Characterization of major structural proteins of measles virus with monoclonal antibodies.

We have prepared and characterized monoclonal antibodies against five major structural proteins, i.e. the HA, P, NP, F and M proteins, of measles virus. At least three non-overlapping antigenic sites were delineated on the HA protein, three on the P, four on the NP, four on the F and five on the M proteins by competitive binding assays. Antigenic sites on the HA and F proteins roughly represented functional domains defined by serological tests. The reactivity of monoclonal antibodies with various measles virus strains including those from subacute sclerosing panencephalitis (SSPE) and other me

Molecular insights from 2023 and 2024 outbreaks reveal exclusive circulation of peste des petits ruminants virus lineage IV in India.

Peste des petits ruminants (PPR) is a highly contagious disease affecting sheep and goats, caused by PPR virus (PPRV), a member of the genus Morbillivirus. In 2023 and 2024, on testing of suspected clinical samples for PPR received through passive surveillance of sheep and goat flocks in various states of India resulted in the documentation of 15 laboratory-confirmed outbreaks of PPR. During the outbreak investigation, morbidity and significant mortality rates were observed, and clinical samples collected from the affected flocks were tested using ELISA and RT-PCR, confirming the presence of P

Vaccination with recombinant adenovirus expressing peste des petits ruminants virus-F or -H proteins elicits T cell responses to epitopes that arises during PPRV infection.

Peste des petits ruminants virus (PPRV) causes an economically important disease that limits productivity in small domestic ruminants and often affects the livestock of the poorest populations in developing countries. Animals that survive PPRV develop strong cellular and humoral responses, which are probably necessary for protection. Vaccination should thus aim at mimicking these natural responses. Immunization strategies against this morbillivirus using recombinant adenoviruses expressing PPRV-F or -H proteins can protect PPRV-challenged animals and permit differentiation of infected from vac

Antigenic analysis of genetic variants of Canine distemper virus.

Canine distemper virus (CDV) is an RNA virus of the genus Morbillivirus within the family Paramyxoviridae. CDV produces multi-systemic disease in dogs and other terrestrial carnivores. With the development of modified live vaccines in the 1950s and 1960s, the disease, with a few exceptions, has been successfully controlled. However, recently the cases of CDV in vaccinated dogs have been increasing throughout the world, including the United States. There are many reasons that can lead to vaccine failure, including antigenic differences between the vaccine strains and the currently circulating w

History of measles.

Measles is a highly contagious viral disease transmitted by aerosols through human-to-human contact. It is often considered as a benign disease, although mortality remains high in developing countries (>5%). Frequent complications (diarrhea, otitis, pneumonia, encephalitis) can be observed. The disease mainly affects young children from 5 to 6 months of age with a mortality peak in the first three years of life. The Persian physician Rhazes gave the first clinical description of the disease in the 10th century, clearly differentiating it from smallpox. Measles spread worldwide from the Renaiss

The Entangled History of Sadoka (Rinderpest) and Veterinary Science in Tanzania and the Wider World, 1891-1901.

Scholarship on the Tanzanian Rinderpest epizootic of the 1890s has assumed that German colonizers understood from the start that they were confronting the same disease that had afflicted Eurasia for centuries. Outward indicators of the epizootic, known locally as sadoka, especially wildlife destruction, were unknown in Europe, leading German veterinarians to doubt that the African disease was Rinderpest. Financial constraints and conflicting development agendas, especially tension between ranching and pastoralism, deterred early colonial applications of veterinary science that might have led t

Post Mortem Findings of Cetaceans Stranded Along the Campania Coast from 2016 to 2022.

The presence of cetaceans along the Campania coast has always been documented. Between 2016 and 2022, out of 65 cetaceans stranded along the Campania coast, 46 were studied for bacteriological, virological, parasitological, and histopathological investigations. The results highlighted that for 59% (n = 27) of the specimens, the cause of death was of natural origin, while for only 2% (n = 1) of animals, the origin of death was ascribed to anthropic causes. Unfortunately, for 39% (n = 18) of the cetaceans, it was impossible to determine the cause of death. All the cetaceans that died of natural

Eradicating the Scourge of Peste Des Petits Ruminants from the World.

Peste des Petits Ruminants (PPR) is a highly contagious viral disease of both domestic (goats and sheep) and wild ruminants. Caused by a morbillivirus, that belongs to the family Paramyxoviridae. The disease is clinically and pathologically similar to rinderpest of cattle and human measles. PPR is one of the most economically devastating viral diseases of small ruminants. In April 2015, the Food and Agriculture Organization of the United Nations (FAO) and the World Organisation for Animal Health (OIE) launched the PPR Global Control and Eradication Strategy (PPR GCES) with the vision for globa

Novel alphaherpesvirus in a wild South American sea lion (Otaria byronia) with pulmonary tuberculosis.

In 2017, an adult male South American sea lion (Otaria byronia), presenting emaciation and a cervical abscess, stranded alive in Florianópolis, southern Brazil. The animal was directed to a rehabilitation center, dying a few days later. On necropsy, the main gross findings were necrotizing lymphadenitis of the right prescapular lymph node and nodular bronchopneumonia. A novel alphaherpesvirus, tentatively named Otariid alphaherpesvirus 1, was amplified in several tissue samples. No histopathologic findings associated with viral infection were observed. Additionally, pulmonary tuberculosis by M

Immunization With Bovine Herpesvirus-4-Based Vector Delivering PPRV-H Protein Protects Sheep From PPRV Challenge.

