A new myeloarchitectonic map of the human neocortex based on data from the Vogt-Vogt school.

The human cerebral cortex contains numerous myelinated fibres, the arrangement and density of which is by no means homogeneous throughout the cortex. Local differences in the spatial organization of these fibres render it possible to recognize areas with a different myeloarchitecture. The neuroanatomical subdiscipline aimed at the identification and delineation of such areas is known as myeloarchitectonics. During the period extending from 1910 to 1970, Oscar and Cécile Vogt and their numerous collaborators (The Vogt-Vogt school) published a large number of myeloarchitectonic studies on the co

Time-dependent changes in ghrelin-immunoreactivity in dissociated neuronal cultures of the newborn rat neocortex.

Ghrelin is a hormone, initially described as a gastric peptide stimulating appetite and growth hormone secretion, which also has an important role in the regulation of many other processes, including higher brain functions. Ghrelin has been described in situ in different parts of the brain, but so far there has been no data about its expression in cell cultures. Therefore, we aimed in this study to investigate the levels of ghrelin in dissociated cortical neurons at various times in culture. We applied the ABC immunocytochemical method for the detection of ghrelin in one-day-, one-week-, and t

Sensitivity to radiation-induced apoptosis and neuron loss declines rapidly in the postnatal mouse neocortex.

Therapeutic brain irradiation can cause progressive decline in cognitive function, particularly in children, but the reason for this effect is unclear. The study explored whether age-related differences in apoptotic sensitivity might contribute to the increased vulnerability of the young brain to radiation. Postnatal day 1 (P1) to P30 mice were treated with 0-16 Gy whole-body X-irradiation. Apoptotic cells were identified and quantified up to 48 h later using the TdT-UTP nick end-labelling method (TUNEL) and immunohistochemistry for activated caspase-3. The number of neuron-specific nuclear pr

Hypothyroidism alters the development of radial glial cells in the term fetal and postnatal neocortex of the rat.

Alterations of thyroid function during human development are known to produce extensive damage to the central nervous system including severe mental retardation. Using immunohistochemistry to identify the intermediate filament nestin, we have studied the possible influence of fetal and neonatal hypothyroidism on neocortical neuronal migration by arresting the normal development of the radial glial scaffold. By embryonic day 21 (E21), hypothyroid animals had a significant decrease in the number of nestin immunoreactive processes in the presumptive visual cortex. By postnatal day 5 (P5), hypothy

Functional autaptic neurotransmission in fast-spiking interneurons: a novel form of feedback inhibition in the neocortex.

Autapses are synapses made by a neuron onto itself. Although morphological evidence for existence of autapses has been reported in several brain areas, it is not known whether such self-innervation in the neocortex is functional and robust. Here we report that GABAergic autaptic activity is present in fast-spiking, but not in low-threshold spiking, interneurons of layer V in neocortical slices. Recordings made with the perforated-patch technique, in which physiological intracellular chloride homeostasis was unperturbed, demonstrated that autaptic activity has significant inhibitory effects on

Fibroblast growth factor 2 is necessary for the growth of glutamate projection neurons in the anterior neocortex.

Basic fibroblast growth factor (Fgf2) is required for the generation of founder cells within the dorsal pseudostratified ventricular epithelium, which will generate the cerebral cortex, but the ganglionic eminences are not affected. We report here that the Fgf2 null mutant mice show an approximately 40% decrease in cortical glutamatergic pyramidal neurons. In contrast, no change in pyramidal or granule cell number is detected in the hippocampus of Fgf2 -/- mice. In addition, the soma of the pyramidal cells in the frontal and parietal cortices are smaller in Fgf2 knock-out mice. The decrease in

Dendritic cytoskeletal protein expression in mental retardation: an immunohistochemical study of the neocortex in Rett syndrome.

