Evaluation of the relationship between muscle mass and serum myostatin levels in chronic hemodialysis patients.

The loss of muscle mass and cachexia is commonly seen in hemodialysis (HD) patients and contribute to morbidity and mortality. The exact mechanism of this fact is multifactorial and still unclear. Myostatin, a transforming growth factor-ß family ligand, is released from the skeletal and heart muscle and may be responsible for muscle degradation and atrophy. The aim of this study is evaluation of the relationship between muscle mass and serum myostatin level in chronic HD patients. One hundred and forty HD patients (79 males, 28 diabetic, mean age; 53.96 ± 13.6) were included in this cross-sect

Expression of myostatin in human hematopoietic cells unveils novel autocrine/paracrine actions for the hormone.

Myostatin is a member of the transforming growth factor β (TGFβ) superfamily that has a well-established role as a mediator of muscle growth and development. However, myostatin is now emerging as a pleiotropic hormone with multiple actions in the regulation of the metabolism as well as several aspects of both cardiac and smooth muscle cells physiology. In addition, myostatin is also expressed in several nonmuscular cells where its physiological role remains to be elucidated in most cases. In this report, we have shown that both myostatin and its receptor system are expressed in blood cells and

Emerging Myostatin and Metabolic Hormone Pathway Modulators in Cardiometabolic Disease: Mechanisms, Evidence, and Therapeutic Potential.

Cardiometabolic disease comprises obesity, type 2 diabetes, heart failure, and sarcopenic obesity, which share interconnected mechanisms including muscle wasting, metabolic dysfunction, fibrosis, mitochondrial impairment, and chronic inflammation. Current therapies primarily target individual risk factors rather than these shared pathways. This review summarizes mechanistic and translational evidence on three emerging pathway modulators-myostatin (MSTN), fibroblast growth factor 21 (FGF21), and growth differentiation factor 15 (GDF15)-and their integration with incretin biology, AMPK/mTOR sign

Pre-Dialysis Serum Myostatin Concentration Is Independently Associated with Intradialytic Muscle Cramps in Hemodialysis Patients.

Muscle cramps are among the most common acute complications of hemodialysis (HD), often occurring even in the absence of electrolyte disturbances. Most cramps are painful and may cause patients to fear future dialysis sessions. We hypothesized that susceptibility to muscle cramps may be related to impaired skeletal muscle myokine secretion. No significant differences in pre-dialysis myokine levels were observed between patients with and without muscle cramps (myostatin 36.2 vs. 86.9 (p = 0.09), fibronectin type III domain-containing protein 5 (FNDC5) 0.6 vs. 0.6 (p = 0.61), interleukin-6 (IL-6

Myostatin-dependent detrusor remodelling mediates contractility impairment in diabetic rats' bladder - a maximal elastance analysis.

The role of myostatin, an endogenous myokine mediating the pathological progression of myopathy, in contractility impairments of the chronically diabetic bladder remains unclear. This study examined whether maximal elastance (mE) reliably and validly assesses bladder contractility in vivo and investigated the role of myostatin-associated tissue remodelling in the contractility deficits in diabetic bladder. Eighty-three female Sprague-Dawley rats were assigned to control (CON), streptozotocin-treated (STZ; 30 mg/kg twice, i.p.) and STZ with non-specific IgG (STZ + IgG; 2 mg/kg, i.p.) or Myo-029

Myostatin gene invalidation does not prevent skeletal muscle mass loss during experimental sepsis in mice.

Loss of muscle mass and function induced by sepsis contributes to physical inactivity and disability in intensive care unit patients. Limiting skeletal muscle deconditioning may thus be helpful in reducing the long-term effect of muscle wasting in patients. We tested the hypothesis that invalidation of the myostatin gene, which encodes a powerful negative regulator of skeletal muscle mass, could prevent or attenuate skeletal muscle wasting and improve survival of septic mice. Sepsis was induced by caecal ligature and puncture (CLP) in 13-week-old C57BL/6J wild-type and myostatin knock-out male

Myostatin knockout induces apoptosis in human cervical cancer cells via elevated reactive oxygen species generation.

Myostatin (Mstn) is postulated to be a key determinant of muscle loss and cachexia in cancer. However, no experimental evidence supports a role for Mstn in cancer, particularly in regulating the survival and growth of cancer cells. In this study, we showed that the expression of Mstn was significantly increased in different tumor tissues and human cancer cells. Mstn knockdown inhibited the proliferation of cancer cells. A knockout (KO) of Mstn created by clustered regularly interspaced short palindromic repeats (CRISPR)/CRISPR-associated protein (Cas) 9 (CRISPR/Cas9) induced mitochondria-depen

Decreasing maternal myostatin programs adult offspring bone strength in a mouse model of osteogenesis imperfecta.

During fetal development, the uterine environment can have effects on offspring bone architecture and integrity that persist into adulthood; however, the biochemical and molecular mechanisms remain unknown. Myostatin is a negative regulator of muscle mass. Parental myostatin deficiency (Mstntm1Sjl/+) increases muscle mass in wild-type offspring, suggesting an intrauterine programming effect. Here, we hypothesized that Mstntm1Sjl/+ dams would also confer increased bone strength. In wild-type offspring, maternal myostatin deficiency altered fetal growth and calvarial collagen content of newborn

Antibody-directed myostatin inhibition improves diaphragm pathology in young but not adult dystrophic mdx mice.

Duchenne muscular dystrophy (DMD) is characterized by progressive skeletal muscle wasting and weakness, leading to premature death from respiratory and/or cardiac failure. A clinically relevant question is whether myostatin inhibition can improve function of the diaphragm, which exhibits a severe and progressive pathology comparable with that in DMD. We hypothesized that antibody-directed myostatin inhibition would improve the pathophysiology of diaphragm muscle strips from young mdx mice (when the pathology is mild) and adult mdx mice (when the pathology is quite marked). Five weeks treatment

Creatine/Creatinine Ratio and Myostatin as Biomarkers to Monitor Muscle Function in Duchenne Muscular Dystrophy Patients.

