Oestrogen modulates stress-induced vagally-dependent gastric responses to oxytocin.

The brain-gut axis is central to gastrointestinal regulation and disorders of gut-brain interaction are more prevalent and often more severe in females. The aim of the present study was to test the hypothesis that brain-gut neurocircuitry is influenced by stress and ovarian hormone levels. Gastric emptying in Sprague-Dawley female rats was slower when oestrogen levels were high, but stress had no further effect. Whole-cell recordings revealed that, unlike previous reports in male rats, the 'anti-stress' peptide, oxytocin (OXT), suppressed GABAergic transmission to dorsal motor nucleus of the v

Electroacupuncture Attenuates Colitis in Mice Through Activation of Vagus Cholinergic Antiinflammatory Pathways.

BACKGROUND: Colitis impairs life quality, and treatments are limited. Acupuncture may engage vagal anti-inflammatory pathways, but the precise circuits are unclear. OBJECTIVE: This study aimed to examine the effects of electroacupuncture (EA) at "Shangjuxu" (ST37) on murine colitis and the involvement of the cholinergic vagus system. METHODS: Mice were divided into control, model, EA (electro-acupuncture), and MLA (EA with α7nAChR antagonist) groups. A colitis model was induced in mice by freely drinking a 2.5% dextran sodium sulfate (DSS) solution for 7 days. Colitis severity was assessed v

Cardiovascular Brain Circuits.

The cardiovascular system is hardwired to the brain via multilayered afferent and efferent polysynaptic axonal connections. Two major anatomically and functionally distinct though closely interacting subcircuits within the cardiovascular system have recently been defined: The artery-brain circuit and the heart-brain circuit. However, how the nervous system impacts cardiovascular disease progression remains poorly understood. Here, we review recent findings on the anatomy, structures, and inner workings of the lesser-known artery-brain circuit and the better-established heart-brain circuit. We

Projection-specific circuits of retrosplenial cortex with differential contributions to spatial cognition.

Retrosplenial cortex (RSC) is a brain region involved in neuropsychiatric and neurodegenerative disorders. It has reciprocal connections with a diverse set of cortical and subcortical brain regions, but the afferent structure and behavioral function of circuits defined by its projection-specific sub-populations have yet to be determined. The corticocortical connections between RSC and secondary motor cortex (M2), as well as corticothalamic connections between RSC and anterodorsal thalamus (AD) have been hypothesized to function as semi-independent, but parallel pathways that impact spatial inf

A spatial-temporal map of glutamatergic neurogenesis in the murine embryonic cerebellar nuclei uncovers a high degree of cellular heterogeneity.

The nuclei are the main output structures of the cerebellum. Each and every cerebellar cortical computation reaches several areas of the brain by means of cerebellar nuclei processing and integration. Nevertheless, our knowledge of these structures is still limited compared to the cerebellar cortex. Here, we present a mouse genetic inducible fate-mapping study characterizing rhombic lip-derived glutamatergic neurons of the nuclei, the most conspicuous family of long-range cerebellar efferent neurons. Glutamatergic neurons mainly occupy dorsal and lateral territories of the lateral and interpos

Retrograde labelling in vivo of vagus nerve nucleus in rat by carbocyanine dye.

To elucidate the patterns of visceral innervation of the vagus nerve in rats, we labelled the severed efferent path at the level of the neck in vivo by applying a solution of 0.1% carbocyanine dye DiI/100% ethanol. Two weeks after dye application, horizontal sections of the brain stem were analysed quantitatively under an epifluorescence microscope. Particles of DiI were present in neuronal cell bodies and the cellular processes in the dorsal motor nucleus (DmoX) of the vagal nerve and nucleus ambiguus (Nam). Approximately 80% of DiI-labelled cells were located in the DmoX whereas 16% were in

Dscam1 Has Diverse Neuron Type-Specific Functions in the Developing Drosophila CNS.

Two key features endow Drosophila Down syndrome cell adhesion molecule 1 (Dscam1) with the potential to provide a ubiquitous code for neuronal arbor self-avoidance. First, Dscam1 contains three large cassettes of alternative exons, so that stochastic alternative splicing yields 19,008 Dscam1 isoforms with different Ig ectodomains. Second, each neuron expresses a different subset of Dscam1 isoforms, and isoform-specific homophilic binding causes repulsion. This results in even spacing of self-arbors, while processes of other neurons can intermingle and share the same synaptic partners. In princ

Neurotransmitters in the cerebral cortex.

This article surveys the conventional neurotransmitters and modulatory neuropeptides that are found in the cerebral cortex and attempts to place them into the perspective of both intracortical circuitry and cortical disease. The distribution of these substances is related, where possible, to particular types of cortical neuron or to afferent or efferent fibers. Their physiological actions, where known, on cortical neurons are surveyed, and their potential roles in disease states such as the dementias, epilepsy, and stroke are assessed. Conventional transmitters that occur in afferent fibers to

Monosynaptic Retrograde Tracing From Prelimbic Neuron Subpopulations Projecting to Either Nucleus Accumbens Core or Rostromedial Tegmental Nucleus.

The prelimbic (PL) region of the medial prefrontal cortex (mPFC) has been implicated in both driving and suppressing motivated behaviors, including cocaine-seeking in rats. These seemingly opposing functions may be mediated by different efferent targets of PL projections, such as the nucleus accumbens (NAc) core and rostromedial tegmental nucleus (RMTg), which have contrasting roles in reward-seeking behaviors. We sought to characterize the anatomical connectivity differences between PL neurons projecting to NAc core and RMTg. We used conventional retrograde tracers to reveal distinct subpopul

[Latency, threshold and frequency selectivity of the crossed medial cochlear efferent pathways].

