A bidirectional network for appetite control in larval zebrafish.

Medial and lateral hypothalamic loci are known to suppress and enhance appetite, respectively, but the dynamics and functional significance of their interaction have yet to be explored. Here we report that, in larval zebrafish, primarily serotonergic neurons of the ventromedial caudal hypothalamus (cH) become increasingly active during food deprivation, whereas activity in the lateral hypothalamus (LH) is reduced. Exposure to food sensory and consummatory cues reverses the activity patterns of these two nuclei, consistent with their representation of opposing internal hunger states. Baseline a

Evolution of cerebral asymmetry.

The human brain is often characterized in terms of a duality, with the left and right brains serving complementary functions, and even individuals are sometimes classified as either "left-brained" or "right-brained." Recent evidence from brain imaging shows that hemispheric asymmetry is multidimensional, comprised of independent lateralized circuits. Cerebral asymmetries, which include handedness, probably arise in phylogenesis through the fissioning of ancestral systems that divided and lateralized with increasing demand for specialization. They also vary between individuals, with some showin

Circuit Mechanisms of Reward, Anhedonia, and Depression.

Pleasure and motivation are important factors for goal-directed behavior and well-being in both animals and humans. Intact hedonic capacity requires an undisturbed interplay between a number of different brain regions and transmitter systems. Concordantly, dysfunction of networks encoding for reward have been shown in depression and other psychiatric disorders. The development of technological possibilities to investigate connectivity on a functional level in humans and to directly influence networks in animals using optogenetics among other techniques has provided new important insights in th

Structural brain network analysis in schizophrenia using minimum spanning tree.

Schizophrenia is a mental disorder in which functional and structural brain networks are disrupted. Classical network analysis has been used by many researchers to quantify brain networks and to study the network changes in schizophrenia, but unfortunately metrics used in this classical method highly depend on the networks' density and weight; the comparisons made by this method are biased. The minimum spanning tree (MST) is an alternative method to solve this problem, but its usefulness in studying the schizophrenic brain network has not been examined yet. In the present study, we quantified

The effects of aerobic exercise on the structure and function of DMN-related brain regions: a systematic review.

Physical activity may play a role in both the prevention and slowing of brain volume loss and may be beneficial in terms of improving the functional connectivity of brain regions. But much less is known about the potential benefit of aerobic exercise for the structure and function of the default mode network (DMN) brain regions. This systematic review examines the effects of aerobic exercise on the structure and function of DMN brain regions in human adulthood. Seven electronic databases were searched for prospective controlled studies published up to April 2015. The quality of the selected st

Multisensory maps in parietal cortex.

Parietal cortex has long been known to be a site of sensorimotor integration. Recent findings in humans have shown that it is divided up into a number of small areas somewhat specialized for eye movements, reaching, and hand movements, but also face-related movements (avoidance, eating), lower body movements, and movements coordinating multiple body parts. The majority of these areas contain rough sensory (receptotopic) maps, including a substantial multisensory representation of the lower body and lower visual field immediately medial to face VIP. There is strong evidence for retinotopic rema

Charles Bonnet syndrome: evidence for a generative model in the cortex?

Several theories propose that the cortex implements an internal model to explain, predict, and learn about sensory data, but the nature of this model is unclear. One condition that could be highly informative here is Charles Bonnet syndrome (CBS), where loss of vision leads to complex, vivid visual hallucinations of objects, people, and whole scenes. CBS could be taken as indication that there is a generative model in the brain, specifically one that can synthesise rich, consistent visual representations even in the absence of actual visual input. The processes that lead to CBS are poorly unde

Strategic control in decision-making under uncertainty.

Complex economic decisions - whether investing money for retirement or purchasing some new electronic gadget - often involve uncertainty about the likely consequences of our choices. Critical for resolving that uncertainty are strategic meta-decision processes, which allow people to simplify complex decision problems, evaluate outcomes against a variety of contexts, and flexibly match behavior to changes in the environment. In recent years, substantial research has implicated the dorsomedial prefrontal cortex (dmPFC) in the flexible control of behavior. However, nearly all such evidence comes

[The reader brain: natural and cultural story].

The report of cases of pure alexia suggests that some regions of the neural system are dedicated to reading-specific visual processing abilities. Pure alexia results from the disruption or the disconnection of the visual word form area, a region reproducibly located within the left occipito-temporal sulcus and encoding the abstract identity of strings of visual letters. The functional specialization of this area suggests that it is initially plastic and becomes attuned to the orthographic regularities that constrain letter combination during the acquisition of literacy. The visual word form ar

Procedural learning difficulties: reuniting the developmental disorders?

During the past 30 years, research into developmental disorders has fragmented, emphasizing differences rather than commonalities. We propose that reunification might be achieved by using a "neural-systems" approach. Deficits in dyslexia are attributed to an intact declarative learning system combined with an impaired procedural learning system--a network that includes prefrontal language systems and basal ganglia, parietal and cerebellar structures. A typology is provided for other prevalent learning disabilities; this framework focuses on different learning skills in the understanding of lea

Emergent mechanisms in multiple pattern generation of the lobster pyloric network.

By using a hard-wired oscillator network, multiple pattern generation of the lobster pyloric network is simulated. The network model is constructed using a relaxation oscillator representing an oscillatory or quiescent (i.e. steady-state) neuron. Modulatory inputs to the network are hypothesized to cause changes in the dynamical properties of each pyloric neuron: the oscillatory frequency, the postinhibitory rebound property, and the resting membrane potential. Changes in each of these properties are induced by changing appropriate parameters of the oscillator. By changing seven parameters of

The development of the nerve network in the fetal human carotid body and its subsequent function in cardiac disease.

