal antibody.

A concentrated bacterial culture supernatant from the hemolytic Moraxella bovis strain UQV 148NF was used to immunize mice and generate monoclonal antibodies (MAbs). One, MAb G3/D7, neutralized the hemolytic activity of M. bovis and recognized a 94-kDa protein by Western blot analysis in hemolytic M. bovis strains representing each of the different fimbrial serogroups. Exposure of corneal epithelial cells to M. bovis concentrated culture supernatants demonstrated a role for an exotoxin in the pathogenesis of infectious bovine keratoconjunctivitis, while neutralization of hemolytic and cytotoxi

Q pili enhance the attachment of Moraxella bovis to bovine corneas in vitro.

Moraxella bovis, the causative agent of infectious bovine keratoconjunctivitis, exhibits several virulence factors, including pili, haemolysin, leukotoxin, and proteases. The pili are filamentous appendages which mediate bacterial adherence. Prior studies have shown that Q-piliated M. bovis Epp63 are more infectious and more pathogenic than I-piliated and non-piliated isogenic variants, suggesting that Q pili per se, or traits associated with Q-pilin expression, promote the early association of Q-piliated bacteria with bovine corneal tissue. In order to better evaluate the role of Q pili in M.

Neisseria meningitidis and Moraxella osloensis: dual infection in blood and peritoneal fluid.

The clinical course of a malnourished alcoholic in which Neisseria meningitidis was isolated from the blood and Moraxella osloensis from the peritoneal fluid is described. Following bacteriologic diagnosis, the patient was treated and responded to a course of penicillin therapy. To our knowledge, this represents the first case of peritonitis associated with M. osloensis. Clinical reports of the isolation of this organism are rare; its pathogenicity is not clearly established, and the presence of the organism may often be unrecognized.

Semiselective medium for isolation of Moraxella bovis from cattle with infectious keratoconjunctivitis.

The incorporation of 2.5 micrograms/ml of cloxacillin into 5% bovine blood agar provided an inexpensive, easily prepared culture medium for the primary isolation of Moraxella bovis from bovine lacrimal and nasal secretions. With this medium, the time required to identify and isolate M. bovis from large numbers of field specimens was substantially reduced, whereas the sensitivity of isolation was increased by 60%.

Characterization of a Heme-Binding Radical SAM Enzyme HemW from Moraxella catarrhalis.

Moraxella catarrhalis is an emerging human respiratory pathogen responsible for various infectious diseases. Recent transposon sequencing analysis identified yggW (renamed hemW) as essential for M. catarrhalis growth under iron-limiting conditions, mimicking host-imposed nutritional immunity. HemW is annotated as a putative radical S-adenosylmethionine (SAM) enzyme and belongs to the HemN-like subfamily, but its biochemical properties remain unclear. Here, we report on the first experimental characterization of M. catarrhalis HemW (McHemW). Our bioinformatic analysis confirmed its evolutionary

Moraxella oculi sp. nov., isolated from a cow with infectious bovine keratoconjunctivitis.

A novel species of the genus Moraxella was isolated from an ocular swab from a cow with infectious bovine keratoconjunctivitis. 16S rRNA gene sequencing suggested this species was Moraxella bovis (99.59 % nucleotide identity). Average nucleotide identity was calculated using a draft whole genome sequence of this strain compared with type strains of closely related Moraxella species and results established that it represents a new species. The genome size was 2 006 474 nucleotides and the G+C content was 42.51 mol%. The species could not be identified by matrix assisted laser desorption/ionizat
