Spontaneously hypertensive rats exhibit altered cardiovascular and neuronal responses to muscle contraction.

We examined the cardiovascular and ventrolateral medullary neuronal responses to muscle contraction in the spontaneously hypertensive rat (SHR) and normotensive Wistar-Kyoto rat (WKY) control. Cardiovascular, respiratory and ventrolateral medullary neuronal responses to muscle contraction evoked by tibial nerve stimulation were recorded. SHRs exhibited significantly larger drops in arterial pressure compared to WKYs in response to muscle contraction (P < 0.05). Basal ventrolateral medulla neuronal discharge rates were similar between the SHR and the WKY groups. A majority of neurons recorded r

Pliometric contraction-induced injury of mouse skeletal muscle: effect of initial length.

For single pliometric (lengthening) contractions initiated from optimal fiber length (Lf), the most important factor determining the subsequent force deficit is the work input during the stretch. We tested the hypothesis that regardless of the initial length, the force deficit is primarily a function of the work input. Extensor digitorum longus muscles of mice were maximally activated in situ and lengthened at 2 Lf/s from one of three initial fiber lengths (90, 100, or 120% of Lf) to one of three final fiber lengths (150, 160, or 170% of Lf). Maximal isometric force production was assessed bef

Does vaginal electrical stimulation cause pelvic floor muscle contraction? A pilot study.

The purpose of the present study was to evaluate whether vaginal electrical stimulation using Conmax and Medicon MS-105 causes pelvic floor muscle contraction. In addition, pain and discomfort described by the participants were registered. Nine women, mean age 37.7 years (range 24-54) participated in the study; five healthy physio-therapists and four patients with diagnosed genuine stress incontinence. All the participants used Conmax and Medicon MS-105 with 10, 20 and 50 Hz in random order. The women increased the current step by step to tolerance level. Two physiotherapists were observing th

Muscle function and protein metabolism after initiation of eccentric contraction-induced injury.

This study was designed to determine the relationship between skeletal muscle function and protein metabolism after initiation of eccentric contraction-induced injury. Mouse anterior crural muscles were injured in vivo, and then either immediately or 3, 6, 24, 48, 72, 120, or 336 h after injury muscles were isolated and studied for indexes of muscle function, injury, phagocyte infiltration, and protein metabolism. A group of mice were administered anti-polymorphonuclear cell and anti-macrophage antisera in an attempt to reduce phagocytic infiltration into injured muscle. Force production in ex

Lower airway smooth muscle contraction induced by Ptychodiscus brevis (red tide) toxin.

Ptychodiscus brevis toxin (PBTX) is produced by the organism Ptychodiscus brevis. This toxin causes a phenomenon that has come to be known as Florida red tide. It also stimulates neuronal sodium channels, resulting in activation of the cholinergic and adrenergic nerve endings of the autonomic nervous system in upper airway smooth muscle and rat vas deferens, respectively, as previously reported. It is the cholinergic stimulating action that has been implicated as a possible "triggering" event in bronchial asthma. This article concerns the investigation of whether PBTX may also affect lower air

Effects of inhibiting carbonic anhydrase on isometric contraction of frog skeletal muscle.

Carbonic anhydrase (CA) activity was determined in a homogenate of frog skeletal muscle by measuring the kinetics of CO2 hydration in a pH stopped-flow apparatus. The results suggest that frog skeletal muscle contains a high-activity CA with properties similar to those of the isoenzyme CA II found in white skeletal muscle tissue of the rabbit. In an attempt to assess the functional significance of CA in skeletal muscle, the maximal isometric force of frog gastrocnemius muscle was measured in response to direct or indirect (ischiadic nerve) single-pulse electrical stimulation before (control) a

The electrical basis for contraction and relaxation in canine fundal smooth muscle.

1. Mechanical and intracellular electrical activities were recorded simultaneously from canine fundal and antral smooth muscle preparations.2. Most fundal preparations displayed no spontaneous electrical or mechanical activity. The tissue had a space constant of 1.5 mm and a time constant of 189 msec and showed outward rectification in response to depolarizing current.3. Transmural nerve stimulation of fundal preparations demonstrated the presence of cholinergic excitatory and non-cholinergic, non-adrenergic inhibitory neural inputs to the tissue. The cholinergic nerve response consisted of a

Prevention of persistent cerebral smooth muscle contraction in response to whole blood.

Using an in vitro system designed to measure arterial constriction, we have demonstrated the importance of platelet function in maintaining cerebral smooth muscle contraction after whole blood injection. We tested two agents, acetyl salicylic acid (ASA) and phthalazinol, both known to interfere with platelet function. In control tests normal rabbit and monkey blood produced a reliable and persistent arterial constriction. In experimental tests blood drawn from animals premedicated with ASA and phthalazinol failed to produce a persistent contraction. These results support the hypothesis that ch

Changes in the kinetics of muscle contraction in vitamin D-depleted rats.