BACKGROUND: Duchenne Muscular Dystrophy (DMD) is characterized by progressive muscle wasting leading to early loss of motor function. Functional tests monitor disease progression and serve as clinical trial endpoints but are influenced by maturation in younger patients, high intra-patient variability and patient motivation. Clinical milestones, instead, have been used to identify modifiers of disease progression but are rarely used as primary endpoints since clinical trials are usually too short to capture them. Blood biomarkers, that can reflect disease progression and objectively evaluate tr

Functional tests monitor disease progression and serve as clinical trial endpoints, but are influenced by maturation in younger patients, variability, and patient motivation. Blood biomarkers, that can predict disease progression and objectively evaluate treatment responses, offer a valuable alternative. In this study, we investigated whether longitudinal observations of biomarkers myostatin and the creatine/creatinine ratio (Cr/Crn) are associated with functional

Correlations of serum myostatin and irisin with sarcopenia and osteoporosis in rheumatoid arthritis patients: a cross-sectional study.

Myostatin and irisin, the key regulators of muscle metabolism, are myokines secreted by skeletal muscles that significantly influence osteoporosis (OP) and sarcopenia. This study investigated the associations of serum myostatin and irisin levels with sarcopenia and OP prevalence in 182 rheumatoid arthritis (RA) versus 142 healthy controls. RA patients exhibited significantly elevated serum myostatin (P = 0.001) and reduced irisin levels (P = 0.009), with markedly higher rates of myostatin/irisin dysregulation compared to controls (both P < 0.0001). Subgroup analyses revealed that RA patients w

Longitudinal Assessment of Creatine Kinase, Creatine/Creatinineratio, and Myostatin as Monitoring Biomarkers in Becker Muscular Dystrophy.

BACKGROUND AND OBJECTIVES: The slow and variable disease progression of Becker muscular dystrophy (BMD) urges the development of biomarkers to facilitate clinical trials. We explored changes in 3 muscle-enriched biomarkers in serum of patients with BMD over 4-year time and studied associations with disease severity, disease progression, and dystrophin levels in BMD. METHODS: We quantitatively measured creatine kinase (CK) using the International Federation of Clinical Chemistry reference method, creatine/creatinineratio (Cr/Crn) using liquid chromatography-tandem mass spectrometry, and myosta

Myostatin is a direct regulator of osteoclast differentiation and its inhibition reduces inflammatory joint destruction in mice.

Myostatin (also known as growth and differentiation factor 8) is a secreted member of the transforming growth factor-β (TGF-β) family that is mainly expressed in skeletal muscle, which is also its primary target tissue. Deletion of the myostatin gene (Mstn) in mice leads to muscle hypertrophy, and animal studies support the concept that myostatin is a negative regulator of muscle growth and regeneration. However, myostatin deficiency also increases bone formation, mainly through loading-associated effects on bone. Here we report a previously unknown direct role for myostatin in osteoclastogene

Mechanisms stimulating muscle wasting in chronic kidney disease: the roles of the ubiquitin-proteasome system and myostatin.

Catabolic conditions including chronic kidney disease (CKD), cancer, and diabetes cause muscle atrophy. The loss of muscle mass worsens the burden of disease because it is associated with increased morbidity and mortality. To avoid these problems or to develop treatment strategies, the mechanisms leading to muscle wasting must be identified. Specific mechanisms uncovered in CKD generally occur in other catabolic conditions. These include stimulation of protein degradation in muscle arising from activation of caspase-3 and the ubiquitin-proteasome system (UPS). These proteases act in a coordina

Cedrol derivative attenuates muscle atrophy through regulation of myostatin transcription via Ca2+-CaMK-FoxO3a signaling pathways.

Sarcopenia is a progressive and generalized muscle wasting syndrome characterized by loss of muscle strength and mass. Although many drug candidates have been developed to treat sarcopenia, their results were unsuccessful due to adverse or off-target effects. In this study, we identified a cedrol derivative which is a bioactive sesquiterpene having substantial suppressive effects on muscle atrophy. We demonstrated that the cedrol analog regulated myostatin expression via transcriptional regulation and that the cedrol derivative regulated this expression more effectively than the original form.

Pharmacological inhibition of myostatin effectively ameliorates osteolytic lesions in syngeneic and xenograft breast cancer mouse models.

Breast cancer (BC)-derived bone metastases colonize bone and drive severe bone degradation through complex interactions with bone-resorbing osteoclasts (OCs). Subsequent bone resorption liberates matrix-stored factors, such as TGF-β and calcium, which further stimulate tumor proliferation and exacerbate bone destruction. Myostatin (Mstn), a member of the TGF-β superfamily, is known to enhance OC differentiation and bone resorption in models of musculoskeletal disease; however, its role in BC-associated bone lesions and metastases remains unknown. Here, we demonstrate that bone metastases from

The aging myostatin null phenotype: reduced adiposity, cardiac hypertrophy, enhanced cardiac stress response, and sexual dimorphism.

The natural aging process results in the physiological decline of multiple tissues and organ systems. Changes commonly occur with middle age and include decreased skeletal muscle mass, bone mineral density, cardiac output, and insulin sensitivity, and increased adiposity, all of which can contribute to the onset of sarcopenia, osteoporosis, heart failure, or type 2 diabetes. Recent studies suggest that myostatin may influence many of these systems. We therefore sought to determine whether they are affected by aging, especially in 'middle-aged' Mstn-/- mice (12-20 months old (m.o.)). Although b