The efferent innervation of guinea pig cochleas was sectioned medially, at the level of the floor of the fourth ventricle, to study the effects of the crossed part of the medial efferent pathway on the compound action potential (CAP) masking phenomenon. Sectioning reduced CAP masking for a masker level varying with the frequency of the masker and the time elapsed between the masker onset and the probe onset. Functional properties of the crossed part of the medial efferent tracts: latency, thresholds and frequency selectivity, could be deduced from these data. This intensification of the maskin

Efferent feedback minimizes cochlear neuropathy from moderate noise exposure.

Although protective effects of the cochlea's efferent feedback pathways have been well documented, prior work has focused on hair cell damage and cochlear threshold elevation and, correspondingly, on the high sound pressure levels (>100 dB SPL) necessary to produce them. Here we explore the noise-induced loss of cochlear neurons that occurs with lower-intensity exposures and in the absence of permanent threshold shifts. Using confocal microscopy to count synapses between hair cells and cochlear nerve fibers, and using measurement of auditory brainstem responses and otoacoustic emissions to ass

Influence of medial olivocochlear efferents on the sharpness of cochlear tuning estimates in children.

The present study objectively quantified the efferent-induced changes in the sharpness of cochlear tuning estimates and compared these alterations in cochlear tuning between adults and children. Click evoked otoacoustic emissions with and without contralateral broadband noise were recorded from 15 young adults and 14 children aged between 5 and 10 yrs. Time-frequency distributions of click evoked otoacoustic emissions were obtained via the S-transform, and the otoacoustic emission latencies were used to estimate the sharpness of cochlear tuning. Contralateral acoustic stimulation caused a sign

Medial olivocochlear efferent activity in awake guinea pigs.

Cochlear outer hair cells receive numerous connections from the medial olivocochlear efferent neurons. Medial olivocochlear efferent activity is highly dependent on the level of anesthesia. The present study was thus designed to investigate the efficiency of contralateral white noise stimulation on the distortion product otoacoustic emissions (DPOAEs) in a large number of awake guinea pigs, and to compare in the same animals the effect of urethane- and pentobarbitone-anesthesia. The monitoring of DPOAEs during contralateral white noise stimulation in awake animals requires the development of a

AAV-Mediated Gene Delivery to the Enteric Nervous System by Intracolonic Injection.

The enteric nervous system of the lower gastrointestinal tract comprises intrinsic neural circuits as well as extrinsic afferent and efferent innervation. The development of strategies for neuronal gene transfer has created new opportunities for functional analysis, circuit mapping, and neuromodulation in the enteric nervous system. Studies of AAV-mediated gene transfer to enteric neurons and dorsal root ganglion neurons (DRG) have provided proofs-of-concept for the utility of AAV vectors for genetic manipulations of the intrinsic and extrinsic components of the enteric nervous system. Here we

[The effect of chronic ototoxicity of gentamicin on the function of auditory efferents].

OBJECTIVE: To study the effect of chronic ototoxicity of gentamicin on the function of the cochlea and auditory efferents. METHOD: The effect of chronic ototoxicity of gentamicin on the function of the cochlea was determined by recording thresholds of the compound action potentials (CAP) of the auditory nerves to tone pip at 4, 6, 8, 10 and 12 kHz. The function of the medial olivocochlear (MOC) system was determined, through the observation of the effect of the contralateral noise (CLN) on CAP, before and after gentamicin administration. RESULT: The suppression effects of CLN on CAP were irr

Facial nerve injury induces facilitation of responses in both trigeminal and facial nuclei of rat.

A study was made of the effects of facial nerve transection on trigeminal stimulation- evoked field potentials in the principal trigeminal (Pr5) and facial nuclei (7) in rats. Although the transected branch of the facial nerve contains pure motoric efferents, it resulted in enhanced responses in both Pr5 and 7. These electrophysiological results suggest a functional circuitry involving the whiskers, trigeminal nerve, Pr5 and 7 and the facial nerve as efferent. The disconnection (opening) of this loop results in enhanced responsiveness of the neurons in both Pr5 and 7.

Noncoding variants alter GATA2 expression in rhombomere 4 motor neurons and cause dominant hereditary congenital facial paresis.

Hereditary congenital facial paresis type 1 (HCFP1) is an autosomal dominant disorder of absent or limited facial movement that maps to chromosome 3q21-q22 and is hypothesized to result from facial branchial motor neuron (FBMN) maldevelopment. In the present study, we report that HCFP1 results from heterozygous duplications within a neuron-specific GATA2 regulatory region that includes two enhancers and one silencer, and from noncoding single-nucleotide variants (SNVs) within the silencer. Some SNVs impair binding of NR2F1 to the silencer in vitro and in vivo and attenuate in vivo enhancer rep

Brain-to-stomach transfer of α-synuclein via vagal preganglionic projections.

Detection of α-synuclein lesions in peripheral tissues is a feature of human synucleinopathies of likely pathogenetic relevance and bearing important clinical implications. Experiments were carried out to elucidate the relationship between α-synuclein accumulation in the brain and in peripheral organs, and to identify potential pathways involved in long-distance protein transfer. Results of this in vivo study revealed a route-specific transmission of α-synuclein from the rat brain to the stomach. Following targeted midbrain overexpression of human α-synuclein, the exogenous protein was capable
