Second Primary Cancer After Chimeric Antigen Receptor-T-Cell Therapy: A Review.

IMPORTANCE: The commercialization of chimeric antigen receptor-T-cell (CAR-T) therapy has changed the landscape of treatment of hematological cancers. Numerous studies from the early 2000s paved the way for cell-based targeted therapeutics, which have been established as practice-changing therapies in lymphoma, leukemia, and multiple myeloma. However, there has been some recent concern about the risk for second primary cancers (SPCs). OBSERVATIONS: Multiple cases of SPCs arising after CAR-T therapy have been reported to the US Food and Drug Administration. Most SPCs have been negative for the

[Analysis of the characteristics of second primary malignancy affecting the survival of lymphoma patients].

Objective: To analyze the characteristics of the second primary tumor affecting the survival of patients with lymphoma, and to explore the risk factors of death from the second primary tumor. Methods: The medical records and related death information of 1 173 lymphoma patients who had already died with known causes were collected. The basic causes of death and the characteristics of patients who died of the second primary tumor were analyzed. Cox regression model was used to analyze the risk factors of lymphoma patients who died of the second primary tumor. Results: Among the 1 173 patients wh

[The risk and management strategies for second primary tumor after CAR-T cell therapy].

Chimeric antigen receptor T (CAR-T) cell therapy has revolutionized the treatment of refractory hematologic malignancies, but its long-term safety has become an increasing concern. Secondary primary malignancies (SPM) following CAR-T-cell therapy have emerged as a growing focus of clinical and translational research. Evidence to date suggests that the incidence of different types of SPMs vary, with a two-year cumulative incidence ranging from approximately 0.08% to 8% and a median time to onset of 9.7 to 26.4 months, predominantly involving myeloid malignancies such as myelodysplastic syndrome

Second Primary Malignancies in Chronic Lymphocytic Leukemia/Small Lymphocytic Lymphoma: A Comprehensive Review.

Patients with chronic lymphocytic leukemia (CLL)/small lymphocytic lymphoma (SLL) have benefited significantly over the last decade from the approval of several novel, targeted therapeutic agents, resulting in prolonged overall survival. However, an excess mortality for CLL/SLL patients remains largely due to nondisease progression events, including second primary malignancies (SPMs). Estimates on the contribution of SPMs to overall excess morbidity and mortality in CLL patients are varied, but recent literature suggests SPMs are responsible for up to 20% of deaths in CLL patients, and studies

Clinical group and modified TNM stage for rhabdomyosarcoma: A review from the Children's Oncology Group.

The Children's Oncology Group (COG) uses Clinical Group (CG) and modified Tumor Node Metastasis (TNM) stage to classify rhabdomyosarcoma (RMS). CG is based on surgicopathologic findings and is determined after the completion of initial surgical procedure(s) but prior to chemotherapy and/or radiation therapy. The modified TNM stage is based on clinical and radiographic findings and is assigned prior to any treatment. These systems have evolved over several decades. We review the history, evolution, and rationale behind the current CG and modified TNM classification systems used by COG for RMS.

Metastatic malignant melanoma: A case of metastasis from foot to mandibular anterior region.

Metastatic malignant melanoma (MMM) of the oral cavity is extremely rare which generally presents in the setting of disseminated disease and have extremely poor prognosis. The most common site for MMM in the oral cavity is tongue followed by buccal mucosa for soft-tissue lesions and posterior mandible for bone involving lesions. Primary tumor sites of oral MMM are usually cutaneous lesions of the trunk, head and neck, limbs, and nasal septum mucosa. Patients bearing this diagnosis face median survival time of approximately 9 months with a probability of surviving 5 years after the initial pres

Synchronous versus metachronous spinal metastasis: a comparative study of survival outcomes following neurosurgical treatment.

PURPOSE: Patients with spinal metastases (SM) from solid neoplasms typically exhibit progression to an advanced cancer stage. Such metastases can either develop concurrently with an existing cancer diagnosis (termed metachronous SM) or emerge as the initial indication of an undiagnosed malignancy (referred to as synchronous SM). The present study investigates the prognostic implications of synchronous compared to metachronous SM following surgical resection. METHODS: From 2015 to 2020, a total of 211 individuals underwent surgical intervention for SM at our neuro-oncology facility. We conduct

Synchronous and metachronous urothelial carcinoma of the upper urinary tract and the bladder: Are they clonally related? A systematic review.