Many syndromes associated with mental retardation (MR) are characterized by cortical dendritic anomalies. Despite their morphological similarity, these changes appear to involve different stages of dendritic development. The neuronal cytoskeleton, which includes microfilaments, neurofilaments and microtubules, is essential for these developmental processes. Levels and phosphorylation of microtubule-associated proteins (MAPs), which stabilize microtubules, seem to determine different stages of dendritic formation with certain MAPs (e.g. MAP-2) appearing to mediate the effects of external modula

Brn-5 is a divergent POU domain factor highly expressed in layer IV of the neocortex.

We have identified rat cDNA clones that encode a POU domain protein, referred to as brain-5 (Brn-5). During embryogenesis in the rat, Brn-5 is widely expressed with highest levels in the developing brain and spinal cord from embryonic day 12.5. In the adult, Brn-5 mRNA is most abundant in the brain, where it is diffusely expressed with the exception of an enrichment in layer IV of the neocortex. However, Brn-5 is also found in multiple adult tissues outside the central nervous system, including kidney, lung, heart, adrenal, skin, testis, and anterior pituitary. This expression pattern contrast

Ultrastructural investigation into the influence of ethanol on synaptic maturation in rat neocortex. I. Qualitative assessment.

Male rats were exposed daily to ethanol vapour from 3 days of age, and their brains were examined at 7, 14, 21, 56 and 76 days postnatally. Control animals were examined at each age, and ethanol-rehabilitated animals were examined at 56 and 76 days postnatally. Tissue from the parietal cortex of each animal stained with osmium tetroxide and with ethanolic-phosphotungstic acid (E-PTA) was analyzed by qualitative ultrastructural techniques. The ethanol flow rate in the incubation chamber was adjusted to maintain the blood ethanol level as close as possible to 0.1 g/100 ml. Ethanol-treated rats w

[Functional state of the CNS at extreme levels of spatial synchronization of background potentials of rabbit neocortex].

Neurophysiological mechanisms of facilitation of excitation transmission from receptors to effectors were studied in the case of maximal correlation between cortical potentials. This condition was accompanied by a rise in excitability of the sensorimotor, visual, auditory cortex and of the dorsal hippocampus as well as by an increase of muscle activity. On the basis of the obtained and literature data it is suggested that maximal synchroneity of cortical potentials reflects a certain state not only of the neocortex, but also of other CNS parts, down to spinal centres. This condition is attende

Top-Down Projections Direct the Gradual Progression of Alzheimer-Related Tau Pathology Throughout the Neocortex.

In sporadic Alzheimer's disease (sAD), tau pathology gradually but relentlessly progresses from the transentorhinal region of the temporal lobe into both the allocortex and temporal high order association areas of the neocortex. From there, it ultimately reaches the primary sensory and motor fields of the neocortex. The brunt of the changes seen during neurofibrillary stages (NFT) I-VI is borne by top-down projection neurons that contribute to cortico-cortical connectivities between different neocortical fields. Very early changes develop in isolated pyramidal cells in layers III and V, and th

Modifications of gamma-aminobutyric acidA receptor subunit expression in rat neocortex during tolerance to diazepam.

We evaluated whether tolerance to the antagonism of bicuculine-induced seizures by diazepam is associated with changes (i) in the content of mRNAs encoding for gamma-aminobutyric acidA (GABAA) receptor subunits, (ii) in the expression density of these subunits, and (iii) in the 1,4-benzodiazepine binding site characteristics in discrete neocortical structures. We found that in diazepam-tolerant rats, the content of the mRNA encoding for the alpha 1 subunit of the GABAA receptor decreased in the frontoparietal motor (FrPaM) cortex and in the hippocampus (42% and 20%, respectively) but not in th

Most Early-Born Subplate Neurons Persist as Layer 6b Neurons in the Adult Mouse Neocortex.

Subplate neurons (SpNs) are early-born neurons in the mammalian neocortex and play a pivotal role in cortical development. It is widely believed that SpNs are predominantly lost due to postnatal cell death, with only a few remnants contributing to layer 6b (L6b) neurons in the adult neocortex, but the extent to which SpNs persist as L6b neurons remains largely unknown. To address this, we conducted a comprehensive birthdate analysis using multiple ethynyl deoxyuridine (EdU) injections over 24 h, thereby precisely tracking the survival and distribution of SpNs in the adult mouse neocortex. We f

DSCAM gene triplication causes excessive GABAergic synapses in the neocortex in Down syndrome mouse models.