The Morbillivirus peste des petits ruminants virus (PPRV) is the causal agent of a highly contagious disease that mostly affects sheep and goats and produces considerable losses in developing countries. Current PPRV control strategies rely on live-attenuated vaccines, which are not ideal, as they cannot differentiate infected from vaccinated animals (DIVA). Recombinant vector-based vaccines expressing viral subunits can provide an alternative to conventional vaccines, as they can be easily paired with DIVA diagnostic tools. In the present work, we used the bovine herpesvirus-4-based vector (Bo

Emerging infectious diseases in cetaceans worldwide and the possible role of environmental stressors.

We reviewed prominent emerging infectious diseases of cetaceans, examined their potential to impact populations, re-assessed zoonotic risk and evaluated the role of environmental stressors. Cetacean morbilliviruses and papillomaviruses as well as Brucella spp. and Toxoplasma gondii are thought to interfere with population abundance by inducing high mortalities, lowering reproductive success or by synergistically increasing the virulence of other diseases. Severe cases of lobomycosis and lobomycosis-like disease (LLD) may contribute to the death of some dolphins. The zoonotic hazard of marine m

Prolonged Visual Evoked Potential Latencies in Dogs Naturally Infected with Canine Distemper Virus.

Canine distemper (CD) is a deadly, multi-system infection caused by a Morbillivirus. The canine distemper virus (CDV) frequently affects the nervous system with demyelinating leukoencephalitis, the most common neurological lesion. The disease has been linked to multiple sclerosis (MS) in humans due to similar clinical presentation and pathophysiological mechanisms. In MS, visual evoked potentials (VEPs) have been identified as a reliable marker for disease progression, enabling the early detection of clinically suspected lesions. The aim of this study was to determine if there are any abnormal

Subacute sclerosing panencephalitis (SSPE) associated with congenital measles infection.

A 13-month-old male presented with repetitive episodes of myoclonic jerks of the head and extremities for two months. His past medical history revealed that his non-immunized mother had measles at the time of delivery. Measles antibody titers in serum and cerebrospinal fluid (CSF) were 1/512 (hemagglutinin inhibition [HI]) and 1/128 HI, respectively. Immunofixation electrophoresis of CSF revealed an oligoclonal IgG band. The magnetic resonance imaging (MRI) of the brain on T2-weighted images showed lesions of high signal intensity in the subcortical white matter. Electroencephalography (EEG) r

[Current aspects of the pathogenesis and clinical characteristics of otosclerosis: possibilities of drug therapy].

Otosclerosis is a multifactorial disease. A number of theories on the pathogenesis of this disease have been established in the last decades. It is important to review recent data on the pathogenesis of otosclerosis as it is a severe inner ear disease leading to deafness in the majority of cases. Surgical therapy is not always successful or feasible. In this review, authors describe the most relevant genetic, infective, immunological, inflammatory factors, as well as the impaired bone metabolism underlying the pathogenesis of otosclerosis. It is likely that genetic predisposition associated wi

In vivo CAR T cell generation using retargeted and functionalized lentiviral vectors with reduced immunogenicity.

Despite striking efficacy against hematologic malignancies, the cost and complexity of CAR T manufacturing present significant barriers to broader patient access. Beyond manufacturing challenges, ex vivo expansion of T cells may be detrimental to their function and persistence. Thus, delivery of CARs to reprogram host cells in vivo would represent a significant advance towards a readily available therapy, but has been limited by low efficiency, low specificity, and immunogenicity of viral vectors. Here, we describe the design of pseudotyped lentiviral vectors (LV) with superior functionality a

Ginsenoside Rh1 suppresses Peste des petits ruminants virus replication by inhibiting autophagy to promote interferon responses and restrict inflammatory responses.

Peste des Petits Ruminants (PPR), a highly contagious disease of domestic and wild small ruminants, is characterized by severe morbidity and mortality. PPRV, the causative agent, is a morbillivirus in the family Paramyxoviridae. The virus poses a significant barrier to sustainable agricultural development in the developing world. Currently, no effective therapeutics agent for PPRV infection is available. Ginsenoside Rh1, a protopanaxadiol ginsenoside, the major pharmacological ingredient in the plants of ginseng, was reported to inhibit the replication of a broad range of human viruses. Howeve

Placentitis and abortion caused by canine distemper virus in a Linnaeus's two-toed sloth.

Canine distemper virus (CDV; family Paramyxoviridae, taxon species Morbillivirus canis) causes systemic disease in a broad range of mammalian hosts. CDV commonly targets epithelial, lymphoid, and nervous tissues. Reproductive disease is reported less frequently. Here, we describe CDV-associated placentitis and abortion in a 4-y-old, female Linnaeus's two-toed sloth (Choloepus didactylus) from a private collection. The animal developed respiratory and gastrointestinal illness, aborted a midgestational fetus, and subsequently died. Gross examination revealed necrotizing placentitis. Histological

When measles is not benign: meningoencephalitis and status epilepticus in an unvaccinated adolescent (case report).

Measles is a highly contagious viral exanthematous disease, caused by a Morbillivirus, that continues to cause outbreaks in under-immunized populations. While neurologic complications are rare, the progression to meningoencephalitis with convulsive status epilepticus is exceptional, particularly in immunocompetent adolescents. We report a 17-year-old unvaccinated male who presented with a febrile, descending asymptomatic maculopapular rash, lacking Koplik spots, followed by rapid onset of severe headache, photophobia, vomiting, and convulsive status epilepticus. Day-7 IgM was negative, and PCR