PURPOSE: Following radical nephroureterectomy for upper urinary tract urothelial carcinoma (UTUC), intravesical recurrence (IVR) is found in 22% to 47% of patients. Patients with a primary urothelial carcinoma of the bladder (UCB) have an increased risk of a future UTUC (1%-5%). Paired UTUC and UCB might represent clonally related tumors due to intraluminal seeding of tumor cells or might be separate entities of urothelial carcinoma caused by field cancerization. We systematically reviewed all the relevant literature to address the possible clonal relation of UTUC and paired UCB. MATERIALS AN

Follow, consider, and catch: second primary tumors in acromegaly patients.

BACKGROUND AND AIM: The risk of second primary tumors is increased in general cancer population, however, there is no data on acromegalic cancer patients in this regard. The aim of this study is to determine the prevalence of patients with two primary tumors among acromegalic cancer patients and to evaluate if patients with two primaries have distinct clinical characteristics or risk factors compared to those with one. METHODS: This is a single-center retrospective cohort study. The study included 63 patients with at least one malignant tumor out of a total number of 394 acromegaly patients.

Risk Factors for Subsequent Primary Cancers in Colorectal Cancer Survivors: A Systematic Review and Meta-analysis.

As colorectal cancer survival improves, survivors face an increased risk of subsequent primary cancers (SPC). This systematic review assessed the associations between cancer treatments, tumor features, and other risk factors and the risk of SPCs for colorectal cancer survivors. We searched four databases for peer-reviewed articles published up to October 2025. We conducted meta-analyses using random-effects models to summarize pooled relative risks (RR) for each SPC. Of 10,577 articles screened, 36 met the inclusion criteria. Radiotherapy was associated with increased risks of uterine [RR, 2.6

Radiation therapy in adolescents and young adults (AYA): contemporary evidence on utilization, outcomes, late toxicities, and second malignant neoplasms - A comprehensive systematic review and synthesis.

Adolescent and young adult (AYA; 15-39 years) cancer survivors face decades of potential life-years, making radiotherapy (RT) modality and technique selection central determinants of long-term health. A PRISMA 2020-reported systematic review was performed using PubMed, Web of Science, and the Cochrane Library (January 2026). Data were synthesised qualitatively using a Synthesis Without Meta-analysis (SWiM) approach. The primary synthesis focused on RT utilization and late outcomes including late toxicity, survivorship morbidity, patient-reported outcomes, and secondary malignant neoplasms (SMN

Rate of Benign and Malignant Secondary Tumors Associated With Nevus Sebaceous: A Systematic Review and Meta-Analysis.

BACKGROUND: Nevus sebaceous (NS) is a rare congenital skin lesion affecting approximately 0.3% of all newborns. Although benign, NS lesions can harbor malignant secondary tumors. The published rate of development of these malignant tumors varies. This meta-analysis aimed to identify the rate of malignant and benign secondary neoplasms occurring in NS. METHODS: A literature search was conducted using PubMed, Embase, and Web of Science from inception to April 2023. Eligible studies reported incidence or risk of secondary neoplasms in patients with NS. Two independent reviewers screened studies,

Transformation From Atypical to Anaplastic Metastatic Meningioma: Evolutionary Turning Point Marked by Strong 18 F-FDG Avidity and Uptake Collapse From 68 GA-DOTATOC.

World Health Organization (WHO) grade III anaplastic meningioma is scarce. In this way, most studies compared WHO grade I and II. Otherwise, some authors are uncertain about using 18 F-FDG as a diagnostic tool to estimate the WHO grade, especially high. We report the case from a man with a grading tumor evolution from WHO grade II atypical to grade III anaplastic metastatic meningioma. This turning point was imaged using 68 Ga-DOTATOC and 18 F-FDG. Diagnostic was confirmed by histology. The case underlines the unparalleled power of molecular imaging characterized by high sensitivity staging an