Down syndrome (DS) is caused by the trisomy of human chromosome 21 (HSA21). A major challenge in DS research is to identify the HSA21 genes that cause specific symptoms. Down syndrome cell adhesion molecule (DSCAM) is encoded by a HSA21 gene. Previous studies have shown that the protein level of the Drosophila homolog of DSCAM determines the size of presynaptic terminals. However, whether the triplication of DSCAM contributes to presynaptic development in DS remains unknown. Here, we show that DSCAM levels regulate GABAergic synapses formed on neocortical pyramidal neurons (PyNs). In the Ts65D

Low-grade tumour over the left temporal neocortex and ictal asystole: network and surgical implications.

We describe a patient with focal epilepsy characterized by ictal asystole episodes and low-grade tumour over the left temporal neocortex. Non-invasive pre-surgical evaluation showed an epileptogenic zone extended beyond the low-grade tumour. This extension was confirmed by intraoperative electrocorticography. One-stage surgery with anterior temporal lobe resection was performed. The patient was seizure-free after one year of follow-up. Detailed electroclinical and therapeutic reasoning with hypotheses defining epileptogenic and symptomatogenic networks are discussed.

Organization of rodent auditory cortex: anterograde transport of PHA-L from MGv to temporal neocortex.

In the present study we analyzed the organization of the thalamocortical projections of the specific auditory relay nucleus of the thalamus, the ventral division of the medial geniculate body (MGv), using the anterograde axonal tracer Phaseolus vulgaris leucoagglutinin. All injections of MGv produced dense labeling of axonal fibers in temporal cortex. In all cases, labeled axons were predominantly concentrated in cortical layers III and IV and, to a lesser extent, at the junction of layers V and VI. Injections confined to the medial regions of MGv, and specifically to the ovoid nucleus of MGv

Circadian Rhythms in Plasticity Markers Are Regulated by the Suprachiasmatic Nucleus and Local Circadian Oscillators in the Rodent Neocortex.

Circadian rhythms in clock gene expression are a global feature of neurons. While clock gene products have been shown to modulate plasticity at the synapse level, little is known about molecular circadian rhythms in neuroplasticity. In this study, we aim to establish circadian regulatory mechanisms of plasticity in the rat cerebral cortex. We show that the expression of the plasticity markers activity-regulated cytoskeleton-associated protein (Arc), brain-derived neurotrophic factor (Bdnf), and early growth response protein 1 (Egr1) exhibits marked circadian rhythms with increasing transcript

Etomidate-induced myoclonus correlates with the dysfunction of astrocytes and glutamate transporters in the neocortex of Sprague-Dawley rats.

OBJECTIVE: Etomidate-induced myoclonus is common in clinical anesthesia. Propofol and lidocaine, as other sedative hypnotic and anticonvulsant drugs, rarely induce myoclonus. The mechanism of the myoclonus remains unclear. MATERIALS AND METHODS: Eighty-four adult male Sprague-Dawley (SD) rats anesthetized intravenously with etomidate, propofol, or lidocaine plus etomidate were observed of the behavioral changes at 0, 1, 2, 3, 4 and 5 min after anesthesia. Five minutes later, glutamate levels were measured in the cerebrospinal fluid (CSF), neocortex and hippocampus. The mRNAs and proteins expr

High-potential defense mechanisms of neocortex in a rat model of transient asphyxia induced cardiac arrest.

Cardiac arrest (CA) is a common cause of disability and mortality and thus an important risk for human health. Circulatory failure has dramatic consequences for the brain as one of the most oxygen-consuming organs. Hippocampus, striatum and neocortex rate among the most vulnerable brain regions. The neocortex is less sensitive to hypoxia/reperfusion in comparison with the hippocampal CA1 region. That implicates the existence of efficient defense mechanisms in the neocortex against hypoxia/reperfusion injury, which we analyzed in a well-established CA rat model. We explored different immunohist