Using an in situ rat soleus neuromuscular preparation, changes in the muscle contraction kinetics in response to vitamine D depletion were studied. For a single isometric contraction, the time-to-peak tension (Tp) and the time-for-recovery-half-way-to-resting tension (T1/2r) were recorded. For a 150 Hz, 300 msec tetanus, the T1/2r was determined. Animals raised on high-calcium, high-phosphate, vitamin D-depleted diets showed prolongation of all parameters. Repletion of vitamin D returned Tp and T1/2r values to normal. Neither dietary calcium deficiency nor thyroparathyroidectomy produced an pr

Cerebellar transcranial static magnetic field stimulation reduces muscle activity during maximum contraction.

BACKGROUND: Transcranial static magnetic field stimulation (tSMS) is a noninvasive technique that modulates neuronal excitability without inducing an electrical current. Although triple-magnet tSMS has been shown to suppress cortical excitability in the motor cortex, its effects on cerebellar circuits and motor output remain unclear. OBJECTIVE: We examined whether cerebellar triple-magnet tSMS modulates voluntary muscle activation, cortical excitability, cerebellar output in healthy adults. METHODS: In this single-blind, randomized, sham-controlled study, 44 healthy participants received real-

Molecular genetics of skeletal muscle diseases associated with abnormal excitation-contraction coupling.

Skeletal muscle excitation-contraction coupling (ECC) is a highly specialized process that converts membrane depolarization into contraction through tightly regulated intracellular Ca²⁺ dynamics. Recent advances in molecular genetics have expanded the spectrum of skeletal muscle disorders associated with ECC-related proteins, including congenital myopathies, malignant hyperthermia susceptibility, exertional rhabdomyolysis, exertional heat illness, and related episodic disorders. Although these disorders have traditionally been classified according to clinical manifestations, pathological findi

Effects of eccentric contraction on force enhancement in rat fast-twitch muscle.

The aim of this study was to elucidate the effects of eccentric contraction (ECC) on force enhancement in rat fast-twitch skeletal muscle. Gastrocnemius (GAS) muscles were subjected to 200 ECCs in situ by electrical stimulation. Immediately before and after the stimulation, isometric torque produced by ankle flexion was measured at an ankle angle of 90°. After the second torque measurement, the superficial regions of the muscles were dissected and subjected to biochemical and skinned fiber analysis. ECC did not induce changes in the amount of degraded titin. After ECC, isometric torques in the

Signaling Pathways Mediating Bradykinin-Induced Contraction in Murine and Human Detrusor Muscle.

Bradykinin (BK) has been proposed to modulate urinary bladder functions and implicated in the pathophysiology of detrusor overactivity. The present study aims to elucidate the signaling pathways of BK-induced detrusor muscle contraction, with the goal of better understanding the molecular regulation of micturition and identifying potential novel therapeutic targets of its disorders. Experiments have been carried out on bladders isolated from wild-type or genetically modified [smooth muscle-specific knockout (KO): Gαq/11-KO, Gα12/13-KO and constitutive KO: thromboxane prostanoid (TP) receptor-K

Mechanisms of enhanced vascular smooth muscle contraction induced by sickle erythrocyte constituents.

The mechanisms of the increased vascular tone associated with vaso-occlusive crisis of sickle cell disease have not been clearly defined. The goal of the present study was to examine the role of vascular smooth muscle membrane Na+-K+-ATPase enzyme activity as well as nitric oxide synthase inhibition on the contractile responses induced by sickle erythrocyte constituents. 2 mm ring segments of rabbit carotid arterial ring preparations were placed in 20 ml organ baths containing physiological salt solution (PSS) bubbled with 95% O2, 5% CO2, at 37oC and pH 7.4 and isometric contractions recorded,

Effect of reperfusion on vascular smooth muscle reactivity in three contraction models.

BACKGROUND: Ischemia and reperfusion remain inseparable elements of numerous medical procedures such as by-pass surgery, organ transplantation or other cardiology and intervention radiology. The contraction of the smooth muscle of the vessel is considered to be one of the basic components leading to impaired perfusion, an increase in the oxygen deficit of the endothelium of the vessel, and subsequently also to tissues vascularized by the vessel. Main aim of this study was to evaluate the effect of ischemia and reperfusion on vascular smooth muscle cells stimulated pharmacologically with mastop

[Contraction mechanism of smooth muscle cells and its relationship with penile erection].

Penile erectile dysfunction (ED) is ascribed to the contraction-relaxation imbalance of smooth muscle cells (SMC), the weakening of their diastolic function and the strengthening of their systolic function. The contraction-related signaling pathways, cell membrane ion channels and SMC phenotypes all participate in the regulation of their contraction and its malfunction may cause a variety of SMC-related diseases. The signaling pathways RhoA/Rock and Raf/MEK/ERK1/2 interact with each other, suppressing the expression of the RhoA protein or reducing the level of Rock2 phosphorylation, which may