Carotid bodies from 17 human fetuses of gestational age ranging from 10 weeks to full term were examined in histological sections stained by the Bodian silver protargol method to demonstrate nerve axons. At 10 weeks gestation the carotid body was contacted by a single nerve bundle at its apical pole but by the 13th week a second bundle had also reached the proximal pole. Thin, pale nerve axons extended from these bundles and surrounded the carotid body to form a plexus from which several small groups of axons entered its superficial regions. With increase of gestational age beyond this point t

In silico model for automated calculation of functional metrics in animal models of peripheral nerve injury repair.

The rat sciatic nerve model is commonly used to test novel therapies for nerve injury repair. The static sciatic index (SSI) is a useful metric for quantifying functional recovery, and involves comparing an operated paw versus a control paw using a weighted ratio between the toe spread and the internal toe spread. To calculate it, rats are placed in a transparent box, photos are taken from underneath and the toe distances measured manually. This is labour intensive and subject to human error due to the challenge of consistently taking photos, identifying digits and making manual measurements.

[Electroneurography and other electric tests on prognostic assessment of Bell's palsy].

Twenty-two patients with Bell's palsy who had complete unilateral facial paralysis were selected for this study. Electroneurography (ENoG), nerve excitability test (NET), maximal stimulation test and facial nerve latency test were done on each patient. The patients were examined within 21 days of onset of facial paralysis and evaluated at least six months after onset to determine the degree of recovery of facial motor function. ENoG was the most accurate test for prognostic assessment of Bell's palsy when it was performed within 3 weeks after onset. When the response loss of ENoG on the involv

Ion transport abnormalities in inflamed rat jejunum. Involvement of mast cells and nerves.

Basal and stimulated changes in ion transport in vitro were examined in jejunal mucosa from rats during inflammation produced after infection with the nematode Nippostrongylus brasiliensis. The gut was acutely inflamed at days 7 and 10 when net secretion of Na+ and Cl- ions was evident. Serum levels of rat mast cell protease II were elevated, providing evidence for mast cell activation. In addition, the magnitude of the short-circuit current responses to electrical transmural stimulation of enteric nerves (but not to histamine in the presence of neural blockade) were significantly reduced (p l

Neuroendocrine Neoplasms.

Neuroendocrine neoplasms (NENs), also known as neuroendocrine tumors (NETs), are rare tumors derived from cells with characteristics of both nerve and endocrine cells. The clinical presentation, diagnosis, and treatment of NENs vary significantly depending on the type, location, whether the neoplasm is hormonally functional, how aggressive it is, and whether it has metastasized to other parts of the body. This article provides an overview of specific types of NETs, clinical presentations and related syndromes, diagnosis, and approach to management of common NENs.

Enhancing diagnosis of Hirschsprung's disease using deep learning from histological sections of post pull-through specimens: preliminary results.

PURPOSE: Accurate histological diagnosis in Hirschsprung disease (HD) is challenging, due to its complexity and potential for errors. In this study, we present an artificial intelligence (AI)-based method designed to identify ganglionic cells and hypertrophic nerves in HD histology. METHODS: Formalin-fixed samples were used and an expert pathologist and a surgeon annotated these slides on a web-based platform, identifying ganglionic cells and nerves. Images were partitioned into square sections, augmented through data manipulation techniques and used to develop two distinct U-net models: one

Usefulness of critical flicker fusion frequency measurement and its laterality for evaluating compressive optic neuropathy due to pituitary neuroendocrine tumors.

Critical flicker fusion frequency (CFF) is a short but sensitive method for evaluating optic nerve function. We measured CFF in patients with pituitary neuroendocrine tumors (Pit-NETs) to assess its usefulness. Data from 184 patients with nonfunctioning Pit-NETs, who had been treated with transsphenoidal surgery and had no medical history of eye diseases, was used in this retrospective study. Visual acuity decline (VAD) was defined as > 0.10 reduction in logMAR visual acuity and CFF decline (CFD) was defined as CFF value < 35 Hz. Visual field defect (VFD) was evaluated by automated perimetry o

Tenascin-R is antiadhesive for activated microglia that induce downregulation of the protein after peripheral nerve injury: a new role in neuronal protection.

Microglial activation in response to pathological stimuli is characterized by increased migratory activity and potential cytotoxic action on injured neurons during later stages of neurodegeneration. The initial molecular changes in the CNS favoring neuronofugal migration of microglia remain, however, largely unknown. We report that the extracellular matrix protein tenascin-R (TN-R) present in the intact CNS is antiadhesive for activated microglia, and its downregulation after facial nerve axotomy may account for the loss of motoneuron protection and subsequent neurodegeneration. Studies on the

Baicalin attenuates HIF-1α-driven histone lactylation and neutrophil extracellular trap (NET) formation in autoimmune uveitis.

Uveitis is a group of vision-threatening intraocular inflammatory disorders that can affect the uveal tract, retina, optic nerve, and vitreous, leading to irreversible structural damage and vision loss. Neutrophil extracellular trap (NET) formation is a key driver of acute uveitis (AU) pathology, but the immunometabolic mechanisms governing NETosis remain unclear. Here, we identified hypoxia-inducible factor-1α (HIF-1α) as a central regulator that links neutrophil metabolism to NET formation. DIA-based lactyl-proteomic profiling revealed widespread nuclear protein lactylation, with histone H3
